유근피가루 첨가가 쌀다식의 품질에 미치는 영향 (Effect of Ulmus Davidiana Powder on the Quality of Rice Dasik)

  • To evaluate the effect of Ulmus davidiana powder on rice dasik, Ulmus davidiana dasik was made after supplementation with Ulmus davidiana powder(0, 3, 6, 9 and 12%(all w/w)). The proximate component of Ulmus davidiana powder was 7.49% moisture content, 2.33% crude protein, 0.84% crude fat, 14.09% crude fiber and 8.50% crude ash. The moisture content of samples were significantly(p<0.05) lower than that of dasiks with Ulmus davidiana powder. The pH values were significantly decreased with increased Ulmus davidiana powder(p<0.05). The lightness(L-value) was significantly decreased as Ulmus davidiana powder increased(p<0.05). Redness(a-value) of dasik with Ulmus davidiana powder was significantly higher than that of the control group. However redness was decreased with the amount of increased Ulmus davidiana powder. The yellowness(b-value) decreased significantly as Ulmus davidiana powder increased(p<0.05). In the mechanical evaluation, hardness in the groups with Ulmus davidiana powder increased significantly(p<0.05). Adhesiveness of the control group was lower than that of the groups with Ulmus davidiana powder. Springness in the control group was lower than that in the groups with Ulmus davidiana powder. But no significant difference in cohesiveness, gumminess and chewiness among groups was found. The intensity of color, odor and astringency of dasik with Ulmus davidiana powder significantly increased as the amount of supplementation increased(p<0.05). And the preference of flavor with 12% Ulmus davidiana powder was significantly lower than that of the control group. In conclusion, rice dasik with less than 9% of Ulmus davidiana powder would be most proper in taste and overall acceptability.

  • P. alba${\times}$glandulosa 교잡종(交雜種)의 지속적(持續的)인 개량(改良)을 위하여 교배양친수(交配兩親樹)의 gene population의 규모가 커야 할 것이나 현재(現在) P. glandulosa는 몇 그루만이 존재(存在)하고 또한 이 수종(樹種)은 P. alda와 P. davidiana의 자연잡종(自然雜種)에서 분리(分離)된 P. davidiana에 가까운 개체(個体)로 인식(認識)하고 있는바 P. glandulosa의 gene population을 넓힐려면 P. alba와 P. davidiana의 여러 교잡종(交雜種)을 만들어 우선 잎의 특성(特性)을 비교(比較)하여 P. glandulosa에 가까운 개체(個体)를 선발(選拔)코저 본시험(本試驗)을 실시(實施)한바 그 결과(結果)는 다음과 같다. 1. P. glandulosa는 엽리면(葉裏面)의 털이 약간 있고 잎 가장자리는 serrate. 엽기부(葉基部)에 gland가 있다. 교잡(交雜)에서 이 특성(特性)에 유사한 개체(個体)는 P. alba davidiana${\times}$P. davidiana에서 44%, P. davidiana alba${\times}$P. davidiana에서 90%가 나타났고 2. 잎의 크기의 비(比) 즉 엽장(葉長), 엽폭(葉幅), 엽기부(葉基部)에서 장폭(帳幅)까지 길이, 엽병장(葉柄長)을 비교(比較)한 결과(結果) P. glandulosa와 유사하여 선발(選拔)한 개체(個体)들은 잎크기의 비(比)가 P. glandulosa와 비슷 하였으며 3. 엽리면(葉裏面)의 은모(銀毛)가 차대(次代)에 유전(遺傳)하는 정도(程度)는 교배양친수종(交配兩親樹種)의 중간(中間) 정도(程度)의 형질(形質)을 나타내며 P. alba를 교배모수(交配母樹) 혹은 화분수(花粉樹)로 사용했을 때 그 차대(次代)에 털의 밀도(密度)는 서로 다르게 나타났다. 즉 화분수(花粉樹)로 사용했을 때 그 차대(次代)에 털의 밀도(密度)가 높았다. 4. P. glandulosa의 엽기부(葉基部)에 gland는 P. davidiana의 gland가 있는 개체(個体)에서 유전(遺傳)된 것으로 생각된다. 즉 P. alba에는 gland가 없고 P. davidiana는 gland가 있는 개체(個体)도 있고 없는 개체(個体)도 있으므로 gland가 있는 P. davidiana로부터 유전(遺傳)된 것으로 생각된다.

