Absorption of Itraconazole from Rat Small Intestine (이트라코나졸의 랫트 소장으로부터의 흡수)

  • The absorption characteristics of itraconazole, which is an antifungal agent, from intestinal segments in the anesthetized rat i1l situ were investigated in order to design an effective oral drug delivery system. The pH-solubility profile of itraconazole, the rate and extent of absorption of itraconazole, the optimal absorption site(s) of itraconazole and the absorption enhancing effect of sodium cholate on itraconazole were examined in the present study. In situ single-pass perfusion method and recirculating perfusion technique using duodenum(D), jejunum(J) and ileum(I) were employed for the calculation of apparent permeability(Pe) and apparent first-order rate constant(Kobs). respectively. The results of this study were as follows; (1) Itraconazole showed appreciable aqueous solubility only at pH values of below 2.0. (2) pe(cm/sec) decreased in the following order: $D(10.24{\pm}1.78{\times}10^{-4})>J(8.86{\pm}0.79{\times}10^{-4})>I(3.78{\pm}0.13 X 10^{-4})$. (3) $Kobs(min^{-1})$ decreased in the following order: $J(17.12{\pm}3.19{\times}10^{-3})>D(13.37{\pm}0.6{\times}10^{-3})>I(11.05{\pm}0.91{\times}10^{-3})$. (4) The solubility of itraconazole markedly increased with the increase of the concentration of sodium cholate. (5) The addition of 10 mM sodium cholate significantly increased the apparent first-order rate constant of itraconazole in the ileum by a factor of 6.8.

  • The present study attempted to impregnate alkali metals to amino acid in order to improve $CO_2$ absorption capacity. A used amino acid was glycine, of which pH increased up to about 11 with the addition of alkalies. $CO_2$ absorption capacity of amino acid salts was evaluated in a batch and a continuous process. The absorption capacity appeared in turns as; Sodium Glycinate ${\geq}$ Lithium Glycinate > Potassium Glycinate. Amino acid salts showed lower absolute capacity of $CO_2$ absorption than primary amine (Monoethanolamine) at $20^{\circ}C$. In a continuous absorption with 10% $CO_2$ flow, the increasing the reaction temperature, the increasing rate of absorption for amino and was higher that of than amino absorbent.

  • The electromagnetic (EM) properties of media, such as propagation, focusing and scattering, strongly rely on the electric permittivity and the magnetic permeability of media. Recently, artificially-created metamaterials (MMs) composed of periodically-arranged unit cells with tailored electric permittivity and magnetic permeability have drawn wide interest due to their capability of adjusting the EM response. MM absorbers using the conventional sandwich structures usually have very high absorption at a certain frequency, and the absorption properties of MMs can be adjusted simply by changing the geometrical parameters of unit cell. In this work, we suggested an incident-angle-independent broadband perfect absorber based on resistive layers. We analyze the absorption mechanism based on the impedance matching with the free space and the distribution of surface currents at specific frequencies. From the simulation, the absorption was expected to be higher than 96% in 1.4-6.0 GHz. The corresponding experimental absorption was found to be higher than 96% in 1.4-4.0 GHz, and the absorption turned out to be slightly lower than 96% in 4.0-6.0 GHz owing to the irregularity in the thickness of resistive layers.

Studies on Drug Absorption Characteristics for Development of Ocular Dosage Forms: Ocular and Systemic Absorption of Topically Applied ${\beta}-Blockers$ in the Pigmented Rabbit

  • The objective of this study was to determine the influence of drug lipophilicity on the extent of ocular and systemic absorption following topical solution instillation in the pigmented rabbit. ${\beta}-Blockers$ of various lipophilicity were chosen as model drugs, $25\;{\mu}l$ of a 15 mM drug solution in isotonic pH 7.4 buffer was instilled, and ocular tissue and plasma drug concentrations were monitored. Ocular absorption was apparently increased in all eye tissues, but non-corneal absorption ratio was decreased by increasing of drug lipophilicity. Systemic bioavailability was ranged from 61% for atenolol to 100% for timolol, and at least 50% of the systemically absorbed drug reached the blood stream from the nasal mucosa. Occluding the nasolacrimal duct for 5 min reduced the extent of systemic absorption of timolol and levobunolol, but did not do so for atenolol and betaxolol. Taken together, the ocular absorption of topically applied ophthalmic drugs would be modest for lipophilic drugs. By contrast, the systemic bioavailability would be modest for drugs at the extremes of lipophilicity, and the nasal contribution to systemically absorbed drug diminished with increasing of drug lipophilicity.

