PLGA and PCL copolymers initiated by carbitol as drug carriers were synthesized by ring-opening polymerization of L-lactide (LA), glycolide (GA), and $\varepsilon-caprolactone(\varepsilon-CL)$. Implantable wafers were simply fabricated by direct compression method after physical mixing of copolymers and bovine serum albumin-fluorescein isothiocyanate (BSA-FITC) as a model protein drug. The release amounts of BSA-FITC from wafers were determined by fluorescence intensity using the fluorescence spectrophotometer. Also, the release behavior of BSA-FITC on wafers was controlled by adding the additives such as collagen, small intestinal submucosa (SIS), poly(vinyl pyrrolidone) (PVP), and poly(thylene glycol) (PEG). The wafer prepared by PLGA and PCL exhibited slow release within $10\%$ for 30 days. But, those prepared by a variety of additives exhibited the controlled BSA release patterns with a dependence on the additive contents. furthermore, the wafers containing natural materials such as collagen and SIS showed more zero-order release profile than that with synthetic materials such as PVP and PEG. It was confirmed that the release of BSA from implantable wafers could be easily controlled by adding natural additives.

Hydrophilic reactive hot-melt polyurethane adhesive(HRHA) using a hydrophilic polyol having different molecular weight and NCO index was synthesized. This HRHA was synthesized using Polyethylene glycol(PEG) as a hydrophilic polyol, Polypropylene glycol(PPG) and Polycaprolactone diol(PCL) as hydrophobic polyols, and Methylene diphenyl diisocyanate(MDI) as an isocyanate. The changes in IR spectrum, viscosity and thermal properties of HRHA with different PEG molecular weights and NCO index were investigated, and the tensile strength and elongation of the HRHA casting film and the peel strength, moisture permeability and water pressure of the HRHA coated fabric were confirmed. In this experiment, as the molecular weight of PEG and NCO index increased, the adhesive strength, tensile strength, elongation and moisture permeability was increased but viscosity and Tg was decreased.
Thermosensitive nanoparticles were prepared via the self-assembly of two different $poly({\varepsilon}-caprolactone)$-based block copolymers of poly(N-isopropylacrylamide)-b-$poly({\varepsilon}-caprolactone)$ (PNPCL) and poly(ethylene glycol)-b-$poly({\varepsilon}-caprolactone)$ (PEGCL). The self-aggregation and thermosensitive behaviors of the mixed nanoparticles were investigated using $^1H-NMR$, turbidimetry, differential scanning microcalorimetry (micro-DSC), dynamic light scattering (DLS), and fluorescence spectroscopy. The copolymer mixtures (mixed nanoparticles, M1-M5, with different PNPCL content) formed nano-sized self-aggregates in an aqueous environment via the intra- and/or intermolecular association of hydrophobic PCL chains. The microscopic investigation of the mixed nanoparticles showed that the critical aggregation concentration (cac), the partition equilibrium constants $(K_v)$ of pyrene, and the aggregation number of PCL chains per one hydrophobic microdomain varied in accordance with the compositions of the mixed nanoparticles. Furthermore, the PNPCL harboring mixed nanoparticles evidenced phase transition behavior, originated by coil to the globule transition of PNiPAAm block upon heating, thereby resulting in the turbidity change, endothermic heat exchange, and particle size reduction upon heating. The drug release tests showed that the formation of the thermosensitive hydrogel layer enhanced the sustained drug release patterns by functioning as an additional diffusion barrier.

Polymeric nanoparticles are recognized as promising drug carriers [1]. Here, novel tri-block copolymers based on poly PPDO, PCL and PEG were synthesized and employed for the formulation of reproducible polymeric nanoparticles [2]. To estimate the feasibility of the polymer to form polymeric nanoparticles, nanoparticles were prepared by co-solvent evaporation technique. Polymerization and structural features of the polymer were analyzed by different physico-chemical techniques. Existence of hydrophobic domains as a core of nanoparticles was characterized by $^{1}H-NMR$ spectroscopy, and further confirmed by fluorescence technique using pyrene as probe.

