Effect of Split Application of Nitrogen Fertilizer on Growth and Yield in Platycodon grandiflorum A. DC. (질소 분시 비율에 따른 도라지의 생육 및 수량)

  • These studies were conducted to investigate the proper method of nitrogen application for increasing the growth stability and quality in ballon flower (Platycodon grandiflorum A. DC.). In growth characteristics by split ratio of nitrogen application, stem diameter, number of branching roots and number of leaves were higher at the split ratio of 50:50 (nitrogen were splitted a half with basal application and the other half in autumn) than at 70:30. Root length, root diameter and number of branching root were higher at 50:50 than those of 70:30. The root yield was increased by 30% (230 kg/10a) at 50:50 compared with that of the 70:30. The crude saponin contents of root from 50:50 was 4.68%.

  • This study was conducted to investigate the effects of different soil texture in agronomic characteristics, root yield and quality of baloon flower (Platycodon grandiflorum A. DC.). Three kinds of soil texture, sandy loam, silt loam and clay loam, were tested for two years. Baloon flowers were seeded with 20cm row space and cultivated in the one square meter pot with 0. 6m depth. Fifty plants per pot were remained. In growth characteristics of one-year-old plants, shooting date was not significantly different according to the changes of soil texture. The ratio of stem growth was highest between July and August. In two-years-old plants, it was showed between May and June. Plants grown on clay loam showed the highest growing characteristics than those of other soil textures. Dry root weight cultivated on clay loam was 274kg/10a in one-year-old and 934kg/10a in two-years-old plants. The content of ethanol extracts from root was highest at those of sandy loam as 47.1 % in one-year-old root. The crude saponin content was not different significantly among the three kinds of soil texture.

Immunostimulatory activity of hydrolyzed and fermented Platycodon grandiflorum extract occurs via the MAPK and NF-κB signaling pathway in RAW 264.7 cells

  • BACKGROUND/OBJECTIVES: Platycodon grandiflorum (PG) has long been known as a medicinal herb effective in various diseases, including bronchitis and asthma, but is still more widely used for food. Fermentation methods are being applied to increase the pharmacological composition of PG extracts and commercialize them with high added value. This study examines the hydrolyzed and fermented PG extract (HFPGE) fermented with Lactobacillus casei in RAW 264.7 cells, and investigates the effect of amplifying the immune and the probable molecular mechanism. MATERIALS/METHODS: HFPGE's total phenolic, flavonoid, saponin, and platycodin D contents were analyzed by colorimetric analysis or high-performance liquid chromatography. Cell viability was measured by the MTT assay. Phagocytic activity was analyzed by a phagocytosis assay kit, nitric oxide (NO) production by a Griess reagent system, and cytokines by enzyme-linked immunosorbent assay kits. The mRNA expressions of inducible nitric oxide synthase (iNOS) and cytokines were analyzed by reverse transcription-polymerase chain reaction, whereas MAPK and nuclear factor (NF)-κB activation were analyzed by Western blots. RESULTS: Compared to PGE, HFPGE was determined to contain 13.76 times and 6.69 times higher contents of crude saponin and platycodin D, respectively. HFPGE promoted cell proliferation and phagocytosis in RAW 264.7 cells and regulated the NO production and iNOS expression. Treatment with HFPGE also resulted in increased production of interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α, C-X-C motif chemokine ligand10, granulocyte-colony-stimulating factor, granulocyte-macrophage colony-stimulating factor, and monocyte chemoattractant protein-1, and the mRNA expressions of these cytokines. HFPGE also resulted in significantly increasing the phosphorylation of NF-κB p65, extracellular signal-regulated kinase, and c-Jun N-terminal kinase. CONCLUSIONS: Taken together, our results imply that fermentation and hydrolysis result in the extraction of more active ingredients of PG. Furthermore, we determined that HFPGE exerts immunostimulatory activity via the MAPK and NF-κB signaling pathways.

The radioprotective effects of radices herbs (대표적 근류 생약의 방사선 방호효과)

