  • Hollow type suppositories inserted polyvinyl alcohol (PVA) hydrogel capsule containing propranolol·HCI (PPH) were prepared using different bases, polyethylene glycol (PEG), Witepsol H-15 (WH-15) and Witepsol W-35 (WW-35) to improve the controlled release of PPH. The release of PPH from the hollow type suppository inserted PVA hydrogel capsule was retarded than that from PEG, WH-15, or WW-35 hollow type suppositories in rat rectal cavity. When the suppositories were administered to rats, the controlled release of PPH was proved by the plasma concentration-time-profiles of PPH. No significant difference (p〈0.05) among the three different hollow type suppositories was observed in terms of AUC and MRT of PPH. WH-15 hollow type suppository inserted 12% of PVA hydrogel capsule caused irritation to rat rectal mucosa. However, the WH-15 hollow type suppository inserted PVA hydrogel capsule caused no severe irritation on rectal mucosa. The application of the hollow type suppositories using PVA in sustained rectal delivery of drugs might be feasible.

Micro pH Sensor Using Patterned Hydrogel with pH Indicators

  • This paper presents a study into pH Indicator-Embedded hydrogel micro-particles which are encoded various shapes according to the captured indicator. We incorporate various pH indicators into a photo-curable hydrogel, PEG-DA(Poly(ethylene glycol) diacrylate). Using the latest fluidic lithography techniques, we can easily fabricate variously patterned hydrogel particles based on in-situ photopolymerization of the PEG-DA in a micro-fluidic channel. The shape of the particle is related to the pH indicator inside the particle. We demonstrate that the micro pH sensors change their colors according to the pH levels. The micro pH sensors have various characteristics that are related to the curing time, particle size, etc. By changing these conditions, we can adjust the long term stability and reaction time of the hybrid micro pH sensors.

Development of a cell-laden thermosensitive chitosan bioink for 3D bioprinting

  • 3D bioprinting is a technology to produce complex tissue constructs through printing living cells with hydrogel in a layer-by-layer process. To produce more stable 3D cell-laden structures, various materials have been developed such as alginate, fibrin and gelatin. However, most of these hydrogels are chemically bound using crosslinkers which can cause some problems in cytotoxicity and cell viability. On the other hand, thermosensitive hydrogels are physically cross-linked by non-covalent interaction without crosslinker, facilitating stable cytotoxicity and cell viability. The examples of currently reported thermosensitive hydrogels are poly(ethylene glycol)/poly(propylene glycol)/poly(ethylene glycol) (PEG-PPG-PEG) and poly(ethylene glycol)/poly(lactic acid-co-glycolic acid) (PEG/PLGA). Chitosan, which have been widely used in tissue engineering due to its biocompatibility and osteoconductivity, can be used as thermosensitive hydrogels. However, despite the many advantages, chitosan hydrogel has not yet been used as a bioink. The purpose of this study was to develop a bioink by chitosan hydrogel for 3D bioprinting and to evaluate the suitability and potential ability of the developed chitosan hydrogel as a bioink. To prepare the chitosan hydrogel solution, ${\beta}-glycerolphosphate$ solution was added to the chitosan solution at the final pH ranged from 6.9 to 7.1. Gelation time decreased exponentially with increasing temperature. Scanning electron microscopy (SEM) image showed that chitosan hydrogel had irregular porous structure. From the water soluble tetrazolium salt (WST) and live and dead assay data, it was proven that there was no significant cytotoxicity and that cells were well dispersed. The chitosan hydrogel was well printed under temperature-controlled condition, and cells were well laden inside gel. The cytotoxicity of laden cells was evaluated by live and dead assay. In conclusion, chitosan bioink can be a candidate for 3D bioprinting.

  • Biocompatible and biodegradable hydrogels based on carboxymethyl cellulose(CMC) and polyethyleneglycol(PEG) were prepared far physical barriers for preventing surgical adhesions. These interpolymeric hydrogels were synthesized by a gamma irradiation crosslinking technique. The 1Scmxl.Scm of cecal serosa and adjacent abdominal wail were abraded with bane burr until tbe serosal surface was disrupted and hemorrhagic but not perforated. and the serosa of tbe cecum was sutured to the abdominal wall in 5mm apart from the injured sire. The denuded cecum was covered with either CMC/PEG hydrogels or solution from CMC/PEG hydrogel. Control rat serosa was not covered. Two weeks later. the rats were sacrificed and adhesion was scored on a 0-5 scale. No treatment showed the significantly higher incidence of adhesions than either CMC/BEC hydrogels or solution from CMC/PEG hydrogel. In conclusion, these studies demonstrate that CMC/BEG hydrogels have a function of prevention of intra abdominal adhesion in a rat model.

