고정화 생물 반응기에 의한 암모니아 제거 (Removal of ammonia by packed bed bioreactor using immobilized nitrifiers)

  • Nitrifier consortium entrapped in Ca-alginate bead were packed into aerated packed bed bioreactor and non aerated packed bed bioreactor and the performances of two bioreactors were evaluated for the removal of ammonia nitrogen from synthetic aquaculture water. Total ammonia nitrogen(TAN) removal rate was decrease in aerated packed bed bioreactor below 0.3hr of hydraulic residence time(HRT), but increased in non aerated packed bed bioreactor until 0.5hr of HRT. At HRT of 0.05hr, TAN removal rate of non aerated packed bed bioreactor was about 335g TAN/㎥/day and the optimum ratio of packing height and inside diameter of reactor (H/D) was 4. The performance of two bioreactors indicated that non aerated packed bed bioreactor was better than aerated packed bed bioreactor in ammonia removal from synthetic aquaculture water.

혐기성 유동층 생물 반응기와 새로운 모델의 AFPBBR에서 유기성폐수 처리시 Biogas 생성과 반응상수에 관한 연구 (A Study on the Kinetics and the Biogas Formation for Organic Wastewater Treatment in Anaerobic Fluidized-Bed Bioreactor and New Model AFPBBR)

  • The anaerobic digestion of organic synthetic wastewater in anaerobic fluidized bed bioreactor (AFBBR) and anaerobic fluidized packed bed bioreactor (AFPBBR) was studied. This study was conducted to evaluate efficiency and reliability of two reactor. Experiment was performed to find the effect of upflow rate with AFBBR and the height of packed bed with AFPBBR. As a result, this program obtained several conclusion. These are given as follows: As applied the upflow rate increased in AFBBR the produced volume of biogas increased, while the gas production and COD removal decreased at above 0.3 m$^3$/h. When a upflow rate is 0.4 m$^3$/h in AFBBR the volatile suspended solid (VSS) became significantly increased. At an organic loading rate from 0.1 to 0.4 of upflow rate in AFBBR, the methane yield was 1.5584 m$^3$CH$_4$/kgCOD removed, and the observed cell yield coefficient was 0.0933 gVSS/gCOD. In case of AFPBBR, the results showed also that 20 cm of height of packed bed was superior to other in the aspect ot biogas production, the content of methane and COD removal. At 20 cm of height, the profile of microorganisms was stable, while at 30 cm the VSS of effluent became higher than AFBBR. Though COD removal of AFPBBR increased with packed bed, COD removal deteriorate with over packing because the loss of pressure became higher in the reactor. At an organic loading rate from 20 to 40 cm of packed bed in-AFPBBR, the methane yield was 2.5649 m$^3$CH$_4$/kgCOD removed, and the observed cell yield coefficient was 0.0506 gVSS/gCOD. Based upon the results obtained, it is suggested that AFBBR and AFPBBR is the most effective conditions at 0.3 m3/h of upflow rate, the 20cm of packed bed, respectively.   • An experimental investigation has been carried out to evaluate the characteristics in wastewater treatment using an anaerobic packed bed bioreactor with ceramics, rubber sponge, soft stone A, and soft stone B as carrier. The results of the work have shown that soft stone A as a carrier was superior to other carriers in methane production, then the content of methane for soft stone A was about 70%. First of all, soft stone A had higher efficiency of the COD removal than the others in response of passing the operation, as well as it had a low volatile acid in reactor. In addition, the slope of methane production with respect to the removal of COD ($m^{3}CH_{4}$/kgCOD) was 0.58 for soft stone A. In biomass hold-up equation for each carriers, the equation of soft stone A was m$_{p}$=714 ($C_{o}/0.41+C_{o}$) and it was the largest in this experimented carriers. Based upon the results obtained, it is suggested that the major effective carrier in wastewater treatments within the packed bed bioreactor used in this experimental work by soft stone A.

Actinoplanes missouriensis KCTC 1780의 응집 균체에 의한 과당생산 (Production of High Fructose Syrup by Flocculated Actinoplanes missouriensis KCTC 1780)

  • The whole cells of Actinoplanes missouriensis KCTC 1780 which produce glucose isomerase was immobillized by flocculation method for the effective production of high fructose syrup using packed-bed bioreactor system. Among the flocculation methods used In this study, the glucose Isomerase activity of flocculated cells using 5% polyethylenimmine and 0.2% glutaraldehyde was the highest as 46.3 unit, and the flocculant was 10.3g(wet weight) per 100m1 of broth, and the residual activity was 92.5%. In the batch operation of glucose isomerization using the flocculated cells, the optimum pH, temperature and isomerization ratio were 7.0, 75$^{\circ}C$ and 31%, respectively. The optimum concentration of Mg2+ which was activator on the glucose isomerization of flocculated cells was 0.1M, and glucose isomerase activity was increased by about 40% compared to none of Mg2+. In the packed-bed bioreactor system with 1.2 hour of residence time at 7$0^{\circ}C$, the reaction stability maintained until 96 hour without toss of activity, and the equilibrium was kept up to 120 hours of the operation.

