This study was carried out to optimize the fermentation conditions for direct alcohol fermentation of xylose by Pichia stipitis CBS 5776. The best cell growth and the ethanol production were obtained under 0.05 VVM aeration and 300rpm agitation at $30^{\circ}C$ using 100 g/l xylose medium of the initial pH 5.0. In the above condition, the maximum specific growth rate and maximum cell concentration were 0.14hr-1 and $1.3 \times109$ cells/ml, respectively. Pichia stipitis CBS 5776 also produced 40.2g/l ethanol utilizing about 96% of 100g/l xylose after 72hr fermentation. At this point, the overall volumetric ethanol productivity was 0.56g/1-hr and the ethanol yield was 0.42 g-ethanol/g-xylose consumed, which corresponds to 82% of the theoretical yield.
The efficiency of acid, enzyme and microbial pretreatment of rice bran was compared based on the content of cellulose, hemicellulose, reducing sugars and xylose in the substrate. An isolate of Aspergillus niger or a strain of Trichoderma viride(MTCC 800) was employed for microbial pretreatment of rice bran in solid state. Acid pretreatment resulted in the highest amount of reducing sugars followed by enzyme and microbial pretreatment. A. niger showed a higher rate of hydrolysis than T. viride. The rice bran hydrolysate obtained from the different methods was subsequently fermented to ethanol either by Zymomonas mobilis(NCIM 806) or by Pichia stipitis(NCIM 3497). P. stipitis fermentation resulted in higher ethanol(37% higher) and biomass production($76{\sim}83%$ higher) than those of Z. mobilis. Maximum ethanol production resulted at 12h in Zymomonas fermentation, while in Pichia fermentation, it was observed at 60h. Microbial pretreatment of rice bran by A. niger followed by fermentation employing P. stipitis was more efficient but slower than the other microbial pretreatment and fermentation.
Glucose and xylose are the most abundant materials in nature which can be used to produce ethanol by yeast fermentation. Three combinations of cultivation with glucose and xylose were carried out; separated, co-culture, and sequential fermentation with Saccharomyces cerevisiae and Pichia stipitis. In the separated fermentation, S. cerevisiae fermented glucose to produce 14.5 g/l ethanol from 29.4 g/l glucose but hardly used xylose. However, P. stipitis utilized not only glucose but also xylose to produce ethanol 11.9 g/l and 11.6 g/l from 29.4 g/l glucose and 29.0 g/l xylose, respectively. In the mixture of glucose and xylose, P. stipitis fermented both sugars, producing 21.1 g/l ethanol while S. cerevisiae fermented only glucose, producing 13.4 g/l ethanol. In the co-culture and sequential fermentation, the co-culture showed more efficient ethanol productivity with 18.6 g/l ethanol than the sequential fermentation with 12.4 g/l ethanol. To investigate the effect of nutrients in the growth of microorganisms and ethanol production, yeast nitrogen base (YNB) was used in the sequential fermentation with S. cerevisiae and P. stipitis. YNB supplemented some nutrients which S. cerevisiae used up in the broth and the culture showed increased growth rate, increased consumption of xylose, and increased ethanol productivity producing 22.5 g/l ethanol from 54.6 g/l sugar with a yield of 0.41 g/g.

The waste seaweed from Gwangalli beach, Busan, Korea was utilized as biomass for ethanol production. Sagassum fulvellum (brown seaweed, Mojaban in Korean name) comprised 72% of the biomass. The optimal hyper thermal acid hydrolysis conditions were obtained as 8% slurry contents, 138 mM sulfuric acid, and $160^{\circ}C$ of treatment temperature for 10 min with a low content of inhibitory compounds. To obtain more monosaccharides, enzymatic saccharification was carried out with Viscozyme L for 48 h. After pretreatment, 34 g/l of monosaccharides were obtained. Pichia stipitis and Pichia angophorae were selected as optimal co-fermentation yeasts to convert all of the monosaccharides in the hydrolysate to ethanol. Co-fermentation was carried out with various inoculum ratios of P. stipitis and P. angophorae. The maximum ethanol concentration of 16.0 g/l was produced using P. stipitis and P. angophorae in a 3:1 inoculum ratio, with an ethanol yield of 0.47 in 72 h. Ethanol fermentation using yeast co-culture may offer an efficient disposal method for waste seaweed while enhancing the utilization of monosaccharides and production of ethanol.