In situ Gel Forming Stereocomplex Composed of Four-Arm PEG-PDLA and PEG-PLLA Block Copolymers

  • Injectable hydrogels are quite promising materials due to their potential to minimize invasive implantation and this provides versatile fitness irrespective of the damaged regions and facilitates the incorporation of bioactive agents or cells. In situ gel formation through stereocomplex formation is a promising candidate for injectable hydrogels. In this paper, a new series of enantiomeric, four-arm, PEG-PLA block copolymers and their stereocomplexed hydrogels were prepared by bulk ring-opening polymerization of D-lactide and L-lactide, respectively, with stannous octoate as a catalyst. The prepared polymers were characterized by $^1H$ nuclear magnetic resonance (NMR) spectroscopy, Fourier-transform infrared (FT IR) spectroscopy, gel permeation chromatography (GPC) and thermal gravitational analysis (TGA), confirming the tailored structure and chain lengths. The swelling and degradation behavior of the hydrogels formed from a selected copolymer series were observed in different concentrations. The degradation rate decreased with increasing polymer content in the solution. The rheological behavior indicated that the prepared hydrogel underwent in situ gelation and had favorable mechanical strength. In addition, its feasibility as an injectable scaffold was evaluated using a media dependence test for cell culture. A Tris solution was more favorable for in situ gel formation than PBS and DMEM solutions were. These results demonstrated the in situ formation of hydrogel through the construction of a stereocomplex with enantiomeric, 4-arm, PEG-PLA copolymers. Overall, enantiomeric, 4-arm, PEG-PLA copolymers are a new species of stereocomplexed hydrogels that are suitable for further research into injectable hydrogels.

Preparation and Characterization of Genetically Engineered Mesenchymal Stem Cell Aggregates for Regenerative Medicine

  • Combining cell- and gene-based therapy is a promising therapeutic strategy in regenerative medicine. The aim of this study was to develop genetically modified mesenchymal stem cell (MSC) aggregates using a poly(ethylene glycol) (PEG) hydrogel micro-well array technique. Stable PEG hydrogel micro-well arrays with diameters of 200 to $500\;{\mu}m$ were fabricated and used to generate genetically engineered MSC aggregates. Rat bone marrow-derived MSCs were transfected with a green fluorescent protein (GFP) plasmid as a reporter gene, and aggregated by culturing in the PEG hydrogel micro-well arrays. The resultant cell aggregates had a mean diameter of less than $200\;{\mu}m$, and maintained the mesenchymal phenotype even after genetic modification and cell aggregation. Transplantation of MSC aggregates that are genetically modified to express therapeutic or cell-survival genes may be a potential therapeutic approach for regenerative medicine.

Poly(L-lysine) Based Semi-interpenetrating Polymer Network as pH-responsive Hydrogel for Controlled Release of a Model Protein Drug Streptokinase

  • With the aim of developing of pH-sensitive controlled drug release system, a poly(Llysine) (PLL) based cationic semi-interpenetrating polymer network (semi-IPN) has been synthesized. This cationic hydrogel was designed to swell at lower pH and de-swell at higher pH and therefore be applicable for achieving regulated drug release at a specific pH range. In addition to the pH sensitivity, this hydrogel was anticipated to interact with an ionic drug, providing another means to regulate the release rate of ionic drugs. This semi-IPN hydrogel was prepared using a free-radical polymerization method and by crosslinking of the polyethylene glycol (PEG)-methacrylate polymer through the PLL network. The two polymers were penetrated with each other via interpolymer complexation to yield the semi-IPN structures. The PLL hydrogel thus prepared showed dynamic swelling/de-swelling behavior in response to pH change, and such a behavior was influenced by both the concentrations of PLL and PEG-methacrylate. Drug release from this semi-IPN hydrogel was also investigated using a model protein drug, streptokinase. Streptokinase release was found to be dependent on its ionic interaction with the PLL backbones as well as on the swelling of the semi-IPN hydrogel. These results suggest that a PLL semi-IPN hydrogel could potentially be used as a drug delivery platform to modulate drug release by pH-sensitivity and ionic interaction.

