Metabolic Engineering of Rational Screened Saccharopolyspora spinosa for the Enhancement of Spinosyns A and D Production

  • Spinosyns A and D are potent ingredient for insect control with exceptional safety to non-target organisms. It consists of a 21-carbon tetracyclic lactone with forosamine and tri-Omethylated rhamnose which are derived from S-adenosyl-methionine. Although previous studies have revealed the involvement of metK1 (S-adenosylmethionine synthetase), rmbA (glucose-1-phosphate thymidylyltransferase), and rmbB (TDP-D-glucose-4, 6-dehydratase) in the biosynthesis of spinosad, expression of these genes into rational screened Saccharopolyspora spinosa (S. spinosa MUV) has not been elucidated till date. In the present study, S. spinosa MUV was developed to utilize for metabolic engineering. The yield of spinosyns A and D in S. spinosa MUV was $244mgL^{-1}$ and $129mgL^{-1}$, which was 4.88-fold and 4.77-fold higher than that in the wild-type ($50mgL^{-1}$ and $27mgL^{-1}$), respectively. To achieve the better production; positive regulator metK1-sp, rmbA and rmbB genes from Streptomyces peucetius, were expressed and co-expressed in S. spinosa MUV under the control of strong $ermE^*$ promoter, using an integration vector pSET152 and expression vector pIBR25, respectively. Here-with, the genetically engineered strain of S. spinosa MUV, produce spinosyns A and D up to $372/217mgL^{-1}$ that is 7.44/8.03-fold greater than that of wild type. This result demonstrates the use of metabolic engineering on rationally developed high producing natural variants for the production.

  • Eight flavonoids were isolated from the BuOH fraction of the 70% EtOH extract of Zizyphus jujuba var. spinosa seeds along with three known compounds, daucosterol (1), butyl ${\beta}$-D-fructofuranoside (2), and magnoflorine (4). On the basis of spectroscopic methods and comparison with literature values their structures were elucidated as 6"'-feruloylspinosin (3), nicotiflorin (5), 6"'-p-coumaroylspinosin (6), isospinosin (7), 6"'-vanilloylspinosin (8), spinosin (9), hovetrichoside C (10), and camelliaside B (11). Compounds 1, 2, 10, and 11 were isolated from this plant for the first time.

Herbicidal and Antifungal Activities of the aqueous extracts of Several Naturalized Plants (수종의 귀화식물 수용성추출물의 제초 및 항균 활성 탐색)

  • The study researched germination of the plants and growth of experimented bacteria according to concentration of water extract in order to provide basic data for developing natural agricultural resources by using naturalized plants including Solidago altissima, Amaranthus retroflexus and Sida spinosa. As concentration of water extract increased, most of test plants showed a decrease in relative germinability. Sida spinosa(r=-0.540, p<0.01), Physalis wrightii(r=-0.693, p<0.01), Amaranthus retroflexu(r=-0.724, p<0.01), Solidago altissima(r=-0.728, p<0.01) and Eclipta prostrata(r=-0.779, p<0.01) showed tendency of decrease in relative germinative power in order, respectively. For average germination period, as concentration of the processed group increased, the time for germination increased (r = 0.769, p<0.01) and according to donor plants and test plants, there was a little difference. Also, as concentration of water extract of donor plant, length of above-aerial part(r=-0.587, p<0.01), length of underground part(r=-0.741, p<0.01), fresh weight(r=-0.574, p<0.01) and generation of root hair decreased. An then, for growth of test fungi according to concentration of water extract of donor plants, growths of Botrytis cinerea(r=-0.266, p<0.05), Diaporthe citri(r=-0.323 p<0.01), Colletotrichum gloeosporioides(r=-0.512, p<0.01), Pythiumultimum(r=-0.581, p<0.01) and Rhizoctonia solani(r=-0.806, p<0.01) were repressed in order, respectively. For total amount of content of phenol with herbicidal and Antifungal activities, S. altissima $17.3{\pm}0.5mg/g$, A. retroflexus $13.1{\pm}0.3mg/g$, P. wrightii $12.0{\pm}0.4mg/g$, S. spinosa $9.5{\pm}0.1mg/g$ and E. prostrata L. $4.1{\pm}0.1mg/g$ showed in order, respectively. As these results are summarized, donor plants which were naturalized, have competitive advantage because they release phenolic compounds with allelopathic effect and affect on germination, growth and fungi growth on underground flora compared to native plants and they have eligibility for natural herbicide and germicide.

  • A systematic study of Korean holaxonians was done with specimens from 15 localities in the coastal seas of Korea for the period of 1965 though 1978. The holaxonians identified in the present study turned out to be 15 species representing seven genera in three familes. They were all new to the Korean gorgonarian fauna: Anthoplexaura dimorpha; Euplexaura anastomosans; E. abietina; E. crassa; E. recta; Ellisella maculata; E. rubra; E. limbaughi; Verrucella umbraculum; v. stellata; Primnoa reseda pacifica; Callogorgia pseudoflabellum; Plumarella spinosa; P. rigida; P. adhaerens.

