Studies on Sesame Diseases in Korea I. Incidences of Phytophthora Blight (참깨 병해연구 I. 참깨 역병의 발생에 대하여)

  • Incidences of Phytophthora blight in plant of sesame (Sesamum indicum L.) were observed in southern sesame production areas, Gochang of Jeonbug, Yeonggwang of Jeonnam, Jinyang of Gyeongnam and Dalseong of Gyeongbug province where disease survy was conducted from July 29 to August 1, 1981. The rate of disease incidence ranged from none to $61\%$ depending upon the field observed. The causal species of the Phytophthora was identified as P. nicotianae var. parasitica (Dastur) Waterhouse based on specific pathogenicity to sesame and morphological characteristics of sporangia. Diseased plants of sesame generally showed dark discoloration on the stem leading to plant death.

  • We have investigated the effects of abiotic and biotic stresses on leaf senescence using transgenic tobacco plants, in which cellular contents of polyamines were increased by introducing the genes of polyamine and ethylene biosynthesis in sense or antisense orientation. These transgenic plants showed accumulations of polyamines at higher levels than were found in wild-type. Stress-induced senescence was attenuated in transgenic plants cpmpared with wild-type plants, in terms of total chlorphyll loss and phenotypic changes after oxidative stress of hydrogen peroxide($H_2O_2$), high salinity, acid stress (pH3.0), ABA and fungal pathogen(phytophothora parasitica pv.Nicotianae). Transcripts for antioxidant enzyme, glutathionine-S-transferase and catalase, were also more abundant in transgenic plants than wild-type plants. These result suggested that higher expression of those genes caused a broad-spectrum resistance to abiotic stress/biotic stress. These phenomena indicate that polyamines may play an important role in contributing to the antioxidant defense function in plants. Our findings suggest that facilitate the improvement of stress tolerance of crop plants.

Studies on Characterization of Active Substances from Antagonistic Streptomyces sp. A-2 Strain against Soil-borne Phytopathogen (토양병원균(土壤病原菌) 길항성(拮抗性) Streptomyces sp. A-2 활성물질(活性物質)의 특성(特性)에 관한 연구(硏究))

  • Antifungal substances against three plant pathogenic fungi, Phytophthora capsici, Phytophthora nicotianae var. parasitica and Rhizoctionaia solani were fractionated cultures of Streptomyces sp. A-2 strain isolated in Korean soils. Characterizations of active substances related with antagonistic effects were follows : 1. The excellent media which showed the transfer efficiency of antagonistic substances from Streptomyces sp. A-2 strain G.Y.B and B.H.I. among four that are glucose yeast broth (G.Y.B), $M\ddot{u}eller$, brain heart infusion(B.H.I.) and Czapek media. Active substances which were transfered into ethylacetate or left in residual aqueous phase did not lose antagonistic activity in spite of autoclavation. This indicated that bonds of these compounds were rigid enough to keep activity under such conditions. 2. Antagonistic substances were extracted according to adjustment of pH 3 or pH12 to 5 day-old B.H.I. broth cultures of Streptomyces sp. A-2 strain. Comparative analysis fluorescent bands on HPTLC to antagonsitic spectra against three phytopathogenic fungi indicated that major substances with antagonistic activity were extracted regardless of different pH adjustment to broth cultures. Since UV spectrum of these fractions scanned from 500nm to 200nm was similiar to that of polyene macrolide, major substances related with antagonistic activities were assumed to be polyene derivatives antibiotics.

A Study on the Production of Yeast Utilizing Ethanol as a Sole Carbon Source (Ethanol 이용 미생물에 의한 단세포 단백질 생산에 관한연구)

  • In order to obtain the basic informations on the production of single cell protein from ethanol, 145 yeast strains utilizing ethanol as a sole carbon source were isolated from 32 soil samples in Korea. A yeast strain showing the highest cell yield among the isolated strains was selected and identified. The optimum culture condition, utilization of other carbon sources and the cultural characteristics for the selected yeast, and the chemical analysis of the yeast cell composition, and utilization of ethanol by the selected yeast were investigated. All the culture was carried out in the shaking flasks. The results obtained were as follows: 1. The selected yeast strain was identified as Debaryomyces nicotianae-SNU 72. 2. The optimum composition of the medium for the selected yeast is : Ethanol 40 ml, Urea 0.5 g, Potassium phosphate (dibasic) 0.5 g, Ammoium phosphate (monobasic) 0.15 g, Magnesium sulfate 0.05 g, Calcium chloride 0.01g, Yeast extract 0.005 g, Tap water 1000 ml. 3. The optimum pH was 5.0-5.5, the optimum temperature $30-33^{\circ}C$ and the aerobic state was unimportant. 4. Utilization of methanol, n-propanol, iso-propanol, n-butanol, iso-butanol, tert-amyl alcohol and acetic acid by the selected yeast was very weak. So substitution of the subtrate was thought to be impossible. 5. Studies on the propagation of the yeast cells showed that the lag phase of the yeast cells lasted 16 hours, and the logarithmic growth phase extended 16 to 28 hours. The specific growth rate was about $0.19\;hr^{-1}$ and the doubling time was 3.6 hours during the logarithmic growth phase. 6. As the result of the chemical analysis of the dry yeast cells, the content rate of the crude protein was 55.19 %, the content of others was similar to the average content of the yeast component. 7. After 34 hours cultivation, under the optimum culture condition investigated, the dry cell yield against the amount of the added ethanol was 53.4 % (W/V%), the dry cell yield against the amount of the utilized ethanol was 73.6 % (W/V%), the evaporation rate of ethanol was about 19.1 %.
