The present study investigated the effect of ammonia load on microbial communities in mesophilic anaerobic digestion of propionic acid. A laboratory-scale continuous anaerobic digester treating propionic acid as a sole organic substrate was operated under non-inhibitory condition and inhibitory conditions with ammonia (1.5 g and 3.5 g ammonia-N/L, respectively), and bacterial and archaeal communities in the steady states of each ammonia condition were analyzed using high-throughput sequencing. Thirteen bacterial families were detected as abundant bacterial groups in mesophilic anaerobic digestion of propionic acid. Increase in ammonia concentration resulted in significant shifts in microbial community structures. Syntorophobacter, Pelotomaculum, and Thermovigra were determined as the dominant groups of (potential) propionate oxidizing bacteria in the non-inhibitory condition, whereas Cryptanaerobacter and Aminobacterium were the dominant groups of (potential) propionate oxidizing bacteria in the ammonia-inhibitory condition. Methanoculleus and Methanosaeta were the dominant methanogens. Acetate-oxidation coupled with hydrogenotrophic methanogenesis might be enhanced with increases in the relative abundances of Methanoculleus and Tepidanaerobacter acetatoxydans under the ammonia-inhibitory condition. The results of the present study could be a valuable reference for microbial management of anaerobic digestion systems that are exposed to ammonia inhibition and propionic acid accumulation.
The objective of the current study was to evaluate the inhibitory and antioxidant activities of powdered katsuobushi (dried bonito) protein hydrolysates and their corresponding fractions. The powdered katsuobushi (dried bonito) hydrolysates were obtained by enzymatic hydrolysis using Alcalase, ${\alpha}$-chymotrypsin, Neutrase, pepsin, papain, and trypsin. The antioxidant efficacy of the respective hydrolysates were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl, superoxide, and alkyl radical-scavenging activities. Among the hydrolysates, the peptic-derived hydrolysate exhibited the highest antioxidant activity compared to other enzymatic hydrolysates. Therefore, the peptic-derived hydrolysate was further analyzed, and was found to contain an active peptide with an amino acid sequence identified as Pro-Met-Pro-Leu-Asn-Ser-Cys (756 Da). The purified peptides from powdered katsuobushi (dried bonito) had an $EC_{50}$ value of $105.82{\mu}M$, and exhibited an inhibitory effect against DNA oxidation induced by hydroxyl radicals. Taken together, these results suggests that powdered katsuobushi (dried bonito) could be used as a natural antioxidant in functional foods and prevent oxidation reactions in food processing.
Objective: This experimental study was performed to investigate the anti-inflammation and anti-oxidant effects of Sopungdojeok-tang(SPDJT) and Samulsopung-san(SMSPS) which were used to treat patient with atopic dermatitis Methods: Anti-inflammation and anti-oxidant effects of SPDJT and SMSPS were measured by the inhibitory ability of Nitric oxide(NO) production and the scavenging for 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical. Results: 1. SPDJT and SMSPS were not showed cell toxicity. 2. In inhibitory effects against NO production, all groups of SPDJT were showed anti-inflammation, but all groups of SMSPS were not showed anti-inflammation. There were statistical significances between $20{\mu}g/m\ell$ and 4, $10{\mu}g/m\ell$ in SPDJT groups. 3. In DPPH radical scavenging activity, all groups of SPDJT and all groups of SMSPS were showed anti-oxidant effects. There were statistical significances between $20{\mu}g/m\ell$ SPDJT group and 4, $10{\mu}g/m\ell$ SMSPS groups. Conclusion: These results suggest that the SPDJT groups are better than the SMSPS groups in antiinflammation and anti-oxidant effects.
Objectives : The purpose of this study was to investigate the anti-atherosclerotic effects of 4 herbal formulas for Sasang constitutional medicine (Yeoldahanso-tang: YDHST, Yanggyeoksanhwa-tang: YGSHT, Cheongsimyeonja-tang: CSYJT and Taeeumjowi-tang: TEJWT). Methods : The antioxidant activities of herbal formulas were studied by measuring free radical scavenging activities on ABTS and DPPH. The inhibitory effects on LDL oxidation was evaluated by the formation of TBARS, REM and fragmentation of apolipoprotein B-100 (ApoB). Effects of herbal formulas on macrophage lipid accumulation were determined in native LDL and LPS co-incubated macrophages using Oil Red O staining. Results : The scavenging activities on ABTS and DPPH of herbal formulas were increased in dose-dependent manner (YDHST>YGSHT>CSYJT>TEJWT). Herbal formulas reduced the oxidation properties of LDL induced by $CuSO_4$. YDHST, YGSHT and CSYJT showed strong suppressive effect on LDL oxidation than TEJWT. In addition, YDHST, YGSHT and CSYJT significantly inhibited foam cell formation in LDL/LPS stimulated RAW 264.7 cells. Conclusions : These results demonstrate that YDHST, YGSHT and CSYJT have potentials on anti-atherosclerosis by antioxidative effect and suppressive effect on LDL oxidation.
