A Strategy Using Photodynamic Therapy and Clofibric Acid to Treat Peritoneal Dissemination of Ovarian Cancer

  • Background: The current study examined the effectiveness of concurrent therapy using photodynamic therapy (PDT) and clofibric acid (CA) to treat peritoneal carcinomatosis resulting from ovarian cancer. Materials and Methods: Nude rats were used to create a model of peritoneal carcinomatosis resulting from ovarian cancer and the effectiveness of PDT with 5-aminolevulinic acid methyl ester hydrochloride (methyl-ALA-PDT) was determined. The survival time of rats receiving that therapy was compared to the survival time of a control group. Rats with peritoneal carcinomatosis resulting from ovarian cancer were divided into 3 groups: a group that received debulking surgery (DS) alone, a group that received DS+methyl-ALA-PDT, and a group that received DS+methyl-ALA-PDT+CA. The survival time of the 3 groups was compared. Protoporphyrin, a metabolite of methyl-ALA, produces a photochemical action when activated by light. The level of protoporphyrin (the concentration) that reached organs in the abdomen was measured with HPLC. Results: Rats receiving methyl-ALA-PDT had a significantly longer survival time compared to the controls. Rats with peritoneal carcinomatosis that received DS+methyl-ALA-PDT+CA had a significantly longer survival time compared to the rats that received DS alone. Some of the rats that received concurrent therapy survived for a prolonged period. Protoporphyrin was highly concentrated in peritoneal metastases, but only small amounts reached major organs in the abdomen. PDT was not found to result in necrosis in the intestines. Conclusions: The results indicated that concurrent therapy consisting of PDT with methyl-ALA and CA is effective at treating peritoneal carcinomatosis resulting from ovarian cancer without damaging organs.

AMC-HN3 인체 두경부 암세포에서 genistein과 photofrin PDT의 병행처리에 의한 세포 독성능의 증가 (The Anticancer Effect of Combination of Genistein and Photofrin PDT in Human AMC-HN3 Head and Neck Cancer Cell Lines)

  • 본 연구에서는 두경부 암세포 주에서 genistein의 증식억제 효과를 알아보고, genistein을 광역학 치료와 병행 치료 시 나타나는 세포 독성능 및 세포고사 유발에 대해 연구 하였다. 두경부 암세포 주에서 genistein의 세포독성능을 확인한 결과, 농도 및 시간 의존적으로 세포 독성능이 나타났다. 이에 genistein을 photofrin을 이용한 광역학 치료와 병행 치료 시 더 많은 세포 독성능이 나타났다. Genistein과 광역학 치료의 병행 치료 시 나타나는 형태적 변화는 광역학 치료시에는 주로 세포괴사의 형태가 관찰 되었으나, genistein과 병행 치료 시에는 세포고사가 관찰 되었다. 시간별로 관찰하였을 때, 광역학 치료를 $IC_{80}$으로 처리하였을 때 3시간 때에 apoptotic body가 약 3배정도 증가하였다. 세포 내 ROS를 확인한 결과 genistein과 광역학 치료를 단독으로 처리하였을 때 세포 내 존재하는 ROS가 각각 증가 하였으며, 병행 치료하였을 때는 더 많은 ROS가 형성되는 것으로 나타났다. 즉, genistein이 세포 내 ROS를 유발하여 mitochondria가 손상되고, 또한 photofrin에 의한 광역학 치료도 mitochondria를 손상시켜 세포고사를 유도하는 것으로 추측되어 진다.

Antibacterial Effect of Photodynamic Therapy using Photogem and a 632 nm Diode Laser on $Helicobacter$ $pylori$

  • Eradication of $H.$ $pylori$, usually carried out by using antimicrobial agents, is essential for preventing gastric ulcers and cancers. The $H.$ $pylori$ isolates, however, have continuously grown antimicrobial resistance, which have caused difficulty in treating the bacteria and in turn, photodynamic therapy (PDT) has been found to be effective in inducing deaths of variety of bacteria. After PDT treatment, the number of colony forming units (CFU), the morphologic changes, and flow cytometry were observed. In the PDT group containing 100 and 200 ${\mu}g$/ml photogem, no live $H.$ $pylori$ was observed, while 10 and 50 ${\mu}g$/ml photogem were only partially effective. $H.$ $pylori$ of the PDT group also displayed distortion and shrinkage in morphology. This study demonstrated that photogem-mediated PDT effectively induces deaths of $H.$ $pylori$.

광역동요법을 병행한 두피 모낭염 한방치험 1례 (A case of scalp folliculitis treated by Korean Medicine with photodynamic therapy(PDT))

  • Objectives : The purpose of this study is to report the effect of combination therapy of photodynamic therapy (PDT) with Korean medical treatments on scalp folliculitis, which has been difficult to treat by using antibiotic.Methods : A 32-year-old male patient with scalp folliculitis, received treatment with acupuncture, herbal medicine and PDT and so on. We used Visual Anologue Scale(VAS) to evaluate recovery from erythema, pruritus and pain. Also, the photos of patient were taken before and after treatments.Results : Visible symptoms to the naked eye and VAS scale effectively decreased after Korean medicine treatments for two months.Conclusions : It is expected that Korean medical treatments and PDT were thouht to be effective for scalp folliculitis.

