A field experiment was conducted to examine the growth of one and two year old American ginseng (Panax guinguefolium L.) in the arid interior of British Columbia, Canada. For both years of plants, early season growth was characterized by rapid stem and leaf dry matter production. Root growth commenced in early June and continued until early September. One year old roots had a dry weight of 0.1 to 0.2g at the end of the growing season. The two years old roots commenced the growing season at 0.Is and increased in dry matter by a factor of ten-fold. Dry to fresh weight ratios for both years were similar for root, leaf and stem samples at the end of the growing season. Leaf area index for both years showed similarities in progression over the growing season. As a consequence of the effectiveness of the microclimate modification employed to permit American ginseng cultivation (elevated shade cloth and surface mulch), plant growth and development was extremely good. This indicates the potential for the cultivation of American ginseng in dryland environments.
This study was conducted to investigate effect of leaf temperature on light response and respiration during short-term exposure to a sequnce of PPFD cycle(100-200-300-400$\mu$ mol m$^{-2}$ s$^{-1}$ ) under increasing leaf temperature sequnce up to 3$0^{\circ}C$ and in dark condition, increasing up to 46$^{\circ}C$ with step size (1$^{\circ}C$) in Panax ginseng C. A. Meyer. When leaflet exposed to low light intensity and temperature, Rubisco activity was higher than remained activity in high condition. Leaves adapted to 100$\mu$mol m$^{-2}$ s$^{-1}$ PPFD had a peak response similar to that of 200$\mu$mol m$^{-2}$ s$^{-1}$ at 18$^{\circ}C$, but in above PPFD cycle(300, 400$\mu$mol m$^{-2}$ s$^{-1}$ ) it represented at 17$^{\circ}C$ and 16$^{\circ}C$, re-spectively. $CO_2$ evolution in dark condition increased rapidly when leaf temperature was increased up to 28$^{\circ}C$ and then 'dipped' below steady-state level from above 4$0^{\circ}C$. Thus, Pananx ginseng is able to take advantage of irradiance increase and decrease of $CO_2$ evolution in dark condition to control leaf temperature.mperature.

The purpose of this study is to prepare black Panax ginseng leaf (PGL) and evaluate its antioxidative effect. In order to make black PGL, the raw PGL was successiely steamed at $95^{\circ}C$ for 3 hr nine times. The antioxidant activities of total saponins (Sa) from PGL and black PGL against peroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. Specific TOSC values for black PGL-Sa against peroxyl radicals and peroxynitrites were 2.3-fold and 2.1-fold of PGL-Sa, respectively, and 2.2-fold and 5.2-fold of glutathione, a positive control antioxidant, respectively. The black PGL-Sa exhibited stronger antioxidative effect than PGL-Sa. The main ginsenosides of black PGL were $Rg_3,\;Rk_1\;and\;Rg_5$. Among the saponins in black PGL, the amount of ginsenoside $Rg_3$ was examined by HPLC. 22.12 mg of ginsenoside $Rg_3$ was obtained from 1g of dried black PGL.
This study was carried out to research microorganisms having the antifungal activity against ginseng Alternaria blight pathogen Alternaria panax and ginseng anthracnose pathogen Colletotrichum gloeosporioides. Eleven Bacillus strains. were isolated from Korean traditional soybean paste and Kimchi. Among the 11 isolates, DJ5, KC1, KC2 and KC4 showing antagonistic activity on the mycelial growth of A. panax and C. gloeosporioides in pairing culture were finally selected as the antagonistic microorganisms. Based on 16s rRNA sequence and phylogenetic tree analysis, they were identified as Bacillus spp.. The selected microorganisms were investigated antagonistic activity by measured leaf-segment colonization in pot test. When Bacillus sp. were injected after A. panax treatment, KC1, KC2 and KC4 showed similar effect to chemical pesticides treated control. To measure preventive effect of Bacillus sp, antagonistic microorganisms were injected and C. gloeosporioides was treated in pot. When measuring the effectiveness for the prevention of Anthracnose, All Bacillus spp. showed approximately 83~90 % degree of superior preventive effect. In general, The four Bacillus spp. isolated from Korean traditional fermented foods showed therapeutic effect of Alternaria blight and preventive effect of Anthracnose.
