Isolation and Analysis of Temperature Sensitive Mutants in Aspergillus nidulans (Aspergillus nidulans 온도감수성 돌연변이주의 분리 및 분석)

  • About 40 temperature-sensitive mutants have been isolated as a preliminary step to study the spore germination, the cell cycle, and the control of macromolecular synthesis in Aspergillus nidulans. To obtain temperature-sensitive mutants rapidly and effectively, the selective enrichment method using antifungal antibiotic nystatin was developed. Based on the data which had applied to the concentration of auxotrophic mutants by the earlier investigators, the optimal concentration and the time of treatment at the nonpermissive temeprature were determined as 50 to 100 units per ml and 4.5 hr., respectively. Out of 41 ts mutants assigned to the strain symbol PK, thirteen that seemed to be arrested at the earlystage of spore germination were subjected to the further cytological and genetic analysis. Elght of these mutants are able to form germ tube and five not. Staining with acid fuchsin for the 5PK strains shows that one irreversible mutant, PK6 strain able to form germ tube, accumulate mitotic spindle, being arrested in mitosis. Another PK15 and PK23 strain have more than one intact nucleolus without germ tube formation at the restrictive temperature. the temperature-senstive mutation in PK12 strain, the onlystrain which is able occurred in certain gene specific for the germination of spore. All of the ts markers are recessive and complement each other in heterokaryon between two different ts markers at the restrictive temperature.

Ammonia Nitrogen Removal by Cation Exchange Resin (양이온 교환수지에 의한 암모니아성 질소 제거)

  • This study was conducted to know the removal characteristics of ammonia nitrogen by commercially available cation exchange resins. Eight acidic cation exchange resins were investigated in batch reactors. Among them, the most effective resin for ammonia removal in solution was PK228, which was a strong acidic resin of $Na^{+}$ type. PK228 was compared with activated carbon and natural zeolite. The effects of cation exchange capacity, ammonia concentration, resin amount, temperature and pH on ammonia removal by PK228 were investigated in batch reactor, and the effect of effluent velocity in continuous column reactor. Strong acidic resins of porous type were more effective than week acidic resins or gel type resins for ammonia removal in solution. PK228 was more effective than activated carbon and natural zeolite for ammonia removal in batch reactor. With increasing initial ammonia concentration, the amount of ammonia removed by PK228 increased, but the proportion of removed ammonia to initial ammonia concentration decreased. The effect or temperature on ammonia removal by PK228 was very slight. The ammonia removal to acidic solution was more effective than that at basic solution. With decreasing effluent velocity of solution through column, breakthrough point extended, and ammonia removal capacity increased.d.

Effects of Dietary Supplementation of Ginkgo Leaf and Pumpkin on the Growth Performance, Intestinal Microflora, Blood Biochemical Profile and Antioxidant Status in Broiler Chickens (사료 내 은행잎과 호박 첨가 급여가 육계의 성장, 소장 내 미생물 균총, 혈액 생화학 분석 및 체조직 내 항산화 작용에 미치는 영향)

  • The current study was performed to develop natural bio-active substances as additives for the production of high quality broiler chickens. A total of 120 male 3 day-old broiler chicks were randomly allocated to CON (control), GK2.5 (ginkgo leaf 2.5%), GK5.0 (ginkgo leaf 5.0%), PK2.5 (pumpkin 2.5%) and PK5.0 (pumpkin 5.0%) of five groups in cages (24 birds per group). All birds were fed corresponding diets from 3 to 35 d of age and determined growth performance and biological parameters including blood biochemical profiles, antioxidant status and intestinal microflora. During the entire feeding trial, GK5.0 and PK5.0 groups resulted in a significantly (P<0.05) higher FCR than GK2.5 and PK2.5 groups. Plasma triglyceride significantly (P<0.05) increased in GK5.0 group compared with the other groups, and the level of alanine transaminase (ALT) increased (P<0.05) in GK5.0 and PK5.0 groups compared with that in PK2.5 group. Dietary addition of ginkgo leaf and pumpkin significantly (P<0.05) increased superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in the small intestine. Also, the addition of 2.5% ginkgo leaf significantly (P<0.05) increased the activities of SOD, GSH-Px and glutathione-S-transferase (GST) in the liver. Futhermore, muscle GST activity significantly (P<0.05) enhanced by dietary addition of ginko leaf and pumpkin. However, the level of lipid peroxidation (MDA) in the small intestine and muscle turned to be higher (P<0.05) in PK5.0 group. The colony forming units (CFU) of E. coli in intestinal digesta significantly (P<0.05) decreased in both ginko leaf and pumpkin supplemented groups compared with CON group. In conclusion, dietary addition of 2.5% ginko leaf and pumpkin as dietary sources can be applicable for the production of high quality broiler chickens.

