복분자 열매의 화학성분 및 휘발성 향기성분

  • 최근 복분자(산딸기)의 효능을 이용한 일반식품 및 기능성 식품 등의 제품 개발이 활발히 추진되고 있다. 따라서 본 연구에서는 식품차원의 연구분야 일환으로서 먼저 복분자의 일반성분, 유리당, 유리아미노산, 무기성분 및 향기성분 등을 보고한 후 앞으로 생리 활성 연구 분야에 초점을 두고 연구를 진행할 하여 학문적 자료 및 산업에 응용할시 기초자료로 활용하고자 한다. 복분자의 수분은 5.39%, 총당은 17.3%, 환원당은 8.6%, 조단백질 10.6%, 조회분은 4.5%, 조지방은1.7%, 조섬유는 3.9%이고, 유리당 함량은 sucrose 1.52%, fructose 3.98%, glucose 1.24%이고, 유기산 함량은 citric acid 10.2%, oxalic acid 6.29%로 가장 많이 함유하고 있었고, 그 다음은 malic acid 1.94%였다. 총 아미노산은 7914.1mg%이고, 필수아미노산 함량은 2622.5mg%로 나타났다. 복분자에 가장 많이 함유되어 있는 아미노산은 aspartic acid 1260.3%, 그 다음은 glutamic acid 1054.3% 순으로 함유하고 있었다. 무기성분 K가 387mg으로 가장 많이 함유되어 있었고, 복분자에서 총 52종의 성분이 화인되었는데 확인된 성분들은 관능기별로 분류하면 amylalcohol를 포함한 alcohol류 11종, valeric acid를 포함한 acid류 13종, hexanal를 포함한 carbonyl류 20종, 2-heptanone를 포함한 hydrocabon류 5종 그리고 methyl palmitate를 포함한 ester류가 3종 이었다. 관능기별로 확인된 성분들의 총 peak area%를 살펴보면 alcohol류 11.56%, acid류 38.87%, hydrocarbon류 2.89%, ester류 0.80%, 총 70.32%가 확인되었다. Alcohol류에서 linalool가 3.78%, acid류에서는 caproic acid류가 14.40%, carbonyl류에서 2-hydroxy-4-methoxyacetophenone이 2.99%, hydrocabon류에서는 aromadendrene가1.59% 그리고 ester류에서는 methyl palmitate가 0.43%으로 가장 많이 함유하고 있었다.

Effect of N-Methyl Acetamide on the Critical Micelle Concentration of Aqueous Solutions of Some Surfactants (수용액상 계면활성제의 임계 미셀 농도에 대한 N-methyl acetamide의 영향)

  • The variations of molar conductivity of various surfactants such as sodium caprylate, sodium laurate, sodium palmitate, sodium stearate, sodium oleate, sodium dodecyl sulphate, and lithium dodecyl sulphate with concentrations of the surfactants for each of the solutions consisting of mixtures of varying concentrations of N-methyl acetamide in water at constant temperature of $30{\pm}0.2^{\circ}C$ were studied. The critical micelle concentration (CMC) for each surfactant is measured. It is found that the CMC values in mixtures of N-methyl acetamide and water solutions of various surfactants are lower than the CMC values in water, and the driving force for micelle formation correlates with solvophobicity. The surfactant-solvent interactions that drive amphiphilic self-organization in N-methyl acetamide in water are discussed. Thermodynamic parameters were evaluated for micellar system to explain the results.

Oxidation Stability of Fish Oil Containing Commercially Available Antioxidants (상업용 천연 항산화제의 사용에 다른 어유의 산화 안정성 연구)

  • The effects of commercially available antioxidants in fish oil were studied. Induction period of fish oil was determined from the oxidation curve by rancin\mat. The longest Induction period was observed with catechin (1,000 ppm). Among the rosemary extracts (Antox1, Antox2, Antox3), the most effective antioxidant effect was observed with Antox3 even though higher amount (5,000 ppm) was needed compared to catechin. Compared to oder of control, catechin rather than Antox3 did not affect much the odor changes. When ascorbic palmitate, vitamin C, gallic acid, EDTA, citric acid, or propyl gallate as a synergist were added with catechin (500 ppm), vitamin C and ascorbic palmitate prolonged the induction period significantly. This effect was also observed with Antox3 (1,000 ppm). Among all combinations of catechin (500 ppm) and Antox3 (1,000 ppm) with synergists, the longest Induction period was obtained from Antox3 with vitamin C (200 ppm), suggesting that this combination is most effective combination for retarding the oxidation in fish oil.

Composition of Lipids Associated with Dense Coat-enriched Fractions of Bovine Milk Fat Globule Membrane (우유 지방구막의 고밀도 표피에 결합된 지질의 조성)

  • To analyze the components of dense coat fractions associated with fat globule membrane, The membrane was treated with various concentrations of Triton X-100, a non-ionic detergent, and the composition of lipids associated to the detergent insoluble material was analyzed. The amount of protein, phospholipid, cholesterol and ganglioside in milk fat globule membrane was reduced consistently with increasing concentrations of Triton X-100. Butyrophilin (band 12), xanthine oxidase (band 3) and band 16 as constituents of insoluble coat materials was revealed after electrophorisis on SDS-polyacrylamide gels. Phosphatidyl ethanolamine, phosphatidyl choline, phosphatidyl serine, phosphatidyl inositol and sphingomyelin were identified as the major phospholipids of the coat materials without selective concentration relative to the original membranes. Percentages of total phospholipid were not changed by any of the treatments. Fatty acids of total lipid were myristate, palmitate, stearate (major saturated acids), oleate and linoleate (major unsaturated acids). Cholesterol contents on a protein basis were slightly reduced with increasing concentrations of Triton X-100. Cholesterol adhered to protein more tightly than other constituents The contents of gangliosides was proportionally refuted with increasing concentration of Triton X-100.

