Neuroprotective Effects of Bee Venom, which Removes High Molecular Elements against $MPP^+$-induced Human Neuroblastoma SH-SY5Y Cell Death ($MPP^+$로 유도된 SH-SY5Y신경세포 사멸에 대한 고분자성분제거 봉독약침액의 신경보호 효과 연구)

  • Objectives : The neuroprotective effects of bee venom (BV) have been demonstrated in many studies, but bee venom has many side effects. So we used sweet bee venom (SBV), which has high molecular elements removed to reduce the side effects. I examined the neuroprotective effect of sweet bee venom in 1-methyl-4-phenylpyridine ($MPP^+$)-induced human neuroblastoma SH-SY5Y cells. Methods : To observe the possible toxicity of SBV itself, SH-SY5Y cells were treated with SBV in various concentrations for 3 h and $MPP^+$ in concentrations (1 and 5mM) for 24h. To investigate the protective effect of SBV against $MPP^+$ toxicity, SH-SY5Y cells were pretreated with vehicle or nontoxic concentrations of SBV for 3h and the cells were not washed, followed by incubation with respective concentrations of SBV and 1 mM $MPP^+$ for 24h. To investigate the protective effect of SBV against $MPP^+$ toxicity, SH-SY5Y cells were pretreated with vehicle or nontoxic concentrations of SBV for 3h and the cells were not washed, followed by incubation with respective of SBV(0.5%), 1 mM $MPP^+$, 5uM AKT inhibitor(LY984002) and 10uM ERK inhibitor(PD98059) for 24 h. The protective effect was measured by cell viability assay. To investigate the degree of apoptosis, caspase-3 enzyme activity was measured in control, $MPP^+$, SBV+$MPP^+$. Results : SBV (0.5%) pretreatment protected the SH-SY5Y cells against $MPP^+$-induced apoptotic cell death. The cell viability was higher in the SH-SY5Y cells that were pretreated with vehicle or nontoxic concentrations of SBV than those not pretreated. The caspase-3 activity was lower in the pretreated groups than these not pretreated. ERK and AKT enzymes have a role in the neuroprotective effects of the sweet bee venom. Conclusions : The results demonstrate that SBV has a protective effect on dopaminergic neurons against $MPP^+$ toxicity. This data suggest that SBV could be a potential therapeutic tool for neurodegenerative diseases such as Parkinson's disease(PD).

  • HAP란 Hydroxylapatite의 준말이며 우리말로는 수산화인회석으로도 불린다. 본 실험에서는 다양한 농도의 염기조건(NaOH $10^{-3}$, $10^{-2}$, $10^{-1}$, 1, 10, 30 M)에서 서로 다른 형태의 HAP를 수열합성법을 통해 합성하였다. XRD (X-ray powder diffraction) 로 관찰한 결과 NaOH 농도 $10^{-1}M$ 이상에서부터 HAP가 합성됨을 확인하였다. Transmission and scaning electron microscopy 를 이용하여 HAP의 모양과 표면을 관찰해본 결과, NaOH의 농도가 진해 질수록 육각기둥의 형태에서 사각형으로 변화하였다. 6개의 각각의 HAP의 표면에 Pd (Palladium)을 도입하고 그 양을 정량화 하였다. 합성된 Pd-HAP를 C-C coupling reaction에 이종상 촉매로 사용하였다.

A Highly Selective Mercury(II) Ion-Selective Membrane Sensor (고 선택성 수은(II) 이온 막 센서)

  • A new ion selective PVC membrane electrode is developed as a sensor for mercury(II) ions based on bis(benzoylacetone) propylenediimine (H2(BA)2PD) as an ionophore. The electrode shows good response characteristics and displays, a linear Emf vs. log[Hg2+] response over the concentration range of 1.0×10-6 to 1.0×10-1 M Hg(II) with a Nernstian slope of 29.8±0.75 mV per decade and with a detection limit of 2.2×10-7 M Hg(II) over the pH range of 2.5-11.5. Selectivity concentrations for Hg(II) relative to a number of potential interfering ions were also investigated. The sensor is highly selective for Hg(II) ions over a large number of cations with different charge. The sensor has been found to be chemically inert showing a fast response time of 60 s and was used over a period of 3 months with a good reproducibility (S = 0.27 mV). The electrode was successfully applied to determine mercury(II) in real samples with satisfactory results.

  • The Homogeneous Charge Compression Ignition (HCCI) engine concept allows for both NOx and particulate matter to be reduced simultaneously, and it is a promising way to meet the next environmental challenges. Unfortunately, HCCI combustion often increases CO and HC emissions. The development of oxidation catalyst (OC) requires high conversion efficiency for CO and HC at low temperature. Conventional oxidation catalyst technologies may not be able to convert these emissions because of the saturation of active catalytic sites. The OC used in this study was 600 cpsi cordierite. Three kinds of OC with different amounts of Pt and Pd were used. The influence of the space velocity (SV), $H_2O$ and $O_2$ concentration was also studied. All types of OCs were found to have over 90% CO conversion efficiencies at $170^{\circ}C$. When in the presence of water vapor, CO conversion was increased, but $C_3H_8$ conversion was decreased. The performance of the OC was not influenced by initial the HC concentration. The 2Pt/Pd catalyst was better in terms of thermal aging than the Pt-only catalyst. The $LOT_{50}$ of both fresh and aged OC was increased with increasing SV and with the presence of $H_2O$.

