Comparison Between Phylogenetic Relationships Based on 18S rDNA Sequences and Growth by Salinity of Chlorella-like Species (Chlorophyta)

  • This study was carried out to understand the correlation between phylogenetic relationships based on 18S rDNA sequences and growth by salinity of Chlorella-like species. The 18S rDNA sequences of 71 Chlorella-like species which were mainly collected from Korean waters were analyzed. The 18S rDNA sequences of Chlorella-like species were divided into three groups (group A, B and C) and group B was further divided into three subgroups (subgroup B-1, B-2 and B-3). Thirty-seven Chlorella-like species in group A grew well at high salinity (32 psu) but the other groups grew well in freshwater. The sequence identities of the species in group A and B were 97.2-99.5%, but those of 6 species in group C ("Chlorella" saccharophila), which contained group I intron sequences region were 75.0-75.4%. Two representative species of each group were cultured at different salinities (0, 16 and 32 psu) to examine the correlation between the molecular phylogenetic groups and the phenotypic characteristics on cell growth and size by different salinities. The size of cell cultured at different salinities varied according to the species of each molecular phylogenetic group. The size of "Chlorella" saccharophila in group C was bigger and more obviously elliptical rather than that of the other Chlorella-like species. Considering the results on molecular and phenotypic characteristics, the group A and B belonged to Chlorellaceae, but group C was distinctly different from them.

The Effects of Environmental Factors on the Growth of Lithophyllum yessoense and Hildenbrandia rubra Sporelings in Laboratory Culture (실내배양에서 납작돌잎(Lithophyllum yessoense)과 진분홍딱지(Hildenbrandia rubra)의 배아 생장에 미치는 환경요인의 영향)

  • The effects of environmental factors, such as irradiance, daylength, salinity, and desiccation, on the growth of Lithophyllum yessoense and Hildenbrandia rubra sporelings were examined. Sporelings of each species were cultured with 10, 50, 80, 120, $150{\mu}mol$ photon $m^{-2}s^{-1}$ for 14 days and their maximum growth occurred under $80{\mu}mol$ photon $m^{-2}s^{-1}$. Germlings of both species survived for 21 days in darkness, and even the L.yessoense germlings grew. In the salinity experiment, sporelings of each species survived for 7 days and died after 14 days under 20 and 25 psu, but the sporelings grew well under 34 psu. Physiological features of each species with respect to the evaluated daylengths (8, 12, 14 and 16 h) were slightly different, and maximal growth occurred at 16 h for L. yessoense and at 14 h for H. rubra sporelings. Mortality of the sporelings increased with the exposure period, but H. rubra was less tolerant to desiccation than L. yessoense. In conclusion, sporelings of the two species showed similar growth responses to various environmental factors with slightly different physiological features with respect to salinity, daylength, and desiccation. However, more ecological and physiological studies on slow-growing crustose algae are required to elucidate the expansion of barren ground around the coastal areas of Korea.

Numerical Simulation on Hydrodynamic Characterization Changes Associated with the Construction of Dikes and Dredging Operations in Saemangeum Lake (새만금호 내 방수제 공사 및 준설에 의한 수리동역학적 특성 변화 수치 모델링)

  • The study area is located on the western coast, and the inner development construction has been ongoing since 2011. The purposes of current study are to effectively simulate and quantitatively predict a temporal and spatial distributions of water temperature and salinity due to the stages of inner development construction in saemangeum reclaimed area. The transient-state numerical modeling using EFDC model is done, and the numerical simulation results are validated reasonably by repetitive numerical model calibration procedures with respect to field measurements of water temperature and salinity. The spatial distributions of water temperature and salinity show similar trends before and after construction of the dikes. In spring season, the salinity has maximum value of 21 psu, while, in summer season, the salinity shows 7 psu in a whole modeling domain. Thus, it is clearly observed that salt water is replaced by freshwater. However, the salinity and temperature reach their initial conditions at the end of the year. The salinity after construction of the dikes is lower than that before construction of them at Mankyeong area. On the other hands, after construction of the dikes, the salinity after dredging operations is higher than that before dredging. Because drastical increasing of water volume in Saemangeum Lake leads to increasing of stagnation time at bottom layer, and salt water is easily intruded to the two estuaries. Therefore, it may be concluded that hydrodynamic characteristics on Saemangeum are dominated by either Mankyeong and Dongjin discharge or sluice gates in/out-flow amounts, and thus they must be properly considered when rigorous and reasonable predictions of water temperature and salinity according to the stages of inner development construction.

