Symptoms of fowl cholera including orofacial edema, swollen and edematous wattles and combs, and severe respiratory disorders were detected in domestic poultry in two broiler breeder farms: one located in Gyeong-gi Province (October, 2000) and the other in Chung-cheong-nam Province (March, 2001). Gram-negative, bipolar staining bacillus was easily found in a direct smear. The biochemical properties of isolates were examined using a standard diagnosis method, proving that they were 99.7% similar to the Pasteurella multocida (P. multocida: PM), a pathogenic and causative agent of fowl cholera (FC). As a result, an FC outbreak in domestic fowls was confirmed for the first time in Korea since 1942. Because FC was detected in broiler breeder farms for the first time in 59 years at the same time as an FC outbreak was confirmed in wild birds (October, 2000), our concern was focused on whether the PM strains that originated in wild birds were transmitted into poultry forms. The possibility was tracked down by comparing phenotypic and genetic properties between the two types of PM strains. PM strains of chicken origin showed prominent differences from the PM strains of wild bird origin in both phenotypic and genetic properties. An examination of the origin of the wild bird bacteria was conducted, but no evidence has been identified that PM strains from the wild bird were introduced into domestic poultry farms.

The immunological role of the Pasteurella multocida toxin (PMT) in mice was examined using a PMT mutant strain. After a nasal inoculation, the mutant strain failed to induce interstitial pneumonia. Moreover, PMT had no significant effect on the populations of CD4+, CD8+, CD3+, and CD19+ immunocytes in blood or on the populations of CD4+ and CD8+ splenocytes (P<0.01). However, there was a significant increase in the total number of cells in the BAL samples obtained from the wild-type P. multocida-inoculated mice. On the other hand, the level of IL-l expression decreased when the macrophages from the bronchio-alveolar lavage were stimulated with PMT. Overall, PMT appears to play some role (stimulating and/or inhibiting) in the immunological responses but further studies will be required to confirm this.

A 12-year-old female small-clawed Asian otter (Anoyx cinereus) with a one-week history of anorexia, chills, and abdominal distension was found dead. Grossly, yellowish-brown turbid fluids accumulated in abdominal cavity of the otter, and yellowish thread-like fibrinous materials were found on the surface of abdominal organs. Several variable sized yellowish-white crystalloids were scattered on the medullary space of kidneys. Histologically, diffuse serositis (peritonitis) characterized by the fibrinous exudates, thickened serosal capsule and the swelling of mesothelial cells were observed in the serosa of liver, spleen, stomach, and intestine. Multifocal necrosis, hemorrhage, infiltration of macrophage, and brown pigments were presented in the liver. Isolated bacteria from ascites and fibrinous materials in abdominal visceral surface were white, smooth and convex with characteristic mousy odor on blood agar plate. These bacteria were confirmed as Pasteurella (P.) multocida type A by polymerase chain reaction analysis. Based on the gross examination, histopathologic findings and bacterial experiments, this otter was diagnosed as severe peritonitis associated with P. multocida and necrotic hepatitis.
This study was conducted to investigate the species-specific gene detection, capsular serogroup and antimicrobial resistance pattern of Pasteurella multocida isolated from pneumonic lung lesion of swine in Gyeongbuk province. P. multocida isolates were typed for capsular serogroups by polymerase chain reaction. Of the 32 strains, 28 (87.5%) were typed serotype A, 3 (9.3%) were typed serotype D, and 1 strain was unknown (3.1%), respectively. In antimicrobial agents resistance test, almost of strains were susceptible to amoxicillin (100%), enrofloxacin (96.9%), ampicillin (93.8%), florfenicol (90.6%), chloramphenicol (90.6%) and were resistant to streptomycin (71.9%), spectinomycin (56.3%). All strains were resistant to clindamycin, erythromycin and lincomycin.