Anti-adipogenic effect of the flavonoids through the activation of AMPK in palmitate (PA)-treated HepG2 cells

  • Background: Flavonoids are natural polyphenols found widely in citrus fruit and peel that possess anti-adipogenic effects. On the other hand, the detailed mechanisms for the antiadipogenic effects of flavonoids are unclear. Objectives: The present study observed the anti-adipogenic effects of five major citrus flavonoids, including hesperidin (HES), narirutin (NAR), nobiletin (NOB), sinensetin (SIN), and tangeretin (TAN), on AMP-activated protein kinase (AMPK) activation in palmitate (PA)-treated HepG2 cells. Methods: The intracellular lipid accumulation and triglyceride (TG) contents were quantified by Oil-red O staining and TG assay, respectively. The glucose uptake was assessed using 2-[N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-d-glucose (2-NBDG) assay. The levels of AMPK, acetyl-CoA carboxylase (ACC), and glycogen synthase kinase 3 beta (GSK3β) phosphorylation, and levels of sterol regulatory element-binding protein 2 (SREBP-2) and 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR) expression were analyzed by Western blot analysis. The potential interaction between the flavonoids and the γ-subunit of AMPK was investigated by molecular docking analysis. Results: The flavonoid treatment reduced both intracellular lipid accumulation and TG content in PA-treated HepG2 cells significantly. In addition, the flavonoids showed increased 2-NBDG uptake in an insulin-independent manner in PA-treated HepG2 cells. The flavonoids increased the AMPK, ACC, and GSK3β phosphorylation levels and decreased the SREBP-2 and HMGCR expression levels in PA-treated HepG2 cells. Molecular docking analysis showed that the flavonoids bind to the CBS domains in the regulatory γ-subunit of AMPK with high binding affinities and could serve as potential AMPK activators. Conclusion: The overall results suggest that the anti-adipogenic effect of flavonoids on PA-treated HepG2 cells results from the activation of AMPK by flavonoids.

  • Experiments were carried out to develop of mutant strain from Pichia ciferrii ATCC 14091 and investigate the effect of sodium acetate on the production of tetraacetylphytosphingosine (TAPS). A high-TAPS producing mutant was simply selected by staining with sudan black B and colony shape after the treatment of UV and NTG. Sodium acetate enhanced the TAPS production.

  • A high performance liquid chromatographic procedure is described for the direct determination of the picomole amount of palmitoyl-Coenzyme A and stearoyl-Coenzyme A, using a stainless steel column packed with C-18 derivatized porous silica ($5{\mu}m$), an isocratic elution with a mixture of 33 mM $KH_2PO_4$/acetonitrile as a mobile phase and a UV detector. The long-chain acyl-Coenzyme A esters were determined in incubated microsomal fractions of a bovine liver to demonstrate the utility of this method for monitoring acyl-CoA synthesis in biological samples. The reaction rate of palmitate was higher than that of stearate. After a 60 minute incubation period, the generated amount of palmitoyl-Coenzyme A and stearoyl-Coenzyme A were approximately 70 and 20 n mol/mg micresomal protein, respectively. The advantage of this method are in that no decomposition of the CoA esters is involved, while the constituent molecular species is detected.

  • Antibiotic activity and antipyretic effect of three synthetic prodrugs, chloramphenicol acetylsalicylate (CAF-ASP), chloramphenicol naproxenate(CAF-NAX) and chloramphenicol acetaminophen succinate (CAF-SUC-ACET), were studied in vitro and in vivo. In the experimental results, the CAF-NAX showed the moat rapid absorption and the highest blood levels, and the CAF-SUC-ACET and CAF-ASP also were more rapid absorption and higher blood levels than controls, chloramphenicol and chloramphenicol palmitate. The synthetic prodrugs also showed the antipyretic effect by modified Roszkowski method using endotoxin.

  • Two ergosterins and two triterpenoids were isolated from the dried fruit bodies of Ascomyce Bulgaria inquinans. By means of chemical (hydrolysis) and spectroscopic methods (NMR, EIMS), their structures were established as betuinic acid (1), cerevisterol (2), (24R)ergosta-7, $22E­diene-3\beta,\;5\alpha,\;6\beta-triol-3-O-palmitate$ (3) and ursolic acid (4). Compound 3 is a new compound.

Purification of Cholesterol Esterase from Aeromoans SP. (Aeromoans SP.가 생산하는 콜레스테롤 에스테라아제의 정제)

  • A cholesterol esterase-producing microorganism, strain CES 506, isolated from soil was identified as Aeromonas sp. This strain produce about 0.023 units of cholesterol esterase per ml of culture broth. The cholesterol esterase produced by this strain was purified, 370 fold to homogeneity in an overall yield of 24% from culture broth. The apparent molecular weight was 64, 000, as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The enzyme showed a high substrate specificity for cholesterylpalmitate and the Km value for the hydrolysis of cholesterylpalmitate by this enzyme were 0.15mM.
