Studies on the MSM (Methyl Sulfonyl Methane) Treatment Method of Soybeans (콩의 MSM (Methyl Sulfonyl Methane) 처리 방법에 관한 연구)

  • This experiment was conducted at the affiliated farm of the Suncheon University from 2022 to 2023 to investigate the growth, yield, and quality characteristics of soybeans based on the treatment method of Methyl Sulfony Methane (MSM) for the establishment of stable production practices. In the initial investigation of growth characteristics in 2022, an increasing trend in characteristics such as plant height, stem thickness, and branching index was observed as the treatment concentration increased from 50% to 200%. Yield components also followed the same trend, with the basal fertilization + top dressing 3 times at 200% treatment showing the highest yield at 355 kg·10a-1, with the highest number of pods. In the subsequent study to determine the optimal concentration exceeding 200% in 2023, growth characteristics showed a trend of 400% > 200% > 800%. The basal fertilization + top dressing 3 times at 400% treatment exhibited the longest plant height (106.7 cm) and the most branches (6). In terms of seed quality, this treatment also had the highest proportion (66.9%) of seeds with a diameter over 6.7 mm. Additionally, in yield components such as pods, seeds per pod, and 100-seed weight, the basal fertilization + top dressing 3 times at 400% treatment showed the highest values, resulting in a maximum yield of 374 kg·10a-1, representing a 23.4% increase compared to the control. Therefore, for the optimal production of high-quality soybeans, it is recommended to apply the treatment of basal fertilization + top dressing 3 times at 400% concentration, with top dressing occurring at 30-day intervals before harvest.

Growth Characteristics of Bay Scallop (Argopecten irradians) reared in the Southern East Sea (동해 남부해역에서 양식된 해만가리비 (Argopecten irradians) 의 성장 특성)

  • Bay scallop (Argopecten irradians) has been farmed only in the South Sea of Korea. East Sea Fisheries Research Institute (ESFRI) has developed bay scallop aquaculture technologies to extend its aquaculture area to the Southeast Sea of Korea. For the artificial spawning, the water temperature was maintained at $23^{\circ}C$. Over 100,000,000 eggs were spawned through artificial spawning inductions, such as air exposure and thermal shock by rising the water temperature. The fertilization rate was over 91% with nearly 94,000,000 fertilized eggs. The shape of fertilized eggs was spherical with an average diameter of $61.7{\pm}0.05{\mu}m(54.1-67.4{\mu}m)$. Five days after fertilization, the eggs developed into prodissoconch shell, and continuously grew into umbo stage and then umbones stage. After 8 days of fertilization, the size of larva became $179.7{\pm}8.4{\mu}m$ on average ($150.4-204.8{\mu}m$), and the larva formed a foot and an eye spot. The larvae grew to $235.4{\pm}9.7{\mu}m$ in 10 days and attached to adherence material, becoming juvenile bay scallop. The shells grew from 22.71 mm to 72.40 mm in 6 month (June-December). The total weight increased from 2.0 g to 32.7 g at the same period. The daily growth rates of young scallop were $0.35mm\;d^{-1}$ (Apr. to Jun.) and $0.41mm\;d^{-1}$ (Jun. to Aug.), which were comparable to those found in the South Sea. These findings suggest that the bay scallop aquaculture may be suitable in the Southeast Sea of Korea and may provide an additional crop to aquaculturists.

A Three-year Study on the Leaf and Soil Nitrogen Contents Influenced by Irrigation Frequency, Clipping Return or Removal and Nitrogen Rate in a Creeping Bentgrass Fairway (크리핑 벤트그라스 훼어웨이에서 관수회수.예지물과 질소시비수준이 엽조직 및 토양 질소함유량에 미치는 효과)

