Antifntique Effect and Improvement of Hematotoxicity by Dongchongxiacno (Paecilomyces japonica ) (누에동충하초(Paecilomyces japonica)의 항피로효과 및 조혈장해 개선효과)

  • Cordyceps is reputed for its broad biological activities and as a tonic for replenishing vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on fatigue and hematotoxicity were investigated. The antifatigue activity of dongchongxiacao (Paecilomyces japonica) was studied in mice using weight loaded forced swimming performance method. The water extract of dongchongxiacao(25 mg/kg of body weight, intraperitoneal injection at 12 and 36 hours before irradiation and 30 minute and 24 hours after irradiation) showed a significant antifatigue effect in normal mice and radiation-induced fatigue mice (p<0.0005). In the experiment on the improvement of hematotoxicity, a significant difference from the irradiated controls was seen at day 3 of the group treated with dongchongxiacao and substantial differences from the irradiated controls at day 7 was also detected.

Cytotoxicity of Acetoxyscirpendiol from Paecilomyces tenuipes (Paecilomyces tenuipes로부터 분리한 Acetoxyscirpendiol의 세포사멸작용)

  • Paecilomyces tenuipes is one of the famous Chinese medicinal entomopathogenic fungi that parasite in the lavae of silkworm. A cytotoxic compound, 4$\beta$-acetoxyscirpendiol (ASD) was isolated from a methanolic extracts of Paecilomyces tenuipes. The ASD compound belongs to scirpenol subfamily of trichothecene mycotoxin. In a continuation of the elucidation of the mechanism of ASD, we report here the evidences of induction of apoptosis by ASD in human Jurkat T cell line. In MTT reduction assay for monitoring cell viability, ASD showed strong toxicity. The 50 percent inhibitory concentrations of ASD against human T lymphoid Jurkat cell was 59.5 ng/$m\ell$. Phosphatidylserine externalization was increased by ASD at 3 and 6 hrs when compared with that of 6 hrs in the cell line showing in a time-dependent manner. When whole lysates of cells treated with ASD were subjected to western blot assay, 113 kDa poly(ADP-ribose) polymerase (PARP) was significantly cleaved to 89 kDa fragment. Time-dependent DNA fragmentation was also observed when Jurkat T cells were treated with ASD at 100 ng/$m\ell$ for 6 hrs and 18 hrs at the ratios of 8.5% and 15.0%, respectively. From these data, Jurkat T lymphocytes treated with ASD from Paecilomyces tenuipes underwent typical cascades of apoptotic cell death.

Nomenclatural Studies on Paecilomyces tenuipes in Korea (국내 자생 눈꽃동충하초 균의 명명 고찰)

  • Paecilomyces tenuipes (Nunkkot Dongchunghacho) was erroneously treated in three different scientific names: Paecilomyces japonica, Isaria japonica and Paecilomyces tenuipes. Since the fungus has been widely used in various fields of research and industry, its nomenclatural uncertainty caused many problems. Therefore, morphological studies of Korean isolate were carried out and compared with published data on other isolates of P. tenuipes to establish its proper taxonomic position.

Optimization for the Preparation of Jeung-Pyun, with Added Paecilomyces japonica Powder, Using a Response Surface Methodology (반응표면분석법에 의한 동충하초 첨가 증편 제조의 최적화)

  • To optimize the preparation process of Jeung-Pyun, an experiment was derived, using a central composite design, to fmd the optimal mixing conditions. The addition of Paecilomyces japonica powder, the volume of Tak-Ju and the second fermentation time were independent variables, with the pH, volume, color, sensory and texture properties of the Jeung-Pyun selected as response variables. As the Paecilomyces japonica powder and fermentation time were increased, the b values and volume after the second fermentation also increased. The color of the Jeung-Pyun became deeper with increases in the Paecilomyces japonica powder and fermentation time, and was deepest on the addition of 30% Tak-Ju. The pH increased with increasing fermentation time of the Jeung-Pyun, and on the addition of 3% Paecilomyces japonica rapidly increased more than with the other groups. The optimal mixing conditions for the best quality Jeung-Pyun, with restricted conditions above a b value of 3, a color intensity above 3, and a volume of 40mQ and a pH of 4.02-4.04 after the second fermentation, were 4.0%, 20% and 45 mins for the Paecilomyces japonica powder, Tak-Ju and second fermentation time, respectively.

  • Dongchunghacho is traditionally believed to be effective against various diseaseas. It includes many different genera such as Cordyceps, Paecilomyces, Torrubiella and Podonectria. The three fungus strains, Cordyceps militairs, Cordyceps scarabaeicola and Paecilomyces tenuipes were individually cultivated as supension cultures in potato dextrose broth. (omitted)

  • While studying Bombyx mori "Zipnuae" inoculated with an entomopathogenic fungus, Paecilomyces tenuipes, a mycoparasitic hymenoascomycete with a long neck was isolated from the silkworm rearing room. The fungus was identified as Syspastospora parasitica which has been recognized as a hyperparasite on various moniliaceous hyphomycetes. A descriptive illustration is presented in this study.

  • Thecodiplosis japonensis is an important insect pest in pine trees and hard to be controlled by chemical pesticides due to its characteristic life cycle. Experiments were conducted to determine the pathogenicity and growth temperature of several entomopathogenic fungi as potential microbial insectcides for T. japonensis. Beauveria bassiana SFB-SC and Paecilomyces fumosoroseus SFP-198 were highly pathogenic against T. japonensis. B. bassiana SFB-SC had also pathogenicity against a beneficial insect Bombyx mori. Growth rate and pathogenicity of P. funmosoroseus SFP-198 under 15$^{\circ}C$ was higher than those of B. bassians SFB-SC. These results indicated that P. fumosoroseus SFP-198 is a potential microbial control candidate for control of T. japonensis.

  • In the present paper, a new variety of Cordyceps gunnii (Berk.) Berk. is reported. It differs from C. gunnii by its much smaller secondary ascospores, (2.0-) 3.3 $(-4.3){\times}1.0{\sim}1.8\;{\mu}m$, as well as thinner asci and cap of the asci. Its anamorph is also reported as a new variety of Paecilomyces gunnii Liang, the ansmorph of C. gunnii. Its differ from P. gunnii by its conidia, mostly subglobose and much bigger, (2.2-) 4.7 $(-11.0){\times}(1.8-)\;2.6\;(-3.3)\;{\mu}m$. The type specimen and dry type culture are deposited at Research Center for Entomogenous Fungi, Anhui Agriculture University, China.

  • In the previous report, we described the in vivo antitumor activity of PJ-4, a protein-polysaccharide fraction prepared from the culture filtrate of an insect-born fungus, Paecilomyces tenuipes DGUM 32001. In the present study, we elucidated the immunomodulating activity of PJ-4 on the BALB/c mouse splenic lymphocytes using flow cytometrical techniques. As a result, PJ-4 was found to stimulate the lymphocytes not only to form lymphoblasts but also to express CD25 (IL-2 receptor $\alpha$ chain) molecule, which is well known as a T cell activation marker. More interestingly, its T cell stimulatory activity was more strongly exerted on CD8$^{+}$ T cells than on CD4$^{+}$ T cells. All these data suggest that PJ-4 exerts its antitumor activity at least partly through stimulation of T cells which play major roles in the cell-mediated immune system.tem.