유근피(楡根皮) 추출물의 활성종 억제 및 염증 촉진 인자 제어 효과 (The Inhibitory Effects of Ulmus davidiana on the Reactive Species and Proinflammatory Proteins)

  • Objectives : This study was to investigate the inhibitory effects of Ulmus davidiana on the generation of peroxynitrite $(ONOO^{-})$, nitric oxide (NO) and superoxide anion radicals $(O_{2}^{-})$ in the endothelial cells of rat vessels. The effects of Ulmus davidiana on the expression of inflammation-related proteins, $NF-{\kappa}B$ (p50, p65), COX-2, and iNOS, were examined by western blotting. Methods : For this study, fluorescent probes, namely dihydrorhodamine 123 (DHR 123), 4,5-diaminofluorescein (DAF-2) and 2',7'-dichlorodihydrofluorescein diacetate (DCFDA) were used. Western blotting was performed via using anti-$NF-{\kappa}B$ (p50, p65), anti-COX-2, and anti-iNOS, respectively. Results : Ulmus davidiana inhibited the generation of $ONOO^{-}$, NO and $(O_{2}^{-})$ in the lipopolysaccharide (LPS)-treated endothelial cells of rat vessels in vitro. Ulmus davidiana inhibited the expression of COX-2 and iNOS genes by means of decreasing the $NF-{\kappa}B$ activation. Conclusions : These results suggest Ulmus davidiana is effective on inhibiting the generation of $ONOO^{-}$, NO and $O_{2}^{-}$, and that therefore it might have a potential role as a treatment for the inflammatory process and inflammation-related diseases.

느릅 분말을 첨가한 요구르트의 이화학적 및 관능적 품질 특성 (Physicochemical, Microbial, and Sensory Properties of Yogurt with Ulmus davidiana var. japonica During Storage)

  • This study evaluates changes in yogurt with Ulmus davidiana var. japonica (U. davidiana) in terms of its physicochemical, microbial, and sensory properties during storage. The pH value and mean microbial counts of this yogurt with 0.2 to 0.8% of U. davidiana added ranged from 4.1 to 4.3 and from $1.4{\times}10^8$ to $2.3{\times}10^8cfu/mL$, respectively, during storage at $7^{\circ}C$ for 16 d. In terms of its color, the L value was significantly higher in the control and U. davidiana yogurt 0.2 to 0.4% up to 4 d, than other yogurt samples, and the b value was influenced by the addition of U. davidiana over 0.6% throughout the storage. According to a sensory test, there was a significant difference in the yellowness score between U. davidiana yogurt and the control, but earthy, and bitterness, and viscosity scores were similar. Overall acceptability was not significantly influenced after 8 d of storage in all yogurt samples, including the control. According to these results, concentrations (0.2 and 0.4%) of U. davidiana yogurt had no significantly adverse effects on its physicochemical, microbial, and sensory properties.

초미세 느릅 분말의 항산화 활성 연구 (Antioxidant Activities of Powdered and Ultra-fine Powdered Ulmus Davidiana var. Japonica)

