  • PL(product liability) is a major social, market, and economic force. The legal obligation of manufacturers and sellers to compensate for injury or damage caused by defective products is not a recent phenomenon. The concept of Product liability has been in existence for many years, but its emphasis has changed recently. This PL prevention program has shown that the two area of product quality assurance one is a PLD(product liability defense) and the other is a PLP(product liability prevention.

  • This paper proposes an algorithm for machine recognition of phonemes in continuous speech. The proposed algorithm is static strategy neural network. The algorithm uses, at the stage of training neuron, features such as PARCOR coefficient and auditory-like perceptual liner prediction . These features are extracted from speech samples selected by a sliding 25.6msec windows with s sliding gap being 3 msec long, then interleaved and summed up to 7 sets of parmeters covering 171 msec worth of speech for use of neural inputs. Perfomances are compared when either PARCOR or auditory-like PLP is included in the feture set.

Bacillus pseudomycoides로 부터 분리된 alanine racemase 유전자의 발현 및 생화학 특성 (Biochemical Characteristics of a Bacteria (Bacillus pseudomycoides) Alanine Racemase Expressed in Escherichia coli)

  • B. pseudomycoides로 부터 alanine racemase로 추정되는 유전자를 분리한 다음 6xHistidine 이 결합된 pET-21 운반체를 이용하여 E. coli BL21(DE3)에서 발현시키고 생화학 특성을 조사하였다. 재조합된 alanine racemase는 affinity chromatography를 이용하여 분리하였으며 SDS-PAGE 분석에서 약 46 kDa의 단일밴드를 나타내었다. 분리된 효소는 여러 아미노산 중에서 L-alanine에 대하여 가장 높은 활성도를 보이고 D-alanine에 대하여 두번째로 높은 활성도를 보였다. 따라서 분리된 효소는 alanine racemase로 판단되었다. 분리된 효소는 D-cysteine에 하여 상당히 저해가 되었다. 효소의 최적 활성도는 pH 9.0 근처에서 관찰되었고 산성의 조건보다는 알칼리의 조건에서 보다 안정하였다. 보효소인 PLP 0.3mM의 첨가에 의해 효소의 활성도는 약 70% 가량 증대되었다.

QUANTITATIVE MONITORING OF TISSUE OXYGENATION BY TIME-RESOLVED SPECTROSCOPY

  • Near-infrared spectroscopy is now being used in clinical diagnosis as a non-invasive monitor of tissue oxygenation state. However, due to lack of the optical pathlength information within tissues, it is still difficult to quantitate the hemoglobin concentration with present CW techniques. Time-resolved spectroscopy (TRS), which measures temporal profiles of emerging light from tissues, enables to estimate the pathlength distribution within tissues by converting time to distance. Consequently, quantitative measurement of tissue oxygenation is possible by analyzing the data with optical diffusion equation 1) or our Microscopic Beer-Lambert law2). Time-Resolved Spectroscopy System : TRS-1O3) Our TRS-10 system consists of a three-wavelength (759, 797, 833 nm) PLP as pulsed light source, a high speed PMT with high sensitivity and three signal-processing circuits for time-resolved measurement (CFD/TAC, A/D converter and histogram memory). Optical pulse train consisting of 759, 797 and 833nm is generated by PLP at 5㎒ repetition rate and irradiated a sample through a single optical fiber. The diffuse-reflected light from the sample is collected by a bundle fiber and then detected by the PMT for single photon measurement. After being amplified by a following fast amplifier, the electrical signals for each wavelength are picked out by CFD/TAC module. Then, a signal processing circuit integrated the TRS data for each wavelength individually. The simultaneous TRS measurement for three wavelengths achieved without any optical or mechanical switch. Experiment and Results Input and detection fibers of TRS-10 were attached at the human forehead with a fiber separation of 3cm. TRS measurements were continuously performed for about 20 minutes including 2 minutes hyper ventilation. It was observed that the total hemoglobin concentration was decreasing during the hyper ventilation and recovered until 2 minutes after hyper ventilation. On the other hand, the deoxy-hemoglobin concentration began to increase after hyper ventilation and had its peak at around 2 minute later, showing 502 drop from 75% to 60% due to inhibition of breathing by performing hyper ventilation. The results showed that this system might be able to quantitate the concentrations of oxy- and deoxy-hemoglobin in the human brain.