Background: Development of a nanosized polymeric delivery system for erlotinib was the main objective of this research. Materials and Methods: Poly caprolactone-polyethylene glycol-polycaprolactone (PCEC) copolymers with different compositions were synthesized via ring opening polymerization. Formation of triblock copolymers was confirmed by HNMR as well as FT-IR. Erlotinib loaded nanoparticles were prepared by means of synthesized copolymers with solvent displacement method. Results: Physicochemical properties of obtained polymeric nanoparticles were dependent on composition of used copolymers. Size of particles was decreased with decreasing the PCL/PEG molar ratio in used copolymers. Encapsulation efficiency of prepared formulations was declined by decreasing their particle size. Drug release behavior from the prepared nanoparticles exhibited a sustained pattern without a burst release. From the release profiles, it can be found that erlotinib release rate from polymeric nanoparticles is decreased by increase of CL/PEG molar ratio of prepared block copolymers. Based on MTT assay results, cell growth inhibition of erlotinib has a dose and time dependent pattern. After 72 hours of exposure, the 50% inhibitory concentration (IC50) of erlotinib hydrochloride was appeared to be $14.8{\mu}M$. Conclusions: From the obtained results, it can be concluded that the prepared PCEC nanoparticles in this study might have the potential to be considered as delivery system for erlotinib.

This study reports water-resistant oil-in-water (O/W) emulsion-based sunscreening formulations prepared using a poly(ethylene glycol)-poly(${\varepsilon}$-caprolactone)-poly(ethylene glycol) (PEG-PCL-PEG) triblock polymeric surfactant. As a result of a variety of outdoor recreational activities such as swimming and hiking, consumer needs for development of advanced water-resistant sunscreen formulations are increasing. Water-resistant sunscreens are mostly based on water-in-oil (W/O) emulsions, because they should not be wiped off by water or sweat. However, the W/O emulsion formulations have a disadvantage in that the feeling of use is oily and difficult to remove. On the other hand, the O/W emulsion formulations are excellent in achieving the better skin feel as well as the easier removal. However, it is difficult to provide the O/W emulsion formulations with the water-repelling performance, since re-emulsification likely occurs upon getting touch with water. To solve this problem, this study proposes a O/W emulsion-based sunscreen formulation, a triblock polymeric surfactant having relatively high interfacial tension HLB value (~ 10). This allows the sunscreen formulations to exhibit the improved water repellence function by preventing their re-emulsification. The sunscreen formation system prepared in this study would be useful for diversification of functional sunscreen products, taking advantages of its excellent emulsion stability, UV protection performance, long lasting water-resistant function and selective cleansing effect with only foam cleanser.

Curcumin (Cum) has been reported to have potential chemo-preventive and chemotherapeutic activity through influencing various processes, inducing cell cycle arrest, differentiation and apoptosis in a series of cancers. However, the poor solubility of Cum limits its further applications in the treatment of cancer. We have previously reported Cum-loaded nanoparticles (Cum-NPs) prepared with amphilic methoxy poly(ethylene glycol)-polycaprolactone (mPEG-PCL) block copolymers. The current study demonstrated superior antitumor efficacy of Cum-NPs over free Cum in the treatment of lung cancer. In vivo evaluation further demonstrated superior anticancer effects of Cum-NPs by delaying tumor growth compared to free Cum in an established A549 transplanted mice model. Moreover, Cum-NPs showed little toxicity to normal tissues including bone marrow, liver and kidney at a therapeutic dose. These results suggest that Cum-NPs are effective to inhibit the growth of human lung cancer with little toxicity to normal tissues, and could provide a clinically useful therapeutic regimen. They thus merit more research to evaluate the feasibility of clinical application.

BACKGROUND: The major challenge of tissue engineering is to develop constructions with suitable properties which would mimic the natural extracellular matrix to induce the proliferation and differentiation of cells. Poly(${\varepsilon}$-caprolactone)-poly(ethylene glycol)-poly(${\varepsilon}$-caprolactone) (PCL-PEG-PCL, PCEC), chitosan (CS), nano-silica ($n-SiO_2$) and nano-hydroxyapatite (n-HA) are biomaterials successfully applied for the preparation of 3D structures appropriate for tissue engineering. METHODS: We evaluated the effect of n-HA and $n-SiO_2$ incorporated PCEC-CS nanofibers on physical properties and osteogenic differentiation of human dental pulp stem cells (hDPSCs). Fourier transform infrared spectroscopy, field emission scanning electron microscope, transmission electron microscope, thermogravimetric analysis, contact angle and mechanical test were applied to evaluate the physicochemical properties of nanofibers. Cell adhesion and proliferation of hDPSCs and their osteoblastic differentiation on nanofibers were assessed using MTT assay, DAPI staining, alizarin red S staining, and QRT-PCR assay. RESULTS: All the samples demonstrated bead-less morphologies with an average diameter in the range of 190-260 nm. The mechanical test studies showed that scaffolds incorporated with n-HA had a higher tensile strength than ones incorporated with $n-SiO_2$. While the hydrophilicity of $n-SiO_2$ incorporated PCEC-CS nanofibers was higher than that of samples enriched with n-HA. Cell adhesion and proliferation studies showed that n-HA incorporated nanofibers were slightly superior to $n-SiO_2$ incorporated ones. Alizarin red S staining and QRT-PCR analysis confirmed the osteogenic differentiation of hDPSCs on PCEC-CS nanofibers incorporated with n-HA and $n-SiO_2$. CONCLUSION: Compared to other groups, PCEC-CS nanofibers incorporated with 15 wt% n-HA were able to support more cell adhesion and differentiation, thus are better candidates for bone tissue engineering applications.