Background: Korean ginseng (Panax ginseng Meyer) is a perennial herb prone to various root diseases, with Phytophthora cactorum being considered one of the most dreaded pathogens. P. cactorum causes foliar blight and root rot. Although chemical pesticides are available for disease control, attention has been shifted to viable, eco-friendly, and cost-effective biological means such as plant growth-promoting rhizobacteria (PGPR) for control of diseases. Methods: Native Bacillus amyloliquefaciens strain HK34 was isolated from wild ginseng and assessed as a biological control agent for ginseng. Leaves from plants treated with HK34 were analyzed for induced systemic resistance (ISR) against P. cactorum in square plate assay. Treated plants were verified for differential expression of defense-related marker genes using quantitative reverse transcription polymerase chain reaction. Results: A total of 78 native rhizosphere bacilli from wild P. ginseng were isolated. One of the root-associated bacteria identified as B. amyloliquefaciens strain HK34 effectively induced resistance against P. cactorum when applied as soil drench once (99.1% disease control) and as a priming treatment two times in the early stages (83.9% disease control). A similar result was observed in the leaf samples of plants under field conditions, where the percentage of disease control was 85.6%. Significant upregulation of the genes PgPR10, PgPR5, and PgCAT in the leaves of plants treated with HK34 was observed against P. cactorum compared with untreated controls and only pathogen-treated plants. Conclusion: The results of this study indicate HK34 as a potential biocontrol agent eliciting ISR in ginseng against P. cactorum.
Cotyledon explants of Korean ginseng (Panax ginseng C.A. Meyer), a perennial medicinal plant, produced direct somatic embryos at a high frequency on MS medium without growth regulators. Cytokinin highly suppressed the somatic embryogenesis but stimulated direct fomlation of adventitious buds. BAP was more effective than kinetin for the formation of adventitious bud. IBA combination with cytokinin enhanced the frequency of adventitious bud formation. The highest frequency of adventitious bud formation were $40\%$ at 0.05 mg/l IBA and 5 mg/l BAP. Adventitious buds were mainly formed near the distal portion of cotyledon, while somatic embryos were only formed near the proximal portion of cotyledon. Adventitious buds were covered with sheath similar to axillary buds in the zygotic embryos, and then leaf-like epicotyls were developed.
The possibility of utilizing greet amount of by-product of ginseng (Panax ginseng C.A. meyer) plant-that is, production of ter from ginseng leaf, was studied and the results are summarized as follows: 1. Ginseng leaf contains more soluble matter than tea leaf (Thea sinensis) and the soluble matter is easily extracted by hot water. 2. Ginseng leaf has less tannin (2.2%) than yea leaf (7.89%). Therefore, it has less astringency than tea. 3. Vitamin C content of ginseng leaf is not compared with that of tea leaf. In fact, ginseng leaf contains Vitamin C $50{\sim}110$ times of tea leaf. 4. Ginseng leaf contains $5.7{\sim}8.5%$ glycoside (dammaranes) and the ratio of panaxadiol to panaxatriol is $0.54{\sim}0.75$ that is, panaxatriol contents is high. 5. For the acceptability of the product related with the soluble matter contents and color the method of extracting 2g of ginseng leaf product in 200ml of water for 3 minutes is recommended. 6. As a result of evaluating the flavor characteristics and effective components of the products, product D which is produced by the process of steaming, drying and roasting is considered to have the best quality.

