Additional mitochondrial DNA sequences from the dung beetle, Copris tripartitus (Coleoptera: Scarabaeidae), an endangered species in South Korea

  • The dung beetle, Copris tripartitus (Coleoptera: Scarabaeidae), is an endangered insect in South Korea. Previously, partial mitochondrial COI and CytB gene sequences have been used to infer genetic diversity and gene flow of this species in South Korea. In this study, we additionally collected C. tripartitus (n = 35) from one previous locality and two new localities, sequenced COI and CytB genes, and combined these with preexisting data for population genetic analysis. Sequence divergence of current samples showed slightly lower values [4.86% (32 bp) for COI and 4.16% (18 bp) for CytB] than that in the previous study. Nucleotide diversity (${\pi}$) ranged from 0.005336 (Gulupdo) to 0.020756 (Seogwi-dong) in COI and 0.009060 (Aewol-eup) to 0.017464 (Seogwi-dong) in CytB. Seogwi-dong samples that showed the highest ${\pi}$ in the previous study also showed the highest ${\pi}$ in this study for both gene sequences. The newly investigated Gulupdo samples had the lowest haplotype diversity for both gene sequences. They also had the lowest ${\pi}$ for COI and the second lowest ${\pi}$ for CytB. On the other hand, the newly added Haean-dong sample had relatively higher diversity estimates. Gene flow among populations was high, although significant difference was only detected between Gulupdo and Anmado or between Gulupdo and Seogwi-dong for COI sequences (P < 0.05). Considering the high genetic diversity and gene flow in C. tripartitus populations, one major issue regarding conservation seems not to be recovery of genetic diversity.

근관치료 시 초음파 세정 기술을 이용한 잔사 제거의 효율성 (The efficacy of ultrasonic irrigation technique on debris removal during root canal treatment)

  • 목적: Passive irrigation (PI)과 Passive ultrasonic irrigation (PUI)의 상아질 잔사 제거 효율성을 비교하는 것이다. 연구 재료 및 방법: 16 mm 길이로 표준화된 하악 소구치 치근을 근관 확대 후 종단으로 이등분 하였다. 분리된 한쪽 상아질벽에는 길이 4 mm, 너비 0.2 mm, 깊이 0.5 mm의 표준화된 구를 형성하였고, 반대쪽에는 직경 0.3 mm, 깊이 0.5 mm인 3개의 반구 형태의 함요를 형성하였다. 각 구와 함요에는 NaOCl과 혼합된 상아질 잔사로 채우고, 퍼티 인상재를 사용하여 재조합하였다. Group 1은 PI를 이용하고 Group 2는 PUI를 이용하여 각 근관내를 세정하였다. 현미경과 디지털카메라로 절편의 근관벽을 촬영하고, 구와 함요에 잔존하는 상아질 잔사의 양을 scoring system을 이용하여 평가하였다. 결과: 근관의 중앙 1/3에서 group 1이 group 2에 비해 잔사를 더 많이 제거하였지만, 치근단부에서는 유의한 차이가 없었다. 결론: 하악 소구치에서 근관에 기구가 닿지 못하는 부위를 세정시, 근관의 중앙 1/3에서 PUI를 사용하는 것이 PI에 비해 상아질 잔사의 제거 효율이 우수했지만, 치근단부에서는 PUI와 PI의 유의한 차이가 없었다.

고추 잎점무늬병.역병 복합 저항성 계통육성 (Breeding Lines with Multiple Resistance to both Gray Leaf Spot and Phytophthora Blight in Pepper(Capsicum annuum L.))

  • 1999년도에는 KC47${\times}$KCB14, KC220${\times}$KC268 조합의 $F_2$ 및 여교배 집단과 KC47-1${\times}$KC263(AC2258), KC47-1${\times}$KCB13-2-1, KC47-1${\times}$KCB13-4-2 조합의 $F_3$집단에 대하여 잎점무늬병과 역병을 차례로 접종, 복합저항성 개체를 선발하여 차세대를 육성하였다. 2000년도에는 같은 조합의 $F_3$$F_4$세대에 대하여 잎점무늬병과 역병에 대한 저항성을 검정하여 복합 저항성인 개체로부터 차세대 종자를 확보하였다. 선발세대의 경과에 따라 두 가지 병에 대한 저항성에서 현저한 진전이 관찰되었다.