  • The antioxidant activity was examined on ultra-fine powdered (UFP) Ulmus davidiana (U. davidiana) var. japonica. The average particle size of UFP or commercial powdered (CP) was $1-3{\mu}m$ or $100{\mu}m$. The extraction was performed using either ethanol (EtOH) or hot-water. Contents of phenolic compound of CP and UFP U. davidiana extracts in EtOH was 40.38 and 65.61 mg/g, respectively. In DPPH, EtOH extract of UFP U. davidiana showed a significantly greater activity than hot-water extract at 40 and $200{\mu}g/mL$. At $200{\mu}g/mL$, the activity was over 90% in all groups. The reducing power of UFP U. davidiana var. japonica in EtOH extraction was 74.3%, which was significantly greater than in other samples (p<0.05). In addition, reducing powder was significantly higher in UFP-EtOH than in other samples at all concentrations except for $0.32{\mu}g/mL$. The above results suggest that EtOH extraction of U. davidiana showed slightly higher DPPH radical scavenging activity, and ultra-fine powder of U. davidiana extracts may show higher antioxidative activities based on reducing power.

  • 자연잡종(自然雑種)으로 알려진 Populus glandulosa와, P. tomentiglandulosa에 대(対)하여 그의 잡종성(雑種性)과 분류학적(分類學的) 위치(位置)를 구명(究明)하기 위(為)하여, 형태(形態), 생리(生理), 및 핵학적(核學的)인 조사(調査)를 하여 다음과 같은 결과(結果)를 얻었다. 1. 접(接), 삽목(揷木)의 활착력(活着力)은 잡종간(雑種間)에 차이(差異)있고, 삽목발근력(揷木発根力)은 불완전(不完全) 우성(優性)이다. 2. 개엽성(開葉性)은 부완전(不完全) 우성(優性)이고 착엽성(着葉性)은 P. alba ${\times}$ glandulosa와 P. tomentiglandulosa가 강(強)하다. 3. $KClO_3$에 대(対)한 항독성(抗毒性)은 잡종간(雑種間)에 차(差)가 있다. 4. 엽(葉)의 많은 외부(外部) 형질(形質)들은 불완전(不完全) 우성(優性)이고, P. tomentiglanaulosa는 P. alba ${\times}$ glanaulosa와, 그리고, P. glandulosa는 P. davidiana와 P. alba와의 정(正), 역(逆) 또는 back cross hybrid와 유사(類似)하다. 5. 웅화수(雄花数)는 불완전(不完全) 우성(優性) 또는 잡종강세(雑種強勢) 현상(現象)을 하고, P. tomentiglandulosa는 P. alba ${\times}$ glandulosa와 그리고, P. glandulosa는 P. alba ${\times}$ davidiana 또는 P. davidiana ${\times}$ alba와 수(数)가 근사(近似)하다. 6. male catkin의 포(苞)의 형태(形態)와 대색(帶色)은 불완전(不完全) 우성(優性)이고, P. glandulosa는 P. alba ${\times}$ davidiana에, 그리고 P. tomentiglandulosa는 P. alba ${\times}$ glandulosa와 유사(類似)하다. 7. 엽병(葉炳)의 유관속(維管束)의 수(数)와 배열형(配列型)은 잡종간(雑種間)에 차이(差異)가 있고 잡종(雑種)들은 복잡(複雜)한 형(型)을 하고 있다. 8. 유경(幼莖)의 해부학적(解剖學的) 성질(性質)은 잡종간(雑種間)에 차(差)가 없으나, 목섬유(木纖維)의 크기는 차(差)가 있다. 9. metaphase I과 II에 있어, 인공(人工) 잡종(雑種)과 P. glandulosa 또는 P. tomentiglandulosa는 염색체(染色體) 행동(行動)이 불규칙(不規則)하였다. 10. metaphase I에서 잡종(雑種)과 P. glandulosa 그리고 P. tomentiglandulosa는 19II의 핵판(核板)이 적고, 일가(一価) 염색체(染色体)가 다수(多数) 출현(出現)한다. 11. anaphase I과 II에 있어, laggard chromosome과 chromosome bridge가 있는 이상핵판(異常核板)이 잡종(雑種)과 P. glandulosa 그리고 P. tomentiglandulosa에서 다수(多数)히 출현(出現)하였다. 12. pollen tetrad와 임성화분(稔性花紛)의 출현율(出現率)이 잡종(雑種)과 P. glanaulosa, 그리고, P. tomentiglandulosa에서 낮았다.

