Post-weaning multisystemic wasting syndrome (PMWS) is a new emerging disease affecting nursery and growing pigs in worldwide. Porcine circovirus type 2 (PCV-2) is a most important pathogen associated with PMWS. This study was carried out to investigate the pathological changes in the pancreas of pigs diagnosed as PMWS. To detect the PCV-2 antigen and nucleic acid in the tissue, immunohistochemistry and polymerase chain reaction (PCR) was conducted, respectively. 24 pigs of 4-10 weeks old showed clinical signs of PMWS such as chronic wasting, respiratory distress and diarrhea were examined. Histopathologically, interstitial and periductular mononuclear cells infiltration were observed in pancreas. Multifocal to diffuse necrosis of acinar tissues or necrotizing to granulomatous pancreastitis with numerous syncytial cells infiltration were examined in severe cases. PCV-2 nucleic acid was detected from all tested pancreas using PCR. The PCV-2 antigen in 12 pancreas sections was detected by immunohistochemical staining. PCV-2 has a tropism for vascular endothelial cells and infiltrated macrophages. Although gross lesions are uncommon in the pancreas of pigs with PMWS, histopathological changes and the presence of PCV-2 in this tissue may be related to clinical signs associated with digestive disorders.
In order to obtain the genetic informations of the Korean isolates of porcine circovirus 2 (PCV2), nucleotide sequences of total genome of three isolates and open reading frame 2 (ORF2) of four isolates were determined and compared with those of other reference PCV2 isolates. Nucleotide sequences of 3 isolates showed over 99% homology with those of reference strain (GenBank accession no. AF027217). Point mutations were mainly determined on ORF2 regions but little on ORF1 regions. The patterns of pointmutated sites and nucleotide substitution on ORF2 regions were generally consistent between Korean isolates, and these mutated sites observed in Korean isolates were also relatively similar to those of foreign isolates. Phylogenetic analysis of nucleotide or amino acid sequences showed that there were minor branches consisting of three clusters; cluster of Korea, Canada and America, cluster of Spain and Taiwan, and the last cluster of French and China isolates. These results suggested that Korean PCV2s were probably originated from North America such as Canada or USA. The genetic informations obtained from this study could be useful for the research of diagnosis and pathogenecity of PCV2.
From 2006 to 2009, 50 pigs suspected of enteritis associated with porcine circovirus type 2 (PCV-2) (EAPC) were collected. Gross and histopathologic examinations and immunohistochemistry (IHC) were performed on the small intestine of 50 pigs. The pigs with EAPC were concentrated in grower pigs (68%), and diarrhea (84%) was the most common clinical sign. Grossly, the walls of the small intestine were thickened, and mesenteric lymph nodes were enlarged. The histopathologic features in the small intestine with EAPC were characterized by lymphoid depletion, histiocytic infiltration, and formation of basophilic grape-like inclusion bodies and multinucleated giant cells in gut-associated lymphatic tissue (GALT) and lamina propria. According to IHC, PCV-2 antigens were more intense and distributed widely in GALT (98%) than lamina propria (82%) of the small intestine. Co-infection with other enteric pathogens was detected in 26 pigs (52%). Twenty-four pigs (48%) were confirmed to be a PCV-2 single infection. Collectively, infected PCV-2 in the small intestine may induce an immunosuppressive status of individuals and then allow secondary co-infections in the digestive system. This study shows that PCV-2 can induce diarrhea and enteric lesions in pigs without a co-infection of other enteric pathogens.
Today swine respiratory disease is one of the most important diseases because of its economic losses and severe infection nationwide, and swine society as well as veterinary service are trying to prevent the diseases in Korea. This study would like to obtain some information useful for the control of the diseases. A total of 174 lung specimens with lesion consisted of 3 sorts; 60 were collected from nursey pigs requested for diagnostic service from March of 2006 to October of 2007, 58 finishing pigs and 56 sows were selected from slaughterhouse from September to November 2007. In the detection test of pathogens by PCR, porcine circovirus 2 (PCV2), porcine reproductive and respiratory syndrome virus (PRRSV) and Mycoplasma hyopneumoniae were positive in 95.4%, 31.6%, and 20.1%, respectively. Double infection rate with PCV2 and PRRS was 30.4%, PCV2 and M hyopneumoniae was 19.5%, triple infection with PCV2, PRRS and M hyopneumoniae was 5.7%, respectively.

