Activation tagging that uses T-DNA vectors containing multimerized transcriptional enhancers from the cauliflower mosaic virus (CaMV) 35S gene is a powerful tool to determine gene function in plants. This approach has been successfully applied in screening various types of mutations and cloning the corresponding genes. We generated an activation tagged hairy root pool of ginseng (Panax ginseng C.A. Meyer) in an attempt to isolate genes involved in the biosynthetic pathway of ginsenoside (triterpene saponin), which is known as the major active ingredient of the root. Quantitative and qualitative variation of ginsenoside in activation tagged hairy root lines were profiled using LC/MS. Metabolic profiling data enabled selection of a specific hairy root line which accumulated ginsenoside at a higher level than other lines. The relative expression level of several genes of triterpene biosynthetic pathway in the selected hairy root line was determined by real time RT-PCR. Overall results suggest that the activation tagged ginseng hairy root system described in this study would be useful in isolating genes involved in a complex metabolic pathway from genetically intractable plant species by metabolic profiling.
To evaluate the anticolitic effect of red ginseng (RG, the steamed root of Panax ginseng CA. Meyer, Araliaceae), RG and its representative constituents, ginsenosides Rg3 and Rh2, were orally administered to dextran sulfate sodium (DSS)-induced colitic mice and inflammatory markers investigated. RG and its constituents, ginsenosides Rg3 and Rh2, inhibited colon shortening and myeloperoxidase activity induced by DSS. The ginsenosides Rg3 and Rh2 inhibited mRNA expression of interleukin (IL)-$1{\beta}$ as well as protein levels of IL-$1{\beta}$ and IL-6. These ginsenosides also inhibited the activation of a transcription nuclear factor (NF)-${\kappa}B$. Ginsenoside Rh2 was a more potent inhibitor than ginsenoside Rg3. The anticolitic effects of these ginsenosides were comparable with sulfasalazine.
Panax ginseng is an important medicinal plant that has been used worldwide for geriatric, tonic, stomachic, and aphrodisiac treatments. Ginsenosides contained in the ginseng root are the main substances having active functions for human body. The price of ginseng is very expensive due to a complex process of cultivation, and the yield of ginseng is limited, which cannot meet the demand of the increasing market. Researchers have applied plant biotechnology to solve the problems but there are still things to be determined towards ginsenoside production by large-scale adventitious root culture. In this experiment, 5 to 20 liter bioreactors were employed to determine optimal conditions for adventitious root culture and ginsenoside production of Panax gineng. Callus was induced from the ginseng root on MS agar medium containing 1.0 mg. $L^{-1}$ 2,4-D and 0.1 mg. $L^{-1}$ kinetin. Then the callus was cultured on MS agar medium supplemented with 2.0 mg. $L^{-1}$ IBA, 0.1 mg. $L^{-1}$ kinetin, and 30 g. $L^{-1}$ to induce adventitious roots. The maximum root growth and ginsenoside production were obtained in 1/2 MS medium. 2.0 mg. $L^{-1}$ naphthalene acetic acid resulted in greater root growth than 2.0 mg $L^{-1}$ indole-3-butyric acid. Ginsenoside content increased with 2.0 mg. $L^{-1}$ benzyl adenin or kinetin. High concentrations of benzyl adenin (above 3.0 mg. $L^{-1}$ ) decreased the adventitious root growth and ginsenoside productivity. N $H_{4}$$^{+}$ inhibited the ginsenoside accumulation, while high concentrations of $K^{+}$, $Mg_{2}$$^{+}$, and $Ca_{2}$$^{+}$ increased it. N $H_{4}$$^{+}$ at 0.5 and 1.0 times of the normal amount in 3/4 SH medium resulted in the greatest biomass increase, but the highest ginsenoside productivity was obtained when N $O_{3}$$^{-}$ was used as the sole nitrogen source in the medium. Most microelements at high concentrations in the medium inhibited the root growth, but high concentrations of MnS $O_4$enhanced the root growth. Root dry weight increased with increasing sucrose concentrations up to 50 g. $L^{-1}$ , but decreased from 70 g $L^{-1}$ Ginsenoside productivity was maximized at the range of 20 to 30 g. $L^{-1}$ sucrose. In the experiment on bioreactor types, cone and balloon types were determined to be favorable for both adventitious root growth and ginsenoside production. Jasmonic acid was effective for increasing ginsenoside contents and Rb group ginsenosides mainly increased. These results could be employed in commercial scale bioreactor cultures of Panax ginseng.x ginseng.

