Genotype by Dietary Lysine Interaction for Growth and Response to Sheep Red Blood Cells and Escherichia coli Inoculation in Commercial Broiler Chicks

  • An experiment was conducted to assess the interaction between genotypes and dietary lysine content in commercial broiler chicks by measuring growth, and response to sheep red blood cells (SRBC) and Escherichia coli (E.coli) inoculation. Female chicks from four genotypes (A=Anak 2000; B=Hubbard; C=Cobb and D=Synthetic broiler) were fed four levels of lysine in diet from d old till the end of experiment. The lysine content of the diet was 9.61, 10.51, 11.41 and 12.31 g/kg. Body weights at 0, 14, 28 and 42 d of age and pen-wise feed intake till 14, 28 and 42 d of age were recorded. Production of antibody against SRBC and resistance to E.coli were measured at 5 d of post inoculation (PI) at 43 d of age. Also, response to phytohemaglutinin-P (PHA-P) was measured at 12 and 24 h of PI at 48 d of age. Genotype by dietary lysine interaction was significant for body weights at 14 and 28 d of age, but not at 42 d of age. Genotype by dietary lysine interaction was not significant for feed efficiency, for antibody titers against SRBC, and for air sac lesion score, relative bodyweight change, and relative weights of bursa and spleen in response to E.coli inoculation. However, a significant interaction was observed between the levels of lysine and dosage of SRBC for antibody titers. There was significant genotype by dietary lysine interaction for cutaneous basophilic hypersensitivity (CBH) response to PHA-P at 12 and 24 h of PI. It may be concluded that to obtain optimum body weight and immunity in commercial broilers the dietary lysine requirement may be recommended specific to the genotype.

Fabrication of 2-layer Flexible Copper Clad Laminate by Vacuum Web Coater with a Low Energy Ion Source for Surface Modification (저 에너지 표면 개질 이온원이 설치된 진공 웹 공정을 이용한 2층 flexible copper clad laminate 제작)

  • In order to fabricate adhesiveless 2-layer flexible copper clad laminate (FCCL) used for COF (chip on film) with high peel strength, polyimide (PI; Kapton-EN, $38\;{\mu}m$) surface was modified by reactive $O_2^+$ and $N_2O^+$ ion beam irradiation. 300 mm-long linear electron-Hall drift ion source was used for ion irradiation with ion current density (J) higher than $0.5\;mA/cm^2$ and energy lower than 200 eV. By vacuum web coating process, PI surface was modified by linear ion source and then 10-20 nm thick Ni-Cr and 200 nm thick Cu film were in-situ sputtered as a tie layer and seed layer, respectively. Above this sputtered layer, another $8-9{\mu}m$ thick Cu layer was grown by electroplating and subsequently acid and base resistance and thermal stability were tested for examining the change of peel strength. Peel strength for the FCCLs treated by both $O_2^+$ and $N_2O^+$ ion irradiation showed similar magnitudes and increased as the thickness of tie layer increased. FCCL with Cu (200 nm)/Ni-Cr (20 nm)/PI structure irradiated with $N_2O^+$ at $1{\times}10^{16}/cm^2$ ion fluence was proved to have a strong peel strength of 0.73 kgf/cm for as-received and 0.34 kgf/cm after thermal test.

Suppression of Migration and Invasion by Alnus hirsuta in Human Hepatocellular Carcinoma Cells

  • Hepatocellular carcinoma (HCC) has a poor prognosis and high metastasis and recurrence rates. Although extracts of Alnus hirsuta (Turcz. ex Spach) Rupr. (AH) have been demonstrated to possess potential anti-inflammatory and anti-cancer activities, the underlying mechanism of AH in HCC treatment remains to be elucidated. We investigated the effects and potential mechanisms of AH on migration and invasion of Hep3B cells. Within the non-cytotoxic concentration range, AH significantly inhibited motility and invasiveness of Hep3B cells in a concentration-dependent manner. Inhibitory effects of AH on cell invasiveness are associated with tightening of tight junctions (TJs), as demonstrated by an increase in transepithelial electrical resistance. Immunoblotting indicated that AH decreased levels of claudins, which form major components of TJs and play key roles in the control and selectivity of paracellular transport. Furthermore, AH inhibited the expression and activity of matrix metalloproteinase (MMP)-2 and MMP-9 and simultaneously increased the levels of tissue inhibitor of metalloproteinase (TIMP)-1 and TIMP-2. These effects were related to inactivation of the phosphoinositide 3-kinase (PI3K)/AKT pathway in Hep3B cells. Therefore, AH inhibits migration and invasion of Hep3B cells by inhibiting the activity of MMPs and tightening TJs through suppression of claudin expression, possibly by suppressing the PI3K/AKT signaling pathway.