Quercetin Sensitizes Human Leukemic Cells to TRAIL-induced Apoptosis: Involvement of DNA-PK/Akt Signal Transduction Pathway (Quercetin 에 의한 사람백혈병 세포의 TRAIL 에 대한 감수성 증가: DNA-PK/Akt 신호전달경로의 관여)

  • Despite the fact that many cancer cells are sensitive to TNF-related apoptosis-inducing ligand (TRAIL)-induced apoptosis, some cancer cells show either partial or complete resistance to TRAIL. Human leukemic K562 and CEM cells also show resistance to TRAIL-induced apoptosis. Novel molecular target and treatment strategies are required to overcome TRAIL resistance of human leukemia cells. Therefore, the purpose of this study was to target key anti-apoptotic molecules deciding TRAIL resistance for sensitization of TRAIL-resistant K562 and CEM cells, and to evaluate the effect of quercetin as a TRAIL sensitizer on these TRAIL-resistant cells. We found that quercetin acted in synergy with TRAIL to enhance TRAIL-induced apoptosis in K562 cells by inhibition of the DNA-PK/Akt signaling pathway, which leads to enhancement of TRAIL-mediated activation of caspases and concurrent cleavage of PARP and up-regulation of Bax. The findings suggest that the DNA-PK/Akt signaling pathway plays an essential role in regulating cells to escape from TRAIL-induced apoptosis, and quercetin could act in synergy with TRAIL to increase apoptosis by inhibition of the DNA-PK/Akt signaling pathway, which overcomes TRAIL-resistance of K562 and CEM cells. This study suggests that DNA-PK might interfere with TRAIL-induced apoptosis in human leukemic cells through activation of the Akt signaling pathway.

On the asymptotic correlationship for some process capability indices Ĉp, Ĉpk and Ĉpm (공정능력지수 Ĉp, Ĉpk와 Ĉpm를 위한 점근적 상관성에 관한 연구)

  • Higher quality level is generally perceived by customers as improved performance by assigning a correspondingly higher satisfaction score. Usually, the quality level is measured by process capability indices. The index is used to determine whether a production process is capable of producing items within a specified tolerance. Some useful process capability indices $C_p$, $C_{pk}$ and $C_{pm}$ have been widely used in six sigma industries to assess process performance. Most evaluations on process capability indices focus on point estimates, which may result in unreliable assessments of process performance. It is necessary to investigate their asymptotic correlationship among process capability indices $\hat{C}_p$, $\hat{C}_{pk}$ and $\hat{C}_{pm}$. In this paper, we study their asymptotic correlationship for some process capability indices $\hat{C}_p$, $\hat{C}_{pk}$ and $\hat{C}_{pm}$ under the normal process.

