Molecular and Cytogenetic Analysis of Transgenic Plants of Rice(Oryza sativa L.) Produced by Agrobacterium-mediated Transformation

  • To demonstrate the importance of transformation efficiency in independent event, molecular and cytogenetic analysis were conducted with genomic DNA and chromosome of transgenic plants produced by Agrobacterium tumefeciens LBA4404 (pSBM-PPGN: gusA and bar). Selection ratios of putative transgenic calli were similar in independent experiments, however, transformation efficiencies were critically influenced by the type of regeneration media. MSRK5SS-Pr regeneration mediun, which contains 5 mgL$^{-1}$ kinetin, 2% (w/v) sucrose in combination with 3% (w/v) sorbitol, and 500 mgL$^{-1}$ proline, was efficient to produce transgenic plant of rice from putative transgenic callus in the presence of L-phosphinotricin (PPT). With MSRK5SS-Pr medium, transformation efficincies of Nagdongbyeo were significantly enhanced from 3.7% to 6.3% in independent callus lines arid from 7.3% to 19.7% in plants produced, respectively. Stable integration and expression of bar gene were confirmed by basta herbicide assay, PCR amplification and Southern blotting of bar gene, and fluorescence in situ hybridization (FISH) analysis using pSBM-PPGN as a probe. In Southern blot analysis, diverse band patterns were observed in total 44 transgenic plants regenerated from 20 independent PPT resistant calli showing from one to five copies of T-DNA segments, however, the transformants obtained from one callus line showed the same copy numbers with the same fractionized band patterns.

Development of an Efficient Screening Method for Resistance of Chili Pepper Plants to Meloidogyne incognita (뿌리혹선충 Meloidogyne incognita에 대한 저항성 고추를 선발하기 위한 효율적인 검정법 확립)

  • Root-knot nematodes (Meloidogyne spp.) are major plant pathogens that cause reductions in yield and quality of several solanaceous crops, including pepper (Capsicum spp.). These losses can be averted through planting of resistant cultivars. Plants are defined as resistant when they suppress nematode reproduction. In this study, the resistance degrees of 102 commercial cultivars of chili pepper (Capsicum annuum) to a root-knot nematode, Meloidogyne incognita, were evaluated by comparing the number of egg masses on their roots to those of 'PR huimangchan', a highly susceptible cultivar that exhibited the most egg masses of the chili pepper cultivars evaluated. Among these cultivars, forty-four (43.1%) showed resistance to M. incognita and eighteen (17.6%) were moderately resistant. The other cultivars (39.3%) were determined to be susceptible. For further study, six chili pepper cultivars (i.e., Gangryeokjosenggeon, Shinsegae, Muhanjilju, PR Bulrocho, PR Huimangchan, and Jjang) with different levels of resistance to the nematode were selected. Changes in resistance of the six cultivars under several conditions, such as inoculum concentration, plant growth stage, and cultivation period after transplanting were investigated. We found that an efficient screening method for resistance of chili pepper to M. incognita is to transplant the chili pepper seedlings 7 days before inoculation, to inoculate 28-day-old plants with M. incognita by loading 5,000 eggs per plant into the pot of soil, to cultivate the plants in a greenhouse ($25{\pm}5^{\circ}C$) for 45-60 days, to measure the number of egg masses on roots of the seedlings, and then to determine the resistance response of the plants by comparing the number of egg masses on the roots with a reference-susceptible cultivar 'PR huimangchan'.

The Optimal Time for Embryo Transfer in Fresh IVF: Comparison between Day 3 and Day 5 on Pregnancy Outcomes (체외수정술 후 난할단계 배아와 포배기단계 배아를 이식했을 때의 임신예후의 비교)

  • Objective: The aim of this study was to compare day 3 embryo transfer (D3ET) with day 5 ET (D5ET) in fresh in vitro fertilization (IVF) cycle on pregnancy outcomes. Methods: We conducted a retrospective matched case control study that included 90 women with D3ET and 90 women with D5ET from January 2007 to June 2009. Subjects were matched for reproductive profiles and IVF cycle characteristics. Two good quality embryos were transferred in both groups. Pregnancy rates (PR), implantation rate, and multiple PR were compared. Results: Demographics, stimulation parameters and embryological data were comparable in both groups. Main pregnancy outcomes with D3ET and D5ET groups were not statistically different: implantation rate (39.4% vs. 32.8%), positive PR (57.8% vs. 46.7%), clinical PR (53.3% vs. 45.6%), ongoing PR (50.0% vs. 42.2%), respectively. Both groups showed high multiple PR (37.5% vs. 34.1). Conclusion: D5ET may not be beneficial and necessary in comparison with D3ET on pregnancy outcomes, and elective single ET should be considered to decrease multiple pregnancies in women with favorable conditions and good quality embryos undergoing IVF.

