The purpose of this study was to investigate the effect of salt formation on the percutaneous absorption of meloxicam through hairless mouse skin from a pressure sensitive adhesive (PSA) matrix. In addition, the influences of enhancers on the permeation of meloxicam or meloxicam-ethanolamine (MX-EA) salts across the hairless mouse skin were evaluated using a flow-through diffusion cell system. The salt formation of meloxicam resulted in lower permeation rate than the parent drug. $Span^{(R)}$ 80 provided the highest enhancing effect for meloxicam and meloxicam monoethanolamine salt. The maximum amount of the drug that can be loaded without retarding permeation rate was different depending on the compound. No relationship was found between the fluxes of meloxicam or MX-EA salts from saturated solutions and those from PSA matrices containing the same enhancer.
The HDD (hard disk drive) using Load/Unload (L/UL) technology includes the benefits which are increased areal density, reduced power consumption and improved shock resistance than those of contact-start-stop (CSS). Dynamic L/UL has been widely used in portable hard disk drive and will become the key technology for developing the small form factor hard disk drive. Main design objectives of the L/UL mechanisms are no slider-disk contact or no media damage even with contact during L/UL, and a smooth and short load and unload process. In this paper, we focus on state matrix, pitch static attitude (PSA), roll static attitude (RSA), loading/unloading contour (LC/ULC), impact force and contact. Stability of slider is mainly determined by PSA and RSA. State matrix by PSA and RSA is also important indicator. Therefore we analyze state matrix of SFF HDD suspension through the LC/ULC.
The effects of different formulation variables including pressure sensitive adhesive (PSA), permeation enhancer, thickness of the matrix and loading amount of drug on the transdermal absorption of galantamine were investigated across the hairless mouse skin. The permeation profile of galantamine was different depending on the types of PSA, loading amount of drug, thickness of the matrix and type of enhancer used. Highest flux of galantamine was obtained from acrylic PSA but crystals were formed in the patch within 72 h. Among the PSAs screened, crystal formation was not observed only in the patches formulated in Styrene Butadiene Styrene (SBS) matrix. Permeation rate increased linearly as the concentration of galantamine in SBS matrix increased from 2.5 to 15% w/w. Among the enhancers screened, Brij$^{(R)}$ 30 provided highest flux of galantamine. Matrix thickness of 80 ${\mu}m$ was optimum for maintaining adhesiveness as well as consistently delivering galantamine for longer period of time.
The effects of various pressure sensitive adhesives (PSA) and enhancers on the percutaneous absorption of tulobuterol were investigated. The permeation rate of tulobuterol through hairless mouse skin from various adhesives was evaluated using a flow-through diffusion cell system at $37^{\circ}C$. The permeability of tulobuterol was variable depending on the physicochemical property of the PSA. The permeation rate of tulobuterol from polyethylene oxide grafted acrylic adhesive matrix was higher than that from other PSA matrices. The flux of tulobuterol was $4.37{\pm}0.34\;{\mu}g/hr/cm^2$ from polyethylene oxide grafted acrylic adhesive matrix. When the effects of various enhancers on the percutaneous absorption of tulobuterol from grafted acrylic adhesive were evaluated, Plurol $oleique^{\circledR}$ showed higher flux than all other enhancers tested.
2-Ethylhexyl acrylate, butyl acrylate, methyl methacrylate, and 2-hydroxyethyl methacrylate were polymerized to synthesize acrylic pressure sensitive adhesive (PSA). Alumina and graphite as a filler were added to acrylic PSA to give thermal conductivity. In case of addition of both graphite and alumina, the thermal conductivity of PSA was increased compared with alumina alone due to enhancement of contact between two fillers followed by increasing thermal path in PSA matrix.
본 연구에서는 U-251-MG 세포를 이용하여 증식, 이동, 침윤 및 단백질분해효소의 분비에 미치는 PSA와 HGF의 효과를 확인하였다. 그 결과, PSA siRNA에 의해 U-251-MG 세포의 증식은 약 37% 억제되었으나, HGF 처리 (10 ng/mL)에 의해 증식이 약 1.4배 증가되었다. PSA siRNA에 의한 U-251-MG 세포의 이동은 약 60%가 억제되었으나, HGF 처리에 의해 이동이 약 1.3배 증가되었다. 또한, PSA siRNA에 의해 U-251-MG 세포의 침윤이 약 67% 억제되었으나 HGF 처리에 의해 세포의 침윤이 약 4.3배 증가되었다. PSA siRNA처리에 의해 MMP-2의 분비는 약 25%, MMP-9의 분비는 약 20% 억제되었으며, HGF에 의해 PSA siRNA에 의해 억제된 MMP-2 및 MMP-9의 분비가 약 2.8배 및 3.5배 증가되었다. HGF 처리에 의해 플라스민의 분비량은 약 14배 증가되었고, PSA를 억제한 U-251-MG 세포에 HGF를 처리한 결과, 플라스민의 분비가 약 1.6배 증가하였다. 또한, PSA siRNA에 의해 MMP-2의 발현은 유의할만한 변화가 없었으나, MMP-9의 발현은 약 85% 억제되었다. PSA를 억제시킨 U-251-MG 세포에 HGF를 처리한 결과, MMP-2의 발현은 약 5.7배, MMP-9의 발현은 약 6.3배 증가되었다. 한편, MMPs의 광범위 억제제인 BB-94 처리에 의해 U-251-MG 세포의 증식, 이동 및 침윤이 유의할만하게 억제 된 것은 MMP-2 및 MMP-9이 U-251-MG 세포의 증식, 이동 및 침윤에 관여할 것임을 시사해준다.
