Optimum Culture Condition on Four Species of Microalgae used as Live Food for Seedling Production of Bivalve (이매패류 먹이생물로 이용되는 미세조류 4종의 적정 배양환경조건)

  • In order to investigate the live food value of microalgae for efficacious rearing of larvae and spats of bivalve, we studied growth rates of four microalgal species (Isochrysis galbana, Pavlova lutheri, Chaetoceros simplex, Tetraselmis tetrathele) cultured in different environmental conditions. These include changes in temperatures (20, 25, 30 and $35^{\circ}C$), salinities (20, 25, 30 and 33 psu) and light intensities (60, 100 and $140{\mu}mol\;m^{-2}s^{-1}$). The growth rate of I. galbana was faster at $25^{\circ}C$ than that of $20^{\circ}C$. At $25^{\circ}C$ the highest growth rate of I. galbana was observed at 33 psu (0.413) and the lowest at 20 psu (0.368) in 10 days of culture (P < 0.05). The growth rate of I. galbana was lower at 25 psu (0.383) than that of 30 psu and higher than that of 20 psu (P < 0.05). Similar temperature and salinity-dependent changes were also found in P. lutheri and T. tetrathele. C. simplex showed faster growth rate at $30^{\circ}C$ than that of $25^{\circ}C$. The highest growth rate of C. simplex was observed at 33 psu (0.428) and the lowest at 20 psu (0.389) in 10 days of culture (P < 0.05). Upon exposure to the light with different intensities, all four microalgal species showed a significantly faster growth rate at $140{\mu}mol\;m^{-2}s^{-1}$ than at $100{\mu}mol\;m^{-2}s^{-1}$ (P < 0.05).

Effects of Salinity Stress on the Composition of Free Amino Acids of the Pacific abalone Haliotis discus discus (염분 스트레스가 둥근전복 Haliotis discus discus의 유리아미노산 조성에 미치는 영향)

  • Changes in 13 free amino acids (FAA) of Haliotis discus discus exposed to various salinities were investigated using a high performance liquid chromatography (HPLC). Taurine, glycine and alanine are three major contributors to the total FAA in the gill tissues of H. discus discus. Concentration of taurine was 114 imol/g dry tissue weights accounting for 76.64% of total FAA in H. discus discus. Levels of most FAAs in H. discus discus exposed to low and high salinities for 24 h decreased dramatically. Taurine concentration was slightly increased in the samples exposed to 20 psu and 25 psu for 48 h, and greatly increased after 120 h exposure. After 48 hrs exposure to 20 psu, 25 psu, 30 psu and 40 psu, methionine in H. discus discus was not detected; no methionine was detected in the sample exposed to 20 psu, 25 psu, 30 psu and 40 psu after 120 h of exposure. Taurine:glycine ratio increased depending upon hyper-hypoosmotic condition as well as period of osmotic stress. These data indicate that taurine, glycine and methionine play important role in regulating osmotic stress in H. discus discus. This study suggesting that FAA analysis is a useful tool to diagnose osmotic stress to H. discus discus.

Survival and Histological Change of Integumentary System of the Juvenile Sea Cucumber, Apostichopus japonicus Exposed to Various Salinity Concentrations (염분 농도에 따른 어린 돌기해삼, Apostichopus japonicus의 생존 및 피부계의 조직학적 변화)