This study was conducted to investigate physiological activity and its skin permeability of Petroselinum crispum extract using polymer micelles and cell penetrating peptide. In the antioxidant test, the total concentrations of polyphenol compounds were determined to be $121.68{\pm}2.49mg/g$ (for ethanol extract and), $72.42{\pm}1.52mg/g$ (for hydrothermal extract.). The DPPH radical scavenging ability was $90.48{\pm}0.46%$ (for ethanol extract) and $83.92{\pm}0.13%$ (for hydrothermal extract) at 2000 mg/L. ABTS radical scavenging ability was $91.08{\pm}0.14%$ for ethanol extract ethanol extract, which is higher than that of hydrothermal extract at 800 mg/L ($69.63{\pm}0.55%$). In the SOD experiments, the P. crispum ethanol extract showed higher SOD activity than that of the P. crispum hydrothermal extract at all concentrations.. At a concentration of 16,000 mg/L, P. crispum ethanol extract showed the highest SOD activity of $128.45{\pm}0.70%$. The elastase inhibitory assay also showed concentration dependence and elastase inhibition of P. crispum ethanol extract was $99.99{\pm}1.54%$, which was the highest at 2,000 mg/L. To solve the problem of insolubility and to improve skin permeability of the extract, PCL-PEG polymer micelle containing P. crispum ethanol extracts and 1% cell permeable peptide, hexa-D-arginine (R6) were successfully prepared with a particle size of 40.10 nm. In the results of 24 hours of skin permeation experiment, total accumulated beta-carotene amounts showed $37.99{\mu}g/cm^2$ in Petroselinum crispum extracts and $68.38{\mu}g/cm^2$ (1.8 times) in P. crispum extract of the particles.

This study was conducted to evaluate physiological activity of Robinia pseudo-acacia leaf and its skin penetration using polymer micelles and skin penetrating peptide. After extraction with Robinia pseudo-acacia using the ethanol and distilled water, various physiological activities were examined. The total concentration of polyphenol compounds was determined to be 47.42 mg/g (ethanol extract), 56.88 mg/g (hydrothermal extract) and DPPH radical scavenging ability at $1,000{\mu}g/mL$ was 44.24% in ethanol extract and it is higher than value(41.50%) in hydrothermal extract. The elastase inhibitory assay showed concentration dependence and elastase inhibition of Robinia pseudo acacia leaf ethanol extract was 54.09%, which was the highest at $500{\mu}g/mL$. In the SOD-like experiments, the concentration-dependent results were showed and the SOD-like activity of the Robinia pseudo-acacia leaf ethanol extract was higher than that of the Robinia pseudo acacia leaf hydrothermal extract at all concentrations. At a concentration of $500{\mu}g/mL$, Robinia pseudo acacia leaf ethanol extract showed the highest SOD-like activity of 76.41%. The tyrosinase inhibition at $20{\mu}g/mL$ was determined to be 56.47% (ethanol extract), 23.05% (hydrothermal extract). In the antimicrobial experiments, the hydrothermal extract had no effect, but ethanol extract represented maximum clear zone of 11.00 mm in Propionbacterium acnes strain and maximum clear zone of 10.50 mm. in Bacillus subtilis strain. To solve the problem of insolubility and to improve skin penetration, PCL-PEG polymer micelles containing Robinia pseudo-acacia leaf ethanol extracts and 1.0% cell permeable peptide, hexa-D-arginine (R6) were successfully prepared with particle size of 108.23 and 126.47 nm and excellent skin permeation effects could be showed.