  • Responses of 'Penncross' creeping bentgrass turf to various fairway cultural practices are not well-established or supported by research results. This study was initiated to evaluate the effects of irrigation frequency, clipping return or removal, and nitrogen rate on leaf and soil nitrogen con-tent in the 'Penncross' creeping bentgrass (Agrostis palustris Huds.) turf. A 'Penncross' creeping bentgrass turf was established in 1988 on a Sharpsburg silty-clay loam (Typic Argiudoll). The experiment was conducted from 1989 to 1991 under nontraffic conditions. A split-split-plot experimental design was used. Daily or biweekly irrigation, clipping return or removal, and 5, 15, or 25 g N $m-^2$ $yr-^1$ were the main-, sub-, and sub-sub-plot treatments, respectively. Treatments were replicated 3 times in a randomized complete block design. The turf was mowed 4 times weekly at a l3 mm height of cut. Leaf tissue nitrogen content was analyzed twice in 1989 and three times in both 1990 and 1991. Leaf samples were collected from turfgrass plants in the treatment plots, dried immediately at 70˚C for 48 hours, and evaluated for total-N content, using the Kjeldahl method. Concurrently, six soil cores (18mm diam. by 200 mm depth) were collected, air dried, and analyzed for total-N content. Nitrogen analysis on the soil and leaf samples were made in the Soil and Plant Analyical Laboratory, at the University of Nebraska, Lincoln, USA. Data were analyzed as a split-split-plot with analysis of variance (ANOVA), using the General Linear Model procedures of the Statistical Analysis System. The nitrogen content of the leaf tissue is variable in creeping bentgrass fairway turf with clip-ping recycles, nitrogen application rate and time after establishment. Leaf tissue nitrogen content increased with clipping return and nitrogen rate. Plots treated with clipping return had 8% and 5% more nitrogen content in the leaf tissue in 1989 and 1990, respectively, as compared to plots treated with clipping removal. Plots applied with high-N level (25g N $m-^2$ $yr-^1$)had 10%, 17%, and 13% more nitrogen content in leaf tissue in 1989, 1990, and 1991, respectively, when compared with plots applied with low-N level (5g N $m-^2$ $yr-^1$). Overall observations during the study indicated that leaf tissue nitrogen content increased at any nitrogen rate with time after establishment. At the low-N level treatment (5g N $m-^2$ $yr-^1$ ), plots sampled in 1991 had 15% more leaf nitrogen content, as compared to plots sampled in 1989. Similar responses were also found from the high-N level treatment (25g N $m-^2$ $yr-^1$ ).Plots analyzed in 1991 were 18% higher than that of plots analyzed in 1989. No significant treatment effects were observed for soil nitrogen content over the first 3 years after establishment. Strategic management application is necessary for the golf course turf, depending on whether clippings return or not. Different approaches should be addressed to turf fertilization program from a standpoint of clipping recycles. It is recommended that regular analysis of the soil and leaf tissue of golf course turf must be made and fertilization program should be developed through the interpretation of its analytic data result. In golf courses where clippings are recycled, the fertilization program need to be adjusted, being 20% to 30% less nitrogen input over the clipping-removed areas. The Effects of Resveratrol on Oocyte Maturation and Preimplantation Embryo Development

  • Biotechnologies for cloning animals and in vitro embryo production have the potential to produce biomedical models for various researches. Especially, pigs are a suitable model for xenotransplantation, transgenic production and various areas of reproductive research due to its physiological similarities to human. However, utilization of in vitro-produced embryos for transfer remains limited. Despite improvement over past few decades, obstacles associated with the production of good quality embryos in vitro still exist which limit the efficiency of cloning. One of major problems includes improper in vitro maturation (IVM) and culture (IVC). Oxidative stress caused from in vitro culture conditions contributes to inadequate IVM and IVC which leads to poor developmental competence of oocytes, failure of fertilization and embryo development. To reduce the oxidative stress, various antioxidants have been used to IVM and IVC system. However, limited information is available on the effects of resveratrol on livestock reproductions. Resveratrol is a polyphenolic natural product and well known as an antioxidant in foods and beverages (e.g. in grapes and red wine). Resveratrol is known to be cardioprotective, anticarcinogenic, anti-inflammatory, antioxidant and antiapoptotic. This paper will review the effects of resveratrol on in vitro maturation of oocytes and embryo development.

