Safflower (Carthamus tinctorius L.) seeds have long been clinically used in Korea to promote bone formation and prevent osteoporosis. In addition, the safflower buds (SB) were found to have more useful functional ingredients than safflower seed. Thus, we investigated the preventive effects of SB diet in ovariectomized (OVX) rats. The rats were divided into five groups; sham operated group, OVX alone group, OVX plus 17β-estradiol (E2 10 μg/kg, i.p.) and OVX plus SB diet feeding group (0.3% or 1%). Feeding of SB diet (0.3% or 3%) to OVX rats markedly increased trabecular formation in femur compared to OVX rats. Feeding of SB diet (0.3% or 3%) to OVX rats also decreased TRAP activity compared to OVX rats. These results suggest that SB diets have bone sparing effects by the decrease of osteoclast activity. We also observed that OVX rats fed with SB diet (0.3% or 3%) exhibited the decrease of calcium and phosphorus in serum compared to OVX-induced rats. Therefore, SB may be beneficial for the patients of osteoporosis, especially in postmenopausal women.

Objectives: The present study aimed at investigating the effects of Schisandrae Fructus (SF) on menopause symptoms in ovariectomized mice. Methods: 6 weeks old female mice were ovariectomized (OVX), and the OVX mice were fed with a three doses of SF (25, 100, and 400 mg/kg, respectively) or $17{\beta}$-estradiol (E2, 0.5 mg/kg) 3 times a week for 6 weeks. Body weight was measured at every week and uterus weight was also measured at the end of experiment. Lipids and osteocalcin levels in serum were analyzed. Results: The uterine weight was not changed in SF treated group, while E2 intake increased the reduced uterine weight by OVX. Although Body weight was increased since two weeks after ovariectomy compared with sham operated group, there was no change in SF or E2 treated group compared with OVX. But LDL was significantly reduced in SF (400 mg/kg) or E2 fed mice and SF or E2 treatment decreased enhanced osteocalcin level by OVX. Conclusion: These results indicated that SF would be effective in the improvement of LDL level and osteoporosis on menopause.
The effect of calcium and boron supplementation on bone strength was determined in growing and ovariectomized (OVX) Sparague-Dawley rats. Rats were divided into 9 groups and fed diet with different intake levels of calcium and boron for 4 weeks. About fifty percentages of rats in each group were OVX and the others were sham-operated. The rats were fed same diets after operation for 8 weeks. The feed intake, body weight gain, and FER were significantly higher in OVX rats than those in sham-operated ones. Serum osteocalcin, bone formation biomarker, was significantly increased with increment in calcium and boron intakes. Serum estradiol was lower in OVX rats than in sham-operated ones. Bone mineral density of femur was significantly lower in OVX rats than in other group. The breaking forces of bones were not significantly different among the groups. The urinary excretion of deoxypyridinoline, osteolytic marker was significantly increased with increment in calcium intake and ovariectomy. The urinary calcium excretion was significantly increased with increment in calcium intake, but decreased with increment in boron intake. According to theses results, the boron supplementation resulted in higher serum osteocalcin and lower urinary calcium excretion. Therefore, it could be suggested that the boron supplementation may be complementary and useful to calcium nutrition for bone health.
This study was carried out to investigate the effects of the Korean Safflower (Carthamus inctorius L) seed powder on serum level of hormones and trabecula area during the recovery from osteoporosis induced ovariectomized rats. Four month-old rats were ovariectomized (OVX), remained untreated for 8 weeks, and were subsequently administered safflower seed (0.03 g/kg) every other day 30 for days. We examined the effects of treated safflower seed every 10 days on ovariectomy-related changes in Insulin-like Growth Factors, Insulin-like Growth Factor binding protein-3 (IGFBP-3), Estrogen, Bone-specific alkaline phosphotase, Calcium, and Phospotase in the serum, and also histomorphology of the proximal fibula metaphysis and femur/body weight rate. Ten and 20 days after safflower seed treatment in OVX rats, serum levels of IGF-I, -II and IGFBP-3 were not different from the Sham and OVX groups. In 30 days, serum levels of IGF-I,-II and IGFBP-3 were higher after safflower seed treatment in OVX rats as compared to the other two groups (p<0.05). Bone alkaline phosphatase levels were increased through safflower seed treatment in OVX rats compared to the other two groups in 30 days. There were no differences between OVX and safflower seed treated OVX rats in serum levels of estrogen and femur/body weight rate, but estrogen levels for the sham group were higher than for the other two groups. The safflower seed is increased to serum levels of IGFs, IGFBP-3 and BALP of osteoporosis induced by ovariectomized rats. Thus, we conclude that the safflower seed is a possible role for improvement of osteoporosis induced-ovariectomized rats.
