Gas Permeation Properties of Polymeric Membranes for Biosensor Prepared from Poly(vinyl chloride) Derivatives (Poly(vinyl chloride) 유도체로부터 제조된 바이오센서용 고분자막의 기체 투과특성)

  • Membranes for biosensor were prepared from poly(vinyl chloride) (PVC)l derivatives using the solution casting method, and their gas permeabilities were studied. The polymer membranes dried slowly in air showed higher permeability coefficients than those dried in vacuum. The permeabilily coefficients of carboxylated poly(vinyl chloride) (CPVC) membranes for $O_2$ and $CO_2$ decreased as the pressure of the feed gas increased. The addition of dioctylphthalate (DOP) enhanced the permeation rates for $O_2$ and $CO_2$. For example, the permeability coefficients of CPVC membranes containing 30 wt. % DOP for $O_2$ and $CO_2$ at 100 psig were 2.03 and 0.96 Barrer, respectively, which were about 4~5 times higher than those of the membranes without DOP. Poly(vinyl chloride-co-vinyl acetate-co-vinyl alcohol) (Syn-PVCAcAl) obtained by hydrolysis of poly(vinyl chloride-co-vinyl acetate (PVCA) showed a higher permeability coefficient for $CO_2$ in the presence of DOP than that for commercial PVCAcAl, but did not show any significant difference in permeability for $O_2$.

Determination of Complex Formation Constant of Sodium-Selective Ionophores in Solvent Polymeric Membranes (용매 고분자막 상에 고정된 나트륨 이온선택성 물질의 착물형성상수 결정)

  • The complex formation constants (${\beta}_{MLn}$) of potassium and various sodium-selective neutral carriers in solvent polymeric membranes have been determined using solvent polymeric membrane-based optodes and ion-selective electrodes (ISEs). Two different types of PVC-based membranes containing the H^+selective chromoionophore (ETH 5294) with and without a sodium ionophore (4-tert-bntylcalix[4]arenetetraacetic acid tetraethyl ester, ETH 2120, bis[(12-crown-4)methyl] dodecylmethylmalonate or monensin methyl ester) were prepared and their optical responses to either the changes in alkali metal cation (e.g., sodium and potassium) concentrations at a fixed pH (0.05 M Tris-HCl, pH 7.2) or varying pH at a fixed alkali metal cation concentration (0.1 M) were measured. The same type of membranes were also mounted in conventional electrode body and their potentiometric responses to varying pH at a fixed alkali metal cation concentration (0.1 M) were measured. The complex formation constants of the ligand could be calculated from the calibration plots of the relative absorbance vs. the activity ratios of cation and proton ($a_{M^+}/a_{H^+}$) and of the emf vs. pH. It was confirmed that the ratio values of the complex formation constants for the primary and interfering ions are closely related to the experimental selectivity coefficients of ISEs.

  • Electron microscope radioautography introduced by Liquier-Milward (1956) is now used routinely in many laboratories. Most of the technical difficulties in specimen preparation have been overcome. This method is modified from loop method for improvement of EM radioautographic techniques. The advantages of this method are: 1. the use of single specimens on small corks and of a large wire loop, allows the experimenter to avoid the blemishes in the membrane; 2. the surfactant dioctyl sodium sulphosuccinate is added to diluted ILford L4, thus greatly prolonging the period of time over which good emulsion layers can be made; 3. corks can be handled in perspex holder which allows about 20 specimens to be developed simultaneously. The steps of the method comprise: 1. Cut ribbons of ultrathin sections of silver interference colour 2. Pick them up on formvar-coated 200 mesh grids 3. Prestaining of tissues 4. Coat the specimens with a thin layer of carbon by evaporation (30-60A) 5. Mount the specimens on corks (about 1cm apical diameter) using double-sided scotch tape 6. Emulsion coating; a. Take a 250m1 beaker, place it on the pan of a sliding weight balance and weigh it. Add 10 grams extra to the beam. Add pieces of ILford L4 emulsion to the beaker until the balance is swinging freely. Add the 20ml of distilled water that was previously measured out. b. Surfactant dioctyl sodium sulphosuccinate is added to diluted ILford L4. 7. Prepare a series of membranes of gelled emulsion with the wire loop and apply one to each cork-borne specimen. 8. Put the specimens away to expose by pushing the corks into short length of PVC tubing, each tube having a small hole in the side 9. Place the tubes in small boxes together with silica gel. 10. Exposure 11. Developer - Kodak Microdol X for 3 minutes 12. Fixer - A perspex holder can be manufactured which allows 20 specimens to be developed simultaneously. 12. Fixer - 30% sodium thiosulfate for 10 minutes 13. Examination with Siemens Elmiskop 1A electron microscope

Properties of Silicon Coated Fabric for Membrane Treated by Low Temperature Plasma (저온플라즈마 처리에 의한 실리콘코팅 막구조 원단의 특성변화)

