  • Protein phosphatase manganese dependent 1D (PPM1D) is one of the Ser/Thr protein phosphatases belongs to the PP2C family. They play an important role in cancer tumorigenesis of various tumors including neuroblastoma, pancreatic adenocarcinoma, medulloblastoma, breast cancer, prostate cancer and ovarian cancer. Even though PPM1D is involved in the pathophysiology of various tumors, the three dimensional protein structure is still unknown. Hence in the present study, homology modelling of PPM1D was performed. 20 different models were modelled using single- and multiple-template based homology modelling and validated using different techniques. Best models were selected based on the validation. Three models were selected and found to have similar structures. The predicted models may be useful as a tool in studying the pathophysiological role of PPM1D.

  • Protein phosphatase manganese dependent 1D (PPM1D), a Ser/Thr protein phosphatise, play major role in the cancer tumorigenesis of various tumors including neuroblastoma, pancreatic adenocarcinoma, medulloblastoma, breast cancer, prostate cancer and ovarian cancer. Hence, analysis on the structural features required for the formation of PPM1D-inhibitor complex becomes essential. In this study, we have performed molecular docking of SL-175 and -176 and protein-protein docking of CDC5L with PPM1D. On analysing the docked complexes, we have identified the important residues involved in the formation of protein-ligand complex. Research concentrating on these residues could be helpful in understanding the pathophysiology of various tumors related to PPM1D.

Determination of Amounts of Benzoic Acid and Propionic Acid in Fermented Soybean Products (장류 중 안식향산과 프로피온산 함량 조사)

  • This study investigated the detection of naturally occurring preservatives in fermented soybean products. Benzoic acid was identified in all of the analyzed samples (gochujang, ganjang, doenjang, and cheonggugjang). The amounts of benzoic acid were in the range of 0.85-11.69 ppm in gochujang, 1.13-7.95 ppm in ganjang, 0.74-17.77 ppm in doenjang, and 1.03-7.64 ppm in cheonggugjang. Propionic acid was found in 24 of 29 gochujang samples, 21 of 30 ganjang samples, 31 of 33 doenjang samples, and 28 of 30 cheonggugjang samples. The amounts of propionic acid ranged from N.D. (not detected) to 49.29 ppm in gochujang, N.D. to 144.67 ppm in ganjang, N.D. to 309.14 ppm in doenjang, and N.D. to 113.07 ppm in cheonggugjang. Among the fermented soybean products, doenjang had the highest average benzoic acid content whereas cheonggugjang had the highest average propionic acid content.

  • Purpose : To investigate the 3-bond and spatial connectivity of human brain metabolites by scalar coupling and dipolar nuclear Overhauser effect/enhancement (NOE) interaction through 2D- correlation spectroscopy (COSY) and 2D- NOE spectroscopy (NOESY) techniques. Materials and Methods : All 2D experiments were performed on Bruker Avance 500 (11.8 T) with the zshield gradient triple resonance cryoprobe at 298 K. Human brain metabolites were prepared with 10% $D_2O$. Two-dimensional spectra with 2048 data points contains 320 free induction decay (FID) averaging. Repetition delay was 2 sec. The Top Spin 2.0 software was used for post-processing. Total 7 metabolites such as N-acetyl aspartate (NAA), creatine (Cr), choline (Cho), lutamine (Gln), glutamate (Glu), myo-inositol (Ins), and lactate (Lac) were included for major target metabolites. Results : Symmetrical 2D-COSY and 2D-NOESY pectra were successfully acquired: COSY cross peaks were observed in the only 1.0-4.5 ppm, however, NOESY cross peaks were observed in the 1.0-4.5 ppm and 7.9 ppm. From the result of the 2-D COSY data, cross peaks between the methyl protons ($CH_3$(3)) at 1.33 ppm and methine proton (CH(2)) at 4.11 ppm were observed in Lac. Cross peaks between the methylene protons (CH2(3,$H{\alpha}$)) at 2.50ppm and methylene protons ($CH_2$,(3,$H_B$)) at 2.70 ppm were observed in NAA. Cross peaks between the methine proton (CH(5)) at 3.27 ppm and the methine proton (CH(4,6)) at 3.59 ppm, between the methine proton (CH(1,3)) at 3.53 ppm and methine proton (CH(4,6)) at 3.59 ppm, and between the methine proton (CH(1,3)) at 3.53 ppm and methine proton (CH(2)) at 4.05 ppm were observed in Ins. From the result of 2-D NOESY data, cross peaks between the NH proton at 8.00 ppm and methyl protons ($CH_3$) were observed in NAA. Cross peaks between the methyl protons ($CH_3$(3)) at 1.33 ppm and methine proton (CH(2)) at 4.11 ppm were observed in Lac. Cross peaks between the methyl protons (CH3) at 3.03 ppm and methylene protons (CH2) at 3.93 ppm were observed in Cr. Cross peaks between the methylene protons ($CH_2$(3)) at 2.11 ppm and methylene protons ($CH_2$(4)) at 2.35 ppm, and between the methylene protons($CH_2$ (3)) at 2.11 ppm and methine proton (CH(2)) at 3.76 ppm were observed in Glu. Cross peaks between the methylene protons (CH2 (3)) at 2.14 ppm and methine proton (CH(2)) at 3.79 ppm were observed in Gln. Cross peaks between the methine proton (CH(5)) at 3.27 ppm and the methine proton (CH(4,6)) at 3.59 ppm, and between the methine proton (CH(1,3)) at 3.53 ppm and methine proton (CH(2)) at 4.05 ppm were observed in Ins. Conclusion : The present study demonstrated that in vitro 2D-COSY and NOESY represented the 3-bond and spatial connectivity of human brain metabolites by scalar coupling and dipolar NOE interaction. This study could aid in better understanding the interactions between human brain metabolites in vivo 2DCOSY study.

