Polycyclic Aromatic Hydrocarbon (PAH) Binding to Dissolved Humic Substances (HS): Size Exclusion Effect

  • Binding mechanisms of polycyclic aromatic hydrocarbons (PAHs) with a purified Aldrich humic acid (PAHA) and its ultrafiltration (UF) size fractions were investigated. Organic carbon normalized binding coefficient ($K_oc$) values were estimated by both a conventional Stern-Volmer fluorescence quenching technique and a modified fluorescence quenching method. Pyrene $K_oc$ values depended on PAHA concentration as well as freely dissolved pyrene concentration. Such nonlinear sorption-type behaviors suggested the existence of specific interactions. Smaller molecular size PAH (naphthalene) exhibited higher $K_oc$ value with medium-size PAHA UF fractions whereas larger size PAH (pyrene) had higher extent of binding with larger PAHA UF fractions. The inconsistent observation for naphthalene versus pyrene was well explained by size exclusion effect, one of the previously suggested specific mechanisms for PAH binding. In general, the extent of pyrene binding increased with lower pH likely due to the neutralization of acidic functional groups in HS and the subsequent increase in hydrophobic HS region. However, pyrene $K_oc$ results with a large UF fraction (>100K Da) corroborated the existence of the size exclusion effect as demonstrated by an increase in $K_oc$ values at a certain higher pH range. The size exclusion effect appears to be effective only for the specific conditions (HS size or pH) that render HS hole st겨ctures to fit a target PAH.

Association of glycophorin A mutant frequency and urinary PAH metabolites influenced by genetic polymorphisms of GSTM1 in incineration workers (소각장 근로자에서 GSTM1의 유전자 다형성이 glycophorin A변이 발현율과 소변내 PAH 대사산물 농도와의 관계에 미치는 영향)

  • Eighty-one workers including 38 employees directly incinerating industry wastes were recruited from a company located in South Korea. To evaluate the association between urinary 1-hydroxypyrene glucuronide (1-OHPG) levels, as internal dose of polycyclic aromatic hydrocarbon (PAH) exposure, and glycophorin A (GPA) mutation frequency, as an early biologic effect indicator. Urinary 1-OHPG levels were measured by synchronous fluorescence spectroscopy after immunoaffinity purification using monoclonal antibody 8E11. Erythrocyte GPA variant frequency (NN or NO) was assessed in MN heterozygotes with a flow cytometic assay. The GSTM1 and GSTT1 genotypes were assessed by a multiplex PCR method. The GPA NN phenotype frequency was higher in occupationally exposed group (n=14, mean$\pm$S.D. 6.6$\pm$12.0 in 10/SUP 6/ erythrocyte cells) than in non-exposed group (n=22, 2.1$\pm$3.5). Similarly, the GPA(NO or NN) phenotype frequency was higher in exposed group (n=14, 9.7$\pm$17.3) than non-exposed group (n=22, 4.2$\pm$6.3). The above differences failed to reach statistical significance, but a significant increase was seen in GPA variant frequency levels with increase in urinary 1-OHPG levels (Spearman's correlation: p=0.06 (NO), p=0.07 (NO or NN)). When this association was evaluated by GSTM1 genotype status, the association between GPA mutation and urinary 1-OHPG levels was stronger in individuals with GSTM1 present genotype (Spearmans correlation; r=0.50, p=0.02). These results suggest that the association between urinary 1-OHPG and GPA mutation is be modulated by the GSTM1 genotype.