Seasonal Changes in Gonadal Development of Manila Clam, Ruditapes philippinarum from Shi-Heung-Ri, Jeju, Korea (제주도 시흥리 연안에 분포하는 바지락, Ruditapes philippinarum의 계절에 따른 생식소발달)

  • Annual gametogenic cycle of Manila clam Ruditapes philippinarum collected from Shi-Heung-Ri beach in Jeju was investigated for the 12 month period from May 2001 to April 2002. Water temperature varied from 10 to 27$^{\circ}C$ and salinity from 27 to 35 psu during the course of study. Most of the clams collected in March were in indifferent or in early developing indicating that clams initiated gametogenesis when water temperature becomes 14 to 15$^{\circ}C$. In early June, most of the clams were in developing, expanding follicles and exhibiting growing oocytes. Fully ripe gonads were observed in clams collected during late June to early August. Spawning clams were observed as early as in early June although the spawning activity reached its maximum in early August. Condition index (CI) was rapidly increased from July to early August and then dropped in late August, indicating that most of clams in Shi-Heung-Ri beach spawn in late summer when water temperature was within a range of 25 to 27$^{\circ}C$.

Variations of Water Environments and Species Compositions of Microalgae during Summer in the Coast of Dokdo, Korea (독도 연안의 하계 수환경과 미세조류의 종조성 변화)

  • This study was carried out to investigate the charateristics of environmental factors and the species compositions of phytoplanktons and periphytons in order to analyze the marine ecosystem (DOK1-3) with freshwater (Mul) in the coast of Dokdo. Mean values of conductivity (TSD) (44.3 mS cm–1), total dissolved solids (30.9 mg L–1), salinity (31.8 psu), total suspended solids (39.2 mg L–1), and ash-free dry matter (AFDM) (8.8 mg L–1) were higher in DOK1- 3 than those in Mul (3.2 mS cm–1, 2.3 mg L–1, 1.9 psu, 2.8 mg L–1 and 2.8 mg L–1, espectively), while mean amounts of soluble reactive phosphorus (SRP) (7.5 μg P L–1) and soluble reactive silicon (SRS) (0.1 μg Si L–1) were significantly lower in DOK1-3 than those in Mul (117.3 μg P L–1 and 4,105 μg Si L–1, respectively). The phytoplanktons was a total of 59 species in Dokdo, which were composed of 1 species (1.7%) for Chrysophyceae, 43 species (72.9%) for Bacillariophyceae and 15 species (25.4%) for Dinophyceae. The biomass (Chl-a, phaeopigment and AFDM) of epilithic algae (ELA), epizooic algae (EZA) and epiphytic algae (EPA) were varied with the kinds of substrates (EPA > ELA > EZA) in the whole stations. Water environments was comparatively clean in the coast of Dokdo as comparing with those of the land. However, the monitoring of marine ecosystem in the Dokdo should be continued for species conservation according to the global warming by the climatic change.

Hypoosmotic shock adaptation by prolactin involves upregulation of arginine vasotocin and osmotic stress transcription factor 1 mRNA in the cinnamon clownfish Amphiprion melanopus

  • We cloned cDNA-encoding arginine vasotocin (AVT) from the brain of the cinnamon clownfish Amphiprion melanopus, and that was predicted to encode a protein of 153 amino acids. We examined changes in the expression of AVT mRNA in the brain and arginine vasotocin receptor (AVTR) mRNA and osmotic stress transcription factor 1 (OSTF1) mRNA in the gills of the cinnamon clownfish using quantitative real-time PCR in an osmotically changing environment (seawater (35 psu) ${\rightarrow}$ brackish water (BW, 17.5 psu) and BW with prolactin [PRL]). The expression of AVT, AVTR, and OSTF1 mRNA in the brain and gills increased after transfer to BW, and the expression was repressed by PRL treatment. AVT-immunoreactive cells were almost consistently observed in the telencephalon. The plasma $Na^+$ and $Cl^-$ levels decreased in BW, but the level of this parameter increased in BW with PRL treatme during salinity change. These results suggest that AVT, AVTR, and OSTF1 play important roles in hormonal regulation in osmoregulation organs, and that PRL improves the hyperosmoregulatory ability of cinnamon clownfish in BW environment.

Na+/K+-ATPase Alpha Subunit in the Monogonont Rotifer, Brachionus koreanus: Molecular Cloning and Response to Different Salinity

  • $Na^+/K^+$-ATPase is a membrane protein and plays a key role in osmotic regulation in living organisms. In the present study, a cDNA sequence encoding the $Na^+/K^+$-ATPase alpha subunit from the monogonont rotifer, Brachionus koreanus was cloned by rapid amplification of cDNA ends technique. To investigate the role of this enzyme in osmotic stress, enzymatic activities of $Na^+/K^+$-ATPase were measured after exposure to different salinities for 48 h. The full-length Bk $Na^+/K^+$-ATPase cDNA was 3069 bp-long, encoding a 1022-amino acid polypeptide. Bk $Na^+/K^+$-ATPase possesses eight membrane spanning regions and five conserved domains. Phylogenetic analysis showed that Bk $Na^+/K^+$-ATPase had high identity with those of other species, and was closely clustered with other Brachionus sp. These findings indicate that this protein was conserved both structurally and functionally. B. koreanus $Na^+/K^+$-ATPase activity was stimulated in both hyposaline (6 psu) and hypersaline (32 psu) conditions, suggesting that this protein may play a role in osmoregulation. This study would provide better understanding of the physiology of B. koreanus and this enzyme may be useful as a molecular marker for evaluation of osmotic stress in aquatic environment.