The purpose of this study was to develop transdermal drug delivery system (TDDS) for the combination of physostigmine and procyclidine. The effects of various pressure sensitive adhesives (PSA) on the percutaneous absorption of procyclidine across hairless mouse skin were evaluated to select an appropriate PSA. In addition, the influences of various vehicles on the percutaneous absorption of procyclidine from PSA matrix across hairless mouse skin were evaluated using flow-through diffusion cell system at $37^{\circ}C$. Physostigmine did not have any influence on the permeation rate of procyclidine. The flux of procyclidine was the highest in silicone and PIB and was relatively lower in SIS, Acryl, and SBS adhesive matrices, however, their use was limited by the crystallization of the drug in the matrix. Among acrylic adhesives, the permeability of procyclidine was the highest from poly (ethylene oxide) grafted acrylic adhesive. Some enhancers show different enhancing effect depending on the drug, however, many of the tested enhancers showed enhancing effect for the permeation of both procyclidine and physostigmine to some extent. $Crovol^{\circledR}$ EP 40 showed the highest enhancing effect on the permeation of both compounds. The size of TDDS to provide required permeation rate was estimated to be $35\;cm^2$ based on available information.
To investigate the feasibility of developing a new quercetin transdermal system, a preformulation study was carried out. Therefore, the effects of vehicles and pressure-sensitive adhesives (PSA) on the in vitro permeation of quercetin across dorsal hairless mouse skin were studied. Among vehicles used, propylene glycol monocaprylate (PGMC) and propylene glycol mono-laurate were found to have relatively high permeation flux from solution formulation (i.e., the permeation fluxes were 17.25$\pm$1.96 and 9.60$\pm$3.87 $\mu\textrm{g}$/$\textrm{cm}^2$/h, respectively). The release rate from PSA formulations followed a matrix-controlled diffusion model and was mainly affected by the amount of PSA and drug loaded. The overall permeation fluxes from PSA formulations were less than 0.30 $\mu\textrm{g}$/$\textrm{cm}^2$/h, which were significantly lower compared to those obtained from solution formulations. The lower permeation fluxes may be due to the decrease of solubility and diffusivity of quercetin in the PSA layer, considering the fact that the highest flux of 0.26 $\mu\textrm{g}$/$\textrm{cm}^2$/h was obtained with the addition of 0.2% butylated hydroxyanisole in PGMC-diethyl-ene glycol monoethyl ether co-solvents (80-85 : 15-20, v/v). Taken together, these observations indicate that improvement in the solubility and diffusivity of quercetin is necessary to realize fully the clinically applicable transdermal delivery system for the drug.
Objective : Skin aging is commonly observed in patients with diabetes mellitus, which can be accessed by the amount of skin collagen and matrix metalloproteinase-1 (MMP-1). In the present study, anti-skin-aging effects of Root Cortex of Paeonia Suffruticosa Andrews (PSA), which has been widely used to treat diabetes mellitus, are investigated. Methods : Streptozotocin (STZ) was intraperitoneally injected to rats to induce diabetes. Body weights, feed intake, organ weights, blood glucose, and other biochemical index are determined in both normal and diabetic rats. In order to study the effect of PSA on skin aging, the amount of skin collagen was measured in diabetic rats after PSA treatments. Also, MMP-1 synthesis in UVB-irradiated human skin fibroblasts was investigated. Results : 1. When PSA was administered to STZ-induced diabetic rats, feed intake was significantly increased and blood glucose and total cholesterol were decreased in a dose-dependent manner. However, there are no differences in individual organ weights, GOT, and GPT. 2. A decrease of skin collagen in diabetic rats was significantly suppressed when PSA was treated. 3. PSA also inhibited MMP-1 synthesis in UVB-irradiated normal human skin fibroblasts, similar to retinoid, a well-known effective anti-skin-aging substance. Conclusion: PSA suppressed a collagen decrease in diabetic rats and inhibited MMP-1 synthesis in UVB-irradiated human skin fibroblasts. Therefore, the treatment of PSA is very effective to slow down the skin aging process.
