Electrochemistry on Alternate Structures of Gold Nanoparticles and Ferrocene-Tethered Polyamidoamine Dendrimers

  • Self-assembled systems with polyamidoamine (PAMAM) dendrimers combined with gold nanoparticles have been widely studied because of their potential applications in molecular electronics, catalyst carriers, chemical sensors, and biomedical devices. In our work, gold nanoparticle monolayers and multilayers with pure and ferrocene-tethered PAMAM dendrimers as cross-linking molecules were deposited on a mixed self-assembled monolayer of gold substrates. The various generations of PAMAM dendrimers can be covalently attached to mercaptoundecanoic acid mixed with a mercaptoundecanol self-assembled monolayer. Cyclic voltammograms show that redox peak currents on the alternate multilayers of gold nanoparticles and ferrocene-tethered PAMAM dendrimers increase as the number of layers increases. Fourier transform IR external reflection spectroscopy and scanning electron microscopy support the results from electrochemical measurements.

Covalent Immobilization of Trypsin on a Novel Aldehyde-Terminated PAMAM Dendrimer

  • Dendrimers are a novel class of nonlinear polymers and due to their extensive applications in different fields, called versatile polymers. Polyamidoamine (PAMAM) dendrimers are one of the most important dendrimers that have many applications in nanobiotechnology and industry. Generally aldehyde terminated dendrimers are prepared by activation of amine terminated dendrimers by glutaraldehyde which has two problems, toxicity and possibility of crosslink formation. In this study, novel aldehyde-terminated PAMAM dendrimer was prepared and used for covalent immobilization of trypsin by the aim of finding a special reagent which can prevent crosslinking and deactivation of the enzyme. For this purpose aminoacetaldehydedimethylacetal (AADA) was used as spacer group between aldehyde-terminated PAMAM and trypsin.The findings of this study showed that immobilization of trypsin not only resulted higher optimal temperature, but also increased the thermal stability of the immobilized enzyme in comparison to the free enzyme.

  • The convergent synthesis of symmetric PAMAM-like dendrimers from azide-functionalized poly(amido- amine) (PAMAM) dendrons and two different multi-alkynes was investigated. The stitching method was based on the click chemistry protocol, i.e., the copper-catalyzed cycloaddition reaction between an alkyne and an azide.

PAMAM Dendrimers Augment Inhibitory Effects of Curcumin on Cancer Cell Proliferation: Possible Inhibition of Telomerase

  • Background: Despite numerous useful anticancer properties of curcumin, its utility is limited due to its hydrophobic structure. In this study, we investigated the comparative antiproliferative effect of PAMAM encapsulating curcumin with naked curcumin on the T47D breast cancer cell line. Materials and Methods: Cytotoxic effects of PAMAM dendrimers encapsulating curcumin and curcumin alone were investigated by MTT assay. After treating cells with different concentrations of both curcumin alone and curcumin encapsulated for 24h, telomerase activity was determined by TRAP assay. Results: While PAMAM dendrimers encapsulating curcumin had no cytotoxicity on cancer cells, they decreased the $IC_{50}$ for proliferation and also increased the inhibitory effect on telomerase activity. Conclusions: Considering the non-toxicity in addition to effectiveness for enhancing curcumin anticancer properties, dendrimers could be considered good therapeutic vehicles for this hydrophobic agent.

Patterning Biological Molecules onto Poly(amidoamine) Dendrimer on Gold and Glass

  • Patterning of biological molecules was attempted on both gold and glass using fourth generation (G4) poly(amidoamine) (PAMAM) dendrimer as an interfacing layer between solid surfaces and biomolecules. As for the patterning of avidin and anti-biotin antibody on gold, PAMAM dendrimers representing amine functionalities were firstly printed onto the 11-mercaptoundecanoic acid SAM by microcontact printing, followed by biotinylation, and reacted with fluorescence-labeled avidin or anti-biotin antibody. Fluorescence microscopic analysis revealed that the patterns of avidin and anti-biotin antibody were well constructed with the resolution of < 2 ㎛. The PAMAM dendrimers were also printed onto aldehyde-activated slide glass and reacted directly with anti-BSA antibodies, which had been oxidized with sodium periodate. As a result, distinct patterns of the anti-BSA antibodies were also obtained with a comparable edge resolution to that of avidin patterns on gold. These results clearly show that PAMAM dendrimers can be adopted as an interfacing layer for the patterning of biological molecules on solid surfaces with micrometer resolution.

