들깨종실단백질 중의 phytate 제거에 관한 연구 (Studies on the Removal of Phytate from Korean Perilla (Perilla ocimoides, L.) Protein)

  • pH를 달리한 수용액 및 농도를 달리한 염류수용액에서 들깨종실의 단백질과 phytate의 용해도를 측정하여 단백질로부터 phytate를 제거할 수 있는 조건을 검토하였다. 들깨종실단백질의 용해도는 pH4.0에서 가장 낮은 9.5%로 등전점을 보였고, 그보다 산성 또는 알칼리성쪽으로 갈수록 증가되었다. 반면에 phytate의 용해도는 pH5.0에서 가장 높았으며 그보다 산성 또는 알칼리성쪽으로 갈수록 감소되었다. NaCl 수용액을 처리하였을 때 단백질의 용해도는 pH $3.0{\sim}4.0$ 범위에서 가장 낮았고 pH 6.0 이상에서는 현저히 증가되었다. Phytate의 용해도는 pH$2.0{\sim}5.0$ 범위에서는 약 90%내외로 높았으나 pH6.0 이상에서는 급격히 감소되었다. $Na_2SO_3$ 수용액처리에서는 단백질 용해도가 $pH2.0{\sim}3.0$ 범위에서 가장 낮았고 phytate의 용해도는 $pH5.0{\sim}6.0$에서 최대치를 보였고, 3%의 경우는 전 pH 구간에 걸쳐서 낮았으나 5%와 7%에서는 전 구간에서 높았다. $CaCl_2$ 수용액처리에서는 단백질 용해도가 3% 수용액에서는 전 pH 구간에서 낮았으나 5%와 7%에서는 $pH5.0{\sim}10.0$에서 높은 값을 보였으며 phytate의 용해도는 $pH2.0{\sim}3.0$ 사이에서 최대값을 나타내고 pH4.0이상에서는 급격히 감소하였다. 이상의 결과에서 3% NaCl 용액을 사용하여 pH9.0에서 단백질을 추출하고 pH4.0에서 침전시켰을 때 단백질 수율이 좋고 phytate 잔존량이 가장 적어, 저(低)phytate 분리단백질을 만드는 가장 좋은 조건이었다..

Inheritance of P34 Allergen Protein in Mature Soybean Seed

  • Soybean proteins are widely used for human and animal feeds worldwide. The use of soybean protein has been expanded in the food industry due to their excellent nutritional benefits. But, antinutritional and allergenic factors are present in the raw mature soybean. P34 protein, referred as Gly m Bd 30K, has been identified as a predominant immunodominant allergen. The objective of this research is to identify the genetic mode of P34 protein for the improvement of soybean cultivar with a very low level of P34 protein. Two $F_2$ populations were developed from the cross of "Pungsannamulkong" ${\times}$ PI567476 and "Gaechuck2ho" ${\times}$ PI567476 (very low level of P34 protein). Relative amount of P34 protein was observed by Western blot analysis. The observed data for the progeny of "Pungsannamulkong" and PI567476 were 133 seeds with normal content of P34 protein and 35 seeds with very low level of P34 protein (${\chi}^2=1.157$, P=0.20-0.30). For the progeny of "Gaechuck#1" and PI567476, the observed data were 177 seeds with normal content of P34 protein and 73 seeds with very low level of P34 protein (${\chi}^2=2.353$, P=0.10-0.20). From pooled data, observed data were 310 seeds with normal content of P34 protein and 108 seeds with very low level of P34 protein (${\chi}^2=0.156$, P=0.50-0.70). The segregation ratio (3:1) and the Chi-square value obtained from the two populations suggested that P34 protein in mature soybean seed is controlled by a single major gene. Single gene inheritance of P34 protein was confirmed in 32 $F_2$ derived lines in $F_3$ seeds, which were germinated from the low level of P34 protein obtained from the cross of "Pungsannamulkong" and PI567476. These results may provide valuable information to breed for new soybean line with low level of P34 protein and identification of molecular markers linked to P34 locus.

