장수진흙버섯 자실체에서 메탄올을 이용해 추출한 물질의 폴리페놀 함량을 측정한 결과 4.2 mg/g으로 확인되었다. 장수진흙버섯 추출물의 B16/F10 melanoma와 NIH3T3 세포주에 대한 독성시험에서 이들 공시세포는 $200{\mu}g/mL$ 농도에서 50% 이상 생존하여 세포독성은 없는 것으로 확인되었다. DPPH 전자공여능 시험에서 장수진흙버섯 추출물의 항산화 효과는 농도 의존적으로 나타났으며 양성대조군인 BHT에 비해 우수하였다. 장수진흙버섯 추출물을 B16/F10 melanoma 세포에 150 mg/mL 처리하여 세포내 티로시나아제의 저해능을 조사한 결과, 양성대조군인 arbutin은 31.2%의 저해능을 보인 반면, 같은 농도의 장수진흙버섯 추출물 처리군의 저해능은 46.0%로 나타나 장수진흙버섯 추출물의 티로시나아제의 저해능이 양성대조군 보다 높았다. 또한 장수진흙버섯 메탄올 추출물을 B16/F10 melanoma 세포에 처리하여 멜라닌 생성량을 조사한 결과 $100{\mu}g/mL$ 농도 처리군의 멜라닌 생성량은 대조군의 31.3%로 같은 농도의 arbutin을 처리한 군의 43.9%에 비해 효과가 높았으며, 투여한 추출물의 농도가 증가함에 따라 멜라닌의 생성량은 농도 의존적으로 감소하였다. 또한 장수진흙버섯 메탄올 추출물을 분광광도계를 이용하여 자외선 영역에서의 흡수 스펙트럼을 조사한 결과 270-370 nm의 자외선 파장에서 흡광도가 높아 자외선 차단효과가 우수하였다. 따라서 장수진흙버섯의 메탄올 추출물은 피부미백이나 자외선차단 기능성 화장품으로 개발의 가능성이 높다고 사료되었다.

Background: Surgery is the corner stone for the management of rectal cancer. The purpose of this study was to demonstrate the optimal time of surgical resection after the completion of neoadjuvant chemo-radiotherapy (CRT) in treatment of locally advanced rectal cancer. Materials and Methods: This study compared 2 groups of patients with locally advanced rectal cancer, treated with neoadjuvant CRT followed by surgical resection either 6-8 weeks or 9-14 weeks after the completion of chemo-radiotherapy. The impact of delaying surgery was tested in comparison to early surgical resection after completion of chemo-radiotherapy. Results: The total significant response rate that could result in functional preservation was estimated to be 3.85% in group I and 15.4% in group II. Some 9.62% of our patients had residual malignant cells at one cm surgical margin. All those patients with positive margins at one cm were in group I (19.23%). There was less operative time in group II, but the difference between both groups was statistically insignificant (P=0.845). The difference between both groups regarding operative blood loss and intra operative blood transfusion was significantly less in group II (P=0.044). There was no statistically significant difference between both groups regarding the intra operative complications (P=0.609). The current study showed significantly less post-operative hospital stay period, and less post-operative wound infection in group II (P=0.012 and 0.017). The current study showed more tumor regression and necrosis in group II with a highly significant main effect of time F=61.7 (P<0.001). Pathological TN stage indicated better pathological tumor response in group II (P=0.04). The current study showed recurrence free survival for all cases at 18 months of 84.2%. In group I, survival rate at the same duration was 73.8%, however none of group II cases had local recurrence (censored) (P=0.031). Disease free survival (DFS) during the same duration (18 months) was 69.4 % for patients in group I and 82.3% for group II (P=0.429). Conclusions: Surgical resection delay up to 9-14 weeks after chemo-radiation was associated with better outcome and better recurrence free survival.