The Characteristics of Structure of Warm Eddy Observed to the Northwest of Ullungdo in 1992 (1992년 울릉도 북서부해역에서 관측된 난수성 소용돌이의 구조특성)

  • A warm eddy was continuously observed to the east of Sokcho, Korea from March to June 1992. This warm eddy had been formed in 1991, wintered to the east of Sokcho, and moved northward a little during April-June 1992. The diameter and the depth of the eddy were respectively about 160 km and about 330 m in March. The homogeneous (mixed) layer of 10$^{\circ}C$ and 34.2 psu water was found at the upper layer with the maximum size of about 130 km and maximum depth of about 230 m in March. The size of the eddy and homogeneous layer decreased in June. Maximum current velocity of the eddy was about 65 cm/s at the surface layer and exceeded20 cm/s at 200 m depth. It is shown that the flow field was nearly in geostrophic balance, but there was a little difference in the current velocity between ADCP and geostrophic calculation in June. The surface velocity of the East Korean Warm Current(EKWC) was 50∼70cm/s which was very similar to the northward current velocity of the eddy. The EKWC water appeared in the layer upper than 200 m depth.

Southward Intrusion of the East Sea Intermediate Water into the Ulleung Basin: Observations in 1992 and 1993

  • Hydrographic data retrieved in the southwestern part of the East Sea in 1992-1993 were analyzed to investigate the probability of southward intrusion of the East Sea Intermediate Water (ESIW) into the Ulleung Basin. The ESIW showed the ranges of 1 to 4$^{\circ}$C in potential temperature, 33.80-34.06 psu in salinity, and 26.9-27.3 kg/m$^3$ in potential density (${\sigma}$$_{\theta}$). The mean depth occupied by the ESIW was 170 m, where the characteristic values of the above three were 2.64$^{\circ}$C , 34.02 psu, and 27.13 kg/m$^3$, respectively. One of the most prominent features of the ESIW was that its salinity changed not only seasonally but also interannually. It was low in summer and high in winter. The salinity within the isopycnal layer of 26.9-27.3 kg/m$^3$ was closely related with the potential vorticity (${\rho}$$_{\theta}^{-1}$ f ${\varrho}$${\rho}_{\theta}$/${\varrho}$z), being in direct proportion to the salinity. This implies that the low-salinity water was thicker than the high-salinity water. The flow path of the ESIW was investigated by tracking the low-salinity or the low-potential vorticity water and by referring to acceleration potential. Careful analysis of the flow path proves that the ESIW intrudes from the north between the Korean coast and Ulleung Island into the Ulleung Basin in summer. Existence of the high-potential vorticity water in the Ulleung Basin is associated with the interruption of the inflow of low-salinity water.

Environmental Factors and the Distribution of Eggs and Larvae of the Anchovy (Engraulis japonica) in the Coastal Waters of Jeju Island (제주도 주변해역의 해양환경요인과 멸치 난자치어 분포)

  • Anchovy spawn from the end of May to mid-October, when the water temperature is $14.8-27.2\;^{\circ}C$ and the salinity is 26.0-33.6 psu. The main spawning season is between July to August, when the water temperature is $21.7-27.2\;^{\circ}C$ and the salinity is between 26.0-32.2 psu. The main spawning grounds of anchovy are coastal areas shallower than 50 m around the islands located in the Jeju Strait. Anchovy larvae are distributed near the fronts between Chuja-do, Jangsu-do, Yeoseo-do, and the open sea rather than in the spawning grounds. Anchovy eggs and larvae density increased in accordance with the high level of $Chlorophyll-{\alpha}$ during the summer season (July-August). In terms of the suspended sediment (SS) levels along the northern coast of the Jeju Strait, high densities of anchovy eggs (12.0-18.0 mg/L) were observed, mainly in the area affected by the coastal waters of the southern sea with high SS levels, while larvae (10.0-19.0 mg/L) tended to be distributed over a wide area with high SS levels, including the open sea. In terms of the dissolved oxygen (DO) content, eggs (5.4-6.8 mg/L) were observed in coastal areas with a high DO content, while larvae (4.2-6.4 mg/L) were distributed widely in areas with a relatively low DO content, from the southern coast to the open sea.
