  • In An invertase (EC 3.2.1.26) was extracted from Korean giseng (Panax ginseng C.A. Meyer) with distilled tvater The ginseng invertase was purified about 62.6 folds purified by procedures including ammonium sulfate fractionation , DEAE-cellulofine chromatography and gelfiltrations through Sephadex G-75 and the recovery of enzyme activity was 11.1%. The homogeneity of the purified enzyme was probed by polyacrylamide gel disc electrophoresis. The purifled enzyme was divided into two different subunits by treating with a mixture of SDS and 2-mercautoethanol, and the molecular weight of the large subunit was estimatedtobe 116,000 and that of the small one to be 14,000. The optimal VH and temperature of the enzyme were pH 6 and 45$^{\circ}C$, respectively. The enzyme hydrolyzed specifically the hydrolyzation of the -fructofuranosides such as sucrose, raffinose and inulin. The Km values of the enzyme for sucrose and raffinose were determined to be 0.85 and 0.6 mM, respectively.

  • This study investigated the relations between the photosynthetic rate and the activities of antioxidatile enzymes, glutathione reductase, ascorbate free radical reductase, ascorbate peroxidase, glutathione peroxidase, and ascorbate oxidase, in the leaves of Panax ginseng. Under the normal cultivation condition, Panax in showed lower g1utathione reductase and ascorbate free radical reductase activities the Glycine max. But P ginseng showed higher 91utathione Peroxidase, ascorbate Peroxidase, and ascorbate oxidase activities than C. Panax. Therefore, P. ginseng showed weak activities of reductases for the reduction of the oxidized antioxidants. Under the light intensity of 25 KLux, the reductases showed a decrease of over 75% after 24 hours. But the peroyoxidases decreased about 40%. These results showed that the decrease of reductases acitivities was consistent with the decrease of photosynthetic rate. Therefore, we consider that the regulation of antioxidative enzymes or the application of exogenous antioxidants will be effective means for the protection of photodamage in p. ginseng.

  • This study was carried out to compare the root yields, root characters, saponin and ginsenosides contents of 6-year-old p. ginseng and p. quinquefolium. These two ginseng species showed difference in the diameter and ten비h of mainroot. The main root length or p. quinquefolium was shorter than that of p. ginseng, whereas Jakyung-jong and Hwangsook-jong of P. ginseng showed similar root length. Proximate composition were similar between the two species, however, crude fibercontent was significantly higher in main and lateral root of Jakyung-jong and Hwang sook-jong of P ginseng than P quinquefolium. In regard to mineral contents of root, P ginseng contained more Ca and Mn and less Fe and Al than P. quinquefolium. P. quinquefolium contained more of Rbl and Rd of protopanaxadiol saponin, and less or Re, $Rg_1$ and $Rg_2$ of protopanaxatriol saponin than P ginseng. However, no Rf was detected in the p. quinquefolium. Key words Panax ginseng, Panax quinquefolium, ginseng character, ginsenoside.

  • We have studied the effect of ouabain, tool ginseng saponin, panax saponin C (protopanaxatriol derivative) and ginsenoside $Rb_{1}$ (protopanaxadiol derivative) on $Na^+,K^+$-ATPase and $Mg^{++}$-ATPase activities were determined by the method of Robinson and ATPase activities were determined by the method of King. The $Na^+,K^+$-ATPase activities were inhibitied by 90.1% and 51.1% respectively at the concentration of $10^{-3}M$ and $10^{-4}M$ ouabain. The results are consistent with those of Robinson. The $Na^+,K^+$-ATPase activities were increased by 14.3% and 10.0% respectively at the concentration of $10^{-4}$g/ml and $10^{-5}$g/ml total ginseng saponin. Panax saponin C obtained by the method of Han and ginsenoside $Rb_{1}$ obtained by the method of Shibata were used. The $Na^+,K^+$-ATPase activities were increased in the presence of panax saponin C and the increased activity with panax saponin C was greater than that with total ginseng saponin. On the other hand ginsenoside $Rb_{1}$ showed an inhibitory effect on $Na^+,K^+$-ATPase. Total ginseng saponin, panax saponin C and ginsenoside $Rb_{1}$ had no effect on $Mg^{++}$-ATPase. Therefore, it may be concluded that total ginseng saponin has dual effects on microsomal $Na^+,K^+$-ATPase, that is, panax saponin C exhibits stimulatory action, whereas ginsenoside $Rb_{1}$ shows inhibitory action.