This study was carried out in order to investigate the characteristics of xylitol fermentation by a xylitol dehydrogenase defective mutant PXM-4 of P stipitis CBS 5776 and to determime optimum conditions for the high yield ofxylitol production from xylose. Gluconic acid was selected as a co substrate for the xylitol fermentation, since gluconic acid neither blocked xylose transport nor repressed xylose reductase expression. An increase of gluconic acid concentration reduced the rates of xylitol production and cell growth by decreasing medium pH, and the optimal concentration of gluconic acid was determined to be 20 gll with approximately 100% xylitol conversion yield. A fed-batch cell culture resulted in a 44.8 g/l xylitol concentration with 100% yield, based on the amount of xylose consumed.

Suspended wood waste was being inflow into the dam and the problem of waste disposal has been occurred. In this study, ethanol production using woody floater wastes was performed to estimate value in use for raw material of renewable energy. To achieve the goal, experiments of acid hydrolysis and ethanol fermentation using dam woody floater as raw materials for bioethanol was carried out. In the results of field survey in the chungju dam, kind of woody floater was mainly Japanese larch (Larix leptolepis) and hybrid poplar (Populus tomentiglandulosa). The results of sugar extraction showed that sugar content was higher in Larix leptolepis than Populus tomentiglandulosa. Extracted sugar from wood waste was effective consumed by yeast(P. Stipitis and S cerevisiae). In the experiment consumption of sugar including glucose, galactose and xylose, the consumption rate of S. cerevisiae is faster than that of P. stipitis. and efficiency for ethanol production is higer in S. cerevisiae than P. stipitis. Also it can be confirmed that resource as ethanol production using wood waste was available.
P. stipitis CBS5776 was cultivated to examine the characteristics of ethanol fermentation for hexoses (mannose, g1ucose, and galactose) and pentoses(xylose and arabinose). Glucose was the best carbon source among the sugars used in terms of ethanol yield. Glucose was used to produce ethanol with an yield coefficient 0.376g ethanol/g glucose, whereas mannose was converted to produce ethanol with an yield coefficient 0.326g ethanol/g mannose. P. stipitis CBS5776 was also grown in a mixture of sugars to study the pattern of carbon utilization. The yeast utilized glucose and mannose firsts and then galastose and xylose as carbon sources. Arabinose was partially used for biomass when it was present as a sole carbon source, but it was not metabolized at all in a mixture of carbon sources. P. stipitis produced $12.2g/\ell$ ethanol with a yield coefficient 0.332 g ethano1/g sugar in a mixture of sugars.
It is indispensable to increase the conversion rate of a reducing sugars such as pentose and hexose derived from cellulosic wastes for a highly efficient bioethanol fermentation from food wastes. The saccharification liquid from cellulosic substrates such as vegetable food wastes contained lots of hexose like glucose and pentose like xylose. Since Saccharomyces-based yeasts could not convert xylose to bioethanol, Pichia stipitis which could directly ferment xylose to ethanol was chosen. After selecting Saccharomyces coreanus and P. stipitis, fermentation characteristics by mixture of two yeasts were investigated. As a result, it was verified the production of ethanol was enhanced by the co-fermentation, although there were somewhat differences between the fermentation characteristics by the aeration methods. Moreover, the consumption of pentose, hexose and disaccharide was obviously observed, and aeration in the process of fermentation seemed to stimulate the activity of P. stipitis.

Among the tested cosubstrates, galactose was selected as an adequate cosub-strate on xylitol production of mutant PXM-4. But with the increase in the concentration of galactose in the medium, xylitol production was decreased because the transport of xylose into cell was inhibited by galactose. The optimal concentration of galactose for the production of xylitol using 20 g/ι xylose was 20 g/ι Under this condition, maximum concentration of xylitol and yield were 14.4 g/ι and 97%, respectively. In order to prevent the inhibitory effect of xylose transport by galactose, galactose was fed with low concentration and the concentration of xylitol produced was increased up to 25 g/ι.
