Survey of Overwintering Inoculum Potential of Anthracnose of Sweet Persimmon Caused by Colletotrichum gloeosporioides (감나무 탄저병균(Collectotrichum gloeosporioides)의 월동 전염원 조사)

  • In 2006 to 2007, the potential inoculum source of the anthracnose of sweet persimmon caused by Colletotrichum gloeosporioides was surveyed. The infected twigs, buds, dead twigs, petiole, leaves, dropped fruits were collected and tested for their possibility as overwintering inoculum. The detection rates of the pathogen from various parts of sweet persimmon tree were varied. When the collected samples were examined in April. Over than 93.3% of infected twig samples were harbored mycelia of C. gloeosporioides, and 46.7% of infected buds, 36.7% of dead twigs, 23.3% of petioles, and 16.7% of leaves were beared pathogenic fungus. No pathogenic fungus were detecded from healthy twigs and buds. Infected twigs and bud was important overwintering sites and formed conidia actively in next spring. The infected twigs, leaves, petioles, and fruits in growing season produced great number of conidia and caused active dissemination of the anthracnose disease in sweet persimmon. In growing season, all of the infected parts, such as twigs, leaves, petioles, and fruits produced pathogenic fungus.

Overwintering of the Pathogen and Factors Affecting Disease Develpoment of Rice Brown Spot Caused by Cochliobolus miyabeanus (벼깨씨무늬병균의 월동과 발병조건에 관한 연구)

  • This study was carried out to investigate factors affecting disease development of rice brown spot caused by Cochliobolus miyabeanus in the field and under the greenhouse conditions. In normal paddy fields, the pathogen was more frequently isolated from dried rice straws than wet straws after overwintering. The pathogen was also readily isolated from wet rice straws of salty paddy fields although infection rate of the fields was low. The disease symptom was first observed at 2 to 3days after inoculation when the seedlings were kept for 6 hours in dew chamber. The number of lesions peaked at 14 hr incubation in cultivars Chucheongbyeo and 24 hr incubation in cultivar Daesanbyeo and Tetep. Sixty three-d-old seedlings were more susceptible than 21 to 49-d-old seedlings regardless of cultivar. There was some difference in number of lesions observed, but the lesions were evenly spread regardless of leaf position. Light on the rice seedlings at 3,000 lux or 6,000 lux showed more severe symptoms than those in the dark.

  • These researches have been conducted to obtain the basic information of the effects of paraquat on plant species and to screen the plant species showing specific responses to paraquat. Paraquat resistance related to ecotype and overwintering capacity. Perennial and biennial species showed higher resistance than annual species. In a family, most species showed higher resistance were overwintering species. Lamiaceae, Brassicaceae, and Caryophyllaceae were tolerant to paraquat, whereas Poaceae and Asteraceae were sensitive. Especially Mosla dianthera of Lamiaceae, Hemistepta lyrata and Aster pilosus of Asteraceae, and Paspalum thunbergii of Poaceae showed higher tolerance than others. The response patterns of plant species on germination stage were different to those on vegetative stage. Germination of Amarathus lividus, Arabis glabra, and Bidens frondosa was not inhibited by paraquat. But their seedling growth were highly inhibited.

  • Studies were carried out to investigate the overwintering densities of spider mites, hatching rate and time of P. ulmi eggs, and seasonal occurrences of spider mites (P. ulmi and T. urticae) in apple orchards of Kyungpook province from 1987 to 1989. Overwintering density of P. ulmi eggs was higher in Kunwi, Andong, Chilgok but lower in Kyungju. Overovintering densities of T. urticae were high in all the regions. With the hatching time and rate for P. ulmi eggs, the first hatching ate was April 14, and the last was May 3, and the average hatching rate was 89.3%. The density of P. ulmi was high from early May to middle July and T. urticae begin to increase rapidly from middle June and then was continuously high upto fruit harvesting time.