Protective Effects of Chungkookjang Extract on High Glucose Induced Oxidative Stress in LLC-PK1 Cells

  • This study was designed to investigate the protective effect of a methanol extract of Chungkookjang (CKJ) on high glucose induced oxidative stress in LLC-$PK_1$ cells (renal tubular epithelial cells), which are susceptible to oxidative stress. Freeze dried CKJ powder was extracted with methanol, and the extract solution was concentrated, and then used in this study. To determine the protective effect of CKJ extract, oxidative stress was induced by exposing of LLC-$PK_1$ cells to high glucose (30 mM) or normal glucose (5 mM) for 24 hr. Exposure of LLC-$PK_1$ cells to high glucose for 24 hr resulted in a significant (p<0.05) decrease in cell viability, catalase, SOD and GSH-px activity and a significant (p<0.05) increase in intracellular ROS level and thiobarbituric acid reactive substances (TBARS) formation in comparison to the cells treated with 5 mM glucose. CKJ extract treatment decreased intracellular ROS level and TBARS formation, and increased cell viability and activities of antioxidant enzymes including catalase, SOD and GSH-px in high glucose pretreated LLC-$PK_1$ cells. These results suggest that CKJ extract may be able to protect LLC-$PK_1$ cells from high glucose-induced oxidative stress, partially through the antioxidative defense systems.

Differential expression patterns of gangliosides in the tissues and cells of NIH-mini pig kidneys

  • Gangliosides are a ubiquitous component of the membranes of mammalian cells that have been suggested to play important roles in various cell functions such as cell-cell interaction, adhesion, cell differentiation, growth control and signaling. However, the role that gangliosides play in the immune rejection response in xenotransplantation is not yet clearly understood. In this study, differential expression patterns of gangliosides in HEK293 (human embryonic kidney cells), PK15 (porcine kidney cells), NIH-kd (NIH-mini pig kidney cells, primary cultured) and the cortex, medulla and calyx of the NIH-mini pig kidney were investigated by high-performance thin-layer chromatography (HPTLC). The results revealed that HEK293, PK15 and NIH-kd contained GM3, GM2 and GD3 as major gangliosides. Moreover, GM3, which are the gangliosides of NIH-kd, were expressed at higher levels than HEK293 and PK15. Especially, GT1b were expressed in HEK293 and NIH-kd but not in PK15. Finally, GM1 and GD1a were expressed in NIH-kd, but not in HEK293 or PK15. These results suggest that differential expression patterns of gangliosides from HEK293, PK15 and NIH-kd are related to the immune rejection response in xenotransplantation.

A Study on Pk(Probability of Kill) Calculation Method of the Direct Fire Weapon System using ANN (인공신경망을 적용한 직사화기 무기체계의 살상확률(Pk) 산출방법론 연구)

  • Until now it has had the limitation of the target in the US JMEM to calculate the Pk with the existing method by our study. In this study, we focused on deriving a method to calculate the Pk of the actual targets except JMEM targets using ANN. We study the initial predictive model of ANN(Artificial Neural Network) from the targets data of the specification and the vulnerable area in the US JMEM(Joint Munitions Effectiveness Manuals), and calculate the actual targets vulnerable area by using this method. Finally, we propose a method to calculate the Pk by applying those data to the existing method of us.

Preparation of Polyketone Micro/nanofiber Membrane based on Electrospinning Condition and Its Application in Oil-Water Separation (전기방사법의 제조 조건에 따른 폴리케톤 마이크로/나노섬유 분리막 제조 및 유수 분리 적용)

  • In this study, a novel material polyketone (PK) was chosen and PK micro/nano fiber membranes were fabricated via electrospinning method under various conditions. After that, the potential application in oil-water separation was thoroughly investigated. The surface of microfiber membrane formed under high humidity especially became much rougher than that formed under low humidity. When salt was added to the spinning solution, the diameter of fibers was reduced up to 90% and the nanofiber membranes could be formed. The oil/water emulsions were prepared and separated under gravity condition using the manufactured rPK-LNC and PK-H membranes. The separation characteristics was evaluated by measuring total organic carbon (TOC) and turbidity. Meanwhile, the changes in the physical properties of fiber membranes under various conditions and with or without salt, as well as the changes in oil water separation characteristics were also studied.