STUDIES ON NATIVE AND IMPROVED NATIVE PASTURES IN SOUTH SULAWESI, INDONESIA-EFFECTS OF SULFUR FERTILIZER AND STOCKING RATE ON ANIMAL PRODUCTION

  • The effects of S fertilization and stocking rate on cattle production from native and sown pastures were studied in South Sulawesi, Indonesia. On the native pasture there was no effect of S application over the three years of the experiment. The per head production was lower at the higher stocking rate (1.0 hd/ha), but the overall production increased by an average of 49%. There was no response to S applied to the clean seedbed pastures in the first year, but significant responses developed in the second and third years. There was an overall higher production from the higher stocked pastures (3.0 hd/ha), but the per head production was lower. There was a mean of about a two-fold increase in animal production from the highest native to the lowest improved pasture and a 3.4 times increase from the low stocking rate native to the high stocking rate sown pasture.

Co-culture of In Vitro Matured and Fertilized Bovine Oocytes with Oviductal Epithelium (체외성숙, 체외수정 우 난포란의 Co-culture에 관한 연구)

  • Bovine oocytes obtained from follicles(2~5mm) of ovaries after slaughter were cultured in TCM 199 medium with 10~20% heat-inactivated estrus cow serum(ECS) for 25~27 hr, at 39$^{\circ}C$ under 5% CO2 in air. At the end of culture period, some oocytes were stained with 1% acetoorcein and examined for the evidence of oocyte maturation. The remainder were used to assess the potential of in vitro fertilization(IVF) with frozen-thawed spermatozoa and subsequent development in media with or without bovine oviduct epithelial cell (BOEC) co-culture. The results obtained were summarized as follows ; 1. The maturation rate of oocyte in vitro in TCM 199 medium with 15% ECS group(76.3) was superior to 10% ECS group(68.3%) and 20% ECS group(64.5%). 2. The IVF rates of oocytes matured in vitro, and formation rate of male and female pronuclei were 63.6%(77/121) and 93.5%(72/77), respectively. The incidence of polyspermy was very low(2.4%). 3. Of 73 oocytes fertilized in vitro and cultured in TCM 199 medium with 10% fetal calf serum for 7 days, 41(56.3%) were cleaved over 2-cell and only 1(2.4%) was developed beyond the 16-cell stage. 4. Of 76 oocytes co-cultured with BOEC, 58(76.3%) were cleavaged and 23(39.7%) were developed to morula and blastocyst stage. The results of this study indicate that co-culture with BOEC deserved a positive effect on the IVF oocyte development through the 16-cell block.

Effects of Zinc Toxicity on Larval Development and Seed Collection of Abalone, haliotis discus hannai (참전복, Haliotis discus hannai 유생발생 및 채묘에 미치는 아연독성)

  • in the present study, the zinc toxicity to larval development and seed attachment of the abalone, Haliotis discus hannai was obtained under continuous flow through system. The zinc concentration melted from zinc coating pipe for 7 months ranged from $89.00\pm2.55 \mu\textrm{g}/\ell to 15.23\pm2.58\mu\textrm{g}/\ell(Y=0.85M^2-19.71+109.96)$. Treatments were carried out with zinc concentration $0~160 \mu\textrm{g}/\ell$. The maximum and minimum of fertilization rate were $87.7\pm5.3%$ in control, $83.7\pm7.6%$ in zinc concentration $160\mu\textrm{g}/\ell$, respectively. The maximum and minimum of hatching rate were $87.5\pm4.5%$ in zinc concentration $10\mu\textrm{g}/\ell$, $79.3\pm5.6%$ in zinc concentration $160\mu\textrm{g}/\ell$, respectively. Both of the results were not significantly different (P>0.05). But the normality rate, setting rate and survival rate of abalone larvae at over zinc concentration TEX>$20\mu\textrm{g}/\ell$ decreased rapidly and showed significantly different from those of the other group(P<0.05).

