Antioxidant Property and Inhibitory Effects of an Water Extract of Hwang-Ryun-Chung-Sim-Um on the Acetylcholinesterase (황련청심음(黃連淸心飮)의 항산화 및 AChE 억제 효과에 관한 연구)

  • Objective: An water extract of the Hwang-Ryun-Chung-Sim-Um (HRC) was assessed to determine the mechanisms of its antioxidant activity. In addition, the HRC was examined in vitro for the inhibitory effect on the acetylcholinesterse (AChE). Methods: The HRC exhibited a concentration-treatment; scavenging ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$ (DPPH) radical, linoleic acid oxidation in a thiocyanate assay system, hydroxyl radical-induced DNA nicking. We investigated mRNA levels such as catalase activity, superoxide-dismutase and glutathione peroxidase. The water extract of HRC showed inhibitory effect on AChE activity. Result: The HRC extract showed dose-dependent free radical scavenging activity, including DPPH radicals and hydroxyl radicals, using different system. The HRC was also found to be effective in protecting plasmid DNA against the strand breakage induced by Hydroxyl radicals in Fenton's reaction mixture. Futhermore, catalase mRNA expression levels increased, but SOD1 and MnSOD was not expressed. HRC in a various concentration-dependent decreased AChE mRNA levels and inhibitory effect showed AChE. Conclusion: According to the above results, it is supposed that HRC is applicable to the Dementia-type of Alzheimer clinically.

Biological Properties of Different Types and Parts of the Dandelions: Comparisons of Anti-Oxidative, Immune Cell Proliferative and Tumor Cell Growth Inhibitory Activities

  • Dandelions have been reported to have medicinal properties and bioactive components that impact human health. However, the precise biological properties of dandelions and the parts of the plants possessing bioactive components remain uncertain. In this study, we evaluated 3 different types of dandelions based on their cultivation origin (Songpa, Uiryung, and native Uiryung types) as well as their 4 different plant parts (leaf, flower, root, skin). Each sample was extracted with $80\%$ methanol and then compared for the biological activities (anti-oxidative, immune cell proliferative and tumor cell growth inhibitory activities). All 3 types of dandelions possessed a degree of biological functions including the hydroxyl radical scavenger activity, immune cell proliferative activity and tumor cell growth inhibitory activity. However, there was no significant difference in these activities between the 3 dandelion types. Leaves of all three dandelion types showed the highest levels of all biological activities. To a lesser degree, the flower and root parts displayed biological activities. In the skin parts, anti-oxidative activity was also detected only at higher doses of dandelion extracts. Heating the dandelion leaf extract did not affect the biological activity, suggesting a heat-stable nature of the biological compounds. Taken together, these collective data suggest that dandelions, in particular their leaves, possess a high concentration of heat-resistant biological compounds, which are responsible for anti-oxidative, immune cell proliferative and tumor cell growth-inhibitory activities.

Inhibitory Effects of Bupleuri Radix on ox-LDL induced Foam Cell Formation (시호 추출물의 oxLDL 유도 Foam Cell 형성 억제 작용)

  • The oxidative modification of low density lipoprotein(LDL) has been implicated in the development of atherosclerosis. Oxidized LDL(oxLDL) is captured into macrophage and stimulates to form macrophage foam cell. And it can induce an inflammation and smooth muscle proliferation in atherosclerotic plaque. Objective : In this study, we aimed to investigate the effect of Bupleuri radix(SH) on the foam cell formation, a critical initiation stage of atherosclerosis. Methods : To achieve the goal, we examined the effect of SH on LDL oxidation, nitric oxide production in RAW264.7, and the effect of SH on cupuric sulfate-induced cytotoxicity, LDH release, and macrophage activity. Results : SH inhibited the formation of oxidized LDL from native LDL in RAW264.7 cell culture, and decreased the release of LDH from cupric sulfate-stimulated RAW264.7 cell. In other experiments, SH activated RAW264.7 cell, and prolonged the survival time, and inhibited foam cell formation induced by oxLDL in Raw 264.7 cells. Conclusion : These results showed that SH might prevent atherosclerosis by controlling the early stages of foam cell formation.