Here, we report the suitability of using a resin-type chitosan formulae as a sustained-releasing form adjuvant in comparison with commercially well-known Freund's adjuvants. To induce the immunological responses, N-terminal region of Pasteurella multocida toxin was used as an antigen, which was found to be protective immunogen against P. multocida infection. Mice immunized with chitosan resin formulae showed statistically significant antibody induction (P<0.001) as much as that of Freund’s adjuvants. As a result, the resin-type sustained-releasing form chitosan formulae is thought to be a good candidate for a new type adjuvant.
The present study was conducted to investigate isolation and antimicrobial drug susceptibility of Pasteurella spp from pneumonic calves and cows (pneumonic lungs) in Gyeongnam south area from January to November 2000. Pasteurella multocida and P haemolytica were isolated 136 (53.8%) and 22(8.7%), respectively, from 158 of 253 pneumonic calves of 1 to 12 months of age and from pneumonic lungs and simultaneous isolation rate of P mutocida and P haemolytica was 4 (1.6%). Seasonal isolation rate of Pasteurella spp in calves and pneumonic lungs varied from 3 to 65.2%, and it was higher in Fall. The majority of biochemical and cultural properties of P multocida and P haemolytica isolated from calves and pneumonic lung were identical to those of the reference strains employed. Pneumonic lesions of the examined lungs were histologically identified as purulent bronchopneumonia and fibrinous pneumonia. All isolates were resistant to penicillin, amikacin, lincomicin, colistin, cloxacilling, sulfonamide, kanamycin, sulfamethoxasoletrimetoprim, and gentamicin(>80%), but some of them were susceptible to amoxicillin, norfloxacin, cephalexin, cefazolin, orbifloxacin, enrofloxacin, and ciprofloxacin(>60%).
A native strain of Pasteurella multocida was isolated from pigs suffering from severe atrophic rhinitis at domestic farms in Gyeonggi Province, Korea, and was identified as capsular serogroup 'D' and somatic serotype '4' by disc diffusion decapsulation and gel diffusion precipitation tests, respectively. The P. multocida (D:4) induced atrophic rhinitis in healthy pigs by the secondary infection. The gene for outer membrane protein H (ompH) of P. multocida (D:4) was cloned in Escherichia coli DH5$\alpha$ by PCR. The open reading frame of the ompH was composed of 1,023 bp, possibly encoding a protein with 341 amino acid residues containing a signal peptide of 20 amino acids at N-terminus, and the gene product with molecular mass of ca. 38 kDa was identified by SDS-PAGE. Hydropathy profiles indicated that there are two variable domains in the OmpH. To express the ompH in E. coli, the gene was manipulated in various ways. Expression of the truncated as well as full-length forms of the recombinant OmpH was fatal to the host E. coli BL21 (DE3). However, the truncated OmpH fused with GST was consecutively expressed in E. coli DH5$\alpha$. A large quantity of the fused polypeptide was purified through GST-affinity chromatography.
Atrophic rhinitis (AR) is the one of important respiratory diseases and causes severe economic losses in pig industry. Severe attempts have been made to reduce the economic losses by preventing the disease. One of the methods is the spraying of antibiotics into nasal cavity of piglets. Recently, the efficacy of the spraying with kanamycin and gentamicin was reduced in the Korean swine industry. Therefore, the preventive methods have been required to be changed based on the antimicrobial susceptibility profiles of causative agents of swine AR. Based on the current situations of this disease, Bordetella (B.) bronchiseptica and Pasteurella (P.) multocida 4D were isolated from pigs with clinical signs of AR. The isolation rates of B. bronchiseptica and P. multocida 4D were 58.5% and 32.9%, respectively. In the antimicrobial susceptibility test, the bacteria were resistant to kanamycin and gentamicin which have been used as the spraying agents, but they were susceptible to amikacin. A new spraying agent was developed using amikacin using ${\beta}$-glucan and yakbaltag as supplementary agents. Field efficacy of the agent was carried out with different schedule. The results from this study suggested that the newly developed spraying agents might be helpful to prevent AR in swine.

