Evaluation of Extruded Pellets and Raw Fish-Based Moist Pellet for Growth of Sub-Adult Flounder Paralichthys olivaceus (미성어기 넙치 사육을 위한 배합사료 및 습사료의 효과 비교)

  • This study was conducted to evaluate extruded pellets(EP) for the growth of sub-adult flounder Paralichthys olivaceus by comparing with raw fish-based moist pellet(MP). Two replicate groups of 150 fish per each tank(initial mean weight $594{\pm}6.5g$) were fed one of two EPs(EP1 and EP2) and a MP for 32 weeks. Survival was not significantly affected by experimental diets. Final mean weight of fish fed the EP1 was not significantly different from that of fish fed the MP, but that of fish fed EP2 was significantly lower than that of fish fed EP1 and MP (P<0.05). Feed efficiency and protein efficiency ratio of fish fed the EP1 and EP2 were significantly higher than those of fish fed MP(P<0.05). Daily feed intake of fish fed the EPs was significantly lower than that of fish fed MP(P<0.05). Condition factor was not significantly different among all groups. The contents of moisture, crude protein and lipid in dorsal muscle were significantly affected by experimental diets(P<0.05). Based on the results of this study, the dietary formulations used in EP1 and EP2 could be applied in the practical extruded pellet feeds for sub-adult flounder(594-1,126 g).

Effects of the Different Fish Meal and Wheat Flour Content in Extruded Pellet on the Growth and Apparent Digestibility for Olive Flounder Paralichthys olivaceus (어분 종류 및 소맥분 함량이 다른 배합사료가 넙치 (Paralichthys olivaceus)의 성장과 소화율에 미치는 영향)

  • A feeding trial was carried out to investigate the effect of different fish meal and wheat flour contents in extruded pellets on the growth and apparent nutrient digestibility of olive flounder Paralichthys olivaceus. Four extruded pellets (HB1, HB1T, HB2 and HB2T) were prepared using two kinds of high quality brown fish meal and low quality tuna fish meal. Two diets (HB1 and HB2) contained two different kinds of brown fish meal, respectively; in HB1T and HB2T, we substituted 50 % of the brown fish meal with 10 % wheat flour. Three replicate groups of fish (initial body weight of 82 g) were fed the experimental pellets to visual satiation for 15 weeks. The highest weight gain was observed in fish fed HB1, and weight gain of fish fed HB2 was significantly higher than that of fish fed HB2T (P<0.05). Feeding efficiency for fish fed HB1 was significantly higher than that of fish fed HB1T and HB2T (P<0.05), but was not significantly different from fish fed HB2. Apparent dry matter digestibility in HB1 and HB2 was significantly higher than in HB1T and HB2T (P<0.05). The highest crude protein digestibility was observed in HB1, and protein digestibility in HB1T and HB2 was higher than in HB2T (P<0.05). The results of this study suggest that fish meal quality considerably affects growth and nutrient digestibility of olive flounder fed extruded pellets, and the use of HB1 is recommended over HB1T, HB2 and HB2T for efficient olive flounder production.

  • This study was conducted to investigate the effect of partially substituting meat meal for fish meal on the growth and body composition of juvenile olive flounder Paralichthys olivaceus during the winter season. Twenty-five fish (initial body weight, 23 g) were distributed into twelve 250 L flow-through tanks. Four experimental diets were prepared in triplicate: the control, MM20, MM40, and MM60 diets. Sixty percent mackerel meal was used as the primary protein source in the control diet. Meat meal was substituted for 20, 40, and 60$\%$ of the mackerel meal in the MM20, MM40, and MM60 diets, respectively. Survival was not significantly affected by the experimental diets. However, the weight gain and specific growth rate of fish fed the control, MM20, and MM40 diets were significantly higher than those of fish fed the MM60 diet (P<0.05). The feed efficiency ratio of fish fed the control, MM20, and MM40 diets was significantly higher than that of fish fed the MM60 diet (P<0.05). The protein efficiency ratio for fish fed the control diet was significantly higher than that for fish fed the MM40 and MM60 diets (P$\%$ substitution of meat meal for fish meal in the diet could be implemented without a reduction in growth or deterioration of the feed efficiency of juvenile olive flounder during the winter season.