Iron Bioavailability in Iron-fortified Market Milk (철분강화 우유의 생이용성 평가)

  • To evaluate iron bioavailability in iron fortified milk, in vitro and in vivo method were used. Low molecular weight components(ILC) from milk was isolated and iron was added, then soluble iron from ILC iron complex was determined. Each iron sources and extrinsically labelled with FeCl3 was used for measuring absorption rate of iron from ILC radiolabelled iron complexes as radioiron absorption into the blood one hour after injection into ligated duodenal loops of iron deficient rats. Iron absorption rate was in the order of ferrous lactate(25.56%)$\geq$ferric citrate(24.71%)$\geq$ferrous sulfate(19.67%) when 100ppm iron was used. In separate experiments, iron fortified milks with each iron sources were gavaged into iron deficient rats. When 25ppm iron was added to milk, the order of iron absorption was ferrous sulfate(12.52%)>ferrous lactate(8.07%)>ferric citrate(6.52%) (p<0.05). When 100ppm iron was added to milk, absorption rate was decreased compared to the treatments with added 25ppm of iron. Absorption rate of ferrous sulfate(5.34%) from milk added 100ppm iron was highly lowered, but ferric citrate(6.45%) was not significantly changed. The absorption rate of ferrous lactate(5.82%) was 70% of 25ppm iron added milk.

  • The reciprocal effects towards the enhanced biological phosphorus removal were performed for anaerobic, aerobic and anoxic phases. The batch experiments showed that the p-absorption in the anoxic phase was 50% lower than aerobic phase. The correlation coefficient between p-back-solution and p-absorption was found to be $R^2=0.557$ however, the coefficient b(b = 8.4049) was relatively higher than the other researchers results. The increase and/or acceptance of the $K^+-,\;Mg^{2+}-$ and $NH_4-N$-concentration was proportional to those of the $PO_4-P$-concentration in the batch test. The relationship between $K^+-,\;Mg^{2+}$ and $PO_4-P$ was determined. The average value of this relation-ship agreed with 0.2 mol $K^+Ion$ / mol $PO_4-P$ ion and 0.21 mol $Mg^{2+}Ion$ / moi $PO_4-P$ ion in the anaerobic phase. The absorbed ratio of $K^+$ to $Mg^{2+}$ over $PO_4-P$ was found to be 1 : 5.

Solubility, Emulsion Capacity, and Emulsion Stability of Protein Recovered from Red Crab Processing Water (홍게 가공회수 단백질의 용해도, 유화력 및 안정성)

  • The functional properties of protein recovered from red crab (Chitinonecetes opiiie) processing in water (RCP) were examined and compared with those of soybean protein isolate at pH 2~10 in water and NaCl solu5ion. The solubilities of RCP and SPI were miniumu at pH 4, the isoelectric point and increased significantly at lower or higher than pH 4. Solubilities in NaCl solution for both proteins decreased with incr NaCl concentration increase at all pH ranges. Emulsion capacity for both proteins was also minimum at pH 4 and increased as protein concentration increased from 2 to 6%. Emulsion capacity of RCP was higher than these of SPI at pH 6∼10 and all protein concentrations. Emulsion stability showed a similar trend to that of emulsion capacity. RCP had higher oft absorption capacity and lower water absorption capacity than SPI.

Simple Autocorrelation Measurement by Using a GaP Photoconductive Detector

  • We developed a simple and real-time readout autocorrelator for several tens and sub-10fs pulses, based on the two photon absorption phenomena of a commercial GaP photodetector including a transimpedance amplifier. With a suitable gain adjustment, we demonstrated that the interferometric autocorrelation for sub-nJ pulses delivered as a high output voltage as to resolve all fringes in an autocorrelation trace with features of low noise and a low offset voltage. By fitting the measured quadratic power dependence of output voltages, we obtained the quantum efficiency of TPA for the GaP detector comparable with those of a GaAsP diode and an SHG with a thin BBO crystal. The autocorrelator of a TPA based GaP photodetector is highly suitable for sensitively measuring a few cycle pulses with a broad spectral distribution from 600 nm to 1100 nm.