우리나라 일부도시의 중 . 고등학교에서 강하분진으로 형성된 축적분진에 함유된 중금속의 분포와 특성에 대한 연구 (Distribution and Characteristics of Heavy Metals in the Fallout Dust Deposits at the Middle and High Schools of Some Cities in South Korea)

  • The purpose of this study was to investigate the distribution and characteristics of heavy metals in the fallout dust deposits at the middle and high schools. We collected the samples at 60 schools in Kongju, Incheon and Daejeon on from September to November, 2001, and analyzed Cadmium(Cd), Copper(Cu), Lead(Pb) and Zinc(Zn) from them. Zn was the highest level as 551 $\pm$ 79.9 ppm, and the next order were Pb, Cu, and Cd as 146.0 $\pm$ 15.2 ppm, 98.7 $\pm$ 14.4 ppm and 5.21 $\pm$ 0.76 ppm, respectively. The rates of schools exceeding the soil environmental standards were 56.3% in Cd, 53.3% in Zn, 51.5% in Pb, and 31.7% in Cu, respectively. Cd and Pb were the highest levels at schools in Daejeon as 6.30 $\pm$ 0.87 ppm and 171 $\pm$ 26.1 ppm, respectively. Cu and Zn were the highest levels at schools in Incheon as 176.2 $\pm$ 55.8 ppm and 919.8 $\pm$ 185.7 ppm, respectively. At Incheon, Cu and Zn levels were significantly higher than Kongju(p=0.04l), and Daejeon(p=0.016), respectively. Total pollution index(PI) of heavy metals was 1.51 $\pm$ 0.16. PI was 2.00 $\pm$ 0.51 on Incheon, 1.50 $\pm$ 0.13 on Daejeon, and 0.92 $\pm$ 0.24 on Kongju. Correlations were 0.675 (p<0.05) between Cu and Cd, 0.663(p<0.05) between Cu and Zn, and 0.477 between Cd and Pb. In conclusion, Among heavy metals in the fallout dust deposits at schools, Cu and Zn were the highest levels at Incheon, Cd and Pb were the highest levels at Daejeon.

  • Nutritional status of cattle in Java in Indonesia was investigated by evaluating P, Ca, Mg, K and Na concentrations in forage and plasma inorganic phosphorus (Pi), Ca and Mg, hematocrit and hemoglobin concentrations of cattle during both rainy and dry seasons in 1988. Investigations were conducted at Jonggol (West Java), Malang (East Java) and Majokerto (East Java). Low P content in forage averaging 0.11% on a dry matter basis, high Ca:P ratio in forage in the rainy season (7.7:1) and low plasma Pi concentrations averaging 4.71 mg/dl were observed in Jonggol. In all three places, Ca content in forages was higher than the requirement according to the NRC standard and plasma Ca concentrations were above the critical level. During the rainy season forty percent of cattle in Malang and Mahokerto showed plasma Mg concentrations below 2mg/dl in spite of normal Mg and K content in forage. Forage samples from each place showed a wide variety in Na content, and Na content of two thirds of forage samples was lower than the requirement according to the NRC standard. Hematocrit and hemoglobin concentrations in Malang and Majokerto showed higher values than the normal range.

  • PD-L1 is expressed in a variety of antigen-presenting cells and provides T cell tolerance via ligation with its receptor PD-1 and B7-1 on T cells. Stimulation with lipopolysaccharide (LPS) can increase the level of PD-L1 expression in B cells and macrophages, which suggests that this molecule plays a role in the immunosuppression observed in severe sepsis. The aim of this study was to identify which of the downstream pathways of TLR4 are involved in the up-regulation of PD-L1 by LPS in macrophages. Flow cytometry was used to examine the expression of PD-L1 in RAW 264.7 macrophages stimulated with LPS. The following chemical inhibitors were used to evaluate the role of each pathway: LY294002 for PI3K/Akt, SB202190 for p38 MAPK, and U0126 for MEK. LPS induced the expression of PD-L1 in a time- and dose-dependent manner. Transfection of siRNA for TLR4 suppressed the induction of PD-L1. Pretreatment with LY294002 and SB202190 decreased the level of PD-L1 expression but U0126 did not. Overall, the PI3K/Akt and p38 MAPK pathways are involved in the up-regulation of PD-L1 expression in RAW 264.7 macrophages stimulated with LPS.