Effect of Water Temperature on the Embryonic Development of Panther Puffer Takifugu pardalis (수온이 졸복 Takifugu pardalis의 난발생에 미치는 영향)

  • We studied the effects of temperature on the egg development and larvae of Takifugu pardalis. The larvae hatched 135-480 hr after fertilization in $13^{\circ}C-24^{\circ}C$ and 32 ppt. The higher the temperature the earlier the larvae hatch within $13^{\circ}C-24^{\circ}C$. The maximum hatching rate was made in $15^{\circ}C$ and 32 ppt. The higher the temperature the larger the newly hatched larvae. The larvae hatched in $24^{\circ}C$ were ca. 3.35 mm in total length. Occurrence of the deformed larvae was lower in $13^{\circ}C-18^{\circ}C$ regime and higher over $21^{\circ}C$ regime.

Study on New Capacitation-method of Bovine Sperm (소 정자의 새로운 수정능 획득방법에 관한 연구)

  • This study was aimed to develope the new method for bovine sperm capacitation through testing and combinating factors relating to sperm capacitation. For verifying the efficiency of this method on inducing capacitation, in vitro fertilization was carried out. The results obtained were as follows: When pHs of HBS /PR for sperm-washing were 5.99, 6.38, 6.78, 7.10, 7.40, 7.69, 8.15, 8.45 and 8.83, variances between light absorhance differences obtained from sperm pre-washing solution and post-washing solution(VADs) were 0.000, 0.001, -0.001, -0.005, -0.005, -0.021,-0.017,-0.016,and-0.036, respectively. There were significant decreases of VADs in pH 7.69 and pH 8.83. When sperm were firstly, secondly, and thirdly washed with HBS /PR, VADs were -0. 024, - 0.006, and - 0.004, respectively. There was significant decrease of VAD in 1st sperm-washing. When washed-sperm were secondly washed with HBS /PR supplemented with no (con-trol), heparin, CSA, PC12, and BSA, VADs were 0.009, -0.024, -0.008, -0.009, and 0.014, respectively. When the sperm were thirdly washed with HBS /PR with no(control), heparin, CSA, PC12 and BSA, VADs were 0.020, -0.007, 0.005, 0.006, and 0.019, respectively. Only heparin treatment showed the negative values of VADs in 2nd and 3rd sperm-washing. Of oocytes cultured with sperm which were repeatedly washed with heparin and high pH, 52% (57/110) were cleaved over 2 cell stage. However, percent of oocytes parthenogenetically divided was 5%(2 /42).

The Effects of Existing Vegetation and Fertilization on the Improvement of Natural Grassland by Oversowing (지표추파법에 의한 목야지 개량시 선점식생과 시비의 영향)

  • A review of factors influencing grass and clover establishment, survival and yield at oversowing was made from the experimental results of home and abroad. The following conclusions are considered: (1) The existing vegetation present at oversowing appeared to be the most critical factor reducing establishment and survival of grass. Therefore, it is essential to check competition from the native vegetation before oversowing. (2) Although lime had comparatively little effect on yield of grassland, the general effect of lime should be more emphasized under our acid soil conditions to promote the availability of all the essential elements and the growth of microorganisms, and to reduce the toxic effects of nutrients. One to two tons of lime per ha at oversowing would be useful. (3) Phosphorus is one of the nutrients most generally deficient in grassland soils of Korea, consequently, this nutrients applied at oversowing is very effective. Application as much as 200kg of phosphorus per ha would be essential. (4) The effect of nitrogen on the establishment and survival of grass depends on the amount and density of the herbage present. The use of nitrogen in dense herbage adversely affected grass establishment and survival, possible because the fertilizer stimulated the growth of the eisting herbage. Around 40kg of nitrogen per ha may be enough at oversowing (5) Potassium is not as universally deficient in soils of native grassland as phosphorus. Therefore, it cannot be over-emphasized at oversowing. Studies determinig the optimum amount of potassium at of oersowing are needed.