Mung beans (Vigna radiata L.) were used to investigate the roles of strontium in hypocolyl elongation under IAA regime during the germination. After imbibition in a medium with or without IAA, $Sr^{2+}$ stimulated IAA oxidation. Three to five fold increasing in IAA oxidase activity seems to be direct evidence of growth inhibition through $Sr^{2+}$. Furthermore, the accumulation of spermidinc and spermine by $Sr^{2+}$ in the range of 1 to 10 mM was observed. Spermidine levels were 2 to 3 fold higher than in control seedling grown without strontium. The increase in polyamine levels was observed on a g fresh weight basis. In conclusion, it was demonstrated that the inhibitory action of $Sr^{2+}$ is closely related with the IAA oxidation and polyamine biosynthesis.

Objectives : This experimental study was performed to investigate the effects of Sulfur extract on anti-inflammation and anti-Propionibacterium acnes. Methods : The cytotoxicity of Sulfur extract about viability of Raw 264.7 cell were tested using a colorimetric tetrazolium assay(MTT assay). To investigate the anti-inflammation effects of Sulfur extract on LPS-induced macrophage Raw 264.7 cell, we used ELISA kit and Western blots. We evaluated anti-oxidation effects of Sulfur extract on HaCaT cell by Enzyme recycling method. And we investigated the inhibitory effects of Sulfur extract on Propionibactrium acnes using paper disk diffusion method. Results: 1. Sulfur extract has a little cytotoxicity in Raw 264.7 cell. 2. Concentration of $100\;{\mu}g/m{\ell}$ Sulfur extract inhibited the production of NO in the Raw 264.7 cell stimulated with LPS. 3. Sulfur extract showed a oxidation inhibition effect by decreasing the DPPH radicals. 4. Sulfur extract has not the significant inhibition effect of Propionibactrium acnes. Conclusions: These results indicate that Sulfur extract has anti-inflammation and anti-oxidation effects. If further study is performed, the use of Sulfur extract will be valuable and benificial in the therapy of Propionibactrium acnes.

Lithospermum erythrorhizon has been known well as one of traditional medicine for fever reduction, detoxication, and blood circulation improvement. This study was carried out to isolate biological active compounds from roots of Lithospermum erythrorhizon and to investigate their low density lipoprotein (LDL) antioxidant and anticancer activities. The hexane extract of Lithospermi Radix has been separated on silica gel chromatography and a naphthoquinone pigment compound 1 has been isolated. The structure of the compound 1 has been identified by spectroscopic technique, including MS and NMR, as acetylshikonin (1). Acetylshikonin showed significantly inhibitory activity on $Cu^{2+}$-induced human LDL oxidation with $IC_{50}$ value of $8.8\;{\mu}M$ and obvious anticancer effect by inhibiting farnesyl:protein transferase (FPTase) activity with $IC_{50}$ value of $23\;{\mu}M$, which suggested that acetylshikonin might be useful for the treatment of atherosclerosis and cancer.
Objectives : This studies were carried out to evaluate the effects on anti-oxidation activities in different parts as trunk bark and root bark of Ulmus devidiana Planchon. var. japonica Nakai and Hemipteleae davidii (Hance) Planchon. Methods : For evaluation of antioxidative effects, scavenging activity on superoxide anion radical and DPPH radical were measured. In this study, we evaluated anti-oxidation, whitening and anti-inflammatory effects of Ardisia crenata for use as the cosmeceuticals. Ardisia crenata extract (70% MeOH) showed a significant free-radical scavenging effect (up to 90% over at the concentration of 0.01 %) against DPPH radical generation and showed a significant inhibitory effect (up to 50% over at the concentration of 0.05 %) on melanin synthesis in B16 meanoma cells. We separated 5 fractions from Ardisia crenata extract (70% MeOH) by MPLC. The 3rd, 4th, and 5th fractions showed the anti-oxidation (DPPH radical scavenging activity and supressive effect on Mn-SOD), whitening (inhibitory effect on melanin synthesis) and anti-inflammatory (supressive effects on $IL-1{\alpha}$, IL-6, COX-2 and total NO synthesis) effects.
The purpose of this study was to investigate the biological activities of water soluble browning reaction products(WS-BRPs) isolated from korean red ginseng. Antioxidative activities of WS-BRPs were examined with the various systems. Three different fractions prepared by os moly tic treatment of WS-BRP(fraction L, S-l and S-2) were found to have an ability to donate hydrogen to DPPH and also exhibited the inhibitory activities in lipid peroxidation, consumption of oxygen and protein oxidation of mitochondrial fraction. Especially, L had the strongest activity of these three WSBRPs in scavenging free radicals. Lipid peroxidation showed the antioxidant effect on linoleic acid oxidation inhibition ratio of $22.5\%,\;31.7\%,\;31.9\%\;and\;33.5\%$, respectivity. And the consumption of oxygen was strongly inhibited by $49.52\%,\;62,44,\;97.54\%$. But three WS-BRPs showed weak inhibitory activity on lipid peroxidation in rat hepatic microsomes.
