미역의 부위별 fucoxantin함량은 미역엽 87.6 mg/100g, 미역줄기에 62.4 mg/100 g그리고 미역귀에 127.7 mg/100 g를 나타내어 미역귀에 함유되어 있는 fucoxanthin함량이 미역엽에 비해 1.4배, 미역줄기에 비해 2.0배 더 함유되어 있는 것으로 나타났다. Fucoxanthin은 444 nm에서 최대흡수극대를 나타내는 전형적인 carotenoids의 분광학적 특성을 나타냈다. 미역에 존재하는 fucoxanthin은 용매 분획, silica gel column chromatography와 HPLC를 이용하여 분리 정제하고, 얻어진 fucoxanthin을 인공생체막인 인지질에서의 자동산화에 이용하였다 인지질에서 50 $\mu$M fucoxanthin을 37$^{\circ}C$에서 72시간 자동산화시킨 결과, 5개의 산화물이 생성되었다. 이러한 산화물은 in uitro상의 산화적 조건하에서 fucoxanthin자동산화에 의해 생성된 산화개열산물로 생각되었다.
활성산소종은 DNA의 손상에서 중요한 역할을 한다. 최근 활성산소를 제어하고 조절하기 위해 천연항산화제를 개발하기 위해 많은 노력이 이루어지고 있다. 느티나무(Zelkova serrate)는 느릅나무과의 식물로 한국 마을 입구에 흔히 심어져 친숙한 식물이다. 하지만 느티나무의 항산화 활성 및 산화적 DNA 손상에 대한 방어효과에 대한 연구는 미흡한 실정이다. 본 연구에서 느티나무 잎의 에틸아세테이트 분획물 및 열수 추출물의 항산화 활성 및 산화적 DNA 손상에 대한 억제활성을 확인하였다. 에틸아세테이트 분획물은 열수 추출물에 비해 DPPH 라디칼 소거활성, ABTS 라디칼 소거활성, $Fe^{2+}$ 킬레이팅 활성 그리고 reducing power에서 높은 항산화 활성을 보였다. 또한, 페놀류 화합물 함량은 각각 에틸아세테이트 분획물은 56.63 mg/g 그리고 열수 추출물은 51.61 mg/g으로 분석됐다. ${\phi}X$-174 RF I plasmid DNA를 이용한 산화적 DNA 손상억제활성은 에틸아세테이트 분획물과 열수 추출물 모두 상당한 방어효과를 나타냈다. 따라서 느티나무 잎의 에틸아세테이트 분획물 및 열수 추출물은 뛰어난 항산화 활성 및 산화적 DNA 손상 억제 효과를 통한 천연 자원으로서의 잠재성을 보였다.
Objectives : Oxidative stress due to excessive accumulation of reactive oxygen species (ROS) is one of the risk factors for the development of several chronic diseases, including neurodegenerative diseases. Methods : In the present study, we investigated the protective effects of cheongnoemyeongsin-hwan (CNMSH) against oxidative stress‑induced cellular damage and elucidated the underlying mechanisms in neuronal-derived SH-SY5Y cells. Results : Our results revealed that treatment with CNMSH prior to hydrogen peroxide (H2O2) exposure significantly increased the SH-SY5Y cell viability, indicating that the exposure of the SH-SY5Y cells to CNMSH conferred a protective effect against oxidative stress. CNMSH also effectively attenuated H2O2‑induced comet tail formation, and decreased the phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V‑positive cells. In addition, CNMSH exhibited scavenging activity against intracellular ROS generation and restored the mitochondria membrane potential (MMP) loss that were induced by H2O2, suggesting that CNMSH prevents H2O2‑induced DNA damage and cell apoptosis. Moreover, H2O2 enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase, a typical substrate protein of activated caspase-3, as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with CNMSH. Furthermore, CNMSH increased the levels of heme oxygenase-1 (HO-1), which is a potent antioxidant enzyme, associated with the induction of nuclear factor-erythroid 2-related factor 2 (Nrf2). According to our data, CNMSH is able to protect SH-SY5Y cells from H2O2-induced apoptosis throughout blocking cellular damage related to oxidative stress through a mechanism that would affect ROS elimination and activating Nrf2/HO-1 signaling pathway. Conclusions : Therefore, we believed that CNMSH may potentially serve as an agent for the treatment and prevention of neurodegenerative diseases caused by oxidative stress.