Morphological Characteristics and Principal Component Analysis of Plums (자두의 형태적 특성과 주성분 분석에 의한 품종군 분류)

  • To examine taxonomic relationships among 53 plums derived from Prunus cerasifera, P. domestica, and P. salicina, principal component analysis (PCA) and cluster analysis on 27 morphological characters were conducted. Of 27 characters, leaf size, leaf shape, and leaf hair were useful characters for plum identification and understanding of taxonomic relationships among them. Leaf length, petiole length, number of leaf nectaries, leaf shape, leaf base, and date of full blooming showed the clear differences between P. salicina group and P. domestica group. Results of cluster analysis using scores of the first three principal components indicated that 53 plums could be grouped into P. salicina-P. cerasifera, P. domestica, and P. spinosa phenon at 1.0 of average distance in UPGMA. Although PCA was useful for rough classification of plums, much more characters were needed for the exact classification.

Density of Arbuscular mycorrhizal spore of plastic film house soil in Yeongnam area and characterestics of AMF in vitro (영남지역 시설재배지에 분포하는 Arbuscular 균근균의 포자 밀도 및 기내조건에서의 포자발아와 균사생장 특성)

  • This study was conducted to obtain the basic data on agricultural use of arbuscular mycorrhizal fungi(AMF) in salt accumulated plastic film house soil by evaluating the density of AMF spores in plastic film house in Yeong Nam area and surface sterility condition, germination rate of AMF spores, and hyphal growth in vitro. The density of AMF spores in plastic film house soils was highest in the site of water melon, and those of cucumber, melon, hot pepper sites were followed in order. The number of AMF was in the range of 101-207 per 100 g dry soil. With decreasing the ratio of bacteria to fungi(B/F), the population density of AMF was increased, and available $P_2O_5$ content of soil was significantly correlated to the population densities of AMF($r=0.416^*$). The surface sterility rate and spore germination of AMF isolated in plastic film house soil were more than 50% in 2% chloramin T and 2% chloramin T + antibiotic and 0.5% NaOCl treatments. The germination rate of Gigaspora margarita in the range of initial pH 5~9 of the medium was more than 56%. Hyphal growth was increased as pH of the medium increased. However the germination rate of Acaulospora spinosa was highest in the medium of pH 9, and hyphal growth in vitro was poor and not related to pH of the medium.

Dynamics of Phytoplankton Community in the Open Water Flowed Through the Shihwa Constructed Wetland from Streams (Banwoul, Donghwa and Samhwa stream) (하천수 (반월천, 동화천, 삼화천)의 시화인공습지 관류에 따른 개방수에서 식물플랑크톤 군집 동태)

  • The Shihwa constructed wetland was established for the treatment of severely polluted water from Banwoul, Donghwa and Samhwa streams, This study was focused on investigating dynamics of phytoplankton community at 5 stations of open waters in the Shihwa constructed wetland from October 2001 to July 2002. The concentration of T-N and T-P of inlet stations from the streams were decreased by flowed through the wetland. However, the TN/TP ratios at all stations were shown as a little over 16 indicating that the T-P plays an important role as a limitation factor. Phytoplankton communities were identified as a total of 413 taxa which were composed of 375 species, 21 varieties, 2 forma and 15 unidentified species. The standing crops of phytoplankton communities and chlorophyll-a concentrations ranged 330 ${\sim}$ 36, 420 cells $mL^{-1}$ and $2.5\;{\sim}\;170.7\;{\mu}g\;L^{-1}$ respectively, and showed a decreasing tendency after flowing through the wetland at almost stations. Dominant species were 14 taxa at all stations which were Euglena oblonga, Synura spinosa, and etc. The species composition, standing crops and chlorophyll concentrations of phytoplankton communities appeared a distinct differences between open waters of inlet from stream and open waters flowed through the wetland. Theses results were affected from decreasing effects of TN, TP and SS by flowed through the wetland from inlet waters of streams.

  • Dynamics of certain $N_2$fixing bacteria such as Rhizobium, Azospirillum and Azotobactor, nodule number in dominant legume, Atylosia trinervia, P-solubilizing bacteria, actinomycetes and fungi were studied in unburned and burned site of natural grassland, southern India. Population of $N_2$- fixing bacteria, P-solubilizing bacteria, fungi and nodule number in legume increased significantly in burned sites. On the other hand, the actinomycetes population remained unchanged. Thirty six species of fungi with tricalcium phosphate solubilizing ability were recorded. The most efficient P-solubilizing fungi recognised in the soils of the study sites are Absidia ramosa, Gongronella butlerii, Mortieralla spinosa, Mucor racemosus, Rhizopus nigricans, R. stolonifer, R. oryzae, Aspergillus fumigatus, A. nidulans, A. niger Theilavia terricola and Cheatomium lunasporium.

Isolation and Enzymatic Characterization of Bacteria from Livestock Manure (축산분뇨에서 분리한 세균의 동정 및 효소학적 특성)

  • To develop the effective composting system, we isolated bacteria that have the abilities to degrade organic matters such as cellulose, carbohydrate, protein and lipid during the compositing of livestock manureAmong 24 strains, 6 bacteria have all the enzymatic activities of protease, amylase, cellulase and lipase.These microorganisms were identified as Corynebacterium varibilis, Bacillus spp., Pseudomonas spinosa,Acetobacter calcoaceticus and Athrobacter cumminsii All the enzymes produced by the bacteria showedactivities at the broad pH range and the maximal activities were obtained at $60^{\circ}C$. It seemed that after theincrease of temperature caused by fermentation of livestock manure, the enzymes started to degrade the rawmaterials, which are added for the control of humidity. However cellulase activity was maximum at $37^{\circ}C$,suggesting that the cellulase-producing bacteria work at an early stage of livestock manure fermentation toprovide the organic material for the growth of other bacteria. The production of the enzymes were growth-associated and maximal activities appeared at the early stationary phase of growth.