  • Juvenile sea cucumber, Apostichopus japonicus(wet weight $1.0{\pm}0.2g$, n=250) acclimated to environmental conditions($17^{\circ}C$, 33 psu) during 4 weeks for observation of survival and structural variation of integumentary system according to various salinity concentration. After that, it was exposed 0, 5, 10, 15, 20, 25, 30, 35, 40 psu in 96h, and recovered during 7 days. In this results, The 96-h lethal concentration(LC50) for sea cucumber was a salinity of 21.05 psu. Integumentary system of the sea cucumber exposed to below the salinity of 20 psu mainly observed thickness reduction of epidermal layer, nucleus condensation of epithelial cells, decreased of mucous cells and loose arrangement of connective tissue in dermal layer. Integumentary system of the sea cucumber exposed a salinity of 40 psu mainly observed nucleus hypertrophy of epithelial cells, increase of mucous cells and tight arrangement of connective tissue in dermal layer. Also, observed secretory cells showing the alcian blue(pH 2.5) positive. During 96-h exposure, this cells decreased after increase below the salinity of 20 psu but increased in the salinity of 40 psu compared with 25-35 psu.

  • Physiological responses and tolerance limit of hybrid striped bass $(Morone\;Chrysops\;male{\times}\;M.\;saxatilis\;female),$ were investigated on salinity changes from 33 psu to 0 psu and from 0 to psu 33 psu in two separate trials, respectively. In the acute salinity decrease from sea water (33 psu) to fresh water (0 psu), plasma level of cortisol, glucose, total protein, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were not affected, however, $Na^+,\;Cl^-$ and osmolality significantly decreased. In the acute salinity increase from fresh water to sea water (33 psu), plasma cortisol level did not change. Glucose and total protein concentrations in plasma and hematocrit (Hct) decreased, whereas AST, ALT, $Na^+,\;Cl^-$ and osmolality increased. The hybrid striped bass can overcome the acute changes of salinity, from 0 psu to 33 psu or from 33 psu to 0 psu, But salinity decrease stressed the fish much more than salinity increase.

  • We investigated growth, survival and stress response depending on salinity changes in hybrid grouper (Epinephelus fuscoguttatus ♀×E. polyphekadion ♂, BCG) juvenile. Experiment I (Exp I), the hybrids were stocked at 32 psu and reared for 30 days under decreasing salinity at a rate of 5-7 psu per day (25, 20, 15, 10, 5 psu). Experiment II (Exp II), the salinity was decreased from 32 psu to 5 psu during a period of 24, 48 and 72 h. Experiment III (Exp III), acclimatized BCGs were exposed from 5 psu to decreasing salinity at 0, 1, 2, 3, 4 psu. In the Exp I, 90% survival only in the 5 psu. Growth were 5 and 32 psu had significantly low growth than other treatment. In the Exp II 100% survived by the end of the experiment. The plasma cortisol level was highest in the treatment the salinity rapidly dropped, but all the treatment recovered quickly after 24 h of salinity reduction. In the Exp III, the lower the salinity, the faster the mortality occurred in salinity below 5 psu. It is therefore concluded that the BCGs are highly tolerant of low salinity and the salinity tolerance threshold is 5 psu.

Development of a L-Band Microwave Radiometer for Remote Sensing of Water Surface Salinity (수면 염분 원격탐사용 L-Band 마이크로파 라디오미터의 개발)

  • The development processes of a L-band microwave radiometer for remote sensing of water surface salinity are described in this paper. Achieving the development aim of the measurement accuracy within 2 psu for water surface salinity of 0~40 psu, the requirements and specifications of the microwave radiometer and its receiver are drawn. The receiver with high gain, high sensitivity is designed and implemented to satisfy these requirements and specifications. The receiver has the bandwidth of 45 MHz, the system gain of 47 dB and the sensitivity of 0.41 K at 1,390 MHz. The effectiveness of the developed L-band microwave radiometer for remote sensing of water surface salinity is demonstrated experimentally. The results show the microwave radiometer can detect water surface salinity for 10~28 psu within the accuracy of 1.4 psu.