This study was conducted to investigate the application possibility of leaf and stem extract (LSE) from the mixture of leaf and stem of Panax ginseng. This study measured the general nutritional composition, aminoacid minerals contents and fatty acid composition of LSE. We conducted analysis of the ginsenoside content by HPLC and the cell cytotoxicity tests in normal liver and kidney cells. The approximate composition of LSE was 2.51% of carbohydrate 0.53% of crude ash,0.20% of crude fat and 0.15% of crude protein, respectively. LSE contained 102.56 mg/100 g of K ion and high contents of acidic amino acids such as glutamic acid and aspartic acid. In addition to this, it contained all essential amino acids. The major compositions of fatty acids were 39.99% of palmitic acid 14.96% of linoleic acid, 13.31% of docosatetranoic acid and 12.91% of linolenic acid, The total ginsenoside was 0.82 mg/mL, and ratio of PD/PT was 0.68. Negative effects were not found from the results of the cell toxicity respection. These results imply that leaf and stem of Panax gineng could be used as possible food resources and functional food material and feed stuff.

Background: Interspecific ginseng hybrid, Panax ginseng ${\times}$ Panax quenquifolius (Pgq) has vigorous growth and produces larger roots than its parents. However, F1 progenies are complete male sterile. Plant tissue culture technology can circumvent the issue and propagate the hybrid. Methods: Murashige and Skoog (MS) medium with different concentrations (0, 2, 4, and 6 mg/L) of 2,4-dichlorophenoxyacetic acid (2,4-D) was used for callus induction and somatic embryogenesis (SE). The embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 Schenk and Hildebrandt (SH) medium. The developed taproots with dormant buds were treated with $GA_3$ to break the bud dormancy, and transferred to soil. Hybrid Pgq plants were verified by random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses and by LC-IT-TOF-MS. Results: We conducted a comparative study of somatic embryogenesis (SE) in Pgq and its parents, and attempted to establish the soil transfer of in vitro propagated Pgq tap roots. The Pgq explants showed higher rate of embryogenesis (~56% at 2 mg/L 2,4-D concentration) as well as higher number of embryos per explants (~7 at the same 2,4-D concentration) compared to its either parents. The germinated embryos, after culturing on $GA_3$ supplemented medium, were transferred to hormone free 1/2 SH medium to support the continued growth and kept until nutrient depletion induced senescence (NuDIS) of leaf defoliation occurred (4 months). By that time, thickened tap roots with well-developed lateral roots and dormant buds were obtained. All Pgq tap roots pretreated with 20 mg/L $GA_3$ for at least a week produced new shoots after soil transfer. We selected the discriminatory RAPD and ISSR markers to find the interspecific ginseng hybrid among its parents. The $F_1$ hybrid (Pgq) contained species specific 2 ginsenosides (ginsenoside Rf in P. ginseng and pseudoginsenosides $F_{11}$ in P. quinquefolius), and higher amount of other ginsenosides than its parents. Conclusion: Micropropagation of interspecific hybrid ginseng can give an opportunity for continuous production of plants.
To compare the growth pattern of ginseng plant under between conventional shading(light transmittance rate 3%) and polyethylene net shading(light transmittance rate 10%), the distribution of leaf area, specific leaf weight (S. L. W), leaf and stem dry weight and changes in light intensity were investigated in 2, 4 and 6 year old ginseng plant populations. Light transmittance rate(L.T.R.) was 3% at front line, 2% at middle line and 1.5% at rear line under conventional shading but it was 12, 10 and 8% under polyethylene net(P.E) shading, respectively. In 2 year old population, there was a little difference in the growth characteristics investigated between conventional and P.E. shading. In 4 year old field, the leaf area, stem and leaf dry weight decreased in large amount in the order of middle, and rear line on ridge under conventional shading, but with a little difference under P.E. shading. And these trends enlarged in 6 year old field with appearance of a large part of shoot over furrow from ridge planted ginseng. Root yield index was much lowered at the rear 3rd, 4th and 5th line of the conventional shading bed, but there was no yield difference among lines except 5th lines under the P.E. shading with higher yield by 28% than conventional shading.