Inhibitory Activity of Flavonoids from Prunus davidiana and Other Flavonoids on Total ROS and Hydroxyl Radical Generation

  • Since reactive oxygen species (ROS) and hydroxyl radicals ($^-OH$) play an important role in the pathogenesis of many human degenerative diseases, much attention has focused on the development of safe and effective antioxidants. Preliminary experiments have revealed that the methanol (MeOH) extract of the stem of Prunus davidiana exerts inhibitory/scavenging activities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, total ROS and peroxynitrites ($ONOO^-$). In the present study, the antioxidant activities of this MeOH extract and the organic solvent-soluble fractions, dichloromethane (CH$_2$Cl$_2$), ethyl acetate (EtOAc), and n-butanol (n-BuOH), and the water layer of P. davidiana stem were evaluated for the potential to inhibit $^-OH$ and total ROS generation in kidney homogenates using 2',7'-dichlorodihydrofluorescein diacetate (DCHF-DA), and for the potential to scavenge authentic $ONOO^-$. We also evaluated the inhibitory activity of seven flavonoids isolated from P. davidiana stem, kaempferol, kaempferol 7-Ο-$\beta$-D-glucoside, (+)-catechin, dihydrokaempferol, hesperetin 5-Ο-$\beta$-D-glucoside, naringenin and its 7-Ο-$\beta$-D-glucoside, on the total ROS, $^-OH$ and $ONOO^-$ systems. For the further elucidation of the structure-inhibitory activity relationship of flavonoids on total ROS and 'OH generation, we measured the antioxidant activity of sixteen flavonoids available, including three active flavonoids isolated from P. davidiana, on the total ROS and 'OH systems. We found that the inhibitory activity on total ROS generation increases in strength with more numerous hydroxyl groups on their structures. Also, the presence of an ortho-hydroxyl group, whether on the Aring or S-ring, and a 3-hydroxyl group on the C-ring increased the inhibitory activity on both total ROS and $^-OH$ generation.

유근피(楡根皮) 약침이 lipopolysaccharide 유발 류마티스 관절염 모델에서 MIF 활성 억제에 미치는 영향 (The Effectiveness of Ulmus Davidiana Planch Herbal Acupuncture to Inhibit MIF Activation on Lipopolysaccharide-induced Rheumatoid Arthritis Model)

  • Objectives : The purpose of this study is to investigate the effect of Ulmus davidiana Planch herbal acupuncture solution in LPS-stimulated RAW 264.7 cells and mouse knee joints, perfom1ed several experimental items: those are MIF mRNA, MIF, $TNF-{\alpha}$, $NF-{\kappa}B$ p65, iNOS mRNA, iNOS, NO, synovial hyperplasia, angiogenesis and fibrosis. Methods : In order to observe mRAN expression of MIF and iNOS in LPS-stimulated RAW 264.7 cells, RT-PCR was used. NO production in LPS-stimulated RAW 264.7 cells was measured by nitrite assay. All the female BALB/c mice were bred and maintained in pathogen-free mouse colonies and were 6 weeks of age on beginning of the experiment. The experimental model of RA was induced by injection of $50{\mu}g/kg$ LPS. Ulmus davidiana Planch herbal acupuncture solution was injected into either S 35 (犢鼻) or EX-LE 202 (內膝眼) of mice in turn daily for 19 days. Immunohistochemical staining was carried out to assess $TNF-{\alpha}$, $NF-{\kappa}B$ p65 and iNOS expression in synovial membrane. Synovial hyperplasia, angiogenesis and fibrosis in synovial membrane was observed with a microscope. Results : 1. Ulmus davidiana Planch herbal acupuncture solution inhibited mRNA expression of MIF and iNOS in dependence on a density of it in LPS-stimulated RAW 264.7 cells. 2. Ulmus davidiana Planch herbal acupuncture solution decreased synovial hyperplasia, angiogenesis and fibrosis in LPS-stimulated mouse knee joints. 3. Ulmus davidiana Planch herbal acupuncture solution curtailed production of MIF, $TNF-{\alpha}$, $NF-{\kappa}B$ p65, iNOS in LPS-stimulated mouse knee joints. Conclusion : On the basis of these results, It was shown that Ulmus davidiana Planch herbal acupuncture solution is significantly able to inhibit the production of MIF as a top in cytokines related to inflammatory or irrlll1une responses. Our results may provide that Ulmus davidiana Planch herbal acupuncture solution has beneficial effect in not only RA but other inflammatory or immune deases.