Blood biochemical and histopathological changes in vital organs of rabbits were studied after 19 wk of feeding composite diets (75 concentrate : 25 roughage) incorporating either urea (2%, wt/wt) ammoniated or alkali (1.5%, wt/wt) treated neem kernel meal (NKM) replacing peanut meal protein of control diet by either 50 or 100%. The blood biochemical constituents (Haemoglobin, Alanine amino transferase, Aspartate amino transferase, Total protein, Blood urea nitrogen &Cholesterol) in rabbits fed on processed NKM diet at either levels, were comparable to the values of thos on control diet except a lowered (p < 0.05) blood glucose concentration in processed NKM fed rabbits as compared to that in control diet fed ones. Histological examination revealed increased goblet cell activity, stunting of jejunal villi, mild tubular degeneration in kidney and hepatic fibro-cellular reaction in rabbits fed on urea ammoniated and alkali treated NKM diets with less marked changes in the latter. Testicular changes with variable degree of disorganization and vacuolation of spermatogonial cells were noticed in rabbits fed higher levels of urea-ammoniated and alkali treated NKM. Thus, alkali treatment and urea-ammoniation were effective in detoxification of meal, but the processing technology is to be further perfected to prevent cumulative effect of residual neem bitters in long term feeding.
This study was undertaken in an effort to product embryos through in vitro maturation(IVM), in vitro fertilization(IVF) and in vitro culture(IVC) after cryopreservation of immature and mature porcine oocytes. The experiments were conducted to investigate IVM rate of oocytes frozen with 3 different cryoprotectants and to examine IVF and IVC of frozen-thawed oocytes. The CEI(cumulus cells expansion index) after IVM of frozen-thawed immature oocytes was higher in oocytes frozen with PG+PEG(propylene glycol plus polyethylene glycol) than those frozen with single cryoprotectant and this index was almost 90% of unfrozen oocyte's index(2.39 vs. 2.66). The IVF rate of all frozen oocytes was very low(68% of unfrozen oocytes) and the IVF rate of frozen immature oocytes was slightly higher than that of frozen mature oocytes(39.0% vs. 34.4%), but polyspermic penetration was higher in frozen immature oocytes(21.9% vs. 19.1%). The cleavage rate after IVF of frozen-thawed oocytes was 9.3% for frozen mature oocytes and 11.3% for frozen immature oocytes and this rate was significantly lower(P<0.05) than that of control(60.7%). The development to 8-cell stage was greatly lower in frozen mature oocytes than in frozen immature oocytes. The results indicate that the use of PG plus PEG as cryoprotectant may be very effective for vitrification of porcine oocytes and the frozen-thawed immature porcine oocytes can be used fro in vitro embryo production based on IVM, IVF and IVC system.
There is a significant amount of experimental evidence suggesting that polysaccharides from mushrooms enhance the host immune system by activating various mechanisms in immune cells, including macrophages. In this study, polysaccharides from Lentinus edodes were found to stimulate the functional activation of macrophages to secrete inflammatory mediators and cytokines and increase the phagocytotic uptake. The chemical properties of the stimulatory polysaccharides, CPFN-G-I, CPBN-G, and CPBA-G, were determined based on their monosaccharide composition, which mainly consisted of glucose and mannose. According to FT-IR and GC/MS, the structure of CPFN-G-I, purified from the fruiting body of L. edodes, was found to consist of a $\beta$-1,6-branched-$\beta$-1,4-glucan, whereas CPBN-G and CPBA-G, purified from the liquid culture broth, were found to be composed of a heteromannan. The configuration of the p-linkage and triple helical conformation of each polysaccharide were confirmed using a Fungi-Fluor kit and Congo red, respectively.
This study was conducted to investigate the specific expression genes in the cloned bovine tissues. Donor cells, cloned tissues were analysed by RAPD-RFLP method. The results were detected three genes (CH-U7B, CH-U7M and CH-U7P) in the cloned fetus. It was found a single copy genes by southern hybridization. Sequence analysis of CH-U7M gene was shown 99% homology to a previously reported EST from a cloned bovine fetus. The putative ORF was encode a protein of hydrophobicity index 0.03. Semi-quantitative RT-PCR by using the CH-LS001 specific primer was remarkably detected in the lung tissue of cloned fetus. Further investigation of these genes may provide one of the key information to explain the early death, abnormal fetus, large off-spring and the low pregnancy rate in the production of cloned bovine.
