Reexamination of the genus Pterocladiella (Gelidiaceae, Rhodophyta) in Korea based on morphology and rbcL sequences

  • Although Pterocladiella is a well-known agar-producing red algal genus, its taxonomy in Korea appears to be confused. In the present study, we demonstrate the occurrence of Pterocladiella nana and P. tenuis, as well as P. capillacea in Korea on the basis of morphological and plastid rbcL sequence comparisons. P. capillacea commonly occurs along the coasts of Korea and has regular pinnate branches with abundant second-order branches, growing up to 10 cm in length. P. nana occurs in the intertidal zone of Jeju and is distinguished by the small size of its thalli (up to 5 cm) and regular branches with up to third-order branches. P. tenuis is distributed mostly subtidally on the southern coast of Jeju and has irregular branches with rarely second-order branches, growing up to 19 cm in length. We determined rbcL sequences from 19 specimens (15 from Korea and four from France) and downloaded 28 sequences from GenBank. Analyses of all 47 rbcL sequences revealed that each of three species was consistently resolved. P. capillacea and P. tenuis always formed a sister clade with P. nana at the base. Given that 12 rbcL haplotypes from 28 specimens of P. capillacea have been found to date, analysis of a fast-evolving gene from across the range of the species should highlight its genetic diversity.

  • Six adult fish of Laemonema nana, belonging to the family Moridae, were collected in a bottom trawl in the southern coastal waters of Korea. These specimens were characterized by having 46-52 second dorsal fin rays (vs. 57-67 for Lotella phycis and 68 for Physiculus japonicus), two pelvic fin rays (vs. 9 for L. phycis and 6-7 for P. japonicus), and lateral line descending abruptly below the anterior part of the second dorsal fin.

Community of Endophytic Fungi from Alpine Conifers on Mt. Seorak

  • Endophytic fungi occupy various ecological niches, which reinforces their diversity. As few studies have investigated the endophytic fungi of alpine conifers, we focused on four species of alpine conifers in this study-Abies nephrolepis, Pinus pumila, Taxus cuspidata var. nana, and Thuja koraiensis-and examined them for endophytic fungi. A total of 108 endophytic fungi were isolated. There were four taxa in A. nephrolepis, 12 in P. pumila, 18 in T. cuspidata var. nana, and 17 in T. koraiensis; these were divided into five classes: Agaricomycetes (3.2%), Dothideomycetes (29.0%), Leotiomycetes (15.0%), Sordariomycetes (41.9%), and Orbiliomycetes (1.6%). The most prevalent fungi were Sydowia polyspora (22.7%) and Xylariaceae sp. (22.7%) in P. pumila, Phomopsis juglandina (16.1%) in T. cuspidate var. nana, and Thuja-endophytes sp. 1 (70.0%) in T. koraiensis. However, there was no dominant species growing in A. nephrolepis. Some host plants were analyzed using next-generation sequencing. We obtained 4618 reads for A. nephrolepis and 2268 reads for T. koraiensis. At the genus level, the top three endophytic fungi were Ophiostomataceae_uc (64.6%), Nectriaceae_uc (15.5%), and unclassified organism (18.0%) in A. nephrolepis and Nectriaceae_uc (41.9%), Ophiostomataceae_uc (41.8%), and Magnaporthaceae_uc (9.2%) in T. koraiensis. Our results show that there are different communities of endophytic fungi among different host plants, even if the host plants are in the same region. Such ecological niches are important in terms of the ecological restoration of alpine conifers.

수온과 염분에 따른 5종 요각류의 성장과 생산력 (Effects of Temperature and Salinity on Productivity and Growth of Five Copepod Species)

  • This study investigated the productivity and growth of copepods, Sinocalanus tenellus 2 strains, Paracyclopina nana 3 strains, Apocyclops royi 2 strains, Tachidius triangularis 1 strain and Tigriopus sp. 1 strain at different temperatures $(24,\;28\;and\;32^{\circ}C)$ under 15 psu and various salinities (10, 20, 30 psu) at $28^{\circ}C$ for an individual culture. And the mass culture was carried out to evaluate the production of these copepods in the 500 mL vessel. For an individual culture of copepod, survival period of a brood female tended to increase with the decrease of temperature, but was not significantly influenced by salinity. With an increase of temperature and a decrease of salinity, maturation period of nauplii in all species tended to decrease. Total nauglii production from a broodo female at $24^{\circ}C$ was higoher than that from a brood .female at either 28 or $32^{\circ}C$ for S. tenellus and at $32^{\circ}C$ than either 24 or $32^{\circ}C$ for A. royi Haenam stram, but was not influenced by temperature for another copepods. And total nauplii production from a brood female at 10 and 20 psu tended to be higher than that from a brood female at 30 psu, expect for Tigriopus sp. and P. nana Hwajinpo strain. Total nauplii production from a brood female was the highest in Tigriopus sp. (350 nauplii) at $28^{\circ}C$ under 20 psu, followed by A. royi Haenam strain (187 nauplii) at $32^{\circ}C$ under 15 psu and P. nana Hwajinpo strain (152 nauplii) at $24^{\circ}C$ under 10 psu. In the mass culture of each copepod, P. nana was the most productive (26.6 mg dry weight) and this value was two and four times higher than productivity of Tigriopus sp. and A. royi, respectively.

