고구마 현탁배양세포의 EST library에 높은 빈도로 존재하는 metallothionein (MT) 유전자를 선별하였다(IbMT3). MT 유전자는 세포와 조직의 스트레스 조절과 연관되어 있다고 알려져 있다. 본 연구에서 IbMT3 cDNA의 전장을 확보하여 염기서열 분석을 한 결과, IbMT3 유전자는 구조적으로 유형 3에 속하는 MT 단백질을 암호화하고 있었다. 식물에서 유형 3에 속하는 MT 단백질의 기능은 명확히 알려지지 않다. Northern blot 분석 결과, IbMT3 유전자는 고구마 식물체 잎보다 현탁배양 세포에서 매우 강하게 발현되었다. 일반적으로 세포배양은 세포에 산화 스트레스 상태를 부과하는 것으로 알려져 있다. 이에, 고구마 식물체에 산화 스트레스를 처리하여 IbMT3 유전자의 발현이 어떻게 조절되는지 조사하였다. 제초제인 methyl viologen (MV)을 6, 12, 24시간 동안 처리했을 때, IbMT3 유전자의 발현은 6시간 후에 아주 강하게 유도되었고 그 이후에는 감소함을 알 수 있었다. 저온 스트레스($15^{\circ}C$)를 24, 48시간 동안 처리했을 때, IbMT3 유전자는 처리시간이 경과함에 따라 발현이 더 많이 유도되었다. 이로써, IbMT3 유전자는 환경 및 산화 스트레스에 반응하여 발현이 조절되는 유전자임을 알 수 있었다. IbMT3 isoform은 고구마 식물체 내에서 항산화제로써 작용할 가능성이 있을 뿐 아니라, 스트레스 하에서의 세포 적응 메커니즘에 중요한 기능을 할 것으로 사료된다.
연구배경 : 노화란 반응성산소종에 의한 산화적 손상이 축적되어 세포의 기능의 저하로 초래된다고 주장되고 있다. 본 연구에서 1, 4, 8, 12개월된 정상 생쥐의 폐에서 반응성 산소종 생성의 초기에 중요한 방어기전인 superoxide dismutases(SODs) mRNA와 hydroxy radical($OH{\cdot}$)에 의한 산화적 손상의 마지막 방어기전인 metallothionein(MT) mRNA의 함량과 paraquat에 의한 유도능을 조사하였다. 방 법 : 산화적손상은 세포내에서 superoxide을 생성하는 paraquat를 투여한 후 SODs와 MT mRNAs을 RT-PCR 방법으로 측정하였다. 결 과 : 정상생쥐의 폐의 노화과정에서 Mn-SOD와 Cu/Zn-SOD, 그리고 MT mRNA의 항정상태 함량을 조사한 결과, Mn-SOD mRNA는 8개월까지는 증가하다가 12개월에서 감소하였으나, Cu/Zn-SOD mRNA는 별 변화가 없었고 12개월에 감소하였다. 그러나 MT mRNA는 어느 연령에도 변화가 없었다. 산화성 스트레스를 유발하는 paraquat에 의하여 Mn-SOD mRNA는 4개월 부터 유도되지 않았으며, Cu/Zn-SOD mRNA는 전혀 유도되지 않았다. MT mRNA는 관찰한 전 연령에서 유도되었다. 결 론 : 노화과정에서 SODs유도능의 이상이 노화의 원인 인자로 작용하고, 반면에 MT의 정상적인 유도능이 산화적 스트레스에 대하여 방어 역할을 할 가능성을 시사해주고 있다. 이와 같은 결과는 연령이 증가함에 따라 Mn-SOD, Cu/Zn-SOD, 및 MT의 발현과 유도능의 차이로 인하여 반응성 산소종에 의한 산화적 손상의 정도의 차이가 노화의 속도를 결정하는 하나의 인자로 작용할 가능성 이 있을 것으로 생각된다.
