Bacteriological Contamination of Water in Han River basin (한강유역의 수중미생물 오염도 조사)

  • In order to investigate the bacteriological contamination of water in Han river, the survey was carried out in eight reservoirs of Seoul water supply during the period from January to December in 1985. 1. The counts by means of total bacteria in eight reservoirs by standard plate count method were as follows: $7.7\times10^2$ per ml in Paldang reservior, $9.6\times10^3$ per ml in Gueiri, $8.4\times10^4$ per ml in Doogdo, $1.6\times10^6$ per ml in Bogwang, $2.5\times10^6$ per ml in Noryangjin, $2.2\times10^6$per ml in Seon yoo, $5.9\times10^6$ per ml in Yungdeungpo and $1.9\times10^7$per ml in Gayang. 2. The average counts of total coliform in eight reservoirs by MPN method were as follows : $2.4\times10$ per 100 ml in Paldang, $5.6\times10^2$ per 100 ml in Gueiri, $2.3\times10^3$ per 100 ml in Doogdo, $5.1\times10^4$ per 100 ml in Noryang-jin, $1.2\times10^5$ per 100 ml in Bogwang, $6.2\times10^4$ per 100 ml in Seonyoo, $1.1\times10^5$ per 100 ml in Yungdeungpo and $2.8\times10^5$ per 100 ml Gayang. 3. The counts by means of fecal coliform in eight reservoirs by MPN method were as follows : non detection per 100 ml in Paldang, 5.2 per 100 ml in Gueiri, $1.2\times10^2$ per 100 ml in Doogdo, $1.6\times10^3$ per 100ml in Bogwang, $2.0\times10^3$per 100ml in Noryangjin, $6.6\times10^2$ per 100ml in Seonyoo, $1.2\times10^3$ per 100 ml in Yungdeungpo and $2.5\times10^3$per 100 ml in Gayang. 4. The counts by means of fecal streptococci in eight reservoirs by MPN method were as follows: non detection per 100 ml in Paldang and Gueiri, $6.9\times10$ per 100 ml in Doogdo, $3.2\times10^2$ 102 per 100 ml in Bogwang, $2.9\times10^2$ per 100 ml in Noryangjin, $3.0\times10^2$ per 100 ml in Seonyoo, $4.0\times10^2$ per 100 ml in Yungdeungpo and $14\times10^3$ per 100 ml in Gayang. 5. The counts means of pseudomonas aeruginosa in eight reservoirs by MPN method were as follows; non detection per 100 ml in Paldang, 2.4 per 100 ml in Gueiri, $1.5\times10$ per 100 ml in Doogdo, $2.0\times10$ per 100ml in Bogwang,$6.2\times10$ per 100mI in Noryangjin, $2.1\times10$ per 100ml in Seonyoo, $6.4\times10$ per 100mI in Yungdeungpo and $7.1\times10$ per 100ml in Gaynag.

Circadian Clock Genes, PER1 and PER2, as Tumor Suppressors (체내 시계 유전자 PER1과 PER2의 종양억제자 기능)

  • Disruptive expression patterns of the circadian clock genes are highly associated with many human diseases, including cancer. Cell cycle and proliferation is linked to a circadian rhythm; therefore, abnormal clock gene expression could result in tumorigenesis and malignant development. The molecular network of the circadian clock is based on transcriptional and translational feedback loops orchestrated by a variety of clock activators and clock repressors. The expression of 10~15% of the genome is controlled by the overall balance of circadian oscillation. Among the many clock genes, Period 1 (Per1) and Period 2 (Per2) are clock repressor genes that play an important role in the regulation of normal physiological rhythms. It has been reported that PER1 and PER2 are involved in the expression of cell cycle regulators including cyclins, cyclin-dependent kinases (CDKs), and CDK inhibitors. In addition, correlation of the down-regulation of PER1 and PER2 with development of many cancer types has been revealed. In this review, we focused on the molecular function of PER1 and PER2 in the circadian clock network and the transcriptional and translational targets of PER1 and PER2 involved in cell cycle and tumorigenesis. Moreover, we provide information suggesting that PER1 and PER2 could be promising therapeutic targets for cancer therapies and serve as potential prognostic markers for certain types of human cancers.