콩 종자에서 쿠니츠트립신인히비터와 P34 단백질의 유전 (Inheritance of Kunitz Trypsin Inhibitor and P34 Protein in Soybean Seed)

  • 인체내에서 소화불량 및 알레르기 반응을 일으킴으로써 콩의 품질을 저하시키는 물질인 Kunitz trypsin inhibitor(KTI) 단백질이 결핍되고, P34 단백질을 적게 함유하는 콩 계통을 선발하기 위해서, 현재까지 보고되지 않은 KTI 단백질의 유무와 P34 단백질 함량간의 유전관계에 대한 정보를 얻기 위하여 본 연구에서 얻어진 결과는 다음과 같다. 1. 07B1과 PI567476의 교배를 통해 얻어진 479개의 $F_2$ 종자를 대상으로 SDS-PAGE를 이용한 KTI 단백질의 유무를 확인한 결과, KTI 단백질이 존재하는 종자의 수는 353개, 결핍된 종자는 126개로 3 : 1로 분리하였다. 2 Western blot을 이용한 P34 단백질의 함량을 확인 한 결과, P34 단백질 함량이 보통 또는 높은 종자가 363개, 함량이 낮은 종자는 116개로 P34 단백질 함량에 대한 유전분리비는 3 : 1로 나타났다. 3. 전체 $F_2$ 종자 479개 중에서 KTI 단백질이 존재하며, P34 단백질 함량이 보통 또는 높은 종자가 266개, KTI 단백질이 존재하고 P34 단백질 함량이 낮은 종자가 88개, KTI 단백질이 결핍이고 P34 단백질함량이 보통 또는 높은 종자가 102개, KTI 단백질이 결핍이며 P34 단백질 함량이 적은 종자가 23개로 9:3:3:1의 분리비에 적합하여 KTI 단백질 유무와 P34 단백질 함량간에는 독립유전을 하였다.

분리 강남콩 단백질의 유화특성에 관한 연구 (A study of the Emulsifying Properties of Kidney Bean Protein Isolate)

  • This study was carried out in order to study the emulsifying properties of kidney bean protein isolate. Kidney bean protein isolate was tested for the purpose of finding out the effect of pH, addition of NaCl, and heat treatment on the solbulity and emulsion capacity, emulsion stability, surface hydropobicity and emulsion viscosity. The results were summarized as follows. 1 The solubility of kidney bean protein isolate was affected by pH and showed the lowest value at pll 4.5 which is isoelectric point of kidney bean isolate. When the kidney bean protein isolate was heated, the highest value observed at pH 2 and pH 7 was 96.11%, 97.41% respectively. 2. The emulsion capacity of kidney bean protein isolate was not significantly different with each pH. With addition of NaCl, emulsion capacity decreased steadily. When heated thr highest value observed at pH 2 and pH 7 was 82.91 ml oil/100 mg protein ($60^{\circ}C$), 82.08 m1 oil/100 mg protein ($80^{\circ}C$) respectively. 3. The emulsion stability was significantly higher at pH 4.5 than that of pH 2 and pH 7 (p 0.05) When NaCl was added, emulsion stability was generally increased after 2hrs. When heated, the highest value observed at pH 2 and pH 7 was 21.25% ($80^{\circ}C$),23.7%($100^{\circ}C$) respectively after 2hrs. 4. Surface hydrophobicity increased sharply as 0.2 M NaCl was added to pH 4.5. When heated, the surface hydrophobicity increased as the temperature increased. 5. The highest value of emulsion viscosity was observed at pH 4.5 and pH 7 when 0.2 M NaCl was added. Under heat treatment, the highest value was 48,000 cps at pH 4.5 ($40^{\circ}C$). In the case of pH 7, the highest value was 105,000 cpa at $100^{\circ}C$.

두경부 편평세포암종에서 p53, p21, EGFR 및 c-erbB-2 발현의 임상적 의의 (Clinical Significance of the Expression of p53, p21, EGFR and c-erbB-2 in Squamous Cell Carcinoma of the Head and Neck)

  • Background and Objectives: Because of squamous cell carcinoma of the head and neck undergoes a generally poor hospital course, the prognostic significance of the squamous cell carcinomas in head and neck have been evaluated to identify those features associated with aggressive biologic behavior according to the immunologic and histopathologic characteristics. Materials and Method: To assess the significance of EGFR, c-erbB-2, p21 and p53 protein in head and neck tumors and their correlation with prognostic factors, samples from 74 patients with squamous cell carcinomas of larynx, pharynx, and oral cavity were studied immunohistochemically. Results: EGFR, c-erbB-2, p21, and p53 protein were expressed 94.6%, 24.3%, 85.1%, and 55.4% in 74 cases of head and neck squamous cell carcinoma, respectively. The positive expression of EGFR was associated significantly with clinical stage and the negative expressions of p21 was associated significantly with histopathologic differentiation. There were no significant relationships between the reactivity of EGFR, c-erbB-2, p21, and p53 protein. Conclusion: The expression of EGFR, c-erbB-2, p21 and p53 protein could be related to oncogenesis, and the expression of p21 and EGFR protein can be used as a prognosticator in head and neck squamous cell carcinoma under certain limitations, but c-erbB-2 and p53 protein expression alone is not enough for evaluating prognosis of the carcinoma.