In the previous observations, it was reported that both total ginseng saponin and panaxadiol revealed the marked secretory effect of catecholamines (CA) from the rabbit adrenal gland and that CA secretion induced by them is due to dual mechanisms, cholinergic action and the direct action. In the present study, an attempt to investigate the effect of panaxatriol-type saponin (PT), which is known as an active component of Korean ginseng, on the secretion of CA from the rabbit adrenal gland was made. PT(200 $\mu$g) administered into adrenal vein evoked significantly secretion of CA from the isolated perfused rabbit adrenal gland. Secretory effect of CA produced by PT was attenuated clearly by treatment with chlorisondamine or adenosine, but was markedly increased by physostigmine. Perfusion of Krebs solution containing PT (200 $\mu$g) for 30 min potentiated greatly secretion of CA induced by acetylcholine. PT-induced CA secretion was weakened considerably by ouabain treatement or perfusion of calcium-free Krebs solution. These experimental data demonstrate that PT releases CA from the isolated perfused rabbit adrenal gland by a calcium-dependentd exocytotic mechanism. It seems that the secretory effect of PT is caused through the release of acetylcholine form cholinergic terminals present in the adrenal gland and a direct action on the chromaffin cell itself.

Ginseng, the root of Panax ginseng CA Meyer, is well known as a tonic medicine for restoring and enhancing human health. In traditional medicine, ginseng is utilized for the alleviation of emesis, which includes nausea and vomiting. However, it has not yet been demonstrated whether ginseng exhibits in vivo anti-nausea and anti-vomiting properties. In this study, we examined the anti-emetic effect of Korean red ginseng total extract (KRGE) on cisplatin-induced nausea and vomiting using ferrets. Intraperitoneal administration (i.p.) of cisplatin (7.5 mg/kg) induced both nausea and vomiting with one-hour latency. The episodes of nausea and vomiting reached a peak after 1.5 h and persisted for 3 h. Treatment with KRGE via oral route significantly reduced the cisplatin-induced nausea and vomiting in a dose-dependent manner. The anti-emetic effect was 12.7 $\pm$ 8.6, 31.8 $\pm$ 6.9, and 67.6 $\pm$ 4.0$\%$ with doses of 0.3, 1.0, and 3.0 g/kg of KRGE, respectively. Pretreatment with KRGE via oral route 1 and 2 h before cisplatin administration also significantly attenuated the cisplatin-induced nausea and vomiting. However this did not occur with a pretreatment 4 h before cisplatin administration. These results are supportive of KRGE being utilized as an anti-emetic agent against nausea and vomiting caused by chemotherapy (i.e. cisplatin).
Red ginseng acidic polysaccharide (RGAP), isolated from Korean red ginseng (Panax ginseng C.A. Meyer), has been shown to have a variety of biological functions such as immunostimulating and anti-tumor activities. In the present study, we investigated whether RGAP inhibited ligand-induced platelet aggregation. The washed platelet-rich plasma was prepared from male SD rats with successive centrifugation. The platelets $(10^8/ml)$ were preincubated with 1 mM of $CaCl_2$ for 2 min either in the presence or in the absence of RGAP $(10{\sim}50\;{\mu}g/ml)$ and were stimulated with collagen (2.5 ${\mu}g/ml$) and thrombin (0.1 U/ml). RGAP dose-dependently inhibited thrombin-induced platelet aggregation with $IC_{50}$ value of $26.2{\pm}2.0$${\mu}g/ml$. In collagen-induced platelet aggregation, RGAP inhibited the reaction with an $IC_{50}$ value of $31.5{\pm}3.0\;{\mu}g/ml$. RGAP potently suppressed the intracellular calcium ion, which was stimulated by thrombin (0.1 U/ ml). Among mitogen-activated protein kinase (MAPK) subtypes, the extracellular signal-regulated kinase (ERK) 1/2 and p38 MAPK were analyzed in the present study. RGAP inhibited the phosphorylation of ERK2 and p38 MAPK, which was activated by collagen (2.5 ${\mu}g/ml$). Finally, these results suggested that besides saponin fraction, RGAP take an important role in the preventive effect of Korean red ginseng against cardiovascular disease such as thrombosis and atherosclerosis.
Background: Ginseng produced by hydroponics can be cultivated without using agricultural chemicals; thus, it can be used as a raw materials for functional foods, medicines, and cosmetics. This study aimed to determine the optimal harvesting time to obtain the highest levels of ginsenoside and ginseng, as this was not previously unknown. Methods and Results: One-year-old organic ginseng seedlings were transplanted and cultivated using hydroponics for 150 days in a venlo-type greenhouse, using ginseng nursery bed soil and a nutrient solution ($NO_3{^-}-N$; 6.165, P; 3.525, K; 5.625, Ca; 4.365, Mg; 5.085, S; $5.31mEq/{\ell}$). Ginsenoside content and fresh and dry weights were higher at 120 days after transplanting than at 30, 60, 90, and 150 days. Total ginsenoside content was 11.86 times higher in the leaf and stem than in the root at 120 days after transplanting. Ginsenosides F1, F2, F3, and F5 were detected in ginseng leaves and stems. These chemical compounds are known to be effective in altering skin properties, including whitening, anti-inflammation, and anti-aging. Conclusions: Optimal harvesting time for ginseng cultivated using hydroponics was 120 days after transplanting when the biomass and ginsenoside content were highest.