  • 막구조는 근래에 와서 대공간 구조 및 지붕구조에 가장 보편적으로 사용되는 경량 인장 구조물로 각광받고 있다. 구조용 막재는 풍하중 및 설하중에 충분히 감당할 수 있도록 강도와 내구성을 가지고 있어야 한다. 일반적으로 막구조 재는 PVC코팅 폴리에스터막, 실리콘코팅 유리섬유막, PTFE코팅 유리섬유막이 있다. 제직되는 원단의 크기가 한정되어 있기 때문에 재단 후 접착하여 제작한다. 이 때문에 이음부분이 나 재단부분에 코팅으로 인한 접착이 어려워 고온고압으로 접착을 한다. 이 연구에서는 실리콘코팅 유리섬유막의 접착시 어려움을 보완하기 위해 저온 Plasma를 이용한 처리법으로 방전에 의해 Plasma를 발생시켜 50w, 100w 출력으로 10분, 20분간 처리하여 그 결과를 접촉각과 SEM 관찰을 통해 표면처리를 관찰하였다. Plasma 처리로 인해 실리콘 표면층에 균열이 발생하고 표면이 갈라짐을 확인할 수 있었다. 접촉각측정 결과 Plasma 출력과 시간의 증가함에 따라 접촉각은 감소하였다. 실리콘코팅 원단에 저온 Plasma 처리한 후 표면 특성을 분석하고 원단을 접착을 시켜 박리 강도를 측정함으로써 막구조 원단의 접착력 향상에 대한 연구를 진행하였다. KS K 0533 접착포의 박리 강도 시험방법으로 실리콘코팅 원단의 박리 강도를 측정한 결과 플라즈마 처리 원단이 플라즈마 미처리 원단보다 박리 강도가 향상된 것을 확인할 수 있었다. 저온 Plasma 처리 시간이 증가할수록 표면의 젖음성을 향상시켜 접촉각을 낮추었다. 이는 곧 표면에너지의 증가를 뜻하는 것으로 접착력을 증가시켜 실리콘코팅 원단의 접착성을 시킴으로써 강한 강도와 내구성을 갖춘 막구조물의 개발에 기대되고 있다.

Effect of Distribution System Materials and Water Quality on Heterotrophic Plate Counts and Biofilm Proliferation

  • The biofilms on pipe walls in water distribution systems are of interest since they can lead to chlorine demand, coliform growth, pipe corrosion, and water taste and odor problems. As such, the study described in this paper is part of an AWWARF and Tampa Bay Water tailored collaboration project to determine the effect of blending different source waters on the water quality in various distribution systems. The project was based on 18 independent pilot distribution systems (PDS), each being fed by a different water blend (7 finished waters blended in different proportions). The source waters compared were groundwater, surface water, and brackish water, which were treated in a variety of pilot distribution systems, including reverse osmosis (RO) (desalination), both membrane and chemical softening, and ozonation-biological activated carbon (BAC), resulting in a total of 7 different finished waters. The observations from this study consistently demonstrated that unlined ductile iron was more heavily colonized by a biomass than galvanized steel, lined ductile iron, and PVC (in that order) and that the fixed biomass accumulation was more influenced by the nature of the supporting material than by the water quality (including the secondary residual levels). However, although the bulk liquid water cultivable bacterial counts (i.e. heterotrophic plate counts or HPCs) did not increase with a greater biofilm accumulation, the results also suggested that high HPCs corresponded to a low disinfectant residual more than a high biofilm inventory. Furthermore, temperature was found to affect the biofilms, plus the AOC was important when the residual was between 0.6 and 2.0 mg $Cl_2/l$. An additional aspect of the current study was that the potential of the exoproteolytic activity (PEPA) technique was used along with a traditional so-called destructive technique in which the biofilm was scrapped off the coupon surface, resuspended, and cultivated on an R2A agar. Both techniques indicated similar trends and relative comparisons among the PDSs, yet the culturable biofilm values for the traditional method were several orders of magnitude lower than the PEPA values.

Tetrahydrofuran-Containing Crown Ethers as Ionophores for NH+4-Selective Electrodes

  • The ammonium ion-selective electrodes ($NH^+_4$-ISEs) based on the tetrahydrofuran(THF)-containing-16-crown-4 derivatives,1,4,6,9,11,14,16,19-tetraoxocycloeicosane ($L^1$) and 5,10,15,20,-tetramethyl-1,4,6,9,11,14,16,19-tetraoxocycloeicosane ($L^2$), were prepared and the electrode characteristics were tested. The conditioned $NH_4^+$-ISEs (E1) based on $L^1$ with TEHP as a plasticising solvent mediator gave best results with near-Nernstian slope of 53.9 mV/decade of activity, detection limit of $10^{-4.9}$ M, and enhanced selectivity coefficients for the $NH^+_4$ ion with respect to an interfering $K^+$ ion (log $K^{pot}_{NH_4^+,K^+}$ = -1.84). This result was compared to other ammonium ionophores reported previously, for example, that of nonactin (log $K^{pot}_{NH_4^+,K^+}$ = -0.92). The proposed electrode showed no significant potential changes in the range of 3.0 < pH < 9.0.

