Process analytical technology (PAT): new paradigm for the state-of-the-art analytical technology (공정분석기술: 첨단 분석기술의 새로운 패러다임)

  • Process analytics has been already widely utilized in a large-scale continuous production line such as petroleum industries for several decades. Although the process analytics has a long history, a concept of "Process Analytical Technology (PAT)" has been rapidly adopted as a new paradigm for the process monitoring in the production process of various industries. In this review, current status and recent developments of PAT in various research bodies have been introduced, including the introduction of various types of analytical instruments, chemometrics tools, and perspectives and future applications of PAT as well as the fundamentals on PAT such as terminology and its historical background.

  • As the size of the internet market grows rapidly, the number of IPv4 addresses available is being exhausted, while transition to IPv6 is being delayed. As the best alternative solution, Network Address Translation(NAT) scheme is being used. It connects the public internet network with the private internet network in order to reduce the waste of IPv4 addresses space. The purpose of this paper is to study the effective example of network based on common virtual network using Packet Tracer with topology designed rather than usual theoretical approach in Dynamic NAT and PAT, which allows more efficient use of address space.

Utility of the pat gene as a selectable marker gene in production of transgenic Dunaliella salina

  • Background: The objective of this study was to develop an efficient selectable marker for transgenic Dunaliella salina. Results: Tests of the sensitivity of D. salina to the antibiotic chloramphenicol and the herbicide Basta$^{(R)}$ showed that cells ($1.0{\times}10^6cells/ml$) treated with 1000 or $1500{\mu}g/ml$ chloramphenicol died in 8 or 6 days, respectively, whereas D. salina cells ($1.0{\times}10^6cells/ml$) treated with 5, 10, 20, or $40{\mu}g/ml$ Basta$^{(R)}$ died in 2 days. Therefore, D. salina is more sensitive to Basta$^{(R)}$ than to chloramphenicol. To examine the possibility of using the phosphinothricin N-acetyltransferase (pat) gene as a selectable marker gene, we introduced the pat genes into D. salina with particle bombardment system under the condition of helium pressure of 900 psi from a distance of 3 cm. PCR analysis confirmed that the gene was stably inserted into the cells and that the cells survived in $5{\mu}g/ml$ Basta$^{(R)}$, the medium used to select the transformed cells. Conclusions: The findings of this study suggest that the pat gene can be used as an efficient selectable marker when producing transgenic D. salina.

Development of Basta Resistant Tobacco Using Artificial Phosphinothricin Acetyltransferase Gene (인공합성 Phosphinothricin Acetyltransferase 유전자에 의한 Basta 내성 연초식물체의 개발)

  • This experiment was conducted to introduce phosphinothricin acetyl -transferase(PAT) gene, resistant to basta and non-selective herbidide, into tobacco(Nicotiana tabacum cv.BY4). For shoot formation,tobacco leaf disks were placed on the MS medium supplemented with 2.0mg/L BA and 0.1mg/L NAA. In this medium condition, tobacco leaf disces were cocultivated with A. tumefaciens MP90 containing NPT IIand PAT resistant to kanamycin and Basta, respectively. Shoots were obtained in the medium containing antibiotics, and those were transferred to rooting medium supplemented with 0.1mg/L NAA and antibiotics. The plants obtaining roots were transplanted into soil. Phenotype of transgenic tobacco plant was mostly as normal plant. However, about 5% was abnormal plant, which did not set seeds. PCR analysis and southern blot were performed to determine transformation. As the results, it was confirmed that PAT gene was stably integrated into tobacco genome.When herbicide, basta, was sprayed to the plants confirmed by PCR, the transgenic plants showed normal growth, whereas normal plants died. Therefore, the result of this experiment show that tobacco transformation for the resistance to basta, non-selective herbicide, was successful because PAT gene was stably integrated into tobacco.

Epistatic Relationships of Two Regulatory Factors During Heterocyst Development

  • The filamentous cyanobacterium Anabaena sp. Strain PCC 7120 produces a developmental patten of single hete- rocysts separated by approximately 10 vegetative cells. Heterocysts differentiate from vegetative cells and are spe- cialized for nitrogen fixation. The patS gene, which encodes a small peptide that inhibits heterocyst differentiation, is expressed in proheterocysts and plays a critical role in establishing the heterocyst pattem. Another key regulator of heterocyst development is the hetR gene. hetR mutants fail to produce heterocysts and extra copies of hetR on a plas- mid cause a multiple contiguous heterocyst phenotype. To elucidate the relationship between these two counter act- ing factors in the genetic regulatory pathway during heterocyst differentiation, the expression patterns of a patS-gfp and a hetR-gfp fusion were examined in a patS deletion and a hetR deletion strain. The results, in combination with the result from a hetR and patS double deletion strain, suggest patS and hetR are mutually antagonistic and the bal- ance between these two factors in tow different cell types (heterocysts and vegetative cells) may be critical during the decision making process on their cell fates.