The aim of this study was to evaluate the effects of three different mulberry cultivars extracts on serum lipid contents in ovariectomized estrogen-deficient rats. Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with $ 80\% $ ethyl alcohol extracts from various kinds of Tajikistan mulberry (OVX-TM), Korea mulberry (OVX-KM) and China mulberry (OVX-CM) at 200mg/kg bw/day, respectively. The mulberry extracts were orally administrated at 1mL per day. The body weights of OVX rats were significantly heavier than the sham-operated rats at all values (p <0.05). The ovariectomy caused an expected increasing in the levels of serum total cholesterol and triglyceride. The serum HDL-cholesterol level on the OVX-TM, OVX-KM and OVX-CM groups were higher than in the OVX-control group.
It is controversial whether low calcium intake, commonly associated with osteoporosis, results in calcium accumulation in soft tissues. This study was conducted to investigate the effects of low calcium (Ca) and oxalate (ox) intake on soft-tissue Ca deposits and bone metabolism in ovariectomized (ovx) rats. Eight week old female Sprague-Dawley rats were ovariectomized and divided into four groups. The rats were fed experimental diets containing low (0.1%, w/w) or normal (0.5%, w/w) Ca with or without sodium oxalate (1%, w/w); Sham/NCa, Ovx/NCa, Ovx/LCa, Ovx/NCa-ox, Ovx/LCa-ox for 6 weeks. All ovx rats showed a remarkable increase in body and tissue weight, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, blood urea nitrogen, alkaline phosphatase, and decreases in weight, ash, and Ca contents, as well as bone breaking force compared to those in sham rats. Serum Ca concentration was not significantly affected by dietary Ca levels or ox intake. Kidney Ca, ox acid content, and microscopic Ca deposition increased remarkably in the Ovx/LCa-ox group compared to those in the other groups. Ca content in the spleen and aorta also increased significantly, but the weight contents, Ca, bone breaking force, and Ca and oxalic acid in feces decreased significantly in the Ovx/LCa-ox group. Serum parathyroid hormone levels were not significantly different among the groups. These results indicate that low Ca intake decreased bone mineral content and increased Ca deposits in soft tissues, which was aggravated by ox intake in ovx rats. Thus, high ox intake may result in a kidney disorder in patients with osteoporosis who eat a low Ca diet.

Much attention has been given to the role of safflower (Carthamus tinctorius L.) seed in preventing and treating osteroporosis recently. Although numerous studies were done on effects of safflower oil, no attention was given to the role of dietary fiber of safflower seeds. This study was aimed at investigating physiological significance of defatted safflower seed as a source of dietary fiber. Sprague Dawley female rats were assigned one of 5 groups: ovariectomized control (Ovx-control) group and ovariectomized rats 15% (Ovx-ss15) and 30% defatted safflower seed (Ovx-SS30), sham-operated (Sham) group and a normal group. Cholesterol was supplemented to all diets at 0.5% except the normal diet. Ovariectomized rats were pair-fed isocalorically to the Sham group. Ovariectomy caused heavier body weight, but feeding 30% safflower seed brought back to the level of Sham group. Activities of disaccharidases of jejunal mucosa were significantly lowered in Ovx-control group compared to those of Sham, and supplementation of safflower seed tended to increase the activities. Fecal weight of Ovx-SS15 and Ovx-SS30 were almost twice as those of Ovx-control Sham. Propionic acid and butyric acid concentrations per g of feces of Ovx-SS15 and daily excretion of these fatty acids were significantly increased as compared to those of Sham and Ovx-control. In concentration defatted safflower seed supplementation significantly increased fecal bulk and short chain fatty acid production in large intestine of rats.

