Identification of Some Phellinus spp.

  • Four strains of Phellinus spp. was identified based on internal transcribed spacer(ITS) region of rDNA sequence analysis and morphological characteristics. Basidiocarps of all strains were effused-reflexed and hymenial surface was poroid. Hyphal system was dimitic and basidiospore was globose to ellipsoid. The amplification of ITS regions produced a DNA fragment of 500 to 780 by in all strains examined. The determined sequences were analyzed for the reconstruction of phylogenetic tree. From these results, Phellinus sp. KM-1, KM-2, and KM-4 was identified as P. hartigii, P. baumii, and P. linteus, respectively.

  • Phellinus genus belonged to Hymenochaetaceae of Basidiomycetes and has been well known as one of the most popular medicinal mushrooms due to high antitumor activity. This study was carried out to obtain the basic information for mycelial culture conditions of Phellinus linteus, P. baumii, and P. gilvus. According to colony diameter and mycelial density, the media for suitable mycelial growth of them were shown in MEA, glucose peptone, and MCM. The optimum temperature for mycelial growth was $30^{\circ}C$. Carbon and nitrogen sources were man nose and malt extract, respectively. The optimum C/N ratio was 10:1 to 5:1 with 2% glucose concentration, vitamin was thiamine-HCl, organic acid was succinic acid, and mineral salt was $MgSO_{4}{\cdot}7H_{2}O$.

Inhibitory Effects of Phellinus linteus and Rice with Phellinus linteus Mycelium on Obesity and Diabetes (상황버섯, 상황버섯균사체배양쌀 추출물의 비만 및 당뇨 억제 효과)

  • The objective of this study was to examine the ability of extracts from Phellinus linteus (PL) and rice with Phellinus linteus mycelium (PLM) to inhibit obesity and diabetes. The efficacy of PL and PLM were evaluated using Oil Red O staining, cholesteryl ester transfer protein (CETP) levels, protein tyrosine phosphate 1B (PTP1B) levels, organ weight, and serum lipid levels. Lipid accumulation significantly decreased by 76% and 59% upon treatment with $300{\mu}g/mL$ of PL and PLM, respectively (P<0.01). The inhibition of CETP activity increased 99% upon treatment with $300{\mu}g/mL$ of PL or PLM. Treatment with 3, 10, 30, 100, and $300{\mu}g/mL$ of PL, changed PTP1B activity by 10, 11, 14, 12, and 18% respectively. Also, treatment with increasing concentrations of PLM led to a significant concentration-dependent inhibition of PTP1B activity (P<0.01). PL and PLM were orally administered for 28 days after a high fat diet (HFD). PL significantly (P<0.05) reduced triglyceride and cholesterol levels. In addition, PLM significantly (P<0.05) reduced triglyceride, cholesterol, and HDL-cholesterol levels. GOT and GPT were not significantly affected. These results indicate that PL and PLM extracts have potent and useful activities for the treatment of obesity and diabetes mellitus.

Survey for Alternative Therapy Used by Cancer Patients Receiving Radiation Therapy (방사선치료 암 환자의 대체요법 경험실태 조사연구)

  • Although it is presumed that cancer patients take various alternative therapies, the present status is not well recognized. The purpose of this study is to survey alternative therapies used by cancer patients receiving radiation therapy and then, find associated factors of taking alternative therapies. The study subjects were composed of those who receiving radiation therapy in the department of radiation oncology in 5 hospitals located at Extended Busan city and who were on follow-up after medical cure. They were 394 male and female patients over 20 years old. The mean age of the subjects was 53.2 years old and the age ranged from 23 to 83 years old. 188 patients($47.7\%$) used alternative therapies. Total 68 different kinds of alterative therapies were used, average 7.3 kinds per patient were experienced, and average total cost expenditure was 2,830,000 won. Among the alternative therapies, black bean($38.8\%$) was the most commonly used and brown rice($38.3\%$), ganoderm lucidum($37.8\%$), elm tree($33.5\%$), and phellinus linteus($30.8\%$) were followed in order of frequency. However in considering the time, cost and effort spent, phellinus linteus was the first. In terms of cost, phellinus linteus was the highest with average expenditure of 2,740,000 won. Among the motivation of using alternative therapies, expecting auxiliary help for the hospital therapy was the highest ($31.4\%$). About half of users($56.9\%$) of alternative therapies were recommended by their relatives to use alternative therapies. In comparing the characteristics of experienced and unexperienced groups, alternative therapy was experienced significantly more in patients of younger age(p=0.001), in patient of higher educational level(p=0.001), and in patients of higher income(p=0.030) The proportion of using alternative therapies was significantly higher in the group treated with chemotherapy(p=0.005), and in the patients who did not satisfy with radiation therapy(p=0.001). The frequency of drinking was significantly higher tendency in the inexperienced group(p=0.046), There was no significant difference in marital status, job, religion, other disease, surgical operation of the cancer and smoking staus between the two groups. Among the unexperienced group, $34.0\%$ of the patients did not take the alternative therapies because they did not have know]edge for the alternatives, and $22.3\%$ worried about negative effects on hospital-based therapy $58.7\%$ of them were willing to take the alternative therapies if the effects and safety were proven by the government or research institutes. $21.9\%$ of the patients wanted to take the alternatives if they were affordable. $72.3\%$ of the patients was willing to take them if their families recommend, but $27.2\%$ responded they would not take them in any situation. Conclusively, various kinds of alternative therapies which were not proved medically were exposed to patients, In these circumstances, it is required to investigate, study and evaluate the medical effects and safety of the alternative therapies.