Circadian Clock Gene Per1 Mediates BMP2-induced Osteoblast Differentiation in MC3T3-E1 Cells (MC3T3-E1 세포에서 BMP2에 의한 조골세포의 분화에 일주기 유전자 Per1이 미치는 영향)

  • Bone morphogenetic proteins (BMPs) are multifunctional cytokines that play important roles in a variety of cellular functions. Among BMP family members, BMP2 efficiently promotes osteoblast differentiation through Smad-mediated runt-related transcription factor 2 (Runx2) expression. Several recent studies suggest that BMPs are associated with clock genes, in particular Bmal1. Bmal1 protein heterodimerizes with Clock protein and then induces period 1 (Per1) expression. However, the role of Per1 on osteoblast differentiation remains unclear. In this study, we investigated whether Per1 is involved in osteoblast differentiation. MC3T3-E1 cells were treated with BMP2 for induction of osteoblastic differentiation. Osteogenic maker gene and Per1 mRNA expression were measured using real-time PCR. Interestingly, BMP2 treatment induced Per1 mRNA expression in MC3T3-E1 cells. To further investigate the function of Per1 on osteoblast differentiation, MC3T3-E1 cells were transiently transfected with pCMV-Per1. Per1 overexpression increased Runx2 mRNA and protein levels. Also, mRNA expression and promoter activity of osteocalcin were upregulated by Per1 overexpression. To investigate the effect of interaction between Per1 and osteogenic condition, MC3T3-E1 cells were cultured in osteogenic medium containing ascorbic acid and ${\beta}$-glycerophosphate. Osteogenic medium-induced ALP staining level and mineralization were synergistically increased by overexpression of Per1. Taken together, these results demonstrate that Per1 is a positive regulator of osteoblast differentiation.

'Survey on Bacteriological Contamination of Moving Tavern in Seoul Area' ('노상주점의 위생상태에 관한 미생물학적 조사')

  • This sanitary survey was carried out to investigate the bacteriological contamination of cooking utensils and foods of moving tavern in eight sample sites of Seoul area. The results of survey were as follows: 1. The counts by means of total bacteria in cooking utensils and food samples by standard plate count method were as follow: $5.6\times10^5$ per gm in dishcloth, $3.1\times10^6$ per ml in dishwater. In food samples, $5.4\times10^5$ per gm in meat was higher than other samples. 2. The average counts total coliform and fecal coliform in samples by MPN method were as follow: $3.4\times10^4$ MPN per 100ml, and $1.3\times10^2$ MPN per 100ml in chopping board, $6.1\times10^4$MPN per gm and $1.0\times10^2$ MPN per gm in dishcloth, $1.8\times10^5$ MPN per 100ml and $6.1\times10^2$ MPN per 100ml in dishwater. In food samples, $3.1\times10^4$MPN per gm and $2.0\times10^2$ MPN per gm in meat was higher than other samples. 3. The counts by means of Pseudomonas in samples by MPN method were as follow: $2.8\times10^3$ MPN per 100ml in chopping board, $4.7\times10^3$ MPN per gm in dishcloth $5.6\times10^3$ MPN per 100ml in dishwater. In food samples, $2.4\times10^3$ MPN per gm in shellfish was higher than other samples. 4. Isolation cases of Food poisoning organisms from samples were as follow: Staphylococci was detected 9 cases $(17.6\%)$ in chopping board, 7 cases $(13.6\%)$ in dishcloth. In food samples, 9 cases $(25.7\%)$ in meat, 1 case $(4\%)$ in fish samples. Salmonella was detected 2 cases $(3.9\%)$ in dishwater, 1 case in meat samples.

Effect of mPER1 on the Expression of HSP105 Gene in the Mouse SCN

  • The suprachiasmatic nucleus (SCN) of the anterior hypothalamus is the circadian pacemaker entrained to the 24-hr day by environmental time cues. Major circadian genes such as mPeriod ($mPer1{\sim}3$) and mCryptochrome ($mCry1{\sim}2$) are actively transcribed by the action of CLOCK/BMAL heterodimers, and in turn, these are being suppressed by the mPER/mCRY complex. In the study, the locomotor activity rhythms of mPer1 Knockout (KO) mice are measured, and the expression profiles of Heat Shock Protein 105kDa (HSP 105) genes in the SCN were measured by in situ hybridization. In agreement with previous reports, the locomotor activity rhythm of mPer1 KO mice was much shorter than that of wildtype. In addition, the total bout of activity of mPer1 KO was less in comparison to control mice. The expression of HSP 105 in the SCN of mPer1 KO mice was ranged from CT6 to CT22, with a peak level at CT14, implying that the gene are under the control of circadian clock. However, the expression of HSP 105 in the SCN of wildtype could not be detected in our study. Further analysis will reveal the direct or indirect regulation by mPer1 on the expression in the SCN and the role of the gene in the circadian clock.

