엉겅퀴, 울금, 개똥쑥 복합 추출물의 지방간 개선효과 (Ameliorative Effects of Cirsium jaonicum, Artemisia annua and Curcuma longa on Non-alcoholic Fatty Liver Disease)

  • In this study, the effect of complex hot water extracts of Cirsium jaonicum, Artemisia annua and Curcuma longa (CAC) on the improvement of non-alcoholic fatty liver disease (NAFLD) was investigated. CAC inhibited fatty acid synthesis and lipid accumulation in HepG2 cells cultured with free fatty acid (FFA). In the NAFLD animal model, CAC extract suppressed the increase in body weight, liver, and epididymis fat weight, and suppressed the increase in hepatocyte fat and blood triglyceride. In addition, by blocking the Nrf2/HO-1 signaling pathway, cells were protected from oxidative stress in hepatocytes. Moreover, CAC inhibited the expression of COX-2, iNOS, TNF-α and IL-17 in hepatocytes. These results suggest the possibility that CAC extract can be applied in the field of health functional foods and pharmaceuticals for improvement and prevention of NAFLD.

형질전환 벼에서 Protoporphyrinogen Oxidase의 발현 위치가 제초제 저항성에 미치는 영향 (Expression Site of Protoporphyrinogen Oxidase Influences on Herbicide Resistance in Transgenic Rice)

  • 과산화계 제초제 oxyfluorfen이 처리된 비형질전환 벼와 형질전환 벼에서 Protox 발현 위치가 제초제 저항성에 미치는 영향을 비교하였다. Arabidopsis protoporphyrinogen oxidase(Protox; AP 계통)를 색소체에만 발현하는 형질전환 벼와 Myxococcus xanthus Protox 유전자를 색소체와 미토콘드리아에 모두 발현하는 형질전환 벼(TTS 계통)가 형질전환 시스템으로 사용되었다. Oxyfluorfen이 처리된 TTS4 계통은 AP 계통이나 비형질전환 벼에 비해 낮은 수준의 세포질 누출 및 malonyldialdehyde를 보여주었고, 높은 5-aminolevulinic acid 합성 능력을 유지하였다. Oxyfluorfen 작용 동안, TTS4 계통은 AP1 계통보다 높은 제초제 저항성을 보여주었는데, 이는 색소체만에서의 Arabidopsis Protox의 발현에 비해 색소체와 미토콘드리아에서의 M. xanthus Protox의 쌍발현이 광역학적인 protoporphyrin IX의 축적을 더 효율적으로 억제하였기 때문일 것이다. 이 결과들은 미토콘드리아 내 Protox의 발현이 Protox 저해형 제초제에 대한 식물의 저항성에 크게 기여함을 의미한다.

Increased Methylation of Interleukin 6 Gene Is Associated with Obesity in Korean Women

  • Obesity is the fifth leading risk for death globally, and a significant challenge to global health. It is a common, complex, non-malignant disease and develops due to interactions between the genes and the environment. DNA methylation can act as a downstream effector of environmental signals; analysis of this process therefore holds substantial promise for identifying mechanisms through which genetic and environmental factors jointly contribute to disease risk. To assess the effects of excessive weight and obesity on gene-specific methylation levels of promoter regions, we determined the methylation status of four genes involved in inflammation and oxidative stress [interleukin 6 (IL6), tumor necrosis factor ${\alpha}$ ($TNF{\alpha}$), mitochondrial transcription factor A (TFAM), and glucose transport 4 (GLUT4)] in blood cell-derived DNA from healthy women volunteers with a range of body mass indices (BMIs) by methylation-specific PCR. Interestingly, the samples from obese individuals ($BMI{\geq}30kg/m^2$) showed significantly increased hypermethylation for IL6 gene compared to normal weight ($BMI<23kg/m^2$) and overweight sample ($23kg/m^2{\leq}BMI<30kg/m^2$) (P = 0.034 and P = 0.026). However there was no statistically significant difference in promoter methylation of the other 3 genes between each group. These findings suggest that aberrant DNA methylation of IL6 gene promoter may play an important role in the etiology and pathogenesis of obesity and IL6 methylation could be used as molecular biomarker for obesity risk assessment. Further studies are required to elucidate the potential mechanisms underlying this relationship.