Effect of Salinity on Survival and Metabolism of ark shell, Tegillarca granosa (꼬막, Tegillarca granosa의 생존과 대사에 미치는 염분의 영향)

  • We examined physiological responses related to the survival, oxygen consumption and filtration rate of the blood cockle, Tegillaarca granosa as a result of salinity changes. The 44-day $LS_{50}$ (median lethal salinity) at $10^{\circ}C$ for adult and juvenile was 15.8 (confidence limits 13.5-18.2 psu) and 16.2 (confidence limits 14.1-18.4 psu) psu respectively, whereas the 11-day $LS_{50}$ at $25^{\circ}C$ for adult and juvenile was 16.8 psu (confidence limits 12.9-21.2 psu) and 22.4 (confidence limits 20.5-24.7 psu) psu respictively. In conditions of decreasing salinity, Oxygen consumption and filtration rates decreased or varied irregularly as salinity decreased. The results of this study could prove important in investigating causes of mass mortality and managing shellfish aquaculture farms.

  • Fertilization rates were over 90% when sperm from black porgy Acanthopagrus schlegelii acclimated and raised in freshwater and black porgy raised in seawater were fertilized with eggs from female black porgy raised in seawater, although cryopreserved sperm were significantly lower in fertilization rates than non-cryopreserved sperm. From the observations of embryos development at different salinities (0, 10, 20 and 32 psu) within 3 hours upon fertilization (16 cell stage), all were dead at 0 psu. However, the development process and speed at 10 and 20 psu were the same as at 32 psu. But, many developing embryos were egg envelope injury at low-salinities of 10 and 20 psu unlike at 32 psu. This is attributable to osmotic shock. Hatching rate of embryos at 32 psu was 60% or so, whereas low as 5% or less were successfully hatched at 10 and 20 psu, implying that osmotic shock could be responsible for the failure of embryo development.

  • This study investigated the possibility of salinity acclimation of freshwater rotifers (Brachionus calyciflorus) as live food for sweetfish (Plecoglossus altivelis) larvae, and also examined the optimal salinity for the growth of sweetfish. Freshwater rotifers cultured in 0 and 4 PSU and seawater rotifers (B. rotundiformis) cultured in 33 PSU were supplied to the larvae with four kinds of enrichment material (condensed freshwater Chlorella, $\omega-yeast,$ baker's yeast, Super Selco) and larval growth at 4 PSU was examined. Growth of the freshwater rotifers positively increased from 0 PSU to 6 PSU, but decreased when over 8 PSU was reached. Growth and survival of the sweet fish larvae reared in 0 PSU were significantly lower than those reared in either 4 PSU or 33 PSU. This indicated that the freshwater rotifers (B. calyciflorus) could be used as live food for sweetfish larvae reared in 4 PSU. The body weight of sweetfish larvae fed on freshwater rotifers enriched with Super Selco was the highest at 0.163 mg, but there was no significant difference in survival and body length of the fish fed with the other enrichment materials. The content of n-3 HUFA of the sweetfish larvae fed on the freshwater rotifers enriched with Super Selco and the condensed freshwater Chlorella was higher than that enriched with $\omega-yeast$ and baker's yeast. These results indicated that B. calyciflorus cultured with the condensed freshwater Chlorella could be used for the sweetfish larvae without enrichment, and the most efficient enrichment material for B. calyciflorus is Super Selco.

  • As an early warning system to reduce the damage of aquacultured mollusks due to low salinity water, we investigated the possibility of a biomonitoring system measuring the shell valve movement (SVM) of Pacific oyster (Crassostrea gigas) by using the Hall element sensor. In high salinity water of 27 psu, SVMs of Pacific oyster showed spikes which mean a relatively fast closing condition after opened condition of average 10-15 mm, and then the SVM showed back to opening condition slower than closing speed. In water salinity of 20-27 psu, the SVMs were similar to that of 27 psu. However, below 17 psu, it showed abnormal valve movements such as spending more time for shell closure. In 10 psu, we could not detected SVMs due to closed condition during experiment periods. Thus, if we quickly detect abnormal environmental variations like low salinity using bio-monitoring of SVM, it may be contribute to increased productivity by dramatically reducing damages in aquaculture.