The Effects of Photodynamic Therapy for Vancomycin-resistant Enterococci

  • The aim of this study was to evaluate the effects of the photosensitizer photogem with light-emitting diode (LED) on vancomycin-resistant enterococci (VRE). Two VRE strains isolated from the feces of patients. that was identificated Enterococcus faecium (vanA) and Enterococcus gallinarum (vanC1) using traditional biochemical tests and confirmed VRE genotyping from using polymerase chain reaction. In addition, three strains were used Enterococcus. faecalis CDC-286 (vanA), E. faecalis CDC-583 (vanB) and E. gallinarum CDC-42 (vanC1). To examine the antimicrobial effect of photogem mediated photodynamic therapy (PDT) against, CFU quantification and Disk diffusion antimicrobial susceptibility test were evaluated. The effects of Photodynamic therapy was not associated with genotype. Photogem mediated PDT perfectly inhibited the colony formation of E. faecalis CDC-286. The number of viable bacteria decreased greatly after PDT application with photogem $50{\mu}g/mL$ and energy density of $15J/cm^2$. The diameter of inhibition zone was increased to after PDT more than before PDT. The case of vancomycin disc on E. faecalis CDC-583 and E. galinanum-Patient were changed from resistant to intermediate resistant, from intermediate resistant to susceptable. These results demonstrate that lethal photosensitization of VRE can be achieved using photogem plus 630 nm LED irradiation.

Synthesis and Photodynamic Activities of Pyrazolyl and Cyclopropyl Derivatives of Purpurin-18 Methyl Ester and Purpurin-18-N-butylimide

  • The synthesis of new pyrazolyl and cyclopropyl derivatives of purpurin-18 methyl ester and purpurin-18-N-butylimide 1a, 1b, 2a, 2b and 8 is described. The new compounds were characterized by NMR, UV-vis spectroscopy and mass spectrometry. UV-vis spectra of the new compounds showed long wavelength absorption of ranges 692 - 708 nm ($\lambda_{max}$). Photodynamic effects of the chlorin derivatives 1a, 1b, 2a and 2b were investigated by WST-1 assay in A549 cells, and showed good photodynamic activities with high photocytotoxicity and low cytotoxicity in the dark. In comparison between pyrazolyl and cyclopropyl derivatives, purpurin-18 methyl ester compounds 1a and 1b showed comparable photocytotoxicity result of the cell viabilities, otherwise, pyrazolyl derivative of purpurin-18-N-butylimide 2a showed better cell viabilities than those of cyclopropyl derivative 2b. And cyclopropyl derivative of purpurin-18-N-butylimide 2b showed higher dark cytotoxicity than that of others.

  • Hypericin was found to exhibit the highest antitumoral activity in treating EAT by photodynamic therapy (PDT): Hypericin>HPde>PII>TPPS$_4$>ALA. Moreover, 25% of mice after Hyp-based PDT survived 4 months, if compare with control group. Antitumor activity of these photosensitizers was in rather clear correlation with accumulation potential.

  • In the past decades, complain about ground vibration and noise induced by pile driving has been quickly increased. Because of that, auger drilled piling methods have frequently used specially in urban area. However, the present auger drilled piling methods induce inevitable ground disturbance as well as a certain degree of vibration and noise due to the final hammering. For these reasons, a new auger drilled piling method is required to be developed. This paper introduces PDT(Pulse Discharge Technology) piling method and presents the characteristics of bearing capacity. The PDT piling method is to install in-situ piles using electric power so called Pulse. The pile installed by PDT appears to be able to develop shaft and end bearing capacity efficiently.

Flow Analysis in Positive Displacement Micro-Hydro Turbine and Development of Low Pulsation Turbine

  • In order to extract micro hydropower in the very low specific speed range, a Positive Displacement Turbine (PDT) was proposed and steady performance was determined experimentally. However, the suppression of large pressure pulsation is inevitable for practical application of PDT. The objective of the present study is to reveal the mechanism and the characteristics of pressure pulsation in PDT by use of CFD and to suppress the pressure pulsation. Unsteady CFD analysis has revealed that large pressure pulsation is caused by large variation of rotational speed of the following rotor, while the driving rotor, which is output rotor, keeps constant speed. Here is newly proposed a 4-lobe helical type rotor which can reduce the pressure pulsation drastically and the performance prediction of new PDT is determined.

  • Risperidone has been reported to have neuroleptic activity. In this study, risperidone was synthesized using a new method involving a stille reaction, in which 2-methyl-3-vinyl-6,7,8,9-tet­rahydropyrido[1 ,2-a]pyrimidin-4-one was synthesized (5). The chemical synthesis process was found to be simple and produced risperidone in a high yield. In addition, can be easily scaled up for large scale synthesis.