Characterization of Thermostable Tyrosine Phenol-Lyase from an Obligatory Symbiotic Thermophile, Symbiobacterium sp. SC-1

  • Tyrosine phenol-lyase of thermophilic Symbiobacterium sp. SC-1, which is obligately and symbiotically dependent on thermophilic Bacillus sp. SK-1, was purified and characterized. The enzyme is composed of four identical subunits and contains approximately 1 mol of pyridoxal 5'-phosphate (PLP) per mol subunit as a cofactor. The enzyme showed absorption maxima at 330 and 420 nm, and lost this absorption profile by treatment with phenylhydrazine. The apparent dissociation constsnt, $K'_D$, for PLP was determined with the apoenzyme to be about $1.2\;{\mu}M$. The isoelectric point was 4.9. The optimal temperature and pH for the $\alpha,\beta$-elimination of L-tyrosine were found to be $80^{\circ}C$ and pH 8.0, respectively. The substrate specificity of the enzyme was very broad: L-amino acids including L-tyrosine, 3,4-dihydroxyphenyl-L-alanine (L-DOPA), L-cysteine, L-serine, S-methyl-L-cysteine, $\beta$-chloro-L-alanine, and S-(o-nitrophenyl)-L-cysteine all served as substrates. D-Tyrosine and D-serine were also decomposed into pyruvic acid and ammonia at rates of 7% and 31% relative to their corresponding L-enantiomers, respectively. D-Alanine, which was inert as a substrate in a, $\beta$-elimination, was the only D-amino acid racemized by the enzyme. The $K_m$ values for L-tyrosine, L-DOPA, S-(o-nitrophenyl)-L-cysteine, $\beta$-chloro-L-alanine, and S-methyl-L-cysteine were 0.19, 9.9, 0.36, 12, and 5.5 mM, respectively.

Biochemical Characteristics of an Alanine Racemase from Xanthomonas oryzae pv. oryzae

  • A gene encoding a putative alanine racemase in Xanthomonas. oryzae pv. oryzae was cloned, expressed and characterized. Expression of the cloned gene was performed in Escherichia coli BL21(DE3)pLys using a pET-21(a) vector harbouring $6{\times}histidine$ tag. Purification of the recombinant alanine racemase by affinity chromatography resulted in major one band by sodium dodecyl sulfate polyacryl amide gel electrophoresis analysis, showing about 45 kDa of molecular weight. The alanine racemase gene, cloned in this experiment, appears to be constitutively expressed in X. oryzae, as analyzed by reverse transcriptase polymerase chain reaction. The enzyme was the most active toward L-alanine and secondly D-alanine, showing a racemic reaction, thus the enzyme is considered as an alanine racemase. The enzyme was considerably activated by addition of pyridoxal-5-phosphate (PLP), showing that 75% increase in activity was observed at 0.3 mM, compared with control. D-Cysteine as well as L-cysteine significantly inhibited the enzyme activity. The inhibitions by cysteines were more prominent in the absence of PLP, showing 9 and 5% of control activity at 2 mM of addition, respectively. The enzyme was the most active at pH 8.0 and more stable at alkaline pHs than acidic pH condition.

Optimization of Direct Lysine Decarboxylase Biotransformation for Cadaverine Production with Whole-Cell Biocatalysts at High Lysine Concentration

  • Cadaverine (1,5-diaminopentane) is an important industrial chemical with a wide range of applications. Although there have been many efforts to produce cadaverine through fermentation, there are not many reports of the direct cadaverine production from lysine using biotransformation. Whole-cell reactions were examined using a recombinant Escherichia coli strain overexpressing the E. coli MG1655 cadA gene, and various parameters were investigated for the whole-cell bioconversion of lysine to cadaverine. A high concentration of lysine resulted in the synthesis of pyridoxal-5'-phosphate (PLP) and it was found to be a critical control factor for the biotransformation of lysine to cadaverine. When 0.025 mM PLP and 1.75 M lysine in 500 mM sodium acetate buffer (pH6) were used, consumption of 91% lysine and conversion of about 80% lysine to cadaverine were successfully achieved.