  • The seasonal movements of Holotrichia morosa and H. diomphalia in soil were investigated during the period from 1984 to 1986 in Suwon. Most cf the eggs, active larvae and adults of the two species could be found at the soil depth of I-IDem. The larvae began downward movement in late October and early November in order for overwintering when soil tempe¬rature at 5cm below ground reached about $10^{\circ}C$. The overwintering depths of the larvae were 10-40cm for H. morosa and 30-80cm for H. diomPhalia. After the overwintering, pupation and adult emergence of H. morosa occurred at the overwintered sites, \vhereas H. diomPhalia larvae returned to near soil surface next April, and resumed feeding. H.diomphalia larvae began to move downward again in late June for pupation and adult emergence, and overwintered thereafter as adults at the depth of 1O-40cm.

  • The contents of flavonol glycosides, $kaempferol-3-O-{\beta}-D-glucoside$(1), $kaempferol-3-O-{\beta}-D-glactoside$ (2), $kaempferol-3-O-{\beta}-D-rhamnoside$ (3), $quercetin-3-O-{\beta}-D-glucoside$ (4) and $quercetin-3-O-{\alpha}-D-rhamnoside$ (5) in the houseleeks controlled by night-break, day-length control, and temperature during overwintering were determined to be compared with those in wild one. The contents of the flavonol glycosides 1-5 in the houseleeks were decreased roughly with warming during overwintering, and increased with longer light duration under the day-length control experiments. While warming functioned negatively on the production of the flavonol glycosides in the houseleek, longer light irradiation did positively during overwintering.

Assessment of environmental impact of vitamin A-enhanced soybeans and hybrid soybeans

  • An understanding of safety problems pursuant to environmental release of GM (Genetically Modified) crops is considered important. Among the recognized safety problems, the possibilities of weediness and ecosystem invasion are constantly being validated. We herein compared the growth characteristics and germination rate of soybeans formed by hybridization with vitamin A-enhanced soybeans carrying an introduced gene that increases β-carotene content. We also examined overwintering, survival, and weed competitiveness to evaluate hybrid ecological impact on long-term unmanaged cultivatable land. These studies revealed that the hybrid soybeans exhibited intermediate growth characteristics and germination rate compared with the vitamin A-enhanced soybeans and wild soybeans, or exhibited traits similar to those of the maternal strain. Overwintering experiments were conducted by planting seeds at depths of 0, 5, 10, and 20 cm and recovering them after three or five months. After five months, all seeds at depths more than 5 cm lost viability. Among seeds recovered after three months, only wild soybeans retained viability at depths of more than 5 cm. Survival and weed competitiveness were assessed by sowing each type of seed and performing no irrigation, or pest or weed control. Quantitative assessment of numbers of individual soybean plants that appeared in the experimental plot revealed that all plants germinated after sowing, but only wild type plants survived overwintering. These studies suggest that both GM soybeans and hybrid soybeans cannot survive in uncultivated land even if they are released into the environment, which indicates less possibility of ecosystem invasion and weediness.

Diapause and Voltinism in Ostrinia furnacalis (Lepidoptera: Crambidae) in Suwon, and Larval Instar Sensitivity to Diapause Induction (수원 지방에서 조명나방 휴면과 발생 세대수 및 휴면유도에 대한 유충의 민감성)

  • The diapause induction season in Ostrinia furnacalis (Lepidoptera: Crambidae) was estimated in Suwon. Three batches of adult generations were observed, the first one from early May to early July, the second from early or mid-July to early or mid-August, and the third from mid-August to October. In outdoor larval rearing, colony rearing occurring from mid-July to mid-August produced both non-overwintering and overwintering larvae, whereas late-reared colonies produced only overwintering larvae. Larvae collected during July and August in maize fields produced both non-overwintering and overwintering larvae, whereas late-collected larvae produced only overwintering larvae. The results indicated that O. furnacalis has a bi- or trivoltine complex life cycle in this area. In the laboratory, when larvae of all instars within 9 h after molting were first treated to a diapause induction condition (11:13 h = light:dark photoperiod and 20℃), almost all larvae were induced to diapause. However, when similar treatments were conducted age-specifically for the 5th instar larvae, diapause induction rates in 3- and 4-day-old larvae of the 5th instar decreased. In contrast, when larvae were subjected to the diapause induction treatment only during the periods from the hatching stage to the 2nd, 3rd, and 4th instar, almost all larvae were not induced to diapause. The results suggest that the early age of the 5th larval instar is the last stage for sensitivity to diapause induction stimuli. In the diapause-induced larvae, hemolymph trehalose content increased and body supercooling points dropped, compared with those in non-diapause larvae.