가축 및 실험동물의 비충류 감염에 대한 프라지관열($Cesocide^{\circledR}$ 주) 주사제의 치료효과 (Efficacy of Praziquantel ($Cesocide^{\circledR}$ injection) in Treatment of Cestode In(ections in Do%erotic and Laboratory Animals)

  • 프라지관텔 주사제의 조충류 피료 효과를 평가하였다. 개, 고양이, 산양, 사슴, 닭, 쥐 및 마우스 등 총 93마리 동물의 장관내에 기성하는 조충류 dipylidium caninum, Taenia pisiformis, Spirometra sp., Taenia taeniaeformis, Moniekia expanse, Moniexia sp., Raillietina sP. 및 인ymenolepis nana에 체중 kg당 프라지콴텔 12 mg, 9 mg, 6 mg, 3 mg, 1.5 mg을 대퇴부에 근육주사한 후 치료효과를 판정하였다. H. nana에 감염된 마우스는 6mg/kg 투여 후 3시 간부터 6시 간 사이에 충체를 배출하였다. 다른 조충류도 약제투여 48시 간 이내에 모두 체외로 배출되었다. H. nana 및 M. expansa를 제외한 모든 충체는 체외 배출시 심하게 손상되어 있었으며, D.cnninum의 충란낭은 붕괴되어 충란이 분리되었다. 조충의 종류와 감염강도 등에 따른 숙주 체중 kg당 투여 용량의 차이는 관찰되지 않았으며 Spirometra sp.에 감염되었던 한 마리의 개를 제외하고는 프락지관텔 6 mg/kg 1회 용량으로 완치되어 97.9% (46/47)의 치유율을 보임으로써 우수한 치료 효과를 나타내었다.

목질계 Cellulose로부터의 Ethanol의 경제적인 생산공정을 위하여 분자진화에 의한 활성이 획기적으로 증가된 Cellulase의 대량 발현공정 개발 (The Development of Expression Process Leading to Ethanol Production with Highly Active Cellulase Modified by Directed Evolution)

  • Cellobiohyolase (CBH) I 유전자의 확보는 CBH I 유전자 cellulase 생산 균주인 Trichoderma reesei를 배양, 수거하고 액체 질소를 이용하여 세포를 파쇄 후 RT-PCR kit를 이용하여 CBH I gene을 합성하였다. 그 후 발현벡터인 pYGAL에 cloning하였다. CBH I 유전자 앞부분에는 CBH I 단백질이 cell 외부로 분비할 수 있도록 하는 ppL이 포함되었다. CBH I 단백질 발현은 Protein gel 결과를 통하여 발현을 확인하였다. Cellulase 활성을 증대시키기 위한 분자진화 방법 개발은 error prone PCR과 DNA shuffling을 수행하였다. 얻은 CBH I 유전자를 발현 벡터에 삽입하고 효모에 transformation하여 이것을 다시 screening하였다. 1차 screening 후 confirm test하기 위해 DNS (Dinitrosalicylic Acid) 환원당 측정법을 이용하였으며, 이 결과 121-D8, 228-G2, 389-E3, 412-B4, 456-D2의 cellulase 변이체를 획득할 수 있으며, 456-D2의 경우 original CBH I과 비교하여 약 510%의 활성이 증가된 것을 확인할 수 있었다. 분자 진화 된 cellulase sequence 분석결과 CBH I wild type과 비교하였을 때 121-D8의 경우 변경된 9개의 염기 중 아미노산의 변화에 영향을 준 염기는 6개, 228-G2의 경우 변경된 7개의 바뀐 염기 중 아미노산의 변화에 영향을 준 염기는 4개, 389-E3의 경우 변경된 13개의 바뀐 염기 중 아미노산의 변화에 영향을 준 염기는 9개, 412-B4의 경우 변경된 9개의 바뀐 염기 중 아미노산의 변화에 영향을 준 염기는 6개, 456-D2의 경우 변경된 10개의 바뀐 염기 중 아미노산의 변화에 영향을 준 염기는 7개이었다. Cellulase 생산 공정 최적화는 5 L 발효기를 이용하여 고농도 배양을 실험한 결과 최종 O.D. 120까지 진행할 수 있었으며, 약 1.2 g/L의 cellulase를 얻을 수 있었다. Cellulase 생산 공정 scale-up 5 L 규모에서 확립된 최적 fed-batch 발효 공정을 300 L로 scale-up하여 실험하였다. 최종 O.D.는 약 280 정도이며 cellulase의 발현양은 약 3.2 g/L 수준임을 확인하였다. Cellulase 정제 공정 최적화 결과 80%의 수율과 95%의 순도를 확보하였다.