Effects of a Hwanggi-tang Ethanol Extract on Glucose Uptake and Metabolism in Murine Myotubes (근육세포주에서 당 흡수 및 대사 조절에 대한 황기탕 에탄올 추출물의 효과)

  • Objectives: The aim of this study was to evaluate the effects of Hwanggi-tang on glucose digestion, uptake, and metabolism in murine C2C12 myotubes. Methods: Hwanggi-tang was prepared according to the Dong-ui-bo-gam (≪東醫寶鑑≫) prescription by 70% ethanol extraction. The effect on glucose digestion was examined by determining the inhibitory effect of Hwanggi-tang on α-glucosidase activity. We also compared and verified the gene and protein expression of genes related to glucose uptake in C2C12 myotubes treated with Hwanggi-tang or insulin. Glucose metabolism was assessed by the expression levels of associated enzymes. Results: Hwanggi-tang caused a dose-dependent inhibition of α-glucosidase activity, induced glucose uptake by activation of the PI3K/Akt/mTOR pathway in the insulin signaling pathway, and promoted glucose oxidation and β-oxidation. Conclusions: Hwanggi-tang exerts an anti-diabetic effect on murine myotubes by inhibiting glucose digestion and inducing glucose uptake and consumption.

  • 80% extract of Gamigwaruhaebaekbaekju-Tang (GGHBT), Gagamgwaruhaebaekbaekju-Tang (GGGHBT) and Gamigwaruhaebaekwhanggum-Tang (GGHWT) remarkably showed inhibitory effects on HMG-CoA reductase, lipid peroxidation of rat liver and LDL oxidation, and DPPH free radical scavenging effect in a dose-dependent manner. Especially, GGHWT which is formulated with Trichosanthis Fructus, Pinelliae Tuber, Aurantii Immatures Fructus, Magnoliae Cortex, Allii Macrostemi Bulbus, Cinnamomi Ramulus and Scutellariae Radix on the basis of Gwaruhaebaekbaekiu-Tang listed on the traditional medicinal references showed more effective hypocholesterolemic activities in vitro bioassay than the other prescriptions.

Anti-oxidative and anti-inflammatory effects of aerial parts of Rumex japonicus Houtt. in RAW 264.7 cells (양제엽(羊蹄葉) 메탄올 추출물의 항산화 및 항염증 효과)

  • Objectives : The aerial parts of Rumex japonicus Houtt. (RF) is used by traditional clinics to treat parasite infection in East asia. This study aims a verification of anti-oxidative and anti-inflammatory effects of RF methanol extract. Methods : Anti-oxidative effects of RF were measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cells were measured. The inhibitory effects against the production of inflammatory mediators including NO, prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and the translocation of nuclear factor (NF)-${\kappa}B$ in LPS-stimulated RAW 264.7 cells by RF were tested. Results : RF scavenged DPPH, superoxide, NO and peroxynitrite radicals, and RF (at $200{\mu}g/m{\ell}$) reduced the inflammatory mediators definitely. Conclusions : These results indicate that RF may be a potential drug source for oxidative stress related inflammatory diseases.

Total Phenolic Contents, Radical Scavenging Capacities and Inhibitory Effects on Lipid Peroxidation and LDL Oxidation of Prunus persica Branch

  • This study was undertaken to elucidate the antioxidant activity of the ethanol (EEPB) and water (WEPB) extracts of Prunus persica branches. The extracts contained a high phenolic content and revealed a potent hydrogen donating activity in DPPH scavenging assay. Compared to $\alpha$-tocopherol, EEPB (p < 0.001) and WEPB (p < 0.05) significantly inhibited $FeCl_2$-ascorbic acid-induced lipid peroxidation, and also exhibited potent antiradical activities against hydroxyl radical, superoxide anion, nitric oxide and peroxynitrite. In copper- and AAPH-mediated human low-density lipoprotein (LDL) oxidation systems, the extracts demonstrated a strong antioxidant function by metal chelating, rather than direct scavenging, action. Furthermore, EEPB at 5 ${\mu}g/mL$ concentration showed 80.77% inhibition of the electrophoretic mobility of LDL, compared to 77.69% for ascorbic acid and 76.92% for BHT. These results suggest that PB branch extracts may protect against oxidative stress-induced diseases.