콩단백질과 고기단백질 식이가 인체내 단백질과 칼슘.인.마그네슘 대사에 미치는 영향 : 중기간 급식 효과 (Effects of Soy Protein and Meat Protein Diets on Protein, Calcium, Phosphorus, and Magnesium Metabolism in Young Korean Adults : Mid-Tern Feeding Study)

  • A 44-day metabolic study was conducted to compare the effects of soy protein and meat protein on N, Ca, P, and Mg metabolism in young Korean males. Meat protein diet(MPD) and soy protin diet(SPD) were provided for 3 weeks each. For MPD, meat protein Supplied 50.3% of total protein and mean daily suplies of Ca, P, and Mg were 33.28mg, 1013.01mg, 226.81mg respectively. For SPD, soy protein supplied 51.8% of total protein and mean contents of Ca, P and Mg were 737.86mg, 1261.76mg, and 451.3mg. Mean daily fecal and urinary excretions of Ca were 350.6mg and 113.3mg for MPD and 515.0mg and 118.3mg for SPD. Apparent absorption rates in MPD and SPD were 4.2mg in MPD and 700.4mg and 475.1mg SPD. Apparent absorption rates were 65.6% in MPD and 44.5% in SPD(p<0.01). For Mg, fecal and urinary excretions were 144.8mg and 81.2mg in MPD and 292.4mg and 65.5mg in SPD. Apparent absorption rates were 37.5% in MPD and 35.2% in SPD. The correlations between urinary Ca, P, and their balance were significant (p<0.001) in MPD, but not in SPD. Urinary Ca was significantly correlated with urinary N, P, Mg(p<0.001) in MPD, but not in SPD. From these results, meat protein seems to increase the excretions of major minerals compared to soy protein.

흰쥐에서 발암물질로 유발된 갑상선 종양과 p21 및 p53 단백질의 발현 (Development of Thyroid Tumors by Carcinogens and Its Expression of p21 & p53 Protein in Rats)

  • Objectives: The development of thyroid tumor has a relationship with carcinogen, oncogene and tumor suppressor gene. With aminotriazole, radioactive iodine and nitrosomethylurea as carcinogens in rat, authors investigate the incidence in type of the thyroid tumors, p21 and p53 protein expression pattern by immunohistochemical stain and the relationship between the tumors and p21-p53 protein expressions. Materials and Methods: 80 experimental animals were divided into four groups; group 1(control, no carcinogen, n=20), group 2(oral administration of aminotriazole for 36 weeks, n=20), group 3(intraperitoneal injection of 131I for one time and oral administration of aminotriazole for 36 weeks, n=20), group 4(oral administration of nitrosomethylurea for 3 days and aminotriazole for 36 weeks, n=20). After 40 weeks they were sacrificed with pathologic examination and we performed immunohistochemical staining with pan-ras monoclonal antibody for p21 protein and CMI polyclonal antibody for p53 protein with paraffin-embedded specimens. Results: 1) No tumors were observed in group I, but 38.3% of nodular goiters, 11.7% of adenomas and 50.0% of carcinomas were observed in carcinogen treated groups(group 2, 3, 4). 2) The incidence of nodular goiter, adenoma and carcinoma were 70%, 20% and 10% in group 2, 40%, 15% and 45% in group 3 and 5%, 0% and 95% in group 4. 3) p21 protein was not expressed in normal thyroid tissues but was expressed in 26.1% of nodular goiters, 42.9% of adenomas and 6.7% of carcinomas. On the other hands, p53 protein was not expressed in normal thyroid tissues, nodular goiters, adenomas and in well differentiated thyroid carcinomas by immunohistochemical stain. Conclusion: The authors suggest that aminotrizole, 131I, nitrosomethylurea can be etiologic agents in the development of thyroid tumor and the p21 protein can be expressed in the early stage and in benign condition of thyroid tumor but p53 protein is not expressed in all conditions of development in rats.