Adding Germinated Brown Rice Soaked in a Mycelial Culture Broth of Phellinus linteus to Muffins: An Assessment Using the Response Surface Methodology (반응표면분석법을 이용한 상황버섯균사체 배양액 침지 발아현미 첨가 머핀의 제조 조건 최적화)

  • The purpose of this study was to develop a muffin recipe that included germinated brown rice powder soaked in a mycelial culture broth of Phellinus linteus (GBRP). Muffins were made using various quantities of GBRP, butter, and milk. An assessment was conducted using the response surface methodology (RSM), which included 16 experimental points with three replicates for the three independent variables: GBRP, butter, and milk. The physical and chemical evaluation using the RSM showed no significant differences; whereas the hardness, cohesiveness, springiness, gumminess, and chewiness indicated significant differences in the relationships between the factors (p<0.05). The sensory evaluation using the RSM was highly effective in describing the relationships between overall acceptability and taste (p<0.001). Thus, the optimal combination of ingredients in a GBRP muffin, as assessed with numerical and graphical methods, was 84.041 g GBRP, 110.224 g butter, and 80.0 g milk.

Fruit body formation on rice brown and timber log using liquid spawn of Phellinus linteus (목질진흙버섯(Phelinus linteus) 액체배양에 의한 현미와 원목에서 자실체 형성)

  • The main objectives of this study were to fruit body of Phellinus linteus using liquid spawn. Highest mycelial growth and in vitro fruiting was observed when $180m{\ell}$ of water was added to 200g of brown rice in $1000m{\ell}$ PP bottle. Optimum inoculum amount was 100ml per bottle. Better mycelial growth and fruiting were observed when the amount of brown rice was lowered. For timber log culture of P. linteus, low contamination was observed when logs were sterilized for as low as 60 mins. But, best mycelial growth was observed when the logs were sterilized for 120mins. Among different log types, highest mycelial growth and fruiting were observed in oak and mulberry. Only few fruiting could be observed in birch, chestnut, alder, while no fruiting was produced in Aspen. Best fruiting was produced from isolate PH-211, although other isolates also produced few fruiting.

Characteristics of the protein and antioxidant contents in soybeans cultured with Phellinus linteus HN00K9 (목질진흙버섯 (Phellinus linteus) HN00K9 균사체 배양 대두의 단백질함량 및 항산화활성 특성)

  • In this study, the protein content and functional changes in soybeans cultured with Phellinus linteus HN00K9 were analyzed. P. linteus HN00K9 was cultured on soybeans. The crude protein content in soybeans cultured with HN00K9 (PMS) was 41.99%, which was higher than that in soybeans not cultured with the mushroom (UCS). The total free amino acid content in PMS increased to 39,963 mg/100 g, which was higher than that in UCS (36,817 mg/100 g). In particular, in PMS, glutamic acid accounted for 18.5% of the total amino acids at 7,413 mg/100 g. The total polyphenol content in PMS was 2.66 mg GAE/g, which was more than 45% higher than the amount in UCS (1.45 mg GAE/g). Additionally, PMS showed a DPPH radical scavenging activity of 33.3%, which was 3 times higher than that exhibited by UCS (11.5%), reflecting its high antioxidant content. Therefore, the PMS in this study has potential for use as a functional food material.