Possibility of Involvement of Porphyromonas gingivalis in Coronary Heart Disease

  • Porphyromonas gingivalis has been implicated in periodontal diseases. Accumulating evidence suggests that cardiovascular disease is the most prevalent medical problem in patients with periodontal diseases. In order to check the possibility that P. gingivalis is involved in coronary heart disease, the present study was performed to observe P. gingivalis adherence and invasion of human coronary artery endothelial cells (HCAEC) and production of cytokines and growth factors by HCAEC upon P. gingivalis infection. $^3H$-labeled P. gingivalis 381 was incubated with HCAEC for 90 min. The radioactivity of the washed HCAEC was a measure of the absorbed (adhering and invading) P. gingivalis. The absorption radioactivity of the HCAEC infected by P. gingivalis was determined to be 59.58% of the input bacterial cells. In contrast, the absorption radioactivity of the cells infected by S. gordonii Challis which was employed as a control was negligible (0.59%). DPG3, a P. gingivalis mutant defective of fimbriae, appeared to be impaired to some extent in capability of adherence/invasion as compared to that of the parental strain 381, showing 43.04% of the absorption radioactivity. The absorption radioactivity of the HCAEC infected by P. gingivalis 381 in the presence of excessive fimbriae at the concentrations of $50\;{\mu}g$ and $100\;{\mu}g/ml$ was 57.27 and 45.44%, respectively. Invasion of HCAEC by P. gingivalis 381 was observed by an antibiotic (metronidazole) protection assay and transmission electron microscopy (TEM). In the antibiotic protection assay, invasion by the bacterium was measured to be 0.73, 1.09, and 1.51% of the input bacterial cells after incubation for 30, 60, and 90 min, respectively. Invasion by DPG3 was shown to be 0.16% after 90-min incubation. In comparison of invasion efficiency at 90 min of the incubation, the invasion efficiency of DPG3 was 0.37% while that of its parental strain 381 was 2.54%. The immunoblot analysis revealed fimbriae of P. gingivalis did not interact with the surface of HCAEC. These results suggest that fimbriae are not the major contribution to the adherence of P. gingivalis to HCAEC but may be important in the invasion of HCAEC by the bacterium. The presence of cytochalasin D ($1\;{\mu}g/ml$) and staurosporine ($1\;{\mu}M$) reduced the invasion of HCAEC by P. gingivalis 381 by 78.86 and 53.76%, respectively, indicating that cytoskeletal rearrangement and protein kinase of HCAEC are essential for the invasion. Infection of P. gingivalis induced HCAEC to increase the production of TNF-${\alpha}$. by 60.6%. At 90 min of the incubation, the HCAEC infected with P. gingivalis cells was apparently atypical in the shape, showing loss of the nuclear membrane and subcellular organelles. The overall results suggest that P. gingivalis may cause coronary heart disease by adhering to and invading endothelial cells, and subsequently damaging the cells.

Pharmacokinetics of Omeprazole-Resin by Crossover Design and the Variation of Absorption upon pH Change in the Guts of the Rat and the Rabbit (Omeprazole 수지염의 흰쥐와 토끼에서의 위장관내 산도변화에 따른 흡수변화 및 교차시험법에 의한 약물동태연구)

  • Pharmacolinetic profiles of omeprazole enteric coated granules including Ramezole$^\circledR$, Losec$^\circledR$, omeprazole-Na and omeprazole-resin salt were studied using the crossover design in rats and rabbits. The absorption variance of the preparations at the altered pH condition of the gastrointestinal tract was also studied. After oral administration of four omeprazole enteric coated pellets (10mg/kg) with and without concomitant administration NaHCO$_{3}$ (5 mg/ml, 60 mM) in the rats, the differences of absorplion rate and extent were evaluated. In the NaHCO$_{3}$, administration group, the T$_{max}$ appeared to be 2~10 times shorter than water administration group, and the $C_{max}$ also increased to about 4 times, and the AUC increased to about 2.5 times. Pharmacokinetic parameters of four omeprazole enteric coated pellets in rats showed no statistical significance (ANOVA, P>0.05) in both groups. In the crossover study, the second dosed drug showed 4~5 times increased bioavailability than first dosed drug, which shows the strong carry-over effect of acid secretion of the first dosed drug. The differences of the pharmacokinetic parameters of the two test formulations (Losec$^\circledR$ and omeprazole-resin) showed no statistical significance.