동일 축사내에서 Aluminum Sulfate 처리와 단백질 수준에 따른 육계의 생산성과 암모니아 가스 발생 및 깔짚내 수용성 P의 함량에 미치는 영향 (Effect of Dietary Protein Levels and Addition of Alumincum Sulfate to Litter on Broiler Performance, Ammonia Production and Soluble Phosphorus Content of Litter)

  • In this study, broilers were raised up to 6 weeks of age in a single room to determine if different levels of dietary protein or addition of aluminum sulfate[alum, $Al_2$(SO$_4$)$_3$ㆍ14$H_2O$] to the litter affected growth performance, production of ammonia(NH$_3$) and soluble phosphorus(SP) content of the litter.The experimental treatments consisted of six treatments in a 2x3 factorial arrangements: T$_1$=23% protein + 0.2% alum to litter; T$_2$=21% protein + 0.2% alum to litter; T$_3$=19% protein + 0.2% alum to litter; T$_4$=23% protein + no alum; T$_{5}$=21% protein + no alum; T$_{6}$=19% protein + no alum. For broiler performance, there was no effect of alum addition to the litter, but the dietary protein levels significantly affected feed intake from days 22 to 42(P<0.05) and day 0 to 42(P< 0.05), weight gain during all periods(P<0.05 or 0.01), and feed:gain from day 0 to 21(P<0.05) and day 0 to 42(P<0.05). Alum addition to the litter did not affect body weight at 21 and 42 days, but dietary protein levels has a significant effect on it at both 21(P<0.0l) and 42 days(P<0.05). 우리나라 폐경전 여성에서 칼슘, 단백질, 인의 섭취상태가 골밀도에 미치는 영향 (Effects of Dietary Calcium, Protein, and Phosphorus Intakes on Bone Mineral Density in Korean Premenopausal Women)

  • Effects of dietary calcium(Ca), protein, and phosphorus(P) intake on bone mineral density (BMD) were investigated in 129 Korean premenopausal women(age 31-54 years) without diagnosed disease. BMD was measured at the spine(vertebrae L2-4) and femur(neck, Ward's triangle and trochanter). By stepwise multiple regression analysis it was shown that protein, Ca, and P intakes affected most significantly on BMD at the vertebrae L2-4, protein and P intakes affected most significantly on BMD at the femoral neck and Ward's triangle, and body mass index(BMI) affected most significantly on BMD at the trochanteric region. When ate-matched BMD % at the vertebrae L2-4 and all femoral sites was grouped by three levels(<90%, 90-99%, >=100%), only at the vertebrae L2-4>=100% and 90-99% groups had higher Ca intakes than <90% groups. When Ca, protein and P intakes of the recommended level for Korean(RDA) were grouped by three levels (Ca or P ; <=650mg/d, 650-750mg/d, >=750mg/d, Protein ; <=55g/d, 55-60g/d, >=65g/d), only at the vertebrae L2-4>55g/d of protein intake had higher age-matched BMD % than <=55g/d intake, >=750mg/d of Ca and P intakes, age-matched BMD % than <=650mg/d. In RDA range of Ca, protein, and P intakes, age-matched BMD % of the vertebrae L2-4 and all femoral sites was greater than 90%. Correlation between Ca intake and vertebral BMD was examined closer. There was more significant linear correlation between vertebral BMD and Ca intake below 800mg/d(r=0.346, p<0.0001)than above(r=0.376, p<0.019), implying a threshold effect and vertebral BMD was better expressed as a function of the logarithm of calcium intake(r=0.3881, p<0.0001). These results suggest that Ca, protein, and P intakes greater than RDA help to maintain proper BMD in middle-aged prementopausal women. Especially dietary Ca have important role in increasing the vertebral BMD and 800mg/d of Ca intake is optimum amount.

단백질 추출 pH가 참깨 농축단백질의 기능적 특성에 미치는 영향 (The Effect of Protein Extraction pH on the Functional Properteis of Seasame Protein Concentrates)

  • Sesame protein concentrate(SPC) was prepared from defatted sesame flour(DSF) at several different pH(2.0, 7.0, 9.0, 11.0) for protein extraction. Some of their functional properties were determined in order to compare the effects of pH during preparation of concentrates. Compared with DSF, nitrogen solubility was markedly improved in all SPC, and SPC extracted at pH 11.0 showed the highest solubility at all pH leaves examined. Fatabsorption was increased in all SPC prepared, but water absorption was decreased as the extraction pH of protein increased. The emulsifying properteis and foaming properties of SPC were remarkably higher than DSF. As the extraction pH of protein was increased, the emulsion activity was also increased, but emulsion stability was decreased. SPC extracted at pH 7.0 showed the highest foaming capacity on the other hand, the highest foaming stability was shown in SPC extracted at pH 2.0. As the protein extraction pH increased, the viscosity of the protein solution was increased. SPC extracted at pH 11.0 showed highest viscosity at all protein concentrations tested.