Physiological Activities of Extracts from Phellinus linteus on Brown Rice added Rice Bran, Pine Needle and Tumeric Powder (미강과 솔잎, 강황 분말을 첨가한 현미에 배양한 상황버섯 균사체 추출물의 생리활성에 관한 연구)

  • This study was carried out to examine Electron donating ability (EDA), nitrite scavenging, tyrosinase inhibition, ACE inhibition activity and fibrinolytic activity of culture extracts from Phellinus linteus which was grown added rice bran, pine needles and turmeric in brown rice. Electron donating ability of Phellinus linteus extract (PLE) was lower in the water extract than the ethanol extract. Nitrite scavenging activity was the highest in PLE from ethanol extract than water extract. Especially, when the pine needles was addition treatment, the nitrite scavenging activity was about 70% at pH 1.2 by ethanol extract. Tyrosinase inhibition activity of PLE was highest in the water extract than ethanol extract, and inhibition rate was the most higher in the extract by hot water added pine needles. ACE inhibition activity were very low effective at water and ethanol extract. Fibrinolytic activities were similarly strong in rice bran, pine needles and turmeric powder. Especially, when rice bran was added, showed the activity was increased about 5% than plasmin. Therefore, It may be used for the food industry as natural source of bioactive compound after further investigation, such as in vivo experiment.

Antioxidant and Anti-inflammatory Activities of Functional Plant Materials (항산화 및 면역 활성 증강을 위한 생약재의 탐색)

  • Eleven functional plant materials were identified via a literature search, and their antioxidant capacity and inhibitory effects on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW264.7 cells were tested. Yields from hot water extracts of the materials were the highest (52.10%) in Lycii fructus, and the yields from Phellinus linteus were the lowest (5.7%). The yields of another were 14.50-42.47%. Total phenol and flavonoids contents were the highest in P. linteus. The $EC_{50}$ values for DPPH and ABTS radical scavenging activities were lower than $100{\mu}g/ml$ for Salvia miltiorrhiza, whereas the values for P. linteus, Scutellaria baicalensis, and Paeonia lactiflora were $100-200{\mu}g/ml$. The $EC_{50}$ value for the superoxide anion radical scavenging activity of all the extracts was higher than $300{\mu}g/ml$. P. linteus for the reducing power was shown the highest activity. $Fe^{2+}$ chelating activity was the highest in the Morus alba extract. In an MTT assay, the cell viability of the RAW264.7 LPS-exposed cells was above 80% in extracts of $50{\mu}g/ml$ and above 77% in extracts of $100{\mu}g/ml$ in all the plant materials except Acanthopanax sessiliflorum. NO production in the RAW264.7 LPS-exposed cells showed a 12-fold increase compared to the control. The NO production level of all the extracts was $6.86-26.18{\mu}M$. Notably, $100{\mu}g/ml$ of S. baicalensis extract showed a remarkable decrease in NO production (72%) compared with the control. The potent antioxidant and anti-inflammatory activities of S.baicalensis, P. linteus, S. miltiorrhiza, M. alba, and P. lactiflora suggest that they are potential candidates as functional materials.

Protective Effect of water extract Phellinus linteus-discard Schisandra chinensis solid fermented extracts on improvement of sarcopenia by Atorvastatin-induced muscle atrophy cell model (Atorvastatin으로 유도된 근위축 세포모델에서 상황-오미자박 고상발효물 열수추출물의 보호효과)

  • Objectives : This study is to effect of improving muscle atrophy through water extract on the solid-phase fermentation extraction with Phellinus linteus of discarded Schisandra chinensis in an atorvastatin-induced atrophy C2C12 cell. Methods : C2C12 myoblast were differentiated into myotube by 2% horse serum medium for 6 days, and then treated solid-phase fermentation(S-P) extract at different concentrations for 24h. To investigate the effect of S-P extract on the induction of muscle atrophy and expression of atrophy-related genes and apoptosis in differentiated C2C12 myotubes using a GSH, ROS, real-time PCR, western blots analysis. Results : As a result of treatment with atorvastatin at concentrations of 5, 10, and 20 uM on the 6th day of differentiation in C2C12 myotube cells, it was confirmed that the cell morphology was damaged in a concentration-dependent manner, and the length and thickness of the myotube also decreased in a concentration-dependent manner. Treatment with S-P extract (50, 100 and 200 ㎍/㎖) increased of GSH and inhibited ROS in the atorvastatin-induced muscle atrophy cell model at a concentration that did not induce toxicity. In addition, it was confirmed that it has an effect on muscle reduction by inhibiting apoptosis of muscle cells as well as being involved in protein production and degradation of muscle cells. Conclusions : Atorvastatin-induced atrophy C2C12 cell, S-P extract activates related to differentiation/generation and proteolysis, and inhibits cell death of atrophy in C2C12 cell. Based on this, it is necessary to prove its effectiveness through animal models and human application test, but it is considered to be discarded Schisandra chinensis can present the potential for development as a recycling industrial material.