토양개량재와 황토가 준설토의 이화학성과 느릅나무의 초기 생장에 미치는 영향 (Effect of Soil Conditioner and Loess on Physio-Chemical Properties of Dredged Soil and Seedling Height Growth of Ulmus davidiana var. japonica)

  • This research was conducted to compare the effect of soil amendment and loess on physial and chemical properties of dredged soil and primary height growth of Ulmus davidiana var. japonica. Three types of research plots: control plot (Dredged soil only), DC treatment plot (Dredged soil + soil amendment) and DCL treatment plot (Dredged soil + soil amendment + Loess) were set at Okgu research site in Saemanguem. 32 trees of U. davidiana var. japonica were planted in each plots after 14 months from the construction. Soil texture, pH, electrical conductivity (EC), organic matter (OM), total nitrogen (TN), available P, exchangeable cation ($K^+$, $Na^+$, $Mg^{2+}$, $Ca^{2+}$) and sodium chloride were measured after 3 years from the construction. Also, survival rate and height of U. davidiana var. japonica were measured 22 months after planting, and the correlation between height of trees and the physico-chemical properties of soil were analyzed. Even though pH in both DC and DCL treatment plots are lower than control plot, they are still alkaline (> pH 8). OM, TN and available P in both DC and DCL treatment were higher than the control. Particularly, the content of available P in both DC and DCL treatment plots are 1.4~5.1 times and 2.0~3.1 times higher than the control respectively. The concentration of exchangeable Mg in DCL treatment plot was 1.1~5.5 times higher than the control (p < .05). The survival rate of the species was the highest in DCL treatment plot (98%) followed by DC treatment plot and the control. The average height of the trees in both DC and DCL treatment plots is 1.1m while the control is 0.8m. OM, TN, available P, K+ were significantly related to the height of U. davidiana var. japonica(p < .01). The results indicate that soil amendment affects on soil physial and chemical properties of dredged soil and height growth of U. davidiana var. japonica.

Antioxidant and Antidiabetic Activities of Ulmus davidiana Extracts

  • The antioxidant activities of water, ethanol, methanol, and chloroform extracts of Ulmus davidiana were evaluated using various antioxidant assays: DPPH (1,1-diphenyl-2-pricrylhydrazyl) free radical scavenging; hydroxyl radical scavenging; lipid peroxidation scavenging; and reducing power assays. All extracts, except the chloroform extract, demonstrated strong antioxidant activity in all assays. The chloroform extract had the highest hydroxyl radical scavenging activity, and its activity was equivalent to $\alpha$-tocopherol at a concentration of 0.5 mg/mL. Additionally, the antidiabetic activity of their extracts was also evaluated using a rat intestinal $\alpha$-glucosidase inhibition assay. Among all extracts investigated, the methanol extract had the highest $\alpha$-glucosidase inhibitory activity, although its activity was less than acarbose at $0.5\;{\mu}g/mL$. This result suggested that U. davidiana extracts may have antidiabetic activity. Total phenolic compounds and flavonoids were also measured. Phenolic compounds such as tannic acid, p-coumatric acid, and kaempferol were detected by high-performance liquid chromatography (HPLC). These results suggest that U. davidiana extracts may be useful as a potential source of antioxidant and antidiabetic materials.