  • HTS power transmission cable is expected to transport large electric power with a compact size. We are developing a 3-core, 22.9 kV, 50 MVA class HTS power cable, and each core consists of a conductor and shield wound with Bi-2223 tapes, electrical insulation with laminated polypropylene paper (LPP) impregnated with liquid nitrogen. This paper describes the design and experimental results of the model cable for the 22.9 kV, 50 MVA class HTS power transmission cable. The model cable was used the SUS tapes instead of HTS tapes because of testing the electrical characteristics only. The model cable was 1.3 m long and electrical insulation thickness was 4.5 mm. The model cable was evaluated the partial discharge (PD), AC and Impulse withstand voltage in liquid nitrogen. The AC and Impulse withstands voltage and PD inception stress was satisfied with the standard of Korea Electric Power Corporation (KEPCO) in the test results. The 3-core 22.9 kV, 50 MVA class HTS power cable has been designed and manufactured based on these experimental results.

  • $V_{2}O_{5}/ThO_{2}/Pd$-doped $SnO_{2}$ sensor has a good selectivity and stability to CO at high sensor temperature of about $500^{\circ}C$, and shows rapid response. In particular, many kinds of interference gases, such as $NO_{x}$, $C_{3}H_{8}$, $CH_{4}$ and $SO_{2}$ have been found to give only a slight influence on the sensor selectivity to CO gas sensitivity by doped $V_{2}O_{5}$ (3.0 wt.%). For the sensor we used well-known thick film technological route with $V_{2}O_{5}$(3.0 wt.%), Pd(1.0 wt.%) and $ThO_{2}$(l.5 wt.%) as catalytic materials. In the case of mixed $NO_{x}$-CO gases, as combustion exhaust gas, only CO detection by $SnO_{2}$ type semiconductor sensor is generally very difficult because of $NO_{x}$ interference. The developed sensors can use to measure the exhausting gas of the automobile or the boiler for the Air-to-Fuel ratio control.

The CD28-B7 Family in Anti-Tumor Immunity: Emerging Concepts in Cancer Immunotherapy

  • The interactions between B7 molecules and CD28-family receptors are crucial in the regulation of adaptive cellular immunity. In cancer, the aberrant expression of co-inhibitory B7 molecules has been attributed to reduced anti-tumor immunity and cancer immune evasion, prompting the development of cancer therapeutics that can restore T cell function. Murine tumor models have provided significant support for the targeting of multiple immune checkpoints involving CTLA-4, PD-1, ICOS, B7-H3 and B7-H4 during tumor growth, and clinical studies investigating the therapeutic effects of CTLA-4 and PD-1 blockade have shown exceptionally promising results in patients with advanced melanoma and other cancers. The expression pattern of co-inhibitory B7 ligands in the tumor microenvironment has also been largely correlated with poor patient prognosis, and recent evidence suggests that the presence of several B7 molecules may predict the responsiveness of immunotherapies that rely on pre-existing tumor-associated immune responses. While monotherapies blocking T cell co-inhibition have beneficial effects in reducing tumor burden, combinatorial immunotherapy targeting multiple immune checkpoints involved in various stages of the anti-tumor response has led to the most substantial impact on tumor reduction. In this review, we will examine the contributions of B7- and CD28-family members in the context of cancer development, and discuss the implications of current human findings in cancer immunotherapy.

A Study of the Formation of Binary Intermediate Layer on Pd-based Hydrogen Separation Membrane Using Various Types of Metal Oxides (다양한 형태의 금속 산화물을 이용한 Pd계 복합 수소분리막의 2원계 중간층 형성에 관한 연구)

  • In this study, the intermediate layer in Pd-based hydrogen separation membrane was synthesized to minimize the surface roughness and defects using powder-type and sol-type metal oxides. The surface properties and gas permeation characteristics were analysed by SEM and $N_2$ gas permeation test. The coating layer composed of sol type metal oxides has smooth surface, especially the layer coated by $TiO_2$ sol has little pin holes, cracks and defects. The binary layer composed of powder type and sol type metal oxides has similar flux characteristics to a single sol type layer. The Pd-based composite membrane improved by the binary intermediate layer exhibited $0.32mol/m^2s$ of the hydrogen permeation flux with a selectivity ($H_2/N_2$) of ~10,890 at 672 K and a pressure difference of 1 bar.

Aquaporin 8 Involvement in Human Cervical Cancer SiHa Migration via the EGFR-Erk1/2 Pathway

  • Overexpression of aquaporins (AQPs) has been reported in several human cancers. Epidermal growth factor receptor (EGFR)-extracellular signal-regulated kinases 1/2 (Erk1/2) are associated with tumorigenesis and cancer progression and may upregulate AQP expression. In this study, we demonstrated that EGF (epidermal growth factor) induces SiHa cells migration and AQP8 expression. Wound healing results showed that cell migration was increased by 2.79-1.50-fold at 24h and 48h after EGF treatment. AQP8 expression was significantly increased (3.33-fold) at 48h after EGF treatment in SiHa cells. An EGFR kinase inhibitor, PD153035, blocked EGF-induced AQP8 expression and cell migration and AQP8 expression was decreased from 1.59-fold (EGF-treated) to 0.43-fold (PD153035-treated) in SiHa. Furthermore, the MEK (MAPK (mitogen-activated protein kinase)/Erk (extracellular signal regulated kinase)/Erk inhibitor U0126 also inhibited EGF-induced AQP8 expression and cell migration. AQP8 expression was decreased from 1.21-fold (EGF-treated) to 0.43-fold (U0126-treated). Immunofluorescence microscopy further confirmed the results. Collectively, our findings show that EGF induces AQP8 expression and cell migration in human cervical cancer SiHa cells via the EGFR/Erk1/2 signal transduction pathway.