  • We performed this study to determine the effect of Jiegeng(Platycodon grandiflorum), Danggui(Angelica sinensis), Gancao(Glycyrrhiza glabla), Chaihu(Bupleurum falcatnosa), Shoudehuang(Rehmannia glutinosa), Huangqi(Satragalus membranaceus), Muxiang(Saussurea lappa), Yuanzhi(Polygala tenuifolia), Rensen(Panax ginseng) and Baishaoyao(Paeonia lactiflolia), as Oriental radices herbs, on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of ${\gamma}-radiation$. Jiegeng(p<0.005), Danggui(p<0.0005), Gancao(p<0.005), Chaihu(p<0.05), Muxiang(p<0.05), Rensen(p<0.005) and Baishaoyao(p<0.005) were effective in intestinal crypt survival. Danggui(p<0.05), Chaihu(p<0.05), Shoudehuang(p<0.05), Huangqi(p<0.05), Rensan(p<0.005) and Baishaoyao(p<0.05) increased the formation of endogenous spleen colony. The frequency of radiation induced apoptosis was also reduced by pretreatment with Chaihu(p<0.05), Muxiang(p<0.005), Yuanzhi(p<0.05), Rensan(p<0.05) and Baishaoyao(p<0.05). Although the mechanisms of this effect remain to be elucidated, these results indicated that Danggui, Chaihu, Muxiang, Rensan and Baishaiyao might be a useful radioprotector, especially since it is a relatively nontoxic natural product.

  • The flowering locus T (FT) gene, of which expression will be controlled at high temperature by heat shock promoter (it printed as to HSproFT), was introduced into spray-type chrysanthemum (Dendranthema grandiflorum (Ramat.) Kitamura) 2 cultivars ('Pink PangPang' and 'Pink Pride' by co-cultivation with Agrobacterium tumefaciens strain C58C1 harboring pCAMBIA2300 containing the HSproFT gene. After leaf segments of the 2 cultivars were infected with the A. tumafaciens with C58C1 as explants, shoots were regenerated from the explants cultured on the $1^{st}$ selection medium (MS basal salts + 1.0 mg/L BA, 0.5 mg/L IAA + 10 mg/L kanamycin + 0.7% plant agar, pH 5.8). The shoots were transferred into the $2^{nd}$ selection medium (MS basal salts + 1.0 mg/L BA, 0.5 mg/L IAA + 20 mg/L kanamycin + 0.7% plant agar, pH 5.8). One hundred seventeen plantlets from 'Pink PangPang' and 5 ones from 'Pink Pride' were confirmed as transformants by PCR analysis. Twenty six of the transformants and non-transformants were acclimatized and established well in a green house. Eights of 26 transgenic lines showed flower bud 1.7~10 days earlier than nontransgenic plants, and 24 of them flowered 1~6 days earlier than non- transgenic plants. The shape and color of flower of all HSproFT-transgenic lines were not different with those of non- transgenic plants.

A Study on Quality Characteristics of Doraji (Platydodon grandiflorum) Yanggeng using by Different Pre-treatment Methods and Amounts Adding Levels of Doraji (전처리 방법과 비율을 달리하여 제조한 도라지 양갱의 품질 특성에 관한 연구)

  • The mechanical characteristics (texture and color) and consumer preference of Doraji Yanggengs using different pre-treatments and added levels of Doraji were presently evaluated. Three kinds of Doraji Yanggengs were made with differing amounts of Doraji powder (DPY; 30, 50, or 70 g), ground raw Doraji (GRDY; 100, 200, or 300 g), and Doraji juice (DJY; 100, 200, or 300 g). Texture characteristics (hardness, adhesiveness, springiness, chewiness, gumminess, cohesiveness) and color values (lightness, redness, yellowness) were measured three times in three experiments conducted with each formulation. Preference was measured with a nine-point hedonic scale for Yanggengs once every three experiments. Consumer preference was measured with color, flavor, taste, appearance, texture, and overall acceptability. Significant differences according to the amount of Doraji powder were evident with DPY for hardness (p<0.001), adhesiveness (p<0.05), and cohesiveness (p<0.001). Hardness (p<0.001), adhesiveness (p<0.001), chewiness (p<0.01), and gumminess (p<0.01) of 100 g GRDY displayed higher scores than 200 g and 300 g GRDY, but displayed significantly lower scores in cohesiveness (p<0.01). Hardness (p<0.001), adhesiveness (p<0.001), chewiness (p<0.001), gumminess (p<0.01), and cohesiveness (p<0.05) of DJY showed significant differences according to amount of Doraji juice. The highest score in a preference test among DPYs was evident for 30 g DPY. There were no significant differences in preference among GRDYs and DJYs. Color, flavor, taste, appearance, texture, and overall acceptability were significantly (p<0.01) correlated in all Yanggengs. DJY was preferred more than DPY and GRDY. Women in their forties and fifties preferred Yanggeng more when compared to women in their twenties and thirties.