동시 추출을 겸한 생물반응기에서 Lithospermum erythrorhizon 배양에 의한 shikonin 생산 (Bioreactor Cultures of Lithospermum erythrorhizon for Shikonin Production with In Situ Extraction)

  • 식물세포인 Lithospermum erythrorhizon을 교반 반응기와 calcium alghinate에 고정화된 상태로 충전층 반응기에서 n-hexadecane으로 동시 추출하면서 shikonin을 생산하여 각각의 생산성을 비교하였다. 교반 반응기에서 shikonin의 비생산과 부피생산성은 각각 1.5mg shikonin/g cell과 400$\mu g$ shikonin/(L.day)였고 충전층 반응기에서는 가각 2.0mg shikonin/g cell과 2857 $\mu g$ shikonin/(L.day)였으며 이는 각각 교반 반응기에 비하여 1.3, 7.1배 높은 것이다. 충전층 반응기에서 shikonin의 생산성이 높은 것은 calcium alginate 입자에 세포가 고농도고 축적되어 단위 반응기 부피당 세로의 부하 능력이 높고 또한 세포가 서로 접촉하기가 쉽고 고정화 입자내의 환경이 세포가 분화하기에 좋은 조건을 형성하기 때문인 것으로 사료된다.

  • Continuous production of hydrogen by Ca alginate-immobilized photosynthetic bacteria was studied in a packed-bed bioreactor. The dilution rate and input concentration of carbonaces substrate were selected as operating parameters. To choose the strain for immobilization, hydrogen productivities of Rhodopseudomonas caposulata 10006 and Rhodospirillum rubrum KS-301 were compared through preliminary batch cultures of their free cells: the former was found to show better hydrogen productivity in spite of its lower specific growth rate. For the continuous production of hydrogen by immobilized R capsulata, the optimum dilution rate was about 0.84 h$^{-1}$ . The Immobilized tells gave better hydrogen yield and conversion efficiency than free ones. And a kinetic parameter K'$_{m}$ was determined for the packed-bed bioreactor, being practically constant for a specific range of dilution rates.s.

  • 본 실험은 질화세균을 고정화한 충전층 생물반응기에서 저농도 암모니아성 질소(total ammonia nitrogen, TAN)의 제거 시 수력학적 체류시간(hydraulic residence time, HRT), 유입수 암모니아성 질소농도 및 온도에 의한 영향을 평가하였다. 수력학적 체류시간이 감소함에 따라 암모니아성 질소의 제거속도는 증가하였고, 암모니아성 질소의 제거 시 최적 수력학적 체류시간은 0.2시간이었다. 수력학적 체류시간이 0.2시간에서 암모니아성 질소의 제거속도는 $226.1\;g/m^3{\cdot}day$, 제거효율은 88.8%을 나타내었다. 유입수 암모니아성 질소 농도가 증가함에 따라 암모니아성 질소의 제거속도는 증가하였다. 온도 $20{\sim}35^{\circ}C$에서 암모니아성 질소의 제거속도와 제거효율은 일정하게 유지했다.

  • 고정화 CGTase를 이용하여 당전이 스테비오사이드를 연속적으로 제조하기 위하여 몇가지 생물반응기의 적용가능성을 조사하였다. 연속형 반응기로는 packed-bed reactor(PBR)가 적합하였다. 이때 specific productivity는 0.4 g/hr-g immobilized enzyme으로 회분식반응기의 50% 정도이나 연속생산이 가능한 장점이 있었다. PBR의 운전조건이고 $10{\sim}60\;cm/hr$의 선속도 범위내에서 스테비오사이드 전환률 70% 이상을 보이는 기질의 유속은 공간속도로 $0.5{\sim}0.7\;hr^{-1}$이었다. 고정화 효소의 운전 안정성은 $47^{\circ}C$에서 30%의 기질로 2주 일간 연속 사용이 가능하고, 반응기의 control방법으로 반응기 상부에서 20%의 효소를 교체하면 20일 이상 전환률이 유지되었다.

Optimization of Tannase Production by Aspergillus niger in Solid-State Packed-Bed Bioreactor

  • Tannin acyl hydrolase, also known as tannase, is an enzyme with important applications in the food, feed, pharmaceutical, and chemical industries. However, despite a growing interest in the catalytic properties of tannase, its practical use is very limited owing to high production costs. Several studies have already demonstrated the advantages of solid-state fermentation (SSF) for the production of fungal tannase, yet the optimal conditions for enzyme production strongly depend on the microbial strain utilized. Therefore, the aim of this study was to improve the tannase production by a locally isolated A. niger strain in an SSF system. The SSF was carried out in packed-bed bioreactors using polyurethane foam as an inert support impregnated with defined culture media. The process parameters influencing the enzyme production were identified using a Plackett-Burman design, where the substrate concentration, initial pH, and incubation temperature were determined as the most significant. These parameters were then further optimized using a Box-Behnken design. The maximum tannase production was obtained with a high tannic acid concentration (50 g/l), relatively low incubation temperature ($30^{\circ}C$), and unique low initial pH (4.0). The statistical strategy aided in increasing the enzyme activity nearly 1.97-fold, from 4,030 to 7,955 U/l. Consequently, these findings can lead to the development of a fermentation system that is able to produce large amounts of tannase in economical, compact, and scalable reactors.

Trends in Monoclonal Antibody Production Using Various Bioreactor Systems

  • Monoclonal antibodies are widely used as diagnostic reagents and for therapeutic purposes, and their demand is increasing extensively. To produce these proteins in sufficient quantities for commercial use, it is necessary to raise the output by scaling up the production processes. This review describes recent trends in high-density cell culture systems established for monoclonal antibody production that are excellent methods to scale up from the lab-scale cell culture. Among the reactors, hollow fiber bioreactors contribute to a major part of high-density cell culture as they can provide a tremendous amount of surface area in a small volume for cell growth. As an alternative to hollow fiber reactors, a novel disposable bioreactor has been developed, which consists of a polymer-based supermacroporous material, cryogel, as a matrix for cell growth. Packed bed systems and disposable wave bioreactors have also been introduced for high cell density culture. These developments in high-density cell culture systems have led to the monoclonal antibody production in an economically favourable manner and made monoclonal antibodies one of the dominant therapeutic and diagnostic proteins in biopharmaceutical industry.