Nrf2 Expression and Apoptosis in Quercetin-treated Malignant Mesothelioma Cells

  • NF-E2-related factor 2 (Nrf2), a basic leucine zipper transcription factor, has recently received a great deal of attention as an important molecule that enhances antioxidative defenses and induces resistance to chemotherapy or radiotherapy. In this study, we investigated the apoptosis-inducing and Nrf2- upregulating effects of quercetin on malignant mesothelioma (MM) MSTO-211H and H2452 cells. Quercetin treatment inhibited cell growth and led to upregulation of Nrf2 at both the mRNA and protein levels without altering the ubiquitination and extending the half-life of the Nrf2 protein. Following treatment with quercetin, analyses of the nuclear level of Nrf2, Nrf2 antioxidant response element-binding assay, Nrf2 promoter-luc assay, and RT-PCR toward the Nrf2-regulated gene, heme oxygenase-1, demonstrated that the induced Nrf2 is transcriptionally active. Knockdown of Nrf2 expression with siRNA enhanced cytotoxicity due to the induction of apoptosis, as evidenced by an increase in the level of proapoptotic Bax, a decrease in the level of antiapoptotic Bcl-2 with enhanced cleavage of caspase-3 and PARP proteins, the appearance of a sub-$G_0/G_1$ peak in the flow cytometric assay, and increased percentage of apoptotic propensities in the annexin V binding assay. Effective reversal of apoptosis was observed following pretreatment with the pan-caspase inhibitor Z-VAD. Moreover, Nrf2 knockdown exhibited increased sensitivity to the anticancer drug, cisplatin, presumably by potentiating the oxidative stress induced by cisplatin. Collectively, our data demonstrate the importance of Nrf2 in cytoprotection, survival, and drug resistance with implications for the potential significance of targeting Nrf2 as a promising strategy for overcoming resistance to chemotherapeutics in MM.

  • 오랫동안 슈퍼옥사이드 음이온 라디칼(superoxide anion radical)은 활성산소(reactive oxygen species, ROS) 화학종으로서 물리화학적 기초 연구에서부터 생명과학(혹은 생명공학) 분야에 이르기까지 상당한 관심의 대상이었다. 최근에는 고도산화공정(advanced oxidation processes, AOP, 혹은 advanced oxidation technologies, AOT)을 이용하는 오염물 제어 분야뿐만 아니라 나노물질에 의한 유해성을 평가하는데 있어서 슈퍼옥사이드 음이온 라디칼이 중요한 화학종으로 주목을 받고 있다. 그럼에도 불구하고 슈퍼옥사이드 음이온 라디칼에 대한 명확한 이해가 부족하여 관련 연구자들 사이에서 불필요한 논쟁과 혼동을 일으키고 있으며 슈퍼옥사이드 음이온 라디칼의 물리화학적 성질에 대한 오해를 가중시키고 있다. 이 글에서는 기존 연구에서 행해진 슈퍼옥사이드 음이온 라디칼의 물리화학적 특성 및 그 반응성에 대해 정리하였고 고도산화공정, 나노물질 및 생명공학 분야 등에서 슈퍼옥사이드 음이온 라디칼이 갖는 중요성을 서술하였다.

  • This study was designed to investigate the effects of silk fibroin(Mw 500) powder (SFP) on oxygen radicals and the scavenger enzymes in brain membranes of rats. Spragu-Dawley(SD) male rats(160${\pm}$10g) were fed basic diet(control group), and experimental diets(SFP-2.5 and SFP-5.0 groups) added 2.5 and 5.0g/kg BW/day for 6 weeks. Hydroxyl radical($.$OH) levels resulted in a decreases(6.6% and 9.7%, 2.8% and 11.9%, respectively) in brain mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group, but were significantly decreased in these membrances of SFP-5.0 group only. Superoxide radical (O2) levels were a slightly decreased (2.0% and 9.1%, respectively) in brain cytosol of SFP-2.5 and SFP-5.0 groups compared with control group. Lipid peroxide(LPO) levels were significantly decreased (12.9% and 21.9%, 13.2% and 22.5%, respectively) in brain mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (16.7% and 15.7%, respectively) in brain microsomes of SFP-2.5 and SFP-5.0 group compared with control group, but significantly difference between in brain mitochondria of these two groups could not be obtained. Mn-SOD activities were remarkably increased (11.2% and 24.2%, respectively) in mitochodria of SFP-2.5 and SFP-5.0 groups. CuZn-SOD activities were effectively increased (7.7% and 19.6%, respectively) in brain cytosol of SFP-2.5 and SFP-5.0 groups, but significant difference between control and SFP-2.5 groups could be not obtained. GSHPx activities were considerably increased (5.3% and 11.7%, respectively) in brain cytosol of SFP-2.0 and SFP-5.0 groups compared with control group. There results suggest that anti-aging effect of silk fibroin may play an effective learning and memory role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in brain membranes.

Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were remarkably decreased about 11.6% in liver mitochondria of SFP-5.0 group compared with control group. Mn-SOD activities were remarkably in creased (17.6% and 28.8%, respectively) in mitochondria of SFP-2.5 and SFP-5.0 groups, and Cu/Zn-SOD activities were also effectively in creased (about 14.4%) in liver cytosol of SFP-5.0 groups, but significant difference between GSHPx activity in liver cytosol of these two groups could be not obtained. These results suggest that anti-aging effect of silk fibroin may play an effective anti-aging role in a aattenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

Hydroxyl radical (.OH) levels resulted in a consistent decreases (4.0% and 7.2%, 5.0% and 14.1%, respectively) in liver mitochondria and microsomes of SWP-200 SWP-400 groups compared with control group, and O2 radical level was significantly decreased about 12% in liver cytosol of SWP-400 group compared with control group. Lipid peroxide(LPO) levels were significantly decreased (14.4% and 9.1%, respectively) in liver mitochondria and microsomes of SWP-400 group only compared with control group. Oxidized protein (OP) levels were remarkably decreased about 12.7% and 16.3% in liver microsomes only of SWP-200 and SWP-400 groups, but significant difference between liver mitochondria could not obtained. Mn-SOD activities were remarkably increased (15.8% and 25.2%, respectively) in mitochondria of SWP-200 and SWP-400 groups, but significant difference between Cu, Zn-SOD activities in these group could be not obtained. GSHPx activity was significantly increased in liver cytosol of SWP-400 group compared with control group. These results suggest that silkworm powder may play an effective role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

Effects of Ginsenoside-$Rg_1$ on Post-thawed Miniature Pig Sperm Motility, Mitochondria Activity, and Membrane Integrity

  • In this study, we used flow a cytometric assay to evaluate plasma membrane integrity and mitochondrial activity in post-thawed sperm that was supplemented with ginsenoside-$Rg_1$. Varying concentrations of ginsenoside-$Rg_1$ (0, 25, 50 and $100{\mu}M/ml$) were used in the extender during cryopreservation to protect the DNA of thawed sperm, thereby increasing the viability and motility rate as evaluated using a computer-assisted sperm analysis (CASA) method. The results derived from CASA were used to compare the fresh, control, and ginsenoside-$Rg_1$ groups. Sperm motility and the number of progressively motile sperm were significantly (p<0.05) higher in the $50{\mu}M/ml$ ginsenoside-Rg1 group ($61.0{\pm}4.65%$) than in the control ($46.6{\pm}7.02%$), $25{\mu}M/ml$ ($46.2{\pm}4.76%$), and $100{\mu}M/ml$ ginsenoside-$Rg_1$ ($52.0{\pm}1.90%$) groups. However, the velocity distribution of post-thawed sperm did not differ significantly. Membrane integrity and MMP staining as revealed using flow cytometry were significantly (p<0.05) higher ($91.6{\pm}0.82%$) in the $50{\mu}M/ml$ ginsenoside-$Rg_1$ group than in the other groups. Here, we report that ginsenoside-$Rg_1$ affects the motility and viability of boar spermatozoa. Moreover, ginsenoside-$Rg_1$ can be used as a protective additive for the suppression of intracellular mitochondrial oxidative stress caused by cryopreservation.

Influence of Functional Food Containing Bacillus polyfermenticus SCD on Lipid and Antioxidant Metabolisms in Rats Fed a High-Fat and High-Cholesterol Diet

  • We evaluated the effects of newly developed functional food containing Bacillus polyfermenticus SCD as the main material on the lipid and antioxidant metabolisms of hypercholesterolemic rats. Thirty male SD rats were divided into three groups after a 1-week adaptation period and were fed with a high fat-cholesterol diet (control), or with a high fat-cholesterol diet supplemented with low or high doses ($3.1\;{\times}\;10^6\;cfu/day$ or $3.1\;{\times}\;10^8\;cfu/day$) of B. polyfermenticus SCD and other physiological active materials for 6 weeks. Both doses of B. polyfermenticus SCD significantly reduced hepatic total cholesterol and triglycerides, while increasing the fecal excretion rates of total lipid, total cholesterol and triglycerides. B. polyfermenticus SCD increased the total radical trapping antioxidant potential (TRAP). The erythrocytic glutathione peroxidase activity in the B. polyfermenticus groups was significantly lower than that in the control group. Plasma TRAP levels exhibited a highly significant negative correlation with hepatic total cholesterol while a significant positive correlation was detected between fecal total cholesterol and plasma TRAP. This hypolipidemic and antioxidative effect of B. polyfermenticus SCD seemed to be unrelated to its dosage. These results suggest that functional food containing B. polyfermenticus SCD can improve oxidative stress and hepatic lipid profiles by enhancing the excretion of cholesterol and triglycerides in feces of rats fed with high fat-high cholesterol diet.