A Follow-up Survey on the Socio-medical Status of the Drop-out Cases of Tuberculosis in Jeonju Health Center (보건소등록결핵환자(保健所登錄結核患者)에 대(對)한 사회의학적(社會醫學的) 조사연구(調査硏究))

  • A socio-medical survey was carried out on 1,108 cases of pulmonary tuberculosis who had registered and 220 cases unregistered at Jeonju health center in 1973, during from June 1 to July 31, 1974. As the results of this survey, the following conclusions were obtained. 1. Of the total 1,108 cases of pulmonary tuberculosis there were 708 new cases, while remaining 400 were old cases and rate of registration was 4.0 per thousand person. 2. The highest rate of registration of the newly diagnosed pulmonary tuberculosis could be found in bacteriological examination while 54.4 per cent in X-ray examination. 3. As for the educational status of the tuberculosis patients, primary school graduates contituted the greatest proportion or 64.6 per cent, middle school graduates 13.6 per cent, high school graduates 6.8 per cent and collge graduates only 0.7 per cent. 4. By age group of the cases, at age of 20 to 29 years occupied 23.6 per cent (262 out of 1,108 cases), 17.9 per cent at age of 40 to 49 years. 5. The greatest proportion or 38.4 per cent of the cases had no occupation. 6. The living standard of the patient's household, low class constituted 60.6 per cent of the total households. 7. By distribution of residential area, farming area was 5.0 per thousand person, 4.0 in downtwon and suburban areas. 8. The greatest proportion or 70.0 per cent of positive cases in X-ray examination was unregistered in August, 63.6 per cent in July 1973. 9. 220 out of 1,108 cases(19.9%) were unregistered pulmonary tuberculosis in X-ray and bacteriological examinations. 10. For age groups of unregistered caes, most prevalent age group was 30 to 39. 11. Regarding on the cases of unregistration, 'indifference for disease' occupied highest rate with 31.9 percent' and 'private secret' with 15.7 per cent. 12. Of the total 457 cases drop-outed, there were 78 complete heald cases while remaining 207 inactive.

Effects of Nitrogen Level and Seedling Number on Panicle Structure in Japonica Rice

  • Four different rice varieties, Sindongjinbyeo, Dongjin #1, Saegyehwabyeo, and Iksan 467, were transplanted under three different nitrogen levels and two different seedling numbers per hill to obtain basic information on panicle traits under different cultural conditions and to propose the ideal panicle structure in Japonica rice. Sindongjinbyeo and Iksan 467 were characterized by more primary rachis branches (PRBs) per panicle and more grains on PRB than other cultivars. The two varieties also had fewer secondary rachis branches (SRBs) per PRB and fewer grains on SRB per PRB. These characteristics, consequently, resulted in higher ripened grain rate, contrary to that of Dongjin #1 and Saegyehwabyeo. In the correlation coefficient analysis, PRB number per panicle and grain number on PRB per panicle were positively correlated with ripened grain rate, while SRB number per panicle, number of grains on SRB per panicle, SRB number per PRB, number of grains on SRB per PRB and grain number per panicle were negatively correlated with ripened grain rate. Therefore, the number of grains on PRB per panicle, SRB number per PRB and the number of grains on SRB per PRB were the appropriate criteria for determining and achieving higher ripened grain rate in rice. High ripened grain rate over 90% was obtainable with over 12.5 PRBs per panicle and 63 grains on PRB per panicle, and with under 1.7 SRBs per PRB, 5 grains on SRB per PRB, 130 grains per panicle, and 14 panicles per hill. The study recommended that for over 90% high ripened grain rate, the critical limiting factors should be under 2 SRBs per PRB, 6 grains per PRB, and 130 grains per panicle, irrespective of the PRB number per panicle and the number of grains on PRB.

Incidence of Nonfatal Unintentional Injuries among Students, and Compensation Payment for Five Years (2000~2004) in School, Seoul (서울시 초.중.고등학교에서 5년간(2000~2004년) 발생한 학생의 비의도적 손상 및 보상급여비 현황)

