  • The quantitative effects of molecular weight and concentrations of two phase-forming polymers-polyethylene glycol and crude dextran on the two phase extractive ethanol fermentation were investigated using a Box-Wilson central composite protocol. The regression model obtained was used in order to determine optimum compositions of aqueous two phase system. In the aqueous two phase extractive ethanol fermentation of Kluyueromyces fragilis CBS 1555 with Jerusalem artichoke juice, it was found from the regression model that the variables influenlcing on ethanol fermentation were PEG concentration, time, Dx concentration, and PEG molecular weight strongly in order. The interaction of PEG concentration and PEG molecular weight was also found, and the effect of PEG concentration decreased with increase in molecular weight of PEG. The ethanol concentration incresed with increase in molecular weight of PEG, and with decrease in concentration of PEG. In conolusion, maximum concentration of ethanol produced was obtained at the following compositions; PEG MW 20000, Dx concentration ranged from 4% to 5%, and PEG concentration ranged from 3% to 7%.

Attenuated Cerebral Ischemic Injury by Polyethylene Glycol-Conjugated Hemoglobin

  • Polyethylene glycol-conjugated hemoglobin (PEG-Hb) has been proposed as a blood substitute for transfusion due to their plasma expansion and oxygen transport capabilities. The protective effect of PEG-Hb on cerebral hypoxic-ischemic injury was investigated in neonatal hypoxia model and adult rat focal cerebral ischemia model. As intravenously administered 30 min before the onset of hypoxia, PEG-Hb markedly protected cerebral hypoxic injury in a neonatal rat hypoxia model. A similar treatment of PEG-Hb largely reduced the ischemic injury ensuing after 2-h middle cerebral artery occlusion followed by 22-h reperfusion. Consistently, neurological disorder was significantly improved by PEG-Hb. The results indicate that the pharmacological blockade of cerebral ischemic injury by using PEG-Hb may provide a useful strategy for the treatment of cerebral stroke.

Phosphoprotein Partitioning in Metal-Affinity Aqueous Two-Phase Systems and Prediction of Partitioning Behavior (금속 친화성 액 이상분계 시스템에서 Phsphoprotein분배 및 분배예측)

  • A mathematical model has been derived and used to describe phosphoprotein partitioning in Fe(III) IDA-PEG/dextran two-phase systems. This model includes the inhibitory effects of hydrogen and hydroxyl ion concentrations on protein partitioning. For aqueous two-phase partitioning experiments, the Al and A2 subcomponents of ovalbumin carrying two and one surface phosphoryl group(s) were purified using an immobilized metal ion affinity chromatography (IMAC). The ratio of partition coefficients in the presence and absence of Fe(III)IDA-PEG, K/Ko, increased in the pH range of 3.0 to 5.0 due to deprotonation of the second oxygen of the phosphoryl group, and above pH 5.0 declined steeply by the inhibitory binding of hydroxyl ions to the metal ion. This partitioning behavior was well described by the mathematical model. The binding constants for formation of the complex between the phosphoryl group and the Fe(III)IDA-PEG were found to be $6.1{\times}10^3M^{-1} and 2.3{\times}10^4M^{-1}$ in the top and bottom phases, respectively. These values are 3-5 times those for interaction of Cu(II)IDA-PEG with a single surface-accessible histidine.

  • Mungbean trypsin inhibitor (MBTI) was isolated and purified from Mung bean which has been used as a galenic and traditional food. MBTI and poly(ethylene glycol) were conjugated by using water soluble carbodiimide. We evaluated the therapeutic value of the MBTI and MBTI-PEG conjugate using animal models, sublethal septic shock model in guinea pig caused by pseudomonal elastase, shock model in rat caused by lipopolysaccharide, and the vascular permeability test by using pseudomonal elastase. In two shock model in guinea p Is and in rat, hypotesion shock was inhibited by pretreatment of MBTI. And also the vascular permeability caused by pseudomonal elastase reduced by pretreatment of MBTI. Also, therapeutic value of the MBTI-PEG conjugate was evaluated by using the sublethal septic shock model caused by pseudomonal elastase. The MBTI-PEG conjugate was more effective than native MBTI against pseudomonal elastase induced septic shock in guinea pig model.

A Study of Dewatering Phenomena of Potato Slice Cytorrhysed by High Molecules (고분자 용액의 세포 압착 현상에 의한 감자 절편의 탈수 현상에 관한 연구)

  • To study simultaneous water and solute transport kinetics during soaking in concentrated solution, the influence of the concentration and molecular weight of the solute(polyethylene glycol(PEG) and NaCl) in the soaking solution and the temperature on the water loss and solute gain rates were observed by using a model vegetable tissue(potato). When potato slices$(4cm{\times}4cm{\times}0.1cm)$ soaked in 60% PEG solutions, the water loss rate of the early phase decreased with increasing of the molecular weight of PEG from 200 to 6,000, while the final water loss increased with increasing the molecular weight of PEG and it reached to 80%. The cell wall of potato tissue was permeable to NaCl and PEGs of which average molecular weight is smaller than 400 but it was not permeable to PEG 600 and larger molecules. PEG which has average molecular weight below 600 induced plasmolysis and those above 600 induced cytorrhysis. The water loss rate of potato sample soaked in smaller molecular weight PEG solution was faster than those soaked in higher molecular weight PEG solution before cytorrhysis happened. The water loss rate was reversed after cytorrhysis happened. The volume change of potato within the first 60 minutes was larger in low molecular PEG solution but the final ratio of decreasing volume was larger in high molecular PEG solutions. In PEG 200 solution, the potato tissue was slightly shrinked without shape change. However, in PEG 4,000 solution, volume of potato was reduced significantly and potato tissue was twisted.