Genetic Differentiation of Overwintering Populations of Oriental Fruit Moth, Grapholita molesta, and Their Movement (월동세대 복숭아순나방(Grapholita molesta)의 지역적 분화 및 이동)

  • Spring phonology of the oriental fruit moth, Grapholita molesta, was monitored using sex pheromone traps in apple cultivating areas. Their occurrence was earlier in southern areas and their population sizes were significantly different among orchards even in a local cultivating zone. The overwintering populations appeared to move between local orchards, based on the fact that monitoring data obtained at the sites between orchards were similar to those of nearby orchards. However, within orchards, these adult movements appeared to decrease and showed skewed occurrences at the side of upwind direction or close to neighboring orchards. At initial occurrence peak(April 20-25), the ovenvintering populations of the different localities were collected and analyzed in their genetic distances. PCR-RAPD analysis indicated that there were significant genetic differences among the overwintering populations of G. molesta. This genetic differentiation of overwinterin populations may be due to genetic bottleneck following differential selection pressures against the subpopulations of G. molesta during winter on the basis of the RAPD analysis that each early spring population was significantly different to its previous fall population in the same locality.

Physiological Response of the Pearl Oyster, Pinctada fucata martensii, to Low Water Temperature: a Preliminary Study for Indoor Overwintering (진주조개 Pinctada fucata martensii의 실내월동을 위한 저온노출에 따른 생리적 변화)

  • With the aim of developing and indoor overwintering technique for Pearl oyster, Pinctada fucata martensii, the metabolic rates of young oysters (52.4-83.0 mm in shell length) were measured for 2 weeks at water temperatures of 8, 10, 12, and $14^{\circ}C$. The filtration rate (FR) ranged 0 to $4.84\;L\;h^{-1}gDW^{-1}$ (mean, $0.02{\pm}0.06 $ to $3.12{\pm}1.45$), with significant changes observed over thme except for the case of a water temperature of $14^{\circ}C$. Respiration rate (R) ranged from 0 to $2.370\;mgO2\;h^{-1}gDW^{-1}$ (mean, 0 to $1.77{\pm}0.37$), with significant respiratory disorders observed at temperatures below $12^{\circ}C$; in contrast, the rate increased on the $14^th$ day of the experiment in the case of a temperature of 14$^{\circ}C$. No significant difference was observed among the different water temperatures in terms of excretion rate (E) or absorption efficiency (Abs.eff), except for a significant decrease in aerobic metabolism in the case of water temperature of $8^{\circ}C$. The estimated scope for growth (SFG) ranged from -9.1 to $126.9\;J\;h^{-1}gDW^{-1}$ (mean. $-4.1{\pm}2.6$ to $82.85{\pm}42.6$). A significant energy Joss was found at $8^{\circ}C$, with negative SFG observed throughout the experiment and a gradual energy decrease observed over time at water temperatures of $10^{\circ}C$ and 120C. However. SFG remained positive throughout the experiment in the case of $14^{\circ}C$. The estimated minimum energy requirement, assessed from energy expenditure, is $8.00-34.24\;J\;h^{-1}gDW^{-1}$ (mean, $17.67{\pm}6.17$). In conclusion, the lowest temperature suitable for indoor overwintering is above $14^{\circ}C$.