감정 변화에 강인한 음성 인식 파라메터 (Robust Speech Recognition Parameters for Emotional Variation)

  • 본 논문에서는 인간의 감정 변화에 강인한 음성 인식 기술 개발을 목표로 하여 감정 변화의 영향을 적게 받는 음성 인식시스템의 특징 파라메터에 관한 연구를 수행하였다. 이를 위하여 우선 다양한 감정이 포함된 음성 데이터베이스를 사용하여 감정 변화가 음성 인식 시스템의 성능에 미치는 영향에 관한 연구와 감정 변화의 영향을 적게 받는 음성 인식 시스템의 특징 파라메터에 관한 연구를 수행하였다. 본 연구에서는 LPC 켑스트럼 계수, 멜 켑스트럼 계수, 루트 켑스트럼 계수, PLP 계수와 RASTA 처리를 한 멜 켑스트럼 계수와 음성의 에너지를 사용하였다 또한 음성에 포함된 편의(bias)를 제거하는 방법으로 CMS와 SBR 방법을 사용하여 그 성능을 비교하였다. 실험 결과에서 RASTA 멜 켑스트럼과 델타 켑스트럼을 사용하고 신초편의 제거 방법으로 CMS를 사용한 경우에 HMM 기반의 화자독립 단어 인식기의 오차가 $7.05\%$로 가장 우수한 성능을 나타내었다. 이러한 것은 멜 켑스트럼을 사용한 기준시스템과 비교하여 $59\%$정도 오차가 감소된 것이다.

Biochemical characterization of Alanine racemase- a spore protein produced by Bacillus anthracis

  • Alanine racemase catalyzes the interconversion of L-alanine and D-alanine and plays a crucial role in spore germination and cell wall biosynthesis. In this study, alanine racemase produced by Bacillus anthracis was expressed and purified as a monomer in Escherichia coli and the importance of lysine 41 in the cofactor binding octapeptide and tyrosine 270 in catalysis was evaluated. The native enzyme exhibited an apparent $K_m$ of 3 mM for L-alanine, and a $V_{max}$ of $295\;{\mu}moles/min/mg$, with the optimum activity occurring at $37^{\circ}C$ and a pH of 8-9. The activity observed in the absence of exogenous pyridoxal 5'-phosphate suggested that the cofactor is bound to the enzyme. Additionally, the UV-visible absorption spectra indicated that the activity was pH independece, of VV-visible absorption spectra suggests that the bound PLP exists as a protonated Schiff's base. Furthermore, the loss of activity observed in the apoenzyme suggested that bound PLP is required for catalysis. Finally, the enzyme followed non-competitive and mixed inhibition kinetics for hydroxylamine and propionate with a $K_i$of $160\;{\mu}M$ and 30 mM, respectively.

  • Alanine racemase, a bacterial enzyme belonging to the fold-type III group of pyridoxal 5'-phosphate (PLP)-dependent enzymes, has been shown to catalyze the interconversion between L- and D-alanine. The alanine racemase from the pathogenic bacterium Enterococcus faecalis v583 has been overexpressed in E. coli and was shown to crystallize an enzyme at 295 K, using polyethylene glycol (PEG) 8000 as a precipitant. X-ray diffraction data to $2.5{\AA}$ has been collected using synchrotron radiation. The crystal is a member of the orthorhombic space group, $C222_1$ with unit cell parameter of a=94.634, b=156.516, $c=147.878{\AA},\;and\;{\alpha}={\beta}={\gamma}=90{\AA}$. Two or three monomers are likely to be present in the asymmetric unit, with a corresponding $V_m\; of\;3.38{\AA}^3\;Da^{-1}\;and\;2.26{\AA}^3\;Da^{-1}$ and a solvent content of 63.7% and 45.5%, respectively.