  • Korea has produced large quantities of Panax Ginseng roots which have a stimulating effect on the metabolisma of protein, lipid and nucleic acids in the body. Authors believe that the lear and trunk of Panax Ginseng might have some components possessing a similar activity to Panax Ginseng root although the quantity and quality of the functional components may be somewhat different. Therefore, this study was designed to observe the nutritional effects of diet supplemented with the leaves or trunks of Panax Ginseng. Weanling(body weight; $82{\pm}3g$) male albino rats were subjected to six different dietary groups as followings; A groups; dietary groups which were treated with steam for 30 min at $115^{\circ}C}$. B Groups; dietary groups which were not treated with steam. A-C (or B-C) dietary group; Control for A groups(or B groups) containing 99% wheat flour. A-1 (or B-1) dietary group; dietary group supplemented with 2% leaf of Panax Ginseng, which replaced 2% wheat flour of control diet. A-2 (or B-2) dietary group; dietary group supplemented with 2% trunk of Panax Ginseng, which replaced 2% wheat flour or control diet. Each group of rats was maintained with the corresponding diet for 40 days. And then they were sacrificed. The growth rate, protein efficiency ratio, and the contents of lipid and cholesterol in organs were determined. The results obtained are summarized as follows;1) The gained body weights of dietary group supplemented with 2% leaf(A-1 and B-1) or 2% trunk(A-2 and B-2) of panax Ginseng were more increased in comparison to the corresponding control group(A-C and B-C). 2) The gained body weight of each group in A-group(A-C, A-I and A-2) was higher than that or each corresponding dietary group in B-group(B-C, B-1 and B-2). 3) The protein efficiency ratios of A-1 and A-2 dietary group, and B-1 and B-2 dietary group were more improved in comparison to the corresponding control group(A-C and BC). 4) The lipid contents in the liver of A-1 and B-1 dietary groups were lower than in that of A-C and. B-C dietary group, respectively. According to the above results, it could be suggested that the nutritional value of the wheat flour can be improved by supplement of 2% leaf or 2% trunk of Panax Ginseng.

  • The disease development of root-rot [pathogen:Cylindrocarpon destruction (Zinssm.) Scholten] occurred in 2-year old ginseng (Panax ginseng C.A. Meyer) was investigated in the continuous (the first cultivation: 1978∼1982, 2nd cultivation: 1990∼1993 and abolished in 1993 due to replanting problem) and replanted cultivation (period of cultivation: 1980∼1984). In the continuous cultivation, incidences of root-rot were 0.7% on May 3, 48.6% on May 24, and 95.8% on June 14, respectively. In the replanted cultivation, no symptom was observed on May 3 and disease incidence was generally lower compared with the continuous cultivation. On the aerial part of the ginseng infected by C. desiccates, the end and/or margin of leaves were changed to dark reddish color that appeared for behind the root-rot symptom. In this field, the longitudinal growth of lateral root was more inhibited than in the case of the replanted cultivation by C. destmctans. The inhibition rate of rootlet growth was 37.3% in the continuous cultivation as compared with that of replanted cultivation at June 14. Though lesions of root-rot were formed all over the roots, 61.2% of the lesions was positioned within 6 cm under the rhizome. Key words Panax ginseng, Cylindrocarpon destmtan, root-rot of ginseng, replanting problem of ginseng.

  • This study was carried out to obtain the basic information for the development of new ginseng varieties. The two variants (violet-stem variant and yellow-berry variant) of Korean ginseng (Panax ginseng C.A. Meyer) and American ginseng (Panax quinquefolium L.) of one to four-year were used for this study. All of the characteristics, such as leaf length, leaf width, petiol length, number of leaves per plant, number of leaflets per plants, stem diameter, stem length, number of stems per plant, root length, primary root length, root diameter, root weight were determined and correlations among them were estimated. The results obtained were summarized as follows. 1. Leaf length, petiol length, number of leaves per plant, and number of leaflets per plant of Panax ginseng, violet-stem variant and yellow-berry variant, were larger than those of Panax quinquefolium at all of the plant ages, while leaf width was wider in Panax quinquefolium. 2. The length of stem of Panax quinquefolium was shorter than that of Panax ginseng, and the frequency of multi-stem plants at 4-year-old ginseng was larger in violet-stem variant than in Panax quinquefolium and yellow-berry variant. 3. In the characteristics of ginseng root, the primary root length of Panax ginseng, violet-stem variant and yellow-berry variant, were less than that of Panax quinquefolium, while root weight, root diameter, and umber of secondary root related to yield were larger in Panax ginseng. 4. The root weight per plant related to the yield had positive and highly significant correlations with stem diameter, leaf length, leaf length, leaf width, number of compound leaves and leaflets in Panax ginseng and Panax quinguefolium. 5. The root weight related to the wield of ginseng had been influenced to stem diameter, leaf length, and leaf width directly, and number of compound leaves and leaflets indirectly. 6. The number, total area and activity of stomate per mm2 of Panax quinquefolium were more, larger and stronger than those of Panax ginseng.

  • The seasonal variation in the activity of sucrose synthase, ADP-glucose pyrophosphorylase and UDP-glucose pyrophosphorylase in roots of Panax ginseng C.A.Meyer have been studied. It was revealed that sucrose synthase and ADP-glucose pyrophosphorylase are adaptive enzymes and can serve as markers of sink strength, while UDP-glucose pyrophosphorylase is the maintenance enzyme. The average day temperature exceeded 24。C appeared to cause the disturbance in refilling process, affecting the starch synthesis. Study on the dependence of oxygen consumption in stele tissue with temperature revealed the sharp accelerating of this process after 24。C.