Synthesis of Dendrimers from Alkyne-focal Dendrons by Oxidative Homo-coupling of Terminal Acetylene

  • General, fast, and efficient fusion methods for the synthesis of dendrimers with 1,3-diynes at a core were developed. The synthetic strategy was employed the oxidative homo-coupling of terminal alkyne. The oxidative homo-coupling reaction of the alkyne-functionalized Frechet-type dendrons 1-Dm was allowed to provide first through fourth generation dendrimers 2-Gm with 1,3-diynes at core. The fusion of the propargylfunctionalized PAMAM dendrons 3-Dm by homo-coupling of terminal alkyne lead to the formation of symmetric PAMAM dendrimers 4-Gm. Their structure of dendrimers was confirmed by $^1H$ and $^{13}C$ NMR spectroscopy, IR spectroscopy, mass spectrometry, and GPC analysis.

Quaternized Polyamidoamine Dendrimers as Novel Gene Delivery System: Relationship between Degree of Quaternization and Their Influences

  • Quaternary ammonium groups were introduced to Starburst polyamidoamine (PAMAM) dendrimers for a gene carrier. These quaternary dendritic carriers exhibited reduced cytotoxicity on 293T cells compared to parent dendrimers examined and their transfection efficiency were similar with parent dendrimers. Quaternization could be a promising tool to improve properties of dendrimers as a gene delivery carrier.

  • The transport of reactant gas, electrons and protons at the three phase interfaces in the catalytic layers of membrane electrode assemblies (MEAs) in proton exchange, membrane fuel cells (PEMFCs) must be optimized to provide efficient transport to and from the electrochemical reactions in the solid polymer electrolyte. The aim of reducing proton transport loss in the catalytic layer by increasing the volume of the conducting medium can be achieved by filling the voids in the layer with small-sized electrolytes, such as dendrimers. Generation 1.5 and 3.5 polyamidoamine (PAMAM) dendrimer electrolytes are well-controlled, nanometer-sized materials with many peripheral ionic exchange, -COOH groups and were used for this purpose in this study. The electrochemically active surface area of the deposited catalyst material was also investigated using cyclic voltammetry, and by analyzing the Pt-H oxidation peak. The performances of the fuel cells with added PAMAM dendrimers were found to be comparable to that of a fuel cell using MEA, although the Pt utilization was reduced by the adsorption of the dendrimers to the catalytic layer.

Modulation of Electroosmotic Flow through Skin: Effect of Poly(Amidoamine) Dendrimers

  • The objective of this work is to evaluate the effect of polyamidoamine (PAMAM) dendrimers on electroosmotic flow (EOF) through skin. The effect of size and concentration of dendrimer was studied, using generation 1, 4 and 7 dendrimer (G1, G4 and G7, respectively). As a marker molecule for the direction and magnitude of EOF, a neutral molecule, acetoaminophen (AAP) was used. The visualization of dendrimer permeation into the current conducting pore (CCP) of skin was made using G4-fluorescein isothiocyanate (FITC) conjugate and confocal microscopy. Without dendrimer, anodal flux of AAP was much higher than cathodal or passive flux. When G1 dendrimer was added, anodal flux decreased, presumably due to the decrease in EOF by the association of G1 dendrimer with net negative charge in CCP. As the generation increased, larger decrease in anodal flux was observed, and the direction of EOF was reversed. Small amount of methanol used for the preparation of dendrimer solution also contributed to the decrease in anodal flux of AAP. Cross-sectional view perpendicular to the skin surface by confocal laser scanning microscope (CLSM) study showed that G4 dendrimer-FITC conjugate (G4-FITC) can penetrate into the viable epidermis and dermis under anodal current. The permeation route seemed to be localized on hair follicle region. These results suggest that PAMAM dendrimers can permeate into CCP and change the magnitude and direction of EOF. Overall, we obtained a better understanding on the mechanistic insights into the electroosmosis phenomena and its role on flux during iontophoresis.

Functionalization of Au surfaces with 4-(carboxymethyl)aniline and amine-terminated dendrimers for enhanced surface density of antibodies on immunosensor Au chips

  • Here, we demonstrate surface functionalization of Au chips with 4-(carboxymethyl)aniline (CMA) and amine-terminated polyamidoamine (PAMAM) dendrimers for immobilization of antibodies on the Au surfaces. Use of the functionalization strategy led to high surface density of the immobilized antibodies on the Au chips. Specifically, we found that the functionalization of Au chips with CMA and amine-terminated $6^{th}$ generation PAMAM dendrimers allowed immobilization of immunoglobulin (IgG) antibodies with high surface density, which is 5 times higher than that obtained with Au surfaces functionalized with CMA and ethylenediamine.