Analysis of Vegetation-Environment Telationships of Main Wild Vegetables on Short-term Income Forest Products, in Korea (단기소득임산물 자생지 주요 산채류 식생과 환경의 상관관계 분석)

  • This study was conducted in 2016-2017 to provide the basic ecological data needed to establish environmental conditions for the cultivation of wild vegetables. It used TWINSPAN to classify the vegetation structure of natural habitats of wild vegetable nationwide and DCCA ordination to analyze the correlation between the by community structure and environmental factors. We performed TWINSPAN on 100 taxa with high importance values in 91 plots of major habitats of wild vegetables. The vegetation was classified into Cirsium setidens and Synurus deltoides group, Ligularia fischeri and Hemerocallis fulva group, Adenophora divaricata var. manshurica group, Platycodon grandiflorum and Aster scaber group, Aralia elata and Pteridium aquilinum group, and Pimpinella brachycarpa and Osmunda japonica group communities. We then performed DCCA ordination of 11 communities classified by TWINSPAN and 11 environmental factors. The results showed that the altitude had the strongest correlation with the vegetation. The Cirsium setidens, Synurus deltoids, and Lifularia fischeri communities were distributed in areas with similar environmental factors such as high altitude, gentle slope, and nutrient. The Aralia elata and Osmunda japonica communities were distributed in the location environment with low altitude, pH, O.M, T-N, $Ca^{2+}$, and C.E.C. The Hemerocallis fulva community was distributed in the location environment with moderate northeastern and northwestern slope, low altitude and pH, and high $P_2O_5$, whereas the Adenophora divaricata var. manshurica community was distributed in the location environment with gentle southeastern and southwestern slope, high altitude and pH, and low $P_2O_5$, which was the opposite tendency of the location environment from Hemerocallis fulva community. The Platycodon grandiflorum community was distributed in the location environment with gentle southwestern slope, low altitude, pH, O.M, T-N, $P_2O_5$, $Ca^{2+}$, and C.E.C., and high $Mg^{2+}$. The Pteridium aquilinum community was distributed in the location environment with southwestern slope, low altitude, O.M, T-N, C.E.C, $P_2O_5$, $Ca^{2+}$, and $K^+$. The Aster scaber and Pimpinella brachycarpa communities were widely distributed in many plots with various location environments.

Immunoadjuvant Effect of Platycodin D from Platycodon grandiflorum (Platycodin D 길경성분의 면역보조효과)

  • In vaccine development, the major points may be induction of effective and increased levels of antibody production. This is especially the case when the antigenic sources are carbohydrates. For many years, thus, we have researched various types of formulations such as liposomal and conjugate vaccines. However, the fastidious formulation process and high costs are a problem. For this reason, there is currently a focus on utilizing immunoadjuvants. In this present study, we tested if platycodin D (PLD) from Platycodon Radix have immunoadjuvant activity against the cell wall of Candida albicans (CACW). The resulting data showed that in the murine model of antibody production, CACW combined with PLD [CACW/PLD/IFA] increased the production of antibodies specific to C. albicans when compared to the antibody production by [CACW/IFA]-induction, which was used as a negative control (P<0.05). In the case of [CACW/PLD/IFA], the antibody production was 1.4 times as that of the CFA. In addition, formulations containing either had a prolonged antibody inducing activity maintaining the initial titers of antibody as compared to the CFA formula. Cytokine profiling with the antisera displayed that the PLD produced both Th1 and Th2 immunoresponses, but Th1 dominant was much greater (P<0.05). Furthermore, [CACW/PLD/IFA] formula enhanced resistance of mice against disseminated candidiasis, whereas the CFA had no such effect. In conclusion, PLD has an immunologic activity, which is protective against the disease. Thus, PLD can be a goof candidate for a new immunoadjuvant in development of the fungal vaccine.

The Changes of Benzo[α]pyrene in Herbal Teas containing Astragalus membranaceus, Schizandra chinensis, Liriope platyphylla and Platycodon grandiflorum Which are affected by the Puffing Conditions (Puffing 조건에 따른 황기, 오미자, 맥문동 및 길경을 첨가한 한방차의 Benzo[α]pyrene 변화)

  • The following study is the result of herbal teas puffed at different temperatures between $140{\sim}220^{\circ}C$. Depending on treatment temperatures, the water contents decreased, while some carbonization occurred and crude ash contents relatively increased. Also, the crude protein and crude fat experienced little changes. B(${\alpha}$)P contents (0.16~0.17 ppb) showed little change according to treatment temperatures. From this result, the B(${\alpha}$)P content differed depending on the treatment temperature and raw materials. Solid elution rate figures of the herbal teas ranged from 0.27~0.45% (w/w) and the rate of solid elution increased along with higher puffing temperatures. The reason for the increase in solid elution rates is due to the breakage of cross bridges between the raw materials in the herbal tea which are carbohydrates, proteins, lipids and etc. after treatments of physical changes rather than chemical ones.