Effects of bis(2-ethylhexyl) phthalate(DEHP) on plant soil-borne pathogenic bacterium Pectobacterium carotovorum in vitro (Bis(2-ethylhexyl) phthalate가 in vitro에서 식물 토양병원성 세균 Pectobacterium carotovorum에 미치는 영향)

  • Bis(2-ethylhexyl) phthalate (DEHP) is one of the plasticizers used in the polyvinyl chloride(PVC) industry. It is known to be easily released into the environment. In this study, we investigated effects of DEHP on growth, metabolic pathway, and virulence gene expression in soil-borne bacterial plant pathogen, Pectobacterium carotovorum SCC1 using in vitro assays. As a result, DEHP at 20 ㎍ mL-1 did not affect the growth, cell membrane permeability, or ATPase activity of P. carotovorum SCC1. However, it decreased succinyl-CoA synthase (SCS) activity in the tricarboxylic acid (TCA) cycle. Relative expression levels of virulence genes encoding pectate lyase and pectin were differentially influenced by DEHP treatment. These results suggest that biological characteristics of P. carotovorum might be influenced by DEHP in soil.

Relationship between Endotoxin Level of in Swine Farm Dust and Cellular Immunity of Husbandry Workers (돈사 분진 함유 내독소 수준과 축사 작업자들의 세포면역력간 상관성 분석)

  • Objectives: Endotoxins in dust generated in occupational settings is known to contribute to the occurrence of respiratory illness among workers. The relationship between the level of endotoxins in total dust or respirable particulates collected from swine farms and immunological markers related with respiratory allergy was evaluated among swine husbandry workers. Materials and Methods: Peripheral blood samples were collected from ten workers at ten swine farms at Gyeonggi province, Korea. Peripheral mononuclear cells were stimulated with phorbol 12-myristate 13-acetate and ionomycin for 48 hours. The levels of various cytokines produced at culture supernatants were determined using a commercially available ELISA kit. The concentration of particulate matter($PM_{10}$) in the indoor air of the swine farms was evaluated using a PVC membrane filter and mini volume air sampler, and endotoxin levels in the dust were measured by Limulus Amebocyte Lysate Kinetic QCL method. Results: Levels of endotoxins in the total dust were categorized into high(geometric mean: $109.35EU/m^3$) and low concentrations (geometric mean: $0.95EU/m^3$) for five swine farms. Interleukin-4 levels were higher in the high endotoxin group than in the low endotoxin group, while interferon-${\gamma}$ levels were lower in the high endotoxin group than in the low endotoxin group. The ratio (interferon-${\gamma}$ to interleukin-4), indicating immunologic skewedness against allergic reactivities, was lower in the high endotoxin group($1.15{\pm}0.60$) than the low endotoxin group($3.09{\pm}2.38$). In addition, the level of interleukin-13, another cytokine contributing to the occurrence of allergic responses, was significantly higher in the at the high endotoxin group($1.12{\pm}0.37ng/m{\ell}$) than in the low endotoxin group($0.37{\pm}0.04ng/m{\ell}$). Hematologic assessment showed significantly lower cellularity in the number of total leukocytes, neutrophils, and eosinophils in the high endotoxin group than in the low endotoxin group. Conclusions: Even though a sufficient number of swine workers and farms were not investigated, this study generlly suggests that the immunological function of swine farm workers exposed to high levels of endotoxin could be modulated toward allergic reactivities.

Coated Wire Lead(Ⅱ) Ion-Selective Electrodes based on Crown Ethers (Crown Ether를 이용한 탐침형 납 이온선택성 전극)

  • Acryloylmethylbenzo-15-crown-5 was prepared from the reaction of 4'-hydroxymethylbenzo-15-crown-5 with acryloyl chloride. And, poly(acryloylmethylbenzo-15-crown-5) [poly(AMB15C5)] was synthesized by radical polymerization using AIBN as initiator in benzene. Coated wire lead(II) ion-selective electrodes ($Pb^{2+}$-CWISEs) using either poly(AMB15C5) or B15C5 as neutral carrier were prepared, respectively. $Pb^{2+}$-CWISEs gave linear responses with slopes of 28$\pm$ 1mV per decade within the concentration range of $10^{-5} M{\sim}10^{-1}$ M, respectively. Also, the detection limits were $10^{-6}$ M and response times were either 3 or 5 min. for B15C5 and poly(AMB15C5), respectively. $Pb^{2+}$-CWISE base on B15C5 was rather unstable than poly(AMB15C5)'s due to solubility of the B15C5 in water. The selectivity coefficients of a variety of interfering ions such as $Mg^{2+},\; Ca^{2+},\; Co^{2+},\; Ni^{2+},\; Cu^{2+},\; Zn^{2+}$ and $Cd^{2+}$ were small ($10^{-4}{\sim}10^{-5}$), while those of $Na^+$ and $K^+$ were large (0.1∼0.01). In addition, the electrode responses depended upon the pH of test solution and the composition of the membrane. In the range pH 3∼6 of test solution, potentials of Pb2+-CWISEs were hardly changed. The optimal contents of B15C5 and poly(AMB15C5) were 7.7 wt% and 13.1 wt%, respectively.