Molecular Target Therapy of AKT and NF-kB Signaling Pathways and Multidrug Resistance by Specific Cell Penetrating Inhibitor Peptides in HL-60 Cells

  • Background: PI3/AKT and NF-kB signaling pathways are constitutively active in acute myeloid leukemia and cross-talk between the two has been shown in various cancers. However, their role in acute myeloid leukemia has not been completely explored. We therefore used cell penetrating inhibitor peptides to define the contributions of AKT and NF-kB to survival and multi drug resistance (MDR) in HL-60 cells. Materials and Methods: Inhibition of AKT and NF-kB activity by AKT inhibitor peptide and NBD inhibitor peptide, respectively, resulted in decreased expression of mRNA for the MDR1 gene as assessed by real time PCR. In addition, treatment of HL-60 cells with AKT and NBD inhibitor peptides led to inhibition of cell viability and induction of apoptosis in a dose dependent manner as detected by flow cytometer. Results: Finally, co-treatment of HL-60 cells with sub-optimal doses of AKT and NBD inhibitor peptides led to synergistic apoptotic responses in AML cells. Conclusions: These data support a strong biological link between NF-kB and PI3-kinase/AKT pathways in the modulation of antiapoptotic and multi drug resistant effects in AML cells. Synergistic targeting of these pathways using NF-kB and PI3-kinase/AK inhibitor peptides may have a therapeutic potential for AML and possibly other malignancies with constitutive activation of these pathways.

  • The wound induction motor can provide high starting torque and reduced starting current simultaneously by inserting large resistor externally when starting. And this technique is one of the well known methods among the induction motor starting methods and generally used for heavy load starting such as crane and cement factories. The conventional PI controller has been widely used in industrial application due to the simple control algorithm and is generally used for control of current torque, position, and speed for the wound induction motor drive system. However, the conventional control system for wound induction motor may result in poor performance because sensors have to be used but are often limited by the environmental condition. Recently, to overcome these problems, many sensorless vector control methods for the wound induction motor have been studied. This paper presents a MRAS method for sensorless vector control of the wound induction motor drive. In the conventional MRAS method, in low frequency, the stator resistance variation may result in poor performance. Therefore, this paper presents a MRAS method with stator and rotor resistance tuning for sensorless vector control of the wound induction motor to overcome several shortages of the conventional MRAS caused by parameter variation and to enhance the robustness of the sensorless vector control. The validity and effectiveness of the proposed method is verified through digital simulation.

Heating Characteristics of Carbon Fiber Polyimide-Coated by Electrophoretic Deposition (전기영동증착법으로 폴리이미드를 코팅한 탄소섬유의 발열 특성 연구)

  • Carbon fiber(CF) with excellent thermal conductivity and electrical conductivity is attracting attention as an alternative material because metal heating elements have problems such as high heat loss and fire risk. However, since CF is oxidized and disconnected at about 200℃ or higher, the application of heating elements is limited, and CF heating elements in the form of vacuum tubes are currently used in some commercial heaters. In this work, polyimide(PI) with high heat resistance was coated on the surface of carbon fiber by electrophoretic deposition to prevent oxidation of CF in the atmosphere without using a vacuum tube, and the coating thickness and heat resistance were investigated according to the applied voltage. The heater made by connecting the PI-coated CF heating elements in series showed stable heating characteristics up to 292℃, which was similar to the heating temperature result of the heat transfer simulation. The PI layer coated by the electrophoretic deposition method is effective in preventing oxidation of CF at 200℃ or higher and is expected to be applicable to various heating components such as secondary batteries, aerospace, and electric vehicles that require heat stability.