Cathepsin B is abundantly expressed peptidase of the papain family in the lysosomes, and closely related to the cell degradation system such as apoptosis, necrosis and autophagy. Abnormal degradation of organelles often occurs due to release of cathepsin B into the cytoplasm. Many studies have been reported that relationship between cathepsin B and intracellular mechanisms in various cell types, but porcine embryos has not yet been reported. Therefore, this study evaluated the effect of cathepsin B inhibitor (E-64) on preimplantation developmental competence and quality of porcine embryos focusing on apoptosis and oxidative stress. The expression of cathepsin B mRNA in porcine embryos was gradually decreased in inverse proportion to E-64 concentration by using real-time RT-PCR. When putative zygotes were cultured with E-64 for 24 h, the rates of early cleavage and blastocyst development were decreased by increasing E-64 concentration. However, the rate of blastocyst development in $5{\mu}M$ treated group was similar to the control. On the other hand, both the index of apoptotic and reactive oxygen species (ROS) of blastocysts were significantly decreased in the $5{\mu}M$ E-64 treated group compared with control. We also examined the mRNA expression levels of apoptosis related genes in the blastocysts derived from $5{\mu}M$ E-64 treated and non-treated groups. Expression of the pro-apoptotic Bax gene was shown to be decreased in the E-64 treated blastocyst group, whereas expression of the anti-apoptotic Bcl-xL gene was increased. Taken together, these results suggest that proper inhibition of cathepsin B at early development stage embryos improves the quality of blastocysts, which may be related to not only the apoptosis reduction but also the oxidative stress reduction in porcine embryos.
Ruthenium(III) unsymmetrical Schiff base complexes, $[Ru(CHBPH-TP)Cl_2]$ and $K[Ru(CHBPH-HB)Cl_2]$ were synthesized, where CHBPH-TP and CHBPH-HB are 5-chloro-2-hydroxybenzophenonethiophencarba aldehydephenylenediimine and 5-chloro-2-hydroxybenzophenonehydroxybenzophe nonephenylenediimine. These Schiff bases were obtained from the reactions of 5-chloro-2-hydroxybenzophenone (CHB) and 2-thiophenecarbaldehyde (TP) or hyroxybenzophenone (HB) and 1,2-diaminobenzene. Elemental analysis, conductivity and infrared studies of the complexes suggest an octahedral geometry around ruthenium. Magnetic moments of the complexes indicate a single unpaired electron in alow spin $d^5$ configuration. The complexes are capable of catalyzing the oxidation of styrene with sodium hypochlorite in the presence of phase transfer agent. Oxidative cleavage of C=C bond is the major reaction pathway to form benzaldehyde for styrene oxidation.
본 연구는 꽈리허리노린재(Acanthocoris sordidus) 추출물이 산화적 스트레스에 의해 유발되는 세포와 DNA의 손상 억제력을 조사하였다. 꽈리허리노린재 추출물의 DPPH 유리 라디칼과 수산화 라디칼 제거능은 양성 대조군에 비해 각각 48.9%, 37.8%로 나타났으며, $Fe^{2+}$-chelating 효과는 80.0%로 조사되었다. 꽈리허리노린재 추출물이 활성산소에 의해 유도되는 세포손상에 미치는 억제 효과를 조사하기 위해 지질과산화의 상대적 수준과 p21 단백질의 발현율을 조사해보면 꽈리허리노린재 추출물은 라디칼 처리군에 비해 지질과산화를 거의 완벽하게 억제하고 있으며, p21 단백질의 발현은 대조군의 92.4%로 회복되는 것으로 조사되었다. 또한 꽈리허리노린재 추출물의 DNA 분절화 억제 활성은 대조군에 비해 53.3%로 나타나 산화적 스트레스에 의해 유발되는 DNA 분절화를 효율적으로 억제하고 있으며, H2AX 단백질의 인산화비는 라디칼 처리군의 39.0%에 해당하는 수준으로 조사되어 꽈리허리노린재 추출물은 히스톤 단백질의 인산화를 매우 효율적으로 억제하는 것으로 확인되었다. 이러한 결과로 보아 꽈리허리노린재 추출물은 활성산소에 대한 항산화 효과뿐만 아니라 세포와 DNA의 손상을 억제하는데 효과적인 것으로 사료된다.