The antioxidative effects of α-tocopherol on the lipid peroxidation and protein oxidation by free radicals (Free radical에 의한 지질과산화와 단백질산화에 대한 α-tocopherol의 항산화효과)

  • This study was carried out to investigate the inhibitory effects of vitamin E on the oxidative damage of cellular lipids and proteins in free radical reaction induced by $FeCl_3$, and ascorbic acid. In this experiment, a vitamin E treated rat group was administered with 100mg/kg body weight of $dl-{\alpha}-tocophery$ 1 acetate and an untreated rat group was administered with the same volume of corn oil. And then assays of malondialdehyde and carbonyl group in total homogenate, mitochondrial and microsomal fraction of rat liver were carried out at the scheduled time. The results obtained from this study were summarized as follows; 1. Lipid peroxidation levels in vitamin E administered rat liver cells were significantly (p<0.05) decreased at the intervals between 1 hour and 4 hours in liver homogenate, at all times except for 1 hour point in mitochondrial fraction, and also at the intervals between 0.5 hour and 3 hours in microsomal fraction compared with those of the control rat liver cell. 2. Protein oxidation levels in vitamin E administered rat liver cell were also significantly (p<0.05) decreased at the intervals between 1.5 hours and 4 hours in liver homogenate, at over 4 hours in liver mitochondrial fraction, and at the intervals between 0.5 hour and 3 hours in liver microsomal fraction compared with those of the control rat liver cells.

Anti-Oxidative and Anti-Inflammatory Effects of Cheongajihwang-Tang Extract on RAW264.7 Cells (청아지황탕(靑蛾地黃湯)의 RAW264.7 Cell에서 항산화 및 항염증 효능 연구)

  • Objectives This study was designed to investigate whether the Cheongajihwang-Tang (CT) has an inhibitory effect association with oxidation or inflammation in RAW264.7 cells. Methods Cytotoxic activity of CT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Nitric oxide production was measured using Griess reagent system. The total phenolic contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the anti-oxidative effects of CT. Dichlorofluorescin diacetate (DCFH-DA) has been used as a substrate for measuring intracellular oxidant production. Results Cheongajihwang-Tang does not impair the cell viability in tested concentration. CT showed anti-oxidative effects in vitro by decreasing electron donating ability, and also showed anti-inflammatory effects suppressing NO and ROS expression in LPS induced RAW264.7 activation. CT inhibited the generation of intracellular ROS production as dose dependant manner. Conclusions CT has anti-oxidative effects and anti-inflammatory activities. These results indicate that CT extract has an anti-inflammatory activities via anti-oxidative effects.

Antioxidative Effects of Sulfur Containing Compounds in Garlic on Oxidation of Human Low Density Lipoprotein Induced by Macrophages and Copper Ion (마크로파아지 및 구리 이온으로 유도한 사람 low density lipoprotein의 산화에 대한 마늘 유황 화합물의 항산화 효과)

  • Sulfur containing compounds in garlic have all be used as one of the traditional folk medicine as well as food source. The present study was performed to investigate the antioxidative compounds of 1-methyl-1-cysteine, dimethyl trisulfide and 2-vinyl-4H-1,3-dithiin. The antioxidative activity of sulfur containing compounds on human LDL was investigated by monitoring a thiobarbituric acid substances (TBARS). Sulfur containing compounds inhibited on oxidation of LDL mediated by $CuSO_4$ and macrophages in dose dependent manner with almost completely inhibition at $80{\mu}g/ml$. Antioxidant activities of sulfur containing compounds on LDL oxidation were 2-vinyl-4H-1,3-dithiin, 1-methyl-1-cysteine, and dimethyl trisulfide in order. Inhibitory effects of sulfur containing compounds on oxidation of LDL mediated by $CuSO_4$ and macrophages were degraded at much greater rate than native LDL, the LDL oxidation process was arrested as shown by the lower conjugated dienes formation at the concentration of $60{\mu}g/ml$. Sulfur containing compounds in garlic revealed at high antioxidative activity at low physiological concentration for human LDL oxidation in vitro specially, it was indicated that the antioxidative activity of 3-viny l-4H-1,2-dithiin was higher than that of the other sulfur containing compounds.