Background : The patient's work of breathing(WOBp) during assisted ventilation may vary according to many factors including ventilatory demand of the patients and applied ventilatory setting by the physician. Pressure-controlled ventilation(PCV) which delivers gas with decelerating flow may better meet patients' demand to improve patient-ventilator synchrony compared with volume-controlled ventilation(VCV) with constant flow. This study was conducted to compare the difference in WOBp in two assisted modes of ventilation, PCV and VCV with constant flow. Methods : Ten patients with respiratory failure were included in this study. Initially, the patients were placed on VCV with constant flow at low tidal volume($V_{T,\;LOW}$)(6-8 ml/kg) or high tidal volume($V_{T,\;HIGH}$)(10-12 ml/kg). After a 15 minute stabilization period, VCV with constant flow was switched to PCV and pressure was adjusted to maintain the same tidal volume($V_T$) received on VCV. Other ventilator settings were kept constant. Before changing the ventilatory mode, WOBp, $V_T$, minute ventilation($V_E$), respiratory rate(RR), peak airway pressure (Ppeak), peak inspiratory flow rate(PIFR) and pressure-time product(PTP) were measured. Results : The mean $V_E$ and RR were not different between PCV and VCV during the study period. The Ppeak was significantly lower in PCV than in VCV during $V_{T,\;HIGH}$. HIGH ventilation(p<0.05). PIFR was significantly higher in PCV than in VCV at both $V_T$ (p<0.05). During $V_{T,\;LOW}$ ventilation, WOBp and PTP in PCV($0.80{\pm}0.37\;J/min$, $164.5{\pm}74.4\;cmH_2O.S$) were significantly lower than in VCV($1.06{\pm}0.39J/mm$, $256.4{\pm}107.5\;cmH_2O.S$)(p<0.05). During $V_{T,\;HIGH}$ ventilation, WOBp and PTP in PCV($0.33{\pm}0.14\;J/min$, $65.7{\pm}26.3\;cmH_2O.S$) were also significantly lower than in VCV($0.40{\pm}0.14\;J/min$, $83.4{\pm}35.1\;cmH_2O.S$)(p<0.05). Conclusion : During assisted ventilation, PCV with decelerating flow was more effective in reducing WOBp than VCV with constant flow. But since individual variability was shown, further studies are needed to confirm these results.
The purpose of this study was to survey seroprevalence of porcine reproductive and respiratory syndrome (PRRS) virus and porcine circovirus-2 (PCV-2) in Gyeongbuk province by enzyme-linked immunosorbent assay (ELISA). A total of 966 samples collected from 21 pig farms were tested. The sero-positive rate of PRRS and PCV-2 were 77.6% (750/966) and 76.4% (738/966), respectively.
Currently, various diseases reside in Korean swine farms and affect production performance of the farms greatly. These damages from disease are further aggravated by the concurrent infection of other disease. In this study, y investigating the distribution of porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Salmonella spp. in farms, correlation between the damage and the prevalence of disease was analyzed. Ten selected Korean swine farms that uses PCV2 vaccine were tested for presence of antibody and antigen of PRRSV, PCV2, Salmonella spp. per ages of pigs, 4weeks, 7weeks, 11weeks and 17weeks, respectively. The results were analyzed by dividing the farms in to groups with MSY above 19, and that with MSY below 19. Then calculating the distribution of disease each ages of pigs. Farms with MSY below 19 showed high prevalence of disease by PRRSV, PCV2 and Salmonella spp.. In this group, the detection rate of PCV2 and Salmonella spp. was increased by the activation/viremia of PRRSV in the young ages of pigs. The results are proved that the correlation between disease prevalence and production performance in Korean swine farms were very significant. The prevalence of PRRSV is more important index which influence to the productivity in current prevalence of diseases.

Porcine circovirus type 2 (PCV2) is an important infectious swine virus causing postweaning multisystemic wasting syndrome (PMWS). PCV2 capsid protein, encoded by ORF2 has type-specific epitopes, is very immunogenic, and is associated with the induction of neutralizing antibodies. For the efficient production of capsid protein, recombinant Autographa californica nucleopolyhedroviruses were generated to express ORF2 fused with two forms of a partial polyhedrin. Recombinant capsid protein was produced successfully with the partial polyhedrin fusion form and the yield was high, as was shown by SDS-PAGE. Production of recombinant capsid proteins in insect cells was confirmed by Western blot analysis using anti-His monoclonal antibody, anti-ORF2 monoclonal antibody, and anti-PCV2 porcine serum. Fusion expression with amino acids 19 to 110 of the polyhedrin increased the production of recombinant capsid protein, but fusion with amino acids 32 to 85 did not. Additionally, PCV2 capsid protein is a glycoprotein; however, the glycosylation of recombinant protein was not observed. The results of an Enzyme-linked immunosorbent assay (ELISA) showed that recombinant capsid proteins could be utilized as antigens for fast, large-scale diagnosis of PCV2-infected pigs. Our results suggest that the fusion expression of partial polyhedrin is able to increase the production of recombinant PCV2 capsid protein in insect cells.

The prevention of porcine respiratory disease complex (PRDC) is very important because of its high infection-rates in the swine farms and the economic impact in swne industry in Korea. To control the prevalence of PRDC, it is important to know about infection patterns of it. Therefore, this study aimed to investigate the infection patterns of PRDC in the northern area of Gyeongsangnam-do. To this end, the infection of porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Actinobacillus pleuropneumoniae (APP), Mycoplasma hyopneumoniae (MH), and Swine influenza virus (SIV) were examined using 120 pig lung tissues by PCR analysis. As a result, single pathogen positive specimens were 25.0% and the others (75.0%) were turned out to be PRDC with at least two pathogens. Among PRDCs, 50 specimens (41.7%) was infected with PRRSV, PCV2, MH and SIV. Ten specimens (8.3%) showed triple infections of PRRSV, PCV2 and MH. Double infected specimens for PRRSV and PCV2 were 10 (8.3%), and for PCV2 and APP were 20 (16.7%).
A great deal of exhaust gas inside a combustion room goes out through exhaust pipe but residual gas, is called "Blowby gas", enters the crankcase through a small gap between the piston and the cylinder wall. Here, if the crankcase isn't vented, this causes many bad effects such as lubricant oil contamination, corrosion by that and crankcase explosion by rising pressure. So, most automobiles are constituted with a PCV (Positive Crankcase Ventilation) system to prevent previous problems. PCV valve is the most important part in this ventilation system. When companies are manufacturing new engines, engineers are designing it depending on their experiments than theoretical knowledge. Mush efforts and times are needed for new development. This study will show quantitative results to increase the possibilities of reduction of developing time.