Preparation and Characterization of PEG-PLA(PLGA) Micelles for Solubilization of Pioglitazone (Pioglitazone 가용화를 위한 PEG-PLA(PLGA) 고분자 미셀의 제조 및 특성분석)

  • We synthesized PEG-PLA (or PLGA) amphiphilic di-block copolymers, which consist of PEG as biocompatible and hydrophilic block and PLA (or PLGA) as biodegradable and hydrophobic block, by ring opening polymerization of LA in the presence of methoxy PEG as a macroinitiator. The compositions and the molecular weights of the copolymers were controlled by changing the feed ratio of LA (and GA) to PEG initiator. The di-block copolymers could self-assemble in aqueous media to form micellar structure. A hydrophobic model drug, pioglitazone, was loaded into the polymer micelle using solid dispersion and dialysis methods, and the drug-loaded micelles were characterized by AFM, DLS and HPLC measurements. The drug loading capacity and in vitro release studies were performed and evaluated under various conditions. These results indicated that the amphiphilic di-block copolymers of PEG-PLA (or PLGA) could solubilize pioglitazone by solid dispersion method and the drug release was modulated according to micellar chemical compositions.

Effects of poly (ethylene glycol-propylene glycol) copolymer on hemostasis and osteogenesis in a rat calvarial defect model

  • This study aimed to evaluate the effects of a bioabsorbable bone hemostatic agent comprising poly (ethylene glycol-propylene glycol) copolymers (PEG-PPG) on hemostasis and osteogenesis. Bilateral 3 mm diameter calvarial defects were created in 99 male Sprague-Dawley rats. The defects were filled with PEG-PPG or bone wax. The defects of control group were left unfilled. Virtual autopsy was performed to evaluate bioabsorption. The calvaria were subjected to x-ray microtomography (microCT) and histological examination. Bone volume fraction (BV/TV) and bone mineral density (BMD) were measured using microCT; furthermore, white blood cell count and histological examination were performed. After application of PEG-PPG and bone wax, immediate hemostasis was achieved. Autopsy revealed that PEG-PPG disappeared within 48 h at the application site; in contrast, bone wax remained until 12 weeks. The PEG-PPG and control groups showed significantly more osteogenesis than the bone wax group with respect to BV/TV and BMD at 3, 6, and 12 weeks (p < 0.05). Histology revealed that the bone wax group exhibited little bone formation with inflammation. In contrast, PEG-PPG and control groups showed significantly more qualitative osteogenesis than the bone wax group (p < 0.01). In conclusion, PEG-PPG showed immediate hemostasis and was absorbed to allow progressive osteogenesis.

Rheology of flocculated kaolinite dispersions

  • Rheological characterisation of flocculated kaolinite pulps has been undertaken to elucidate particle interactions underpinning the dewatering behaviour induced by flocculation with polyethylene oxide (PEO), anionic polyacrylamide (PAM A) and their blends. Shear yield stress $(\tau_y)$ analysis indicated that polymer mediated particle interactions were markedly amplified upon shear of PEG based pulps. In contrast, PAM A based pulps showed a significant decrease in yield values upon shear. Steady stress measurements analysed using a modified Ellis model indicated subtle differences between the respective linear viscoelastic plateaus of the pulps. Furthermore, modified shear thinning behaviour was evident in PEG based pulps. Estimation of elastic and viscous moduli (G', G') was made using dynamic stress analysis for comparison with values determined from vane measurements. Despite a noticeable difference in the magnitude of G' between the two methods, similar trends indicating sheared PEG-based pulps to be more elastic than PAM based pulps, were observed. Floc microstructural observations obtained in support of rheological properties indicate that PEG flocculant induces significantly more compact particle aggregation within the clay pulps under shear consistent with the yield stress data, in contrast to PAM A, or indeed unsheared PEG based pulps. Consequentially, sheared PEG based pulps show significantly improved dewatering behaviour. The implications of the results, potential benefits and drawbacks of flocculation with PEG and PAM A are discussed with respect to improvements in current dewatering processes used in the minerals industry.

Effect of Methoxy PEG-45 Thioctate (LA-PEG) against Oxidative Protein Damage and Anti-glycation (Methoxy PEG-45 Thioctate (LA-PEG)의 항노화 효과에 대한 연구)

  • Aging is a physiological and irreversible, progressive process involving changes in the ability to maintain cellular functionality. It affects tissues, organs and the whole organism and thus finally cause to death. Oxidative stress has been postulated to contribute significantly to the accelerated accumulation of advanced glycation endproducts (AGEs) in collagen, which is implicated in the process of skin aging. In the present study, glycation inhibitory activity of methoxy PEG-45 thioctate (LA-PEG), and its inhibitory effect of cellular oxidation and senescence was investigated. Treatment of LA-PEG significantly showed lower fluorescent intensity induced by AGEs. In addition, LA-PEG was significantly reduced the formation of ROS induced by AGEs. High antioxidant and anti-glycation activities of LA-PEG in glycated collagen model indicated its contribution to anti-aging process. Cellular senescence leads to an increase in senescence-associated ${\beta}$-galactosidase ($SA-{\beta}-gal$) activity, which can be used as a biomarker to identify senescent cells. Treatment with LA-PEG showed a dose-dependent, statistically significant decreased in $SA-{\beta}-gal$ indicating reduced senescence. These results suggest that LA-PEG may have potent anti-aging effects and can be used as new functional materials against cellular accumulation of AGEs.