  • 본 논문에서는 현재 Jazz에서 사용되어지는 화성기법을 Pat Metheny의 작품인 "Bright Size Life"를 통해서 알아보았다. D Major Scale이 선율에 주로 사용된 본 작품은 A 부분에서는 선율이 완전 5도의 음정으로 발전 되었고 B부분에서는 장7도의 음정을 이용하였다. 화성은 중간에 D Minor Scale의 Bb/A가 사용되어서 Modal InterChange기법도 나타나고 있다. B부분에서는 G/A, F/G의 Constant Structure가 나오고 있다.

  • 본 논문에서는 현재 Jazz에서 사용되어지는 화성기법을 Pat Metheny의 작품인 “Question & Answer”를 통해서 알아보았다. D Minor Pentatonic Scale이 선율에 주로 사용된 본 작품은 Intro부분에서는 Quater Voicing이 사용되었다. A 부분에서는 Line Cliche와 D Minor Scale과 D Harmonic Minor Scale의 Diatonic Chord를 사용하였지만 A Aeolian Mode적인 기법이 A부분 셋 번째 단에서 사용되어 Modal적인 분위기도 연출 하였으며 B부분에서는 John Coltrane이 자주 사용하였던 3 Tonic 기법이 사용되면서 동질의 화성이 일정간격으로 움직이는 Constant Structure기법도 사용되었다.

Ultrasonographic Findings of Both Knee in Hemiplegic Ambulators with Recent Stroke (초기 편마비 보행 환자에서 양측 무릎의 초음파 검사 소견)

  • To evaluate of the ultrasonographic changes in hemiplegic and unaffected knee joints of hemiplegic ambulators with recent onset stroke. Twenty patients (40 knees) with prevalence duration of 14-39 weeks (mean 24.70 weeks) after a stroke insult were included. All participants were walking independently without leg dragging, had no previous knee injury. There were significant differences in intercondylar cartilage thickness and pes anserinous tendinopathy (PAT) between hemiplegic and unaffected knees (p<0.05). There were no differences in cartilage thickness of medial condyle and lateral condyle, patellar tendinitis/bursitis, suprapatellar effusion, synovitis, joint space narrowing, and Baker's cyst of ultrasonographic findings and x-ray findings in hemiplegic knee compared to the control except PAT (p<0.05). In hemiplegic knee, medial or lateral condylar cartilage thickness was significantly correlated with body weight, intercondylar cartilage thickness, and PAT (p<0.05). PAT was significantly correlated with Brunnnstrom stage and condylar cartilage thickness (p<0.05). Knee pain was significantly correlated with lateral condylar cartilage thickness (p<0.1). The hemiplegic knee had more ultrasonographic abnormalities including PAT and more cartilage thickness preservation in patient with recent onset stroke. Intraarticular sonographic findings including cartilage thickness was significantly correlated with extraarticular findings including PAT and a symptom of knee pain.

Efficacy of Proximal Aspiration Thrombectomy for Using Balloon-Tipped Guide Catheter in Acute Intracranial Internal Carotid Artery Occlusion

  • Objective : Mechanical thrombectomy (MT) for acute intracranial internal carotid artery (ICA) occlusion is often complicated by difficult revascularization and non-involved territory embolization possibly related with larger clot-burden. This study aims to evaluate the efficacy of proximal aspiration thrombectomy (PAT) using a balloon-tipped guide catheter for clot-burden reduction in such cases with period-to-period analysis (period 1 : standard MT without PAT; period 2 : PAT first, then standard MT for the remaining occlusion). Methods : Eighty-six patients who underwent MT for acute intracranial ICA occlusion were included in this analysis from the prospectively maintained stroke registry (33 patients in period 1 and 53 in period 2). In period 2, 'responder' was defined as a case where some amount of clot was retrieved by PAT and the following angiography showed partial or full recanalization. Results : Fifteen of fifty-three patients in period 2 (28.3%) were 'responders' to PAT. There was a significantly higher incidence of atrial fibrillation in the 'responder' subgroup. Period 2 showed a significantly shorter puncture-to-reperfusion time (94.5 minutes vs. 56.0 minutes; p=0.002), a significantly higher Thrombolysis in Cerebral Infarction of 2b-3 reperfusion (45.5% vs. 73.6%; p=0.009), but only a trend for better 3-month favorable outcome (mRS 0-2; 36.4% vs. 54.7%; p=0.097). There was no increase in the incidence of procedure-related complications or intracranial hemorrhage in period 2. Conclusion : A strategy of PAT before standard MT may result in shorter puncture-to-reperfusion time and better angiographic outcome than a strategy of standard MT for acute intracranial ICA occlusion.