Oenothera biennis, commonly known as evening primrose, a potential source of natural bioactive substances: flavonoids, steroids, tannins, fatty acids and terpenoids responsible for a diverse range of pharmacological functions. However, whether extract prepared from aerial part of O. biennis (APOB) protects skin against oxidative stress remains unknown. To investigate the protective effects of APOB against oxidative stress-induced cellular damage and elucidated the underlying mechanisms in the HaCaT human skin keratinocytes. Our results revealed that treatment with APOB prior to hydrogen peroxide ($H_2O_2$) exposure significantly increased viability, and the highest DPPH radical-scavenging activities and reducing power of HaCaT cells. APOB also effectively attenuated H2O2-induced comet tail formation and inhibited the $H_2O_2$-induced phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V-positive cells. In addition, APOB exhibited scavenging activity against intracellular reactive oxygen species (ROS) accumulation and restored the mitochondrial membrane potential loss by $H_2O_2$. Objective: p66Shc, a 66 kDa protein isoform encoded by the proto-oncogene SHC, is an essential intracellular redox homeostasis regulatory enzyme that is involved in the regulation of cellular oxidative stress, apoptosis induction and the occurrence of multiple age-related diseases. This study investigated the expression profile and functional characteristics of p66Shc during preimplantation embryo development in sheep. Methods: The expression pattern of p66Shc during preimplantation embryo development in sheep at the mRNA and protein levels were studied by quantitative real-time polymerase chain reaction (RT-qPCR) and immunofluorescence staining. The effect of p66Shc knockdown on the developmental potential were evaluated by cleavage rate, morula rate and blastocyst rate. The effect of p66Shc deficiency on reactive oxygen species (ROS) production, DNA oxidative damage and the expression of antioxidant enzymes (e.g., catalase and manganese superoxide dismutase [MnSOD]) were also investigated by immunofluorescence staining. Results: Our results showed that p66Shc mRNA and protein were expressed in all stages of sheep early embryos and that p66Shc mRNA was significantly downregulated in the 4-to 8-cell stage (p<0.05) and significantly upregulated in the morula and blastocyst stages after embryonic genome activation (EGA) (p<0.05). Immunofluorescence staining showed that the p66Shc protein was mainly located in the peripheral region of the blastomere cytoplasm at different stages of preimplantation embryonic development. Notably, serine (Ser36)-phosphorylated p66Shc localized only in the cytoplasm during the 2- to 8-cell stage prior to EGA, while phosphorylated (Ser36) p66Shc localized not only in the cytoplasm but also predominantly in the nucleus after EGA. RNAi-mediated silencing of p66Shc via microinjection of p66Shc siRNA into sheep zygotes resulted in significant decreases in p66Shc mRNA and protein levels (p<0.05). Knockdown of p66Shc resulted in significant declines in the levels of intracellular ROS (p<0.05) and the DNA damage marker 8-hydroxy2'-deoxyguanosine (p<0.05), markedly increased MnSOD levels (p<0.05) and resulted in a tendency to develop to the morula stage. Conclusion: These results indicate that p66Shc is involved in the metabolic regulation of ROS production and DNA oxidative damage during sheep early embryonic development.
Ferulic Acid (FA) is a metabolite of phenylalanine and tyrosine, a phenolic compound commonly found in fruits and vegetables. Several studies have shown that FA has various functions such as antioxidant effect, prevention of cell damage from irradiation, protection from cell damage caused by oxygen deficiency, anti-inflammatory action, anti-aging action, liver protective effect and anti-cancer action. In this study, we investigated the maturation rate, intracellular glutathione (GSH) and reactive oxygen species (ROS) of porcine oocytes by adding FA to the in vitro maturation (IVM) medium and examined subsequent embryonic developmental competence at 5% oxygen through parthenogenesis. There is no significant difference between the control group ($0{\mu}M$) and treatment groups ($5{\mu}M$, $10{\mu}M$, $20{\mu}M$) on maturation rates. Intracellular GSH levels in oocyte treated with $5{\mu}M$ of FA significantly increased (P < 0.05), and $20{\mu}M$ of FA revealed significant decrease (P < 0.05) in intracellular ROS levels compared with the control group. Oocytes treated with FA exhibited significantly higher cleavage rates (79.01% vs 89.19%, 92.20%, 90.89%, respectively) than the control group. Oocytes treated with $10{\mu}M$ showed significantly higher blastocyst formation rates (28.3% vs 40.3%, respectively) after PA than the control group. Total cell numbers in blastocyst of $10{\mu}M$ FA displayed significantly higher (39.4 vs 51.9, respectively) than the control group. In conclusion, these results suggested that treatment with FA during IVM improved the developmental potential of porcine embryos by increasing intracellular GSH synthesis and reducing ROS levels. Also, there was an improvement of cleavage rate, blastocyst formation and total cell numbers in blastocysts. It might be associated with Keap1-Nrf2 pathway as an antioxidant regulate pathway that plays a crucial role in determining the sensitivity of cells to oxidative damages by regulating the basal and inducible expression of enzymes which is related to detoxification and anti-oxidative effects, stress response enzymes and/or proteins and ABC transporters.