  • Glyphosate and 2,4-D were introduced into the vascular systems of Robinia pseudo-acacia L. (3-5cm diameter) with a syringe-type pressure injector in May, 1985. Robinia pseudo-acacia L. were completely controlled by 3 or 6 ml of 3050ppm glyphosate per tree and 6ml of 1000ppm 2,4-D per tree. Herbicidal efficacy was more effective by 6m1 of combined solutions of 1525ppm glyphosate and 500ppm 2,4-D per tree.

  • Embryo tissue from Pinus koraiensis was established in culture on G.D. and W.S. basal medium supplemented with the auxin (NAA, 2,4-D, IBA) and kinetin. The various combinations of growth regulators were studied in order to determine the specific requirements of the callus tissue in vitro. The inorganic nutrient combination of G.D. medium was found to be better than that of W,S. medium. G,D. basal medium supplemented with 0.1 ppm NAA and 0.1 ppm kinetin was the most successful nutrient combination for the growth of callus induced from the embryo of P. koraiensis.

A Study on the Status of Working Environment of Some Rubber and Chemical Products Manufacturing Industries in Busan (고무와 화학제품 제조업 산업장의 작업환경실태에 관한 조사연구)

  • This study was conducted in order to investigate the status of harmful working environ ment on twelve rubber and ten chemical products manufacturing industries in Busan area over a period of five months from lune 1 to October 31, 1980. The summarized results were as follows: 1. The highest and lowest mean values of harmful environmental elements in workroom of rubber products manufacturing industries were noted in twisting (98.7dB) and coating department (77.3dB) to noise, molding ($6.43mg/m^3$) and forming ($1.33mg/m^3$) to dust, bonding (toluene 463.7ppm, xylene 457.8ppm and benzene 111.8ppm, respectively) and splicing (toluene 90.0ppm, xylene 83.3ppm and benzene 6.7ppm, respectively) to organic solvents, respectively. Also in chemical products manufacturing, they were noted in grinding (95.1dB) and shining department (76.8dB) to noise, packing ($4.30mg/m^3$) and staining ($3.20mg/m^3$) to dust, shining (393.3ppm and 375.0ppm, respectively) and varnishing(125.5ppm and 121.7ppm, respectively) to toluene and xylene, and scattering (51.8ppm) and mixing (23.9ppm) to benzene, respectively. 2. The mean values of harmful elements in workroom of rubber products manufacturing were 86.3dB to noise, $4.16mg/m^3$ to dust, 258.2ppm to toluene, 230.3ppm to xylene, and 5 4.0ppm to benzene, respectively. Also in chemical products manufacturing, they were 85.2dB to noise, $3.69mg/m^3$ to dust, 227.9ppm to toluene, 213.2ppm to xylene, and 36.3ppm to benzene, respectively. 3. Number of workers exposed to harmful working environment, over TLV, of a total 10,195 workers in rubber products manufacturing were 1,002(9.8%) to noise, 212 (2.1%) to dust, 1,581(15.5%) to toluene, 1,509(14,8%) to xylene, and 1,524(15.0%) to benzene, respectively. Number of workers exposed to harmful working environment, over TLV, of a 1,913 workers in chemical products manufacturing were 112(5.9%) to noise, 132(6.9%) to each organic solvent, respectively. 4. The values of noise and dust of rubber and chemical products manufacturing in 1980 were lower then those in 1977, but the value of organic solvent in 1980 was similar with that in 1977.