To study the cause of physiological disorder in leaf of ginseng cultivated at paddy soil, the degree of brown-colored symptom (BCS) and the contents of inorganic matter in leaf were investigated by irrigating the solution of ferric and ferrous iron of 0.1 ~ 2.0%, and citric acid of 1.0 ~ 4.0% on bed soil, respectively. Ratio of BCS by variety was as high as 85.0% in Yoenpoong, while it was as low as 5.4%, 7.5% in Chunpoong and Hwangsook, respectively. The contents of inorganic matter of leaf in Yoenpoong were lower in $P_2O_5$, Ca, and Mg, while it were higher in K, Fe, and Mn than other variety. Iron solution caused BCS more distinctly when each ferric and ferrous iron were dissolved with 1.0% citric acid than when each iron was dissolved without citric acid. Ferric iron caused BCS more effectively than ferrous iron. BCS occurred in 4.0% citric acid was as same as 2.0% ferric iron mixed with 1.0% citric acid. Low $P_2O_5$ and high Fe content in leaf appeared in both of artificial and natural symptoms. We concluded that excessive Fe uptake caused BCS to leaf because the solubility of iron was increased in condition of low soil pH.
Panax ginseng CA Meyer, a herb from the Araliaceae, has traditionally been used as a medicinal plant in Asian countries. Ginseng extract fermented by ginsenoside-${\beta}$-glucosidase treatment is enriched in ginsenosides such as Rh2 and Rg3. Here we show that a fermented ginseng extract, BST204, has anti-proliferative and anti-invasive effects on HT-29 human colon cancer cells. Treatment of HT-29 cells with BST204 induced cell cycle arrest at $G_1$ phase without progression to apoptosis. This cell cycle arrest was accompanied by up-regulation of tumor suppressor proteins, p53 and p21$^{WAF1/Cip1}$, down-regulation of the cyclin-dependent kinase/cyclins, Cdk2, cyclin E, and cyclin D1 involved in $G_1$ or $G_1/S$ transition, and decrease in the phosphorylated form of retinoblastoma protein. In addition, BST204 suppressed the migration of HT-29 cells induced by 12-O-tetradecanoylphorbol-13-acetate, which correlated with the inhibition of metalloproteinase-9 activity and extracellular signal-regulated kinase activity. The effects of BST204 on the proliferation and the invasiveness of HT-29 cells were similar to those of Rh2. Taken together, the results suggest that fermentation of ginseng extract with ginsenoside-${\beta}$-glucosidase enhanced the anti-proliferative and the anti-invasive activity against human colon cancer cells and these anti-tumor effects of BST204 might be mediated in part by enriched Rh2.
본 실험에서는 인삼 씨의 식품 유용자원으로서의 가능성을 알아보기 위해, 각국 인삼 씨의 무게, 일반성분, 무기질, 지방산 조성, 총 당, 산성다당체 함량을 비교해보았다. 먼저 인삼 씨의 백립중은 AGS가 $4.21{\pm}0.31g$으로 가장 무거웠다. 일반성분의 경우 조단백질 함량은 AGS가 $17.00{\pm}1.04%$로 가장 높았고, 조회분 함량은 KGS3가 $8.65{\pm}0.31%$로 매우 높은 값을 나타내 무기질이 풍부한 식품임을 확인할 수 있었다. 이는 저연근의 인삼일수록 무기성분이 많았던 연구결과와 일치했다. 조지방 함량은 AGS가 $24.06{\pm}0.57%$로 가장 높은 값을 나타냈고 조섬유소 함량은 43.21-47.49%로 전체 씨의 절반 정도가 섬유소로 이루어진 섬유질이 매우 풍부한 식품 자원임을 확인할 수 있었다. 무기질은 칼륨과 인이 가장 많이 함유된 것으로 나타났다. 지방산 조성은 96% 이상이 불포화 지방산으로, oleic acid (C18:1, n9c)가 79.93-87.09%로 가장 높게 나타났고, linoleic acid (C18:2, n6c)가 8.94-15.82%로 다음으로 많은 함량을 보였다. 품종이 다른 AGS는 유의적으로 oleic acid와 linoleic acid 조성에 차이가 났고, linolenic acid가 전혀 검출되지 않아 품종에 따른 지방산 조성의 차이를 확인할 수 있었다. 총 당 함량은 15.00-26.17 mg glucose/g으로 인삼에 비해서는 적었지만, 다른 씨와 비교했을 때 매우 높은 수치로 비교적 당을 많이 함유하고 있는 씨라는 것을 확인할 수 있었다. 인삼 산성다당체는 0.56-0.80 mg ${\beta}$-D-galacturonic acid/g을 함유해 소량이지만 최근 유효성분으로 알려진 인삼 산성다당체를 함유하고 있음을 확인하였다.