Development of Microlens Array for Maskless Lithography Application (Maskless lithography 응용을 위한 마이크로렌즈 어레이 개발)

  • A microlens array (MLA) was developed based on the wet-etched quartz substrate and coating of UV adhesive on the substrate for maskless lithography application. The developed MLA has the focal length of ${\sim}45\;{\mu}m$ and the spot size of ${\sim}1\;{\mu}m$. The spot size of the focused beam passing through the MLA was detected by CCD camera, and its intensity was monitored by beam profiler. Uniform spots with nearly identical intensities were observed on the focal plane when a beam passes through the fabricated MLA. The focal length was varied depending on thickness of the coated UV adhesive. The thicker the thickness of the UV adhesive was, the shorter the focal length of the MLA was. With a general mask aligner, UV beam focusing was tested onto photoresist (PR). The beams were well focused onto PR when UV passes through the MLA. Depending on the variable distances from the MLA, beam sizes onto PR were controlled. Even at high temperature for a long time, the performances of the MLA were not changed.

Radiological Downstaging with Neoadjuvant Therapy in Unresectable Gall Bladder Cancer Cases

  • Background: Gall bladder cancer (GBC) usually presents as unresectable or metastatic disease. We conducted a feasibility study to evaluate the effect of neoadjuvant therapy (NAT) on radiologic downstaging and resectability in unresectable GBC cases. Materials and Methods: Patients with locally advanced disease were treated with chemoradiotherapy [CTRT] ( external radiotherapy (45Gy) along with weekly concurrent cisplatin $35mg/m^2$ and 5-FU 500 mg) and those with positive paraaortic nodes were treated with neoadjuvant chemotherapy [NACT (cisplatin $25mg/m^2$ and gemcitabine $1gm/m^2$ day 1 and 8, 3 weekly for 3 cycles). Radiological assessment was according to RECIST criteria by evaluating downstaging of liver involvement and lymphadenopathy into complete response (CR), partial response (PR), stable disease (SD) and progressive disease (PD). Results: A total of 40 patients were evaluated from January 2012 to December 2014 (CTRT=25, NACT=15). Pretreatment CT scans revealed involvement of hilum (19), liver infiltration (38), duodenum involvement (n=22), colon involvement (n=11), N1 involvement (n=11), N2 disease (n=8), paraaortic LN (n=15), and no lymphadenopathy (n=6). After neoadjuvant therapy, liver involvement showed CR in 11(30%), PR in 4 (10.5%), SD in 15 (39.4%) and lymph node involvement showed CR in 17 (50%), PR in 6 (17.6%), SD in 4 (11.7 %). Six patients (CTRT=2, NACT=4) with 66.6 % and 83% downstaging of liver and lymphnodes respectively underwent extended cholecystectomy. There was 16.6 % and 83.3% rates of histopathological CR of liver and lymph nodes. All resections were R0. Conclusions: Neoadjuvant therapy in unresectable gall bladder cancer results in a 15% resectability rate. This approach has a strong potential in achieving R0 and node negative disease. Radiologic downstaging (CR+PR) of liver involvement is 40.5% and lymphadenopathy is 67.5%. Nodal regression could serve as a predictor of response to neoadjuvant therapy.

Differences in Polyspermy and Penetration Rate Following In Vitro Fertilization of In Vitro Matured Porcine Oocytes by Morphologically Normal Sperm Ratio of Epididymal Sperm and Frozen-Thawed Ejaculated Sperm (정소상체 미부정자와 동결 융해된 사출정자의 형태학적 정상정자 비율이 체외성숙 돼지난자의 체외수정후 정자침입율과 다정자침입의 차이에 미치는 영향)

  • The objective of this study was to evaluate the variation of fertilizing ability following the morphologically normal sperm ratio in porcine IVF using epididymal sperm The results obtained in this experiment were summarized as follows: 1. When the penetration rate (PR), polysper my rate (PSR), pronuclei formation (2PNF) and mean number of sperm (MNS) per oocyte were evaluated according to the percentage of morphologically normal epididyrnal sperm at insemination($\leq$lO%, 10~30% and $\geq$50%). the PR and PSR of $\leq$50% group (82.4, 87.4%) were significantly higher than those of other two groups ($\leq$lO%; 29.7%, 22.6% and 10~30%; 20.3, 37.0%) (p<0.01). Also, the 2PNF per examined oocytes was significantly high in $\geq$ 50% group (p<0.01). 2. When the $\geq$50% group in epididymal sperm was adjusted to 100% (5x1$^5$ cells/ml) , the PSR and 2PNF were not different between epididymal sperm (86.7, 35.1%) and frozen-thawed ejaculated sperm (86.0. 39.4%) although the PR in epididymal sperm (79.7%) was significantly lower than that in frozen-thawed ejaculated sperm (95.5%)(p<0.01). 3. Also. when the PR, PSR, 2PNF and MNS of epididymal sperm were evaluated according to the oocyte: sperm ratio (1:6000, 1:6650. 1:7700 and 1: 10000) at insemination. the PR, PSR and MNS were increased as the oocyte:sperm ratio increases. However, this result indicated that the 2PNF was high in the oocyte:sperm ratio (1:6000 and 1:6650). Therefore. these results suggested that when the percentage of morphologically normal epididymal sperm was more than 50. the fertilizing a ability was very similar to that of frozen-thawed ejaculated sperm and that the detailed evalu¬a ation of morphological normality in porcine IVF using epididymal sperm should be prerequisite to obtain the more effective fertilizing ability.