  • This study evaluated the concentrations of urinary metabolites of polycyclic aromatic hydrocarbons (PAHs) in industrial waste incineration workers. The effect of genetic polymorphisms of xenobiotic metabolism enzymes on urinary concentration of PAH metabolites was assessed. And, aromatic DNA adduct levels were also determined in total white blood cells. Fifty employees were recruited from a company handling industrial wastes located in Ansan, Korea: non-exposed group (n=21), exposed group (n=29). Sixteen ambient PAHs were determined by GC/MSD (NIOSH method) from personal breathing zone samples of nine subjects near incinerators. Urinary 1-hydroxypyrene glucuronide (1-OHPG), a major pyrene metabolite, was assayed by synchronous fluorescence spectroscopy after immunoaffinity purification using monoclonal antibody 8E11 (SFS/IAC). Multiplex PCR was used for genotyping for GSTMI/TI and PCR-RFLP for genotyping of CYP1A1 (MspI and Ile/Val). PAH-DNA adducts in peripheral blood WBC were measured by the nuclease P1-enhanced postlabeling assay. Smoking habit, demographic and occupational information were collected by self-administered questionnaire. The range of total ambient PAH levels were 0.00-7.00 mg/㎥ (mean 3.31). Urinary 1-OHPG levels were significantly higher in workers handling industrial wastes than in those with presumed lower exposure to PAHs (p=0.006, by Kruskal-Wallis test). There was a statistically significant dose-response increase in 1-OHPG levels with the number of cigarettes consumed per day (Pearson correlation coefficient=0.686, p<0.001). Urinary 1-OHPG levels in occupationally exposed smoking workers were highest compared with non-occupationally exposed smokers (p=0.053, by Kruskal-Wallis test). Smoking and GSTMI genotype were significant predictors for log-transformed 1-OHPG by multiple regression analysis (overall model R²=0.565, p<0.001), whereas smoking was the only significant predictor for log-transformed aromatic DNA adducts (overall model R²=0.249, p=0.201). Aromatic DNA adducts was also a significantly correlation between log transferred urinary 1-OHPG levels (pearson's correlation coefficient=0.307, p=0.04). However, the partial correlation coefficient adjusting for Age, Sex, and cigarette consumption was not significant (r=0.154, p=0.169). The significant association exists only in individuals with the GSTMI null genotype (pearsons correlation coefficient=0.516, p=0.010; partial correlation coefficient adjusting for age, sex, and cigarette consumption, r=0.363, p=0.038). Our results suggest that the significant increase in urinary 1-OHPG in the exposed workers is due to higher prevalence of smokers among them, and that the association between urinary PAH metabolites and aromatic DNA adducts in workers of industrial waste handling may be modulated by GSTMI genotype. There results remain to be confirmed in future larger studies.

Bioassays of Polycyclic Aromatic Hydrocarbons in ZR-75-1 Human Breast Cancer Cells

  • Polycyclic aromatic hydrocarbons (PAH) are frequently detected in food, water, soil, and sediment and are widespread environmental pollutants formed by the incomplete combustion of fossil fuels, woods and other organic matter. PAHs are considered to be probable human carcinogens. The mechanism of action of PAHs has been studied extensively, however it is not clear how PAHs turn on CYP1A1 in human breast cancer. Our laboratory have been studied the effect of PAHs in the human breast cancer cell MCF7. In this study, we examined the ZR-75-1 human breast cancer cells as a new system to evaluate bioactivity of PAHs. ZR-75-1 human breast cancer cell line responses to estrogen and progesteron. We have been able to estbilish long term culture system of this cells then used for the study to observe the effect of PAHs. We demonstrate that PAHs induced the CYP1A1 promoter and 7-ethoxyresolufin O-deethylase (EROD) activity in a concentration-dependant manner. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1A1 mRNA. Some of PAHs showed stronger stimulatory effect on CYP1 gene expression than TCDD. Apparently, ZR-75-1 cells have Aryl hydrocarbon receptors, therefore it would be good experimental tool to study the cross-talk between PAHs and steroid actions.

Influence of Roasting Conditions on Polycyclic Aromatic Hydrocarbon Contents in Ground Coffee Bean (원두커피의 로스팅 조건이 polycyclic aromatic hydrocarbons 생성에 미치는 영향)

  • Roasting may lead to the formation of undesired compounds, such as polycyclic aromatic hydrocarbons (PAHs). In this study, green coffee beans were roasted under controlled conditions and the formation of PAHs during the roasting process was monitored. Roasting was performed in a hot air roaster, with an inlet air temperature varying from 150 to $250^{\circ}C$ for 5, 10, and 20 min. The PAH content of the roasted coffee was then evaluated by HPLC-FLD. The levels of total PAHs in Arabica (Colombia, Brazil) and Robusta (India) coffee samples were 1.26-215.07, 1.85-178.14, and 0.18-2.61 ${\mu}g$/kg, respectively.

  • 대향류 확산 화염의 매연 생성 특성에 대한 실험적 연구가 수행되었으며, 그 결과 에틸렌 ($C_2H_4$)-프로판($C_3H_8$) 혼합 연료의 경우 매연 생성 상승 효과 (synergistic effect)가 관측되었다. 프로판과 에틸렌의 PAH 생성 양상이 상이하게 나타났으며, 소량의 프로판을 에틸렌 확산 화염에 첨가할 경우 순수 연료에 비하여 매연 및 PAH (다중 고리 방향족 탄화수소; polycyclic aromatic hydrocarbon) 생성이 증대되었다. 단조적으로 변화하는 아세틸렌($C_2H_2$) 농도와 단열 화염 온도를 고려할 때, 이러한 결과는 HACA (H-abstraction-$C_2H_2$-addition) 반응만으로는 확산 화염에서의 매연 발생 및 성장을 설명할 수 없음을 의미한다. 수치해석과 실험 결과의 비교로부터 초기 PAH의 생성 과정을 규명하였으며 이 과정에서 C3 화학종의 재결합 반웅이 매우 중요함을 확인할 수 있었다. 또한, 이러한 C3 화학종과 C2 화학종의 상호 보완적인 역할에 의하여 에틸렌-프로판 혼합 연료에서 매연 생성이 증대됨을 밝혔다.