Selection of Copepods as Live Food for Marine Fish Larvae Based on Their Size, Fecundity, and Nutritional Value

  • Copepods are a major food source for marine fish larvae in nature. Many studies on copepods culture have been conducted to develop a new live food for the seedling production of marine fish larvae. But fish farmers still depend on rotifer and Artemia nauplii. This study was carried out to find suitable copepods as live food for the larvae in hatchery. Eight species of copepods (1 calanoid, 2 cyclpoid, 5 harpacticoid) that were fed Isochrysis galbana were examined in terms of the size of nauplii, fecundity, amino acids, and fatty acids contents. These species were divided into small (nauplii length 46-86 ${\mu}m$) and large (nauplii length 120-188 ${\mu}m$) size group. Nitokra spinipes in the small group and Tigriopus japonicus in the large group showed the highest fecundity with 151.1 and 139.6 nauplii production per gravid female, respectively. With regard to nutrients, essential amino acids were the highest with 21.2% in cyclopoid Paracyclopina nana in the small group and n-3 HUFA were the highest in calanoid Pseudodiaptomus inopinus (8.5 ${\mu}g/mg$) in the large group and P. nana (8.8 ${\mu}g/mg$). In terms of the size, fecundity, and nutritional value of copepods examined in this study, N. spinipes and P. nana seem to be suitable copepod species to develop as a new live food for small mouth fish larvae.

山城山 山火跡地의 植生再生과 二次邊의 (Revegetation and Secondary Succession of the Burned Area in Mt. Sanseung)

  • This report is an investigation of the revegetation and secondary succession in the burned area of Mt. Sanseung in Taegu region. The forest fire took place in December, 1977. The survey was conducted eight times, -three times from October 1, 1983 to October 3, the same year and five times from August 10, 1984 to August 15, the same year. The floristic compositions in the sampled sites constituted 25 kinds of vascular plants and 21 kinds in the burned area and the unburned area respectively. The biological type in both the burned and unburned areas was H-D1-R5-e type, which is generally common to other areas (Taegu, Kyungpook, Chung Buk and Kangweon areas). In the burned area dominant species were Carex humilisvar. nana, Arundinella hirata and Quercus serrata and on the other hand, in the unburned area Pinus densiflora, Carex humilis var. nana, Rhododendron mucrfonulatum var. ciliatum and Quercus serrata. The species diversity diversity index(H) and eveness index(e) of the burned area were higher than those unburned area. Degree of succession (DS) was 650 in the burned area at the 6th years after the fires and 962 in the unburned area. THe vegetation of the burned area was slowly recovered as of 1984 compared with each other. According to the analysis of the soil preperties, pH, available phosphorus and exchangeable potassium were increased, but organic matter, total nitrogen and total organic carbon were decreased. It is assumed that these results were due to the forest fire.

Intestinal Parasites among Wild Rodents in Northern Gangwon-do, Korea

  • To determine geographical patterns of natural parasite infections among wild rodents, a total of 46 wild rodents from 3 different localities in northern Gangwon-do (Province), Korea were examined for intestinal parasite infections. Along with nematodes such as hookworms and Syphacia spp., Plagiorchis muris (2 specimens) (Trematoda) were collected from striped field mice, Apodemus agrarius. In a Korean wood mouse, Apodemus peninsulae, the overall nematode infections were similar to A. agrarius, but an adult worm of Echinostoma hortense (Trematoda) was collected. In addition, 2 species of cestodes, i.e., Hymenolepis nana and Hymenolepis diminuta, were collected from A. agrarius. Through this survey, A. agrarius and A. peninsule were confirmed as the natural definite hosts for zoonotic intestinal helminths, i.e., P. muris, E. hortense, H. nana, and H. diminuta, in northern Gangwon-do, Korea. Considering increased leisure activities around these areas, seasonal and further comprehensive surveys on wild rodents seem to be needed to prevent zoonotic parasite infections.