Microalgal biofuel production from wastewater has economic and environmental advantages. This article investigates the lipid production from high chemical oxygen demand (COD) bioethanol wastewater without dilution or additional nutrients, using a newly isolated heterotrophic microalga, Chlorella vulgaris LAM-Q. To enhance lipid accumulation, the combined effects of important operational parameters were studied via response surface methodology. The optimal conditions were found to be temperature of $22.8^{\circ}C$, initial pH of 6.7, and inoculum density of $1.2{\times}10^8cells/ml$. Under these conditions, the lipid productivity reached 195.96 mg/l/d, which was markedly higher than previously reported values in similar systems. According to the fatty acid composition, the obtained lipids were suitable feedstock for biodiesel production. Meanwhile, 61.40% of COD, 51.24% of total nitrogen, and 58.76% of total phosphorus were removed from the bioethanol wastewater during microalgal growth. In addition, 19.17% of the energy contained in the wastewater was transferred to the microalgal biomass in the fermentation process. These findings suggest that C. vulgaris LAM-Q can efficiently produce lipids from high-concentration bioethanol wastewater, and simultaneously performs wastewater treatment.
To develop potent anitumor polysac-charides from higher hungi of Korea the alcohol precipitated polysaccharides, which were obtained from hot water extracts of shake-cultured mycelia of eight species of basidiomycetes, were subjected to antitumor screening tests, using sarcoma 180 as a tumor cell. When administered i. p, at the dose level of 20mg/kg/day for ten days into the mice which were implanted with $1{times}10^{8}$ cells of sarcoma 180 twenty-four hour before the start of polysaccharide injection, the polysaccharides of Plutcus cervinus ISA-Pc-1004, Pleurotus pulmonarius, Hydnum repandum ISA-Hr-1006, and Laccaria laccata ISA-LI-1008 respectively showed inhibition ratios of 62.0%, 51.7%, 64.8%, and 57.8%. They were selected for further study, while those of Pancillus serotina ISA-Ps-1012, Laccaria amethystina ISA-La 1001, Pholiota adiposa ISA-Pa-1010, and Lepista spISA-Ls-1007 respectively showed inhibition ratios of -9.3%, 25.3%, 27.3% and 33.0%.
Seven essential oils were tested for in vitro cytotoxicity against the cancerous cell lines A-549, HEp-2 and DLA and normal BRL-3A, NRK-49F and Vero cell lines using standard MTT, SRB and dye exclusion techniques. The A-549 cell line was found to be the most susceptible to all the essential oils. The essential oils of A. nilagirica, A. calamus and O. sanctum were found to be the more active against these cells with mean $CTC_{50}$ values of 17.75, 19.00 and $24.37\;{\mu}g/ml$, respectively. The essential oil of Acorus calamus was found to be the most potent with low $CTC_{50}$ values against the cancerous and comparatively higher $CTC_{50}$ values against the normal cell lines. Artemisia pellens and Pelargonium graveolens oils also showed potent activity. These oils merit further investigation to identify the active principles and nature of the anti tumor activity in animal models.
Synurus deltoides (Aiton) Nakai, belonging to the Compositae family, is an edible plant widely distributed in Northeast Asia. In this study, we examined the mechanisms underlying the immunomodulative effects of the ethanol extract of S. deltoides (SDE). The SDE extract strongly down-regulated the mRNA expression of the inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and tumour necrosis factor (TNF)-${\alpha}$, thereby inhibiting the production of nitric oxide (NO), prostaglandin E2 (PGE2), and TNF-${\alpha}$ in the lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Furthermore, SDE also suppressed the nuclear translocation of the activation protein (AP)-1 and the nuclear factor-${\kappa}B$ (NF-${\kappa}B$), and simultaneously decreased the phosphorylation of extracellular signal-regulated protein kinases (ERK), p38, and Akt. In agreement with the in vitro observations, the orally administered SDE ameliorated the acute inflammatory symptoms in the arachidonic acid-induced ear edema and the EtOH/HCl-induced gastritis in mice. Therefore, S. deltoides have a potential anti-inflammatory capacity in vitro and in vivo, suggesting the potential therapeutic use in the inflammation-associated disorders.