Compensatory Growth in Juvenile Olive Flounder (Paralichthys olivaceus) in the Spring

  • A feeding trial was conducted to determine compensatory growth of juvenile olive flounder in the spring. Five treatments of fish with 3 replicates were prepared: the control group fish (C) fed twice daily for 8 weeks, the Sl, S2, S3 and S4 fish fed for 7, 6, 5 and 4 weeks after 1, 2, 3 and 4 weeks of starvation, respectively. Survival of olive flounder was not significantly different among treatments. Weight gain of flounder in S2 was significantly (P<0.05) higher than that of fish in S3 or S4, but not significantly different from that of fish in C or Sl. The poorest weight gain was observed in fish of S4 treatment. Specific growth rate (SGR) for flounder in S2 was significantly (P<0.05) higher than that for fish in C or S4, but not significantly different from that of fish in Sl or S3. Feed intake (g/fish) was proportional to duration of days of feeding except for flounder in S2, but not significantly different among C, Sl or S2. Feed efficiency ratio (FER) and protein efficiency ratio (PER) for flounder in S2 were significantly (P<0.05) higher than for fish in C, but not significantly different from those for fish in Sl, S3 or S4. Hepatosomatic index (HSI) and condition factor (CF) for flounder in Sl, S3 and S4 were not significantly different from those for fish in C, but significantly (P<0.05) lower than S2 except for CF in Sl at the end of the experiment. Proximate composition of the whole body of flounder was not significantly different among treatments. In considering results of the experiment, juvenile olive flounder achieved compensatory growth when properly fed after starved up to 2 weeks in the spring. Compensatory growth of fish was supported by improvement in SGR, FER and PER in fish starved.

The effects of supplementary diets on the water temperture stress in olive flounder, Paralichthys olivaceus (넙치, Paralichthys olivaceus의 수온 자극 스트레스에 대한 사료첨가제 투여 효과)

  • The effect of each dietary supplements (Undaria 5%, Undaria 10%, Obosan 0.5%, Wasabi leaf 2%, Wasabi stem 2%) on the water temperature fluctuation in juvenile olive flounder (Paralichthys olivaceus) was investigated. The response to stress was assessed in terms of effects on haematological and immunological, and resistance against Edwardsiella tarda infection. Plasma glucose and cortisol levels were significantly lower in 5% undaria and 2% wasabi leaf supplement groups than the controls after the first change of water temperature (P<0.05). The plasma lysozyme activities and the survival rates from E. tarda infection were significantly higher in the 5% undaria supplement groups than the control (P<0.05). These results suggest that the 5% undaria supplement seems to be contributable to the increased disease resistance on olive flounder.

  • This study was conducted to evaluate the effect of extruded pellets (EP) containing different levels of protein (51%-55%) and lipid (9%-15%) for growth of flounder (Paralichthys olivaceus) comparing with raw fish-based moist pellet (MP). Two replicate groups of 40 fish per each tank (initial mean weight 106 g) were fed one of three experimental EP (EP1, EP2 and EP3) containing different protein and lipid levels, a commercial EP (EP4) and MP for 16 weeks. Survival was not significantly different among all groups. Final mean weight of fish fed MP was significantly lower than that of fish fed EP1, EP2 and EP4 (P<0.05), but not significantly different from fish fed EP3. Feed efficiency of MP-fed fish was significantly lower than fish fed all EP formulations (P<0.05), but no significant difference was observed among the EP groups. Daily feed intake of MP-fed fish was significantly higher than fish fed all EP formulations (P<0.05). Condition factor was not significantly different among all groups. Whole body moisture and crude lipid contents were significantly affected by diet (P<0.05). Growth and feed efficiency of flounder was not affected by EP protein and lipid levels. Dietary formulation used in EP1, EP2 and EP3 can be applied to the practical feeding of flounder.

Proteases in Cell Lysate of Uronema marinum (Ciliata: Scuticociliatida), an Opportunistic Pathogen of Cultured Olive Flounder (Paralichthys olivaceus)

  • The effects of pH, temperature and various inhibitors on the proteolytic activity of the cell lysate of Uronema marium were investigated using colorimetric and substrate gel electro­phoretic methods. The cell lysate of U. marinum showed proteolytic activity over a wide range of pH, and pH optima ranged from pH 5 to 7. The proteolytic activity was increased according to a rise of temperature but decreased at $40^{\circ}$. The proteolytic activity of the parasite lysate was significantly inhibited by protease inhibitors including trans-epoxysuccinyl -L-leucylamido-(4-guanidino) butane (E-64), pepstatin A, phenyl-methanesulfonyl fluoride(PMSF), and ethylenediamine-tetraacetic acid (EDTA). Preincubation of the lysate with E-64 showed the maximum inhibition of the caseionolytic activity. Four protease bands (152, 97, 67 and 40 kDa) were detected by gelatin SDS-PAGE. Significant inhibition of caseinolytic activity and complete abolition of a 152 kDa band in gelatin SDS-PAGE by EDTA indicated that the cell lysate of U. marinum had a metalloprotease Another three proteolytic bands were inhibited by E64, a cysteine protease inhibitor. Preincubation of the cell lysate with pepstatin or PMSF had no effects on the protease bands.