Non-alcoholic fatty liver disease (NAFLD), especially including non-alcoholic steatohepatitis (NASH) is one of the common diseases with 25% of prevalence globally, but there is no thera-peutic access available. Amomum villosum var. xanthioides (Wall. ex Baker) T.L.Wu & S.J.Chen (AX), which is a medicinal herb and traditionally used for treating digestive tract disorders in Asia countries. We aimed to examine pharmacological effects of ethyl acetate fraction of AX (AXEF) against ER stress-induced NASH mice model using C57/BL6J male mice by tunicamycin (TM, 2 mg/kg) injection focusing on the oxidative stress. Mice were orally administrated AXEF (12.5, 25, or 50 mg/kg), silymarin (50 mg/kg) or distilled water daily for 5 days, and outcomes for fatty liver, inflammation, and oxidative stress were measured in serum or liver tissue levels. AXEF drastically attenuated hepatic ER stress-induced NASH which were evidenced by decreases of li-pid droplet accumulations, serum liver enzymes, hepatic inflammations, and cell death signals in the hepatic tissue or serum levels. Interestingly, AXEF showed potent antioxidant effects by quenching of reactive oxidative stress and its final product of lipid peroxide in the hepatic tissue, specifically increase of metallothionein (MT). To confirm underlying actions of AXEF, we ob-served that AXEF increase MT1gene promoter activities in the physiological levels. Collectively, AXEF showed antioxidant properties on TM-induced ER stress of NASH by enhancement of MTs.
Nickel(II) compounds are carcinogenic metals which induce genotoxicity and oxidative stress through the generation of reactive oxygen species. In search of new molecular pathways toward understanding the molecular mechanism of nickel(II)-induced carcinogensis, we performed mRNA differential display analysis using total RNA extracted from nickel(II) acetate-treated normal rat kidney cells (NRK-52E). Cells were exposed for 3 days to 160 and 240 uM nickel(II) concentrations. cDNAs corresponding to mRNAs for which expression levels were altered by nickel(II) were isolated, sequenced, and followed by a GenBank Blast homology search. Specificity of differential expression of cDNAs was determined by RT-PCR and Western blot analysis. Two of them (SH3BGRL3 and FHIT) were down-regulated and one (metallothionein) was up-regulated by nickel(II) treatment. The expression of these mRNAs were nickel(II) concentration-dependent. The levels of FHIT and metallothionein proteins were also consistent with the results for mRNAs. Overall, although the fundamental questions related to function of these genes in nickel(II)-mediated carcinogenicity are not answered, our study suggests that they can be interesting candidates for studies of molecular mechanisms of nickel(II) carcinogenesis.

This study was conducted to investigate whether dietary chlorella intake could have an effect on antioxidative capacity in rats oxidatively stressed with cadmium (Cd). Sprague-Dawley rats fed dietary chlorella (0, 5, and 10%) for 4 weeks after induction of oxidative stress by exposing to Cd (200 ppm) for 8 weeks. After the oxidative stress applied, plasma and liver malondialdehyde concentrations and xanthine oxidase activities were decreased in 5% chlorella fed group compared to chlorella free group. Although liver heme oxygenase-1 protein expression was not affected by chlorella, the enzyme activity was improved in 5% chlorella fed group. Erythrocyte superoxide dismutase activity and hepatic metallothionein concentration were increased in 5% chlorella fed group. However, 10% chlorella intake had no effect on the improvement of oxidative stress-related enzymes and proteins. These findings suggest that, after induction of oxidative stress with Cd, 5% chlorella intake might improve antioxidative capacity against oxidative stress.

Metallothioneins(MTs) belong to the class of low molecular weight proteins. Recently, it has been suggested that MTs may playa direct role in cellular defense against oxidative stress by functioning as antioxidants. Oxidative damage to different cellular components makes a major contribution to many pathogenenesses. Several studies have demonstrated that MT is able to quench a wide range of reactive oxygen species at a higher efficiency than other well known antioxidants such as superoxide dismutate(SOD). The present study was designed to evaluate the effect of MT on the activities of the reactive oxygen species removal system. MT showed the scavenging of superoxide in the SOD assay system in the presence or absence of SOD. When MT was added to nicotinamide adenine dinucleotide phosphate(NADPH) oxidation system in presence of fixed amount of SOD increase the breakdown rate of superoxide. When MT was added to the system that form nitrite from hydroxylammonium chloride, the formation of nitrite was inhibit. We concluded that the function of MT as antioxidant might have an effect on the level of superoxide scavenging.