Phosphatidylinositol 3-kinase (PI3KCA) Oncogene Mutation Analysis and Gene Expression Profiling in Primary Breast Cancer Patients

  • Background: The phosphatidylinositol 3-kinase (PI3K) pathway plays a significant role in apoptosis, cellular proliferation and motility. The aim of the present study was to analyze mutations and gene expression profiles of the PI3KCA gene to determine any role in breast carcinomas. Materials and Methods: We analyzed 38 breast cancers for mutations in the two PIK3CA hotspots in exons 9 and 20 by direct sequencing of DNA obtained from biopsy samples. We have also analyzed expression of the PI3KCA gene in 38 breast carcinoma tumor and corresponding control tissue samples at the mRNA level by RT-PCR. The Fisher's exact test ($2{\times}2$ only) was performed using MedCalc software for to examine associations with mRNA levels. Results: In the present study a total of 13 cases demonstrated somatic mutations. In 9/13 cases 1633 G>A (E545K) were found in exon 9, whereas in exon 20, 4/13 cases had 3140A>G mutation. Our combined analysis showed PI3KCA mutations present in 34% of human breast cancer patients. In our study, we have also clearly found significantly higher expression in breast cancer tissues in comparison with control tissues (p=0.001). Conclusions: PIK3CA mutation is an emerging tumor marker that, in the future, might be used in the process of choosing a treatment. The detection of PI3KCA mutation might have important clinical implications for diagnosis, progression and therapy.

옥수수 뿌리로부터 분리한 Membrane-bound ATPase의 특성에 관한 연구 (Characterization of the Membrane-bound Adenosine Triphosphatase from Corn Roots)

  • 옥수수 뿌리로부터 분리한 13,000g pellet과 13,000~80,000g pellet 내에 있는 membrane-bound ATPases의 특성을 구명하였다. 13,000g pellet과 13,000~80,000g pellet의 membrane-bound ATPases의 최적 pH는 5와 9였다. Discontinuous sucrose gradient centrifugation에 의한 13,000g pellet의 분획중 Fraction C는 pH 5에서, Fraction D, E 및 F는 pH 5에서보다 pH 9에서 더높은 활성을 나타냈다. 13,000~80,000g pellet의 분획에서 보면, Fraction A, C는 pH 9보다 pH 5에서, Fraction B, D, E 및 F는 pH 5보다 pH 9에서 더 높은 활성을 가겠다. pH 5와 pH 9에서 membrane-bound ATPases의 기질포화 농도는 3~5 mM이며 ATP에 대한 Km 값은 모두 0.25 mM이였다. Vmax 값은 8.0~55.6 $\mu$M Pi/mg membrane protein/hr의 범위에 있었다. Membrane-bound ATPase의 활성은 $K^+$ 이온에 의해 증가되었다.

  • Analyses of $_1$H-NMR, infrared and electronic spectral data for $[Ir(CH_2 = CHCN)(CO)(P(C_6H_5)_3)_2]ClO_4 (1)$prepared by the reaction of $Ir(OClO_3)(CO)(P(C_6H_5)_3)_2$ with $CH_2 = CHCN$, agree with the suggestion that 1 is a mixture of the nitrogen-bonded acrylonitrile complex, $[(CO)(P(C_6H_5)_3)_2Ir-NCCH = CH_2]ClO_4$ and other compound which may be the C = C ${\Pi}$ -system-bonded acrylonitrile complex, "[(CO)(P(C6H5)3)2Ir-CHCN = CH2]ClO4.

Detection of Co-Infection of Notocactus leninghausii f. cristatus with Six Virus Species in South Korea

  • Co-infection with two virus species was previously reported in some cactus plants. Here, we showed that Notocactus leninghausii f. cristatus can be co-infected with six different viruses: cactus mild mottle virus (CMMoV)-Nl, cactus virus X (CVX)-Nl, pitaya virus X (PiVX)-Nl, rattail cactus necrosis-associated virus (RCNaV)-Nl, schlumbergera virus X (SchVX)-Nl, and zygocactus virus X (ZyVX)-Nl. The coat protein sequences of these viruses were compared with those of previously reported viruses. CMMoV-Nl, CVX-Nl, PiVX-Nl, RCNaV-Nl, SchVX-Nl, and ZyVX-Nl showed the greatest nucleotide sequence homology to CMMoV-Kr (99.8% identity, GenBank accession NC_011803), CVX-Jeju (77.5% identity, GenBank accession LC12841), PiVX-P37 (98.4% identity, GenBank accession NC_024458), RCNaV (99.4% identity, GenBank accession NC_016442), SchVX-K11 (95.7% identity, GenBank accession NC_011659), and ZyVX-B1 (97.9% identity, GenBank accession NC_006059), respectively. This study is the first report of co-infection with six virus species in N. leninghausii f. cristatus in South Korea.
