A Duplex PCR Assay for Differentiating Native Common Buckwheat and Tartarian Buckwheat, and Its Application for the Rapid Detection of Buckwheat Ingredients in Food

  • One of the major allergenic proteins in common buckwheat (Fagopyrum elculentum) was found to be a BW10KD. In this work, allergenic BW10KD genomic DNAs from the native common buckwheat 'Pyeongchang' and Tartarian buckwheat 'Clfa47' were cloned by polymerase chain reaction (PCR), and their nucleotide sequences were determined. In addition, a novel PCR assay targeting the allergenic BW10KD gene was developed to detect and differentiate both buckwheat species in food. The nucleotide sequences of the BW10KD genomic DNA from 'Pyeongchang' and 'Clfa47' were 94% identical. Base differences in the nucleotide sequences of the BW10KD genes are probably useful as a molecular marker for species-specific identification. The 'Pyeongchang'-specific primer set 154PF/400PR and the 'Clfa47'-specific primer set 154DF/253DR generated 247 and 100 bp fragments in singleplex PCR, respectively. A duplex PCR assay with 2 species-specific primer sets simultaneously differentiated the 'Pyeongchang' and 'Clfa47' in a single reaction. The PCR assay also successfully allowed for the rapid detection of buckwheat ingredients in foods.

The Manufacturing Technique of Rapid Products using Filling Process (충진공정을 이용한 쾌속시작품 제작 기술)

  • In order to reduce lean-time and cost, recently the technology of Rapid Prototyping and Manufacturing(PR/M) has been used widely. So various RP/M methods have been developed and these systems commercialized several years ago. The machining process is one of these methods. It also offers advantages such as precision and versatility. But there are some considerations during machining. The most important problem among them is the fixturing. So we have to overcome the limitation because the fixturing time is depend on the complexity of geometry to be machined. In this paper, we have developed the fixturing technique using filling process that can be widely useful for rapid products within a short time. So we have carried out some kinds of rapid products such as plastic knob and metal fan using our fixturing process. In fixturing step, the filling material might chosen a resin or a alloy according to wether the work material is plastic or metal respectively. Also we developed the set-up equipment attachable on the table of the milling machine that provided practicable quality during a series of machining operations, named by two step milling process.

Bi-Criteria Process Routing Based on COMSOAL Approach

  • This paper investigates the application of the computer method COMSOAL (Computer Method of Sequencing Operations for Assembly Lines) to the process routing (PR) problem with multiple objectives. In any computer aided process planning (CAPP) system, one of the most critical activities for manufacturing a part could be to generate the sequence that optimizes production time, production cost, machine utilization or with multiple these criteria. The COMSOAL has been adopted to find the optimum sequence of operations that optimizes two major conflicting criteria : production cost and production quality. The COMSOAL is here slightly modified to simultaneously generate and evaluate a set of possible solutions (called as population) instead of processing a solution stepwise in each iteration. The significant features of the COMSOAL include : no parameters settings needed, and a guarantee of feasible solutions. Experimental results show that COMSOAL is a simple but powerful method to quickly generate multiple feasible solutions which are as good as the ones obtained from several other well-known process routing algorithms.

  • The sub-Nyquist nonuniform sampling (SNNS) and the perfect reconstruction (PR) formula are proposed for the development of a systematic method to obtain minimal representation of a speech signal. In the proposed method, the instantaneous sampling frequency (ISF) varies, depending on the least upper boundary of spectral support of a speech signal in time-frequency domain (TFD). The definition of the instantaneous bandwidth (IB), which determines the ISF and is used for generating the set of samples that represent continuous-time signals perfectly, is given. Also, the spectral characteristics of the sampled data generated by the sub-Nyquist nonuniform sampling method is analyzed. The proposed method doesn't generate the redundant samples due to the time-varying property of the instantaneous bandwidth of a speech signal.

  • In order to reduce lead-time and cost, recently the technology of Rapid Prototyping and Manufacturing (PR/M) has been used widely. But we have carried out rapid product, such as sphere, by the milling process instead of RP system. in the case of sphere with three-dimensional shape. the machining method using conventional milling machine has resulted in some troubles because of its deformation and lack of stiffness which is due to usual work piece set up method. In this paper, the feasibility of milling process which is divided into two steps such as the-upper-and-1ower-face milling process using supporting material were investigated and suggested.

  • The purpose of this study is to analyze thinking process in problem solving and to get some teaching materials to improve students' problem solving abilities. For this study, 14 girl and boy students in highschool were tested with 7 testing questions. The whole process of students' problem solving was observed by using 'Thinking aloud', recorded by Audio Tape and finally drawn up to Protocol. On the basis of that Protocol, coding system was set up and characteristics of thinking process in each stage were analyzed. -In the stage of planning, successful problem solvers tried to check the properties of words included in problems(Pr) and made it clear that they were seeking(O) -In the stage of planning, students used abstraction strategy(Ab, making equation(E) or using variable(V)) appropriately could solve more difficult problems. Successful problem solvers turned used unsystematical trial into systematical method and were good at using partial objects, assistant factors. - In the stage of carring out the plan, successful problem solvers to reduce the error, check the purpose, used formula, knowledge and calculation. -In the looking back stage, successful problem solvers generalized the answer and checked the total process.

  • The functional equation related to a distance measure f(pr, qs) + f(ps, qr) = M(r, s)f(p, q) + M(p, q)f(r, s) can be generalized a sum form functional equation as follows $${\frac{1}{n}}{\sum\limits_{i=0}^{n-1}}f(P{\cdot}{\sigma}_i(Q))=M(Q)f(P)+M(P)f(Q)$$ where f, g is information measures, P and Q are the set of n-array discrete measure, and σi is a permutation for each i = 0, 1, ⋯, n-1. In this paper, we obtain the hyperstability of the above type functional equation.

  • Adaptive responses to diverse microbial pathogens might be limited in relatively few types. Host cell responses to pathogens are believed to be patterned or stereotyped along with species or class. We tried to compose the host response to Mycoplasma in terms of cellular gene expression. Although gene expression profile of two host HeLa and 293 cells were quite different each other, 30 genes were differentially expressed by mycoplasma infection in both of HeLa and 293 cells. Six of them (PR48, MADH4, MKPX, CRK, RBM7, NEK3) were related to cell cycle or proliferation. Another category of genes like IL1 HY1, KLRF1, TNFSF14, GBP1 were host defense to elicit immune responses. With this set of genes, we establish the prediction model for mycoplasma contamination.

Bayesian and maximum likelihood estimation of entropy of the inverse Weibull distribution under generalized type I progressive hybrid censoring

  • Entropy is an important term in statistical mechanics that was originally defined in the second law of thermodynamics. In this paper, we consider the maximum likelihood estimation (MLE), maximum product spacings estimation (MPSE) and Bayesian estimation of the entropy of an inverse Weibull distribution (InW) under a generalized type I progressive hybrid censoring scheme (GePH). The MLE and MPSE of the entropy cannot be obtained in closed form; therefore, we propose using the Newton-Raphson algorithm to solve it. Further, the Bayesian estimators for the entropy of InW based on squared error loss function (SqL), precautionary loss function (PrL), general entropy loss function (GeL) and linex loss function (LiL) are derived. In addition, we derive the Lindley's approximate method (LiA) of the Bayesian estimates. Monte Carlo simulations are conducted to compare the results among MLE, MPSE, and Bayesian estimators. A real data set based on the GePH is also analyzed for illustrative purposes.