Polymorphysims of CYP17-I Gene in the Exons Were Associated with the Reproductive Endocrine of Japanese Flounder (Paralichthys olivaceus)

  • The cytochrome P450c17-I (CYP17-I) is one of the enzymes critical to gonadal development and the synthesis of androgens. Two single nucleotide polymorphisms (SNPs) were detected within the coding region of the CYP17-I gene in a population of 75 male Japanese flounder (Paralichthys olivaceus). They were SNP1 (c.C445T) located in exon2 and SNP2 (c.T980C (p.Phe307Leu)) located in exon5. Four physiological indices, which were serum testosterone (T), serum $17{\beta}$-estradiol ($E_2$), Hepatosomatic index (HSI), and Gonadosomatic index (GSI), were studied to examine the effect of the two SNPs on the reproductive endocrines of Japanese flounder. Multiple comparisons revealed that CT genotype of SNP1 had a much lower T level than CC genotype (p<0.05) and the GSI of individuals with CC genotype of SNP2 was higher than those with TT genotype (p<0.05). Four diplotypes were constructed based on the two SNPs and the diplotype D3 had a significantly lower T level and GSI. In conclusion, the two SNPs were significantly associated with reproductive traits of Japanese flounder.

Comparison of Free and Dipeptide Lysine Utilization in Diets for Juvenile Olive Flounder Paralichthys olivaceus

  • We compared the utilization efficiency of free lysine (FL) and dipeptide lysine-glycine (LG) in terms of growth performance and whole-body amino acid composition in olive flounder Paralichthys olivaceus. A basal experimental diet was formulated to contain 0.5% (basal) lysine from fish meal, and four other diets were prepared by supplementing 0.5% or 1.0% of either FL or LG. The experiment was performed in triplicate, and 20 randomly selected fish averaging $5.41{\pm}0.16g$ were fed one of the test diets at the rate of 3% BW/day twice daily for 6 weeks. At the end of the feeding trial, fish fed the basal diet showed significantly less weight gain than did the other groups. The results of a two-way ANOVA showed that both lysine level (P = 0.001) and type (P = 0.034) influenced growth rate; however, we found no significant interaction between lysine level and form (P > 0.05). Our results revealed a significant improvement in protein efficiency ratio (PER) with each increment of dietary lysine, and the groups fed LG-supplemented diets showed higher PER than did those offered FL. Hepatosomatic and viscerosomatic indices were significantly influenced by lysine level and form, and higher values were recorded in fish fed diets containing LG. Significantly higher whole-body arginine levels were found in LG-fed groups, and a significant interaction was observed between lysine level and form (P = 0.009). Whole-body valine and aspartic acid contents were affected by lysine level, and alanine concentration was influenced by both lysine level and form. Our findings indicate that juvenile olive flounder can utilize LG more efficiently than FL for protein synthesis.

Effects of Dietary Wood Vinegar Supplementation on Growth and Immune Responses of Juvenile Olive Flounder, Paralichthys olivaceus (치어기 넙치 사료내 목초액 첨가에 따른 성장 및 면역반응에 미치는 효과)

  • A 6-week feeding trial was conducted to investigate effects of dietary supplementation with wood vinegar (WV) as a feed additive for juvenile olive flounder, Paralichthys olivaceus. Eight experimental diets supplemented with 0(control), 0.01, 0.025, 0.05, 0.1, 0.25, 0.5 and 2.0% WV/kg diet($WV_0,\;WV_{0.01},\;WV_{0.025},\;WV_{0.05},\;WV_{0.1},\;WV_{0.25},\;WV_{0.5},\;and\;WV_{2.0}$, respectively) were prepared by mixing with basal diet. After the 6-week feeding trial, fish fed the $WV_{0.1}$ diet had significantly higher weight gain, feed efficiency, and specific growth rate than those of fish fed the other diets(P<0.05). No mortality was observed in the treatment groups fed the experimental diets for 6 weeks. The liver antioxidant activity of fish fed the $WV_{0.01},\;WV_{0.025},\;and\;WV_{0.05}$ diets was significantly higher than that of fish fed the other diets. The lysozyme activity of fish fed the $WV_{0.25}$ diet was significantly higher than that of fish fed the other diets. Fish fed diets supplemented with WV had significantly lower cumulative mortality than that of fish fed the control diet throughout the challenge test, and fish fed the $WV_{0.05}\;and\;WV_{0.1}$ diets had the highest resistance to disease caused by Edwardsiella tarda at day 7 of the challenge test. Therefore, feeding 0.05-0.1% WV/kg diet improves the growth and immune responses of juvenile olive flounder, Paralichthys olivaceus.