Application of Silver Nanoparticles for the Control of Colletotrichum Species In Vitro and Pepper Anthracnose Disease in Field

  • Pepper anthracnose caused by Colletotrichum species is one of the most important limiting factors for pepper production in Korea, its management being strongly dependent on chemicals. The aim of this work was to evaluate the possibilities of using silver nanoparticles instead of commercial fungicides. In this study, we evaluated the effect of silver nanoparticles against pepper anthracnose under different culture conditions. Silver nanoparticles (WA-PR-WB13R) were applied at various concentrations to determine antifungal activities in vitro and in the field. The application of 100 ppm concentration of silver nanoparticles produced maximum inhibition of the growth of fungal hyphae as well as conidial germination in comparison to the control in vitro. In field trials, the inhibition of fungi was significantly high when silver nanoparticles were applied before disease outbreak on the plants. Scanning electron microscopy results indicated that the silver nanoparticles caused a detrimental effect on mycelial growth of Colletotrichum species.

Effect of Low-Level Laser to Oral Microorganisms (저출력 레이저가 구강미생물에 미치는 영향)

  • This study is to observe the bacteriocidal effect of the Low Level Laser (LLL) against oral microorganisms which are related to the occurrence of periodontal diseases and oral malodors. The Porphyromonas gingivalis 2561 (P. gingivalis 2561) and Prevotella intermedia (Pr. intermedia) were treated with photosensitizing substance-toluidine blue O (TBO; C.I. 52040) and then radiated with the LLL which has 650nm wavelength for 1, 2, 3 and 5mins. continuously upon varying distances of 1, 2 and 3cm for each experimental groups. The results are as follows; 1. The P. gingivalis 2561 which was treated with TBO and then radiated with LLL at a distance of 3cm for 1min. showed 99.99% higher antibacterial effect in comparison to the experimental group treated only with TBO. 2. The Pr. intermedia which was treated with TBO and then radiated with LLL at a distance of 3cm for 1min. showed 99.8% higher antibacterial effect in comparison to the experimental group treated only with TBO. 3. The bacteriocidal effect of the P. gingivalis 2561 treated with TBO was found to gradually increase as the radiation time of LLL extended from 1min. to 3min. at 1min. intervals. 4. A slight decrease in bacteriocidal effect of the P. gingivalis 2561 was found as the radiation distance of LLL increased from 1cm to 3cm at 1cm intervals. 5. The bacteriocidal effect of the Pr. intermedia was found to slightly decrease as the radiation distance of LLL increased from 1cm to 3cm at 1cm. intervals. As the results shown above suggest, the bacteriocidal effect of LLL was found to increase as the radiation time extended and the distance shortened. Moreover, even the experimental group radiated with LLL at 3cm distance for 1min. which showed the lowest level of bacteriocidal effect, was found to have 99.8% higher bacteriocidal effect than the experimental group which was treated only with TBO and, therefore, this clearly shows the bacteriocidal effect of LLL against oral microorganisms. Thus, the use of LLL is thought to become very useful for suportive treatment for periodontitis and implantitis, and controlling oral malodors as long as it is used within the limits where there is no side effect.

New Chiral Borohydrides. 2. Preparation of Potassium B-Methoxydiisopinocampheylborohydride and Its Asymmetric Reducing Properties

  • In order to prepare new chiral borohydrides (4) possessing chirality on dialkyl moieties, a series of B-alkoxydiisopinocampheylborinates (3) were synthesized by treatment of diisopinocampheylborane ($Ipc_2BH$) with alcohols (R in ROH: Me, Et, i-Pr, t-Bu) and reacted with excess of potassium hydride. Of these chiral borinic esters, only B-methoxydiisopinocampheyl borinate (3a) was converted into the corresponding dialkylmonoalkoxyborohydride (4a). For the other borinic esters, hydride uptake reactions were very slow at room temperature, accompanying disproportionation products at $65^{\circ}C$. The hydride (4a) formed is stable at $0^{\circ}C$ and can be stored over potassium hydride for few months. In the asymmetric reduction of the selected ketones, 4a provided the corresponding alcohols, such as 21% ee for 3-methyl-2-butanone, 11% ee for 2,2-dimethylcyclopentanone, 24% ee for acetophenone, 32% ee for 3-acetylpyridine, 30% for methyl benzoylformate, 31% ee for 4-phenyl-3-butyn-2-one, 39% ee for 3-butyn-2-one, and 34% ee for 3-hexyn-2-one.