미생물 고정화를 위한 광경화성 하이드로겔의 합성과 특성 (Synthesis and Characteristics of Photo-crosslinkable Hydrogel for Microbial Immobilization)

  • 산업폐수중의 질소 인 제거에 유용한 미생물을 고정화할 수 있는 담체의 제조를 목적으로 하여, 양 말단이 methacryl기로 치환된 poly(ethyleneglycol)(PEG) 및 Poly(tetramethylene glycol)(PTMG)을 합성하였다. 합성된 프레폴리머와 UV조사하에서 가교한 하이드로겔의 구조는 $^1H$-NMR, FT-IR 분광기로 확인하였다. 프레폴리머 사슬의 길이(M.W. 1,000~8,000) 또는 PEG/PTMG의 혼합비를 달리하면서 광경화된 하이드로겔을 제조하였다. 광경화물의 친수성, 함수율, 기계적 강도 등과 같은 광경화 특성 및 pore size 등을 조사한 결과, 분자량 1000인 PEG 및 PEG(MW1000)와 PTMG (MW2900)를 7:3의 비율로 제조한 하이드로겔에서 최적의 결과를 나타내었다.

Effects of Extracellular Stimulation of Different Niche Condition on the Transcriptional Regulation of Matrix Metalloproteinase Genes in the Mouse Embryonic Stem Cells

  • Matrix metalloproteinases (MMPs) have been known to affect to cell migration, proliferation, morphogenesis and apoptosis by degrading the extracellular matrix. In the previous studies, undifferentiated mouse embryonic stem cells (ESCs) were successfully proliferated inside the extracellular matrix (ECM) analog-conjugated three-dimensional (3D) poly ethylene glycol (PEG)-based hydrogel. However, there is no report about MMP secretion in ESCs, which makes it difficult to understand and explain how ESCs enlarge space and proliferate inside 3D PEG-based hydrogel constructed by crosslinkers containing MMP-specific cleavage peptide sequence. Therefore, we investigated what types of MMPs are released from undifferentiated ESCs and how extracellular signals derived from various niche conditions affect MMP expression of ESCs at the transcriptional level. Results showed that undifferentiated ESCs expressed specifically MMP2 and MMP3 mRNAs. Transcriptional up-regulation of MMP2 was caused by the 3D scaffold, and activation of integrin inside the 3D scaffold upregulated MMP2 mRNAs synergistically. Moreover, mouse embryonic fibroblasts (MEFs) on 2D matrix and 3D scaffold induced upregulation of MMP3 mRNAs, and activation of integrins through conjugation of extracellular matrix (ECM) analogs with 3D scaffold upregulated MMP3 mRNAs synergistically. These results suggest that successful proliferation of ESCs inside the 3D PEG-based hydrogel may be caused by increase of MMP2 and MMP3 expression resulting from 3D scaffold itself as well as activation of integrins inside the 3D PEG-based scaffold.

PEG를 포함한 실리콘 수화젤 렌즈의 제조 및 특성 (Preparation and Characterization of Silicone Hydrogel Lens Containing Poly(ethylene glycol))

  • Poly(ethylene glycol)(PEG)이 콘택트렌즈에 미치는 영향을 조사하기 위하여 PEG가 도입된 실리콘 수화젤 렌즈를 제조하고 특성을 평가하였다. 실리콘 수화젤 렌즈는 methacryloxypropyl tris(trimethylsiloxy) silane(TRIS), methyl methacrylate(MMA), N,N'-dimethyl acrylamide(DMA) 등과 PEG 함유 단량체인 poly(ethylene glycol) methyl ether methacrylate(PEG-MEM)를 공중합하여 제조하였다. 특성평가는 Fourier transform infrared spectroscopy(FT-IR), electron spectroscopy of chemical analysis(ESCA), scanning electron microscopy(SEM) 등과 물 흡수도, 물 접촉각, 광투과도, 인장강도 등을 평가하였고 단백질 흡착 실험을 행하였다. FT-IR 및 ESCA 분석결과 C-O 결합피크의 증가를 보여 PEG 도입을 확인하였고 SEM 분석결과 상분리 현상은 나타나지 않았다. 물흡수도는 PEG-MEM의 도입에 따라 1차적으로 증가하였고 물 접촉각 및 광투과도는 감소하는 현상을 나타내었다. 단백질흡착 실험결과, PEG의 도입에 따라 눈물 속에 있는 단백질인 albumin, lysozyme, $\gamma$-globulin 등의 흡착이 감소하는 현상을 나타내었다. 또한 PEG 분자량의 증가에 따라 흡착되는 단백질의 양이 감소하는 것을 확인할 수 있었다. 인공눈물을 사용한 단백질 흡착실험에서도 PEG의 도입에 따라 단백질 흡착이 감소하는 현상을 나타내었다.