Hormonal Effect on the Callus Induction from Perennial Weeds (다년생잡초(多年生雜草)로부터 Callus 유도(誘導)와 생장조절제(生長調節劑)의 영향(影響))

  • This experiment was conducted to evaluate effect of various hormones on callus induction, and on plantlet formation on various media, and to detect of Londax [Methyl 2-[[[[[(4,6-dimethoxy pyrimidin-2-yl) amino] carbonyl] amono] sulfonyl) methyl] benzoate] and Basta[Ammonium-(3-amino-3-carboxy-propyl)-methyl phosphinate] on callus growth and reaction of succinate dehydrogenase in callus against TTC, using various species such as Eleocharis kuroguwai, Cyperus serotinus, Oryza sativa (samgangbyeo) and Echinochloa crusgalli P. Beauv. var. caudata Kitagawa. The optimal levels of 2,4-D in MS medium seems to be different among species tested, 2.0 ppm for rice and E. crusgalli, 1.0 ppm for Eleocharis kuroguwai, and 4.0 ppm for C. serotinus derived callus from shoot-tip. In case of combination of 2,4-D with BA, 1.0 plus 0.3 ppm appeared the most appropriate level to induce callus from rice and E. kuroguwai, and I.0 plus 0.1 ppm for C. serotinus and E. crusgalli. When 2,4-D treated with TIBA, 1.0 plus 0.5 ppm appeared the most appropriate rates to induce callus derived from seeds of rice, E. crusgalli, seeds of C. serotinus and E. kuroguwai, 1.0 plus 0.3 ppm for shoot-tip of C. serotinus. Positive reaction of succinate dehydrogenase against TTC was observed regardless of calli and herbicides tested, indicating that they all are alive, and these herbicides were not able to kill the calli tested within the short period of time 20 hrs treatment. Regardless of plant species used, the rate of plantlet formation from callus was very low. However, some plantlet formed from E. crusgalli at 0.8 ppm of 2,4-D plus 8.0 ppm of kinetin, and from E. kuroguwai at 1.6 ppm of 2,4-D plus 16.0 ppm of kinetin, showing effectiveness of 2,4-D with kinetin mixture treatment. No callus was induced from C. serotinus treated with Basta from $10^{-6}M$ to $10^{-3}M$. In general, rice was the least susceptible to Basta among plant species tested, followed by E. crusgalli, and E. kuroguwai. In Londax treatment, rice showed the least inhibition rate in callus growth. Callus was induced from rice even at $10^{-3}M$ of Londax. However, $10^{-3}M$ of Londax completely inhibited callus induction from the test species. Rice showed most tolerant to both herbicides, indicating the existence of different responses among plant species.

Effects of the Phytohormones on the Organ Differentiation and the Callus Induction from the Meristem Tip and the Segments of the Leaf and Stem of Potato by in vitro Culture (기내배양시 몇가지 생장조절물질이 감자의 생장점 및 경엽조직편으로부터의 Callus 및 기관분화에 미치는 영향)

  • This study was conducted to define the effect of 2.4-D, NAA, Benzyladenine, and basic mediums on the callus induction and the organ differentiation from the meristem tips and the stem and leaf segments of the potato. Benzyladenine promoted the induction and growth of shoot from the meristem tip of potato but inhibited initiation of roots and induction of callus. At higher concentration of NAA than 0.5 ppm and of 2.4-D than 1.0 ppm the shoots were not initiated but the callus was induced from the meristem. The callus growth was significantly promoted on the medium containing NAA than 2.4-0. The initiation and growth of the shoots from the potato meristem was significantly increased in the medium containing 2.4-D and BA, or NAA and BA, compared with those containing BA, NAA or 2.4-D alone. The callus was more easily induced from the stem segments than the leaf segments of potato. And the 2.4-D was more effective for the induction and growth of the callus than the NAA. MS medium diluted its concentration to 1/2 was more suitable for the initiation and growth of the shoots from the potato meristem than the MS standard medium. For the initiation and growth of the shoots from the potato meristem, the most desirable medium was the diluted MS medium containing 1.0 ppm BA and 0.1 ppm NAA or 0.1 ppm 2.4-D.

Analysis of Trace Metal in Agricultural Products (농산물 중의 미량금속함량 분석)

  • The seven trace metals contents of agricultural products in Kwang-ju were investigated to provide the basic data for its residual limits. Ten kinds of agricultural products (onion, welsh onion, garlic, lettuce, cabbage, pepper, spinach, radish, pear, peach) collected within from Kwang-ju in 1999 were analyzed by Mercury Analyzer for mercury. Inductively Coupled Plasma spectrometer and Atomic Absorption spectrophotometer equipped with Vapor Generation Accessory for Cu, Cd, Mn, Pb, Zn and As. Hg contents were not detected in all samples and As content were detected N.D. ∼0.005 ppm. Cd and Pb contents were detected N.D ∼0.088 ppm and N.D.∼0.096 ppm, respectively, Cu, Mn and Zn contents were 0.109∼1.072 ppm. 0.829∼6.195 ppd and 0.731∼6.176 ppm, respectively. The trace metal concentrations of the agricultural products in Kwang-ju called clean area were within the background level which growth is facilitated.