Analysis for 16 Polynuclear Aromatic Hydrocarbons (PAH) in Sewage Sludge and Soil (하수슬러지와 토양 중 다핵방향족 탄화수소의 정량)

  • The polycyclic aromatic hydrocarbon (PAH) content in sewage sludge and in farm soils were determined by gas chromatography linked to mass spectrometry (GC/MS) with use 2-ethylantracene as internal standards. Twelve PAH were identified in both sludges with naphthalene ($0.78{\mu}g/g$) being the most predominant in industrial sludge and pyrene ($0.26{\mu}g/g$) in municipal sludge. The total PAR content in industrial sludge and in municipal sludge were $1.74{\mu}g/g$ and $1.19{\mu}g/g$ respectively. PAH were contained in paddy soils and the concentration were very low in the range of $0.01-0.04{\mu}g/g$. The total PAH content in industrial and in municipal sludge were about 9.2 times and 6.3 times greater than in paddy soils ($0.19{\mu}g/g$).

Vertical Distribution and Potential Risk of Particulate Polycyclic Aromatic Hydrocarbons in High Buildings of Bangkok, Thailand

  • Vertical variations of polycyclic aromatic hydrocarbon (PAH) concentrations in $PM_{10}$ were investigated in order to assess the factors controlling their behavior in the urban atmosphere of Bangkok City, Thailand. Air samples were collected every three hours for three days at three different levels at Bai-Yok Suit Hotel (site-1 and site-2) and Bai-Yok Sky Hotel (site-3) in February $18^{th}-21^{st}$, 2008. The B[a]P concentration showed a value 0.54 fold, lower than the United Kingdom Expert Panel on Air Quality Standard (UK-EPAQS; i.e. 250 pg $m^{-3}$) at the top level. In contrast, the B[a]P concentrations exhibited, at the ground and middle level, values 1.50 and 1.43 times higher than the UK-EPAQS standard respectively. PAHs displayed a diurnal variation with maximums at night time because of the traffic rush hour coupled with lower nocturnal mixing layer, and the decreased wind speed, which consequently stabilized nocturnal boundary layer and thus enhanced the PAH contents around midnight. By applying Nielsen's technique, the estimated traffic contributions at Site-3 were higher than those of Site-1: about 10% and 22% for Method 1 and Method 2 respectively. These results reflect the more complicated emission sources of PAHs at ground level in comparison with those of higher altitudes. The average values of incremental individual lifetime cancer risk (ILCR) for all sampling sites fell within the range of $10^{-7}-10^{-6}$, being close to the acceptable risk level ($10^{-6}$) but much lower than the priority risk level ($10^{-4}$).

Exploit the Unexploited : the Potential of the PAH 3.3mm emission feature as a star formation rate proxy and beyond

  • Polycyclic aromatic hydrocarbon (PAH) features have emerged as one of the most important infrared (IR) features since these PAH features dominate mid-IR spectra of galaxies and are ubiquitous within galactic and extragalactic objects. These PAH features have the potentials to become reliable star formation rate (SFR) proxies and diagnostics of physical conditions of interstellar medium, such as ionization states of dust grains and grain sizes. While constructing an unbiased library of 44 sample galaxies selected from 5MUSES sample, AKARI mJY Unbiased Survey of Extragalactic Sources in 5MUSES (AMUSES) intends to measure and to calibrate the PAH 3.3 mm emission feature which has not been studied extensively due to its weak strength and dearth of capable instruments. Out of 20 target galaxies, we detected the 3.3mm feature from eight galaxies and measured their line strengths, line widths and line ratios with other PAH emission features. Sample galaxies whose spectral energy distributions (SEDs) are classified as starburst-type have clearly stronger 3.3mm emission features than ones with AGN-type SEDs. We also found that there is a correlation between the PAH 3.3mm luminosity and total IR luminosity within our sample galaxies, albeit a large scatter. We further discuss implications of our results.