We cloned a gene of ompH(D:4) from pigs infected with P. multocida D:4 in Korea [16]. The gene is composed of 1,026 nucleotides coding 342 amino acids (aa) with a signal peptide of 20 aa (GenBank accession number AY603962). In this study, we analyzed the ability of the ompH(D:4) to induce protective immunity against a wild-type challenge in mice. To determine appropriate epitope(s) of the gene, one full and three different types of truncated genes of the ompH(D:4) were constructed by PCR using pET32a or pRSET B as vectors. They were named ompH(D:4)-F (1,026 bp [1-1026] encoding 342 aa), ompH(D:4)-t1 (693 bp [55-747] encoding 231 aa), ompH(D:4)-t2 (561 bp [187-747] encoding 187 aa), and ompH(D:4)-t3 (540 bp [487-1026] encoding 180 aa), respectively. The genes were successfully expressed in Escherichia coli BL21(DE3). Their gene products, polypeptides, OmpH(D:4)-F, -t1, -t2, and -t3, were purified individually using nickel-nitrilotriacetic acid (Ni-NTA) affinity column chromatography. Their $M_rs$ were determined to be 54.6, 29, 24, and 23.2 kDa, respectively, using SDS-PAGE. Antisera against the four kinds of polypeptides were generated in mice for protective immunity analyses. Some $50{\mu}g$ of the four kinds of polypeptides were individually provided intraperitoneally with mice (n=20) as immunogens. The titer of post-immunized antiserum revealed that it grew remarkably compared with pre-antiserum. The lethal dose of the wild-type pathogen was determined at $10{\mu}l$ of live P. multocida D:4 through direct intraperitoneal (IP) injection, into post-immune mice (n=5, three times). Some thirty days later, the lethal dose ($10{\mu}l$) of live pathogen was challenged into the immunized mouse groups [OmpH(D:4)-F, -t1, -t2, and -t3; n=20 each, two times] as well as positive and negative control groups. As compared within samples, the OmpH(D:4)-F-immunized groups showed lower immune ability than the OmpH(D:4)-t1, -t2, and -t3. The results show that the truncated-OmpH(D:4)-t1, -t2, and -t3 can be used for an effective vaccine candidate against swine atrophic rhinitis caused by pathogenic P. multocida (D:4) isolated in Korea.
For the epidemiological studies of the respiratory infection from 1,918 heads of adult rabbits from January 1980 to December 1985 at an integrated farm, the causative agents were. Isolated samples 92 from of lungs and nasal discharges, and identified by serological and non-serololgical tests, the loss rates by the infection were also investigatyed, and the antimicrobial sensitivities of the isolates were determined. The results obtained were as fellows : 1. The loss rate by respiratory infection was 11.9% among 1,819 heads which raised and 41.7% among 518 heads which lost by diseases, and the rate increased annually. 2. The loss rate of rabbits in Spring (45.0%), Summer (45.5%) and Winter (53.5%) showed significantly higher (P < 0.01) than that in Autumn(29.1%). 3. The loss rate of Rex (56.3%) showed significantly higher (P<0.05) than New Zealand White (40.7%) and Californian (42.0% ). 4. P. multocida.(37.9%), B. brouchiseptica (16.0%), E. coli (10.6%) and so forth were isolated from 92 samples of lungs and nasal discharges. 5. Sixty fours trains of P. multocida were typed by serological or non-serological tests and their all serotypes were A type (62 strains) except 2 strains of untypable. 6. In antimicrobial sensitivity test for 64 isolates of P. multocida, all the strains tested were highly sensitive to chloramphenicol(95.3%), colistin (95.3%), gentamicin (92.2%), kanamycin (90.6%), neomycin (82.8%) and tetracycline (81.3%), but exhibited low sensitivity to streptomycin (26.6%), and all strains were resistant to lincomycin. 7 Twenty seven isolates of B. brouchiseptica showed higher sensitivity to cephalothin (100%), erythromycin (100% ), gentamicin (100%). trimethoprim+sulfamethoxazole (100%), chloramphenicol(88.9%), kanamycin(88.9%), neomycin (88.9%) and colistin (85.2%), but lower sensitivity to nitrofurantoin (18.5%), penicillin(18.5%). streptomycin(18.5%) and ampicillin (14.8%), and all strains were resistant to lincomycin.