  • In order to find out the properties in flow resistance of trawlR=1.5R=1.5\;S\;v^{1.8}\;S\;v^{1.8} nets and the exact expression for the resistance R (kg) under the water flow of velocity v(m/sec), the experimental data on R obtained by other, investigators were pigeonholed into the form of $R=kSv^2$, where $k(kg{\cdot}sec^2/m^4)$ was the resistance coefficient and $S(m^2)$ the wall area of nets, and then k was analyzed by the resistance formular obtained in the previous paper. The analyzation produced the coefficient k expressed as $$k=4.5(\frac{S_n}{S_m})^{1.2}v^{-0.2}$$ in case of bottom trawl nets and as $$k=5.1\lambda^{-0.1}(\frac{S_n}{S_m})^{1.2}v^{-0.2}$$ in midwater trawl nets, where $S_m(m^2)$ was the cross-sectional area of net mouths, $S_n(m^2)$ the area of nets projected to the plane perpendicular to the water flow and $\lambda$ the representitive size of nettings given by ${\pi}d^2/2/sin2\varphi$ (d : twine diameter, 2l: mesh size, $2\varphi$ : angle between two adjacent bars). The value of $S_n/S_m$ could be calculated from the cone-shaped bag nets equal in S with the trawl nets. In the ordinary trawl nets generalized in the method of design, however, the flow resistance R (kg) could be expressed as $$R=1.5\;S\;v^{1.8}$$ in bottom trawl nets and $$R=0.7\;S\;v^{1.8}$$ in midwater trawl nets.

  • In this work, the merged PiN Schottky(MPS) diodes based silicon carbide(SiC) have been optimized and designed for 1200V diodes by 2D-atlas simulation tool. We investigated the optimized characteristics of SiC MPS diodes such as breakdown voltage and specific on-resistance by varying the doping concentrations of P-Grid/epi-layer and space of P-Grid, which are the most important parameters. The breakdown voltage and specific on-resistance, based on Baliga's Figure Of Merit (BFOM), have been compared with and the SiC-based MPS diodes show improved BFOMs with low values of specific on-resistance and high breakdown voltage. It has been demonstrated 1,200 V SiC MPS diodes will find useful applications in high voltage energy-efficient devices.

Effect of Microstructure on the Environmentally Induced Cracking Behavior of Al-Zn-Mg-Cu-Zr Aluminum Alloy

  • AA7010 is an Al-Zn-Mg-Cu alloy containing Zr, developed as an alternate to traditional AA7075 alloy owing to their high strength combined with better fracture toughness. It is necessary to improve the corrosion resistance and surface properties of the alloy by incorporating plasma electrolytic oxidation (PEO) method. AA7010-T7452 aluminum alloy has been processed through the forging route with multi-stage working operations, and was coated with $10{\mu}m$ thick $Al_2O_3$ ceramic aluminina coating using the plasma electrolytic oxidation (PEO) method. The corrosion, stress corrosion cracking (SCC) and nano-mechanical behaviours were examined by means of potentiodynamic polarization, slow strain rate test (SSRT) and nano-indentation tests. The results indicated that the additional thermomechanical treatment during the forging process caused a fully recrystallized microstructure, which lead to the poor environmental cracking resistance of the alloy in 3.5% NaCl solution, despite the overaging treatment. Although the fabricated PEO coating improved general corrosion resistance, the brittle nature of the coating did not provide any improvement in SCC resistance of the alloy. However, the hardness and elastic modulus of the coating were significantly higher than the base alloy.

Genomic Approaches for Understanding the Characteristics of Salmonella enterica subsp. enterica Serovar Typhimurium ST1120, Isolated from Swine Feces in Korea

  • Salmonella enterica subsp. enterica serovar Typhimurium, one of the most common foodborne pathogens, is transmitted mainly through contaminated food derived from infected animals. In this study, S. Typhimurium ST1120, an isolate from pig feces in Korea, was subjected to whole-genome analysis to understand its genomic features associated with virulence. The genome of ST1120 was found to have a circular chromosome of 4,855,001 bp (GC content 52.2%) and a plasmid of 6,863 bp (GC content 46.0%). This chromosome was predicted to have 4,558 open reading frames (ORFs), 17 pseudogenes, 22 rRNA genes, and 86 tRNA genes. Its plasmid was predicted to have three ORFs. Comparative genome analysis revealed that ST1120 was phylogenetically close to S. Typhimurium U288, a critical isolate in piggery farms and food chains in Europe. In silico functional analysis predicted that the ST1120 genome harbored multiple genes associated with virulence and stress resistance, including Salmonella pathogenicity islands (SPIs containing SPI-1 to SPI-5, SPI-13, and SPI-14), C63PI locus, ST104 prophage locus, and various antibiotic resistance genes. In accordance with these analysis results, ST1120 showed competence in invasion and survival abilities when it was added to host cells. It also exhibited robust resistance against antibiotics in comparison with other S. Typhimurium strains. This is the first report of the complete genome sequence of S. Typhimurium isolated from swine in Korea. Comparative genome analysis between ST1120 and other Salmonella strains would provide fruitful information toward understanding Salmonella host specificity and developing control measures against S. Typhimurium infection.