  • Purpose: The purpose of this study was to identify incidence of nonfatal unintentional injuries (accidents) among students, and to investigate compensation payment for five years (2000~2004) in schools located in Seoul. Methods: Subjects were 14,783 students injured among elementary, middle and high school students. The accumulated data for 5 years (2000~2004) was from the Seoul School Safety and Insurance Association. Chi-square, ttest, logistic regression, and MANCOVA were conducted. Results: The most frequent accidents occurred among males (73.2%), and in special school students (3.86 per 1,000) followed by middle school students (3.05 per 1,000), in May (0.25 per 1,000) followed by June and October, in physical education classes (0.73 per 1,000) followed by special activities (0.40 per 1,000), recess (0.31 per 1,000) and lunch (0.29 per 1,000). Fractures (0.86 per 1,000) were followed by mild injuries (0.39 per 1,000), joint and ligament injuries (0.31 per 1,000), and teeth injuries (0.26 per 1,000). After adjusting for potential confounding variables, the highest means of final compensation payment was for burns (810 thousand KRW) followed by the teeth injuries (506 thousand KRW), cleaning injuries (550 thousand KRW) followed by injuries incurred during special activities (510 thousand KRW) and injuries incurred at special schools (556 thousand KRW). Conclusion: In school, the highest incidences of nonfatal unintentional injuries were in special schools, among males, fractures and physical education. However, the highest compensation payments were for burns, and injuries taking place at special schools and during cleaning hour. Proper health education including teaching healthy habits for safety/injury prevention is needed to prevent injuries and decrease compensation.

Medical Care Expenditure and Its Determinants in Rural Areas (농촌주민(農村住民)의 의료비지출(醫療費支出)에 관(關)한 연구(硏究))

  • This study was conducted for the purpose of obtaining basic information on the patterns of medical care expenditures, and identifying some determinants of medical care expenditures in the rural Korea. Nine guns were chosen from the eight provinces, excluding Cheju island. One gun in each province and two villages were selected from the each myon or ub within the selected guns. The total number of households was 1,789 and the sample size was 9,826 non-institutionalized people. Followings are the major findings of the study : 1) Medical care expenditures increase proportionally with age in terms of cost per patient, per episode of illness, per treated case, and per person. Averagely, it cost 2,756 won per patient, 2,614 won per spell of illness, 4,361 won per treated case, and 413 won per person. 2) Medical care expenditures increase proportionally with educational level of patients. College graduates spent the most, 4,726 won per patient, 5,987 won per treated case, and 670 won per person. 3) The male spent a little more than the female in terms of per patient, per episode, and per person. For example, a male spent 23 won more than a female. 4) Those who were suffering from illnesses longer than 1 year spent three times more than that had illnesses of less than 1 year duration. 5) The simple correlation coefficient between activity restriction and medical care expenditures was the highest among others, 0.491. The next was 0.294 between duration of illness and medical care expenditures. 6) Attempts are made to identify the explanatory variables in medical care expenditures. Thirty one per cent of the variances in tile expenditures can be accounted for by the selected 15 predictors. Those predictors belonged to clinical renditions, such as activity restriction, duration of illness, and nature of conditions, are proved to be the most potent independent variabes. Level of education and monthly family income are also significant in terms of beta coefficient. Further studies are called for to unreveal the determinants of medical expenditures.

Heritability and Correlation Coefficients in Edible Oil Crops in Korea (우리나라 식용유지 자원식물의 상관 및 유전력)

  • This experiment were carried out to study the heritability, genotypic, pheno-typic and environmental correlation in four edible oil crops. Heritabilities of flowering date, maturing date, plant height and weight of 1, 000 grains in rape were high. A highly significant positive correlation was found between flowering date and maturing date, plant hight and ear length, number of pods per ear and flowering date, maturing date and plant hight, The number of seed per pod showed also significant positive correlation with flowering or maturing date as well as with the relationship between weight of 1, 000 grains and plant height or ear length inrapeseed, respectively. Heritabilities of maturing date, length of stem with eapsule and number of seed per capsule were high, in sesame. Genotypic correlation between plant height and length of stem with capsule, length of stem with capsule and number of capsule per plant, number of capsule per plant and weight of 1, 000 grains, weight of 1, 000 grains and yield were highy positive in sesame. Heritabilities of flowering date, length of main stem, weight of 100 grains and number of shells per square meter in peanut were high, There was positive genotypic correlation between length of main stem and yield, number of shells per quare meter and matured seed ratio, number of shells per square meter and yield, 100 grains weight and yield. On the other hand, a significant negative correlation appeared between flowering date and yield. Heritabilities of days to flowering, stem length, stem diameter and weight of 1, 000 grains in perila were high. There was positive genotypic correlation between stem length and stem diameter, number of pods per plant, stem weight etc, between number of internodes and number of pods per plant, stem weight, number of valid branches and number of pods plant as well, respectively. While, a significant negative correlation was observed between number of valid branches and weight of 1, 000 grains, between number of pods per plant and weight of 1, 000 grains.