Objective : Osteoporosis result from age-related decline in the number of osteoblast progenitors in the bone marrow. Probiotics have beneficial effects on the host, when administered in appropriate amounts. This study investigated the effects of probiotics expressing specific genes, especially the effects of genetically modified bone morphogenetic protein (BMP)-2-expressing Lactobacillus plantarum CJNU 3003 (LP) on ovariectomized rats. Methods : Twenty-eight female Wistar rats (250-300 g, 12 weeks old) were divided into four groups : the sham (control), the ovariectomy (OVX)-induced osteoporosis group (OVX), the OVX and LP (OVX/LP), OVX and genetically modified BMP-2-expressing LP (OVX/LP with BMP) groups. The three groups underwent bilateral OVX and two of these groups were administered two different types of LP via oral gavage daily. At 16 weeks post-OVX, blood was collected from the heart and the bilateral tibiae were extracted and were scanned by ex-vivo micro-computed tomography and stained with hematoxylin-and-eosin (H&E) and Masson's trichrome stain for pathological assessment. The serum levels of osteocalcin (OC), rat C-telopeptide of type I collagen (CTX-I), BMP-2, and receptor activator of nuclear factor-ĸB ligand (RANKL) were measured. Results : The 3D-micro-computed tomography images showed that the trabecular structure in the OVX/LP with BMP group was maintained compared with OVX and OVX/LP groups. No significant differences were detected in trabecular thickness (Tb.Th) between control and OVX/LP with BMP groups (p>0.05). Furthermore, a tendency toward increased BMD, trabecular bone volume, Tb.Th, and trabecular number and decreased trabecular separation was found in rats in the OVX/LP with BMP groups when compared with the OVX and OVX/LP groups (p>0.05). The H&E and Masson's trichrome stained sections showed a thicker trabecular bone in the OVX/LP with BMP group compared with the OVX and OVX/LP groups. There was no difference in serum levels of OC, CTX and RANKL control and OVX/LP with BMP groups (p>0.05). In contrast, significant differences were found in OC and CTX-1 levels between the OVX and OVX/LP with BMP groups (p<0.05). Conclusion : Our results showed that the expression of genetically modified BMP-2 showed inhibition effect for bone loss in a rat model of osteoporosis.

A decrease in the circulating levels of estrogen, occuring as a consequence of post menopausal decline or from surgical ovariectomy, results in an accelerated loss of bone. Estrogen has been shown to stimulate lysyl oxidase activity, and the treatment with estrogen increased the pyridinium content of cortical bone. a trivalent mature cross-links collagen fibrils named pyridinoline, which is especially abundant in collagen of cartilage and bone, markedly increases with growth in humans and rats. The main aim of this study was to examine the increased bone loss caused by ovariectomy through monitoring the concentrations of the collagen and the pyridinium cross-links of collagen, pyridinoline. The ovariectomized rats, 4 weeks old, were divided at random into two or three groups of 5. Ovariectomies were carried out on both of the saline-treated group(OVX(NH)) and the estrogen-treated group(OVX(H)) using the dorsal approach and sham operations were performed on the sham-operated group(sham). They were maintained under identical conditions for 4 or 8 weeks and were allowed free access to food and water. it was observed that there was no significant difference between the control group and the sham-operated group, however, the control group had a higher content of collagen than the saline-treated group after 4 weeks and 8 weeks. Based on these results, iot is supposed that estrogen can enhance collagen synthesis and affects the pyridinoline formation in collagen fibrils through stimulating lysyl oxidase activity.
This study attempted to investigate if the soy isoflavone, genistein, influences bone metabolism in ovariectomized, 4-week-old female Wister rats. All the rats were divided into sham (SH) and ovariectomized (OVX) groups consisting of OVX-17${\beta}$-estradiol($10\;{\mu}g/kg$ b.w.), OVX-1mg or 5 mg or 10mg of genistein/kg b.w. The rates were allowed ad libitum access to foods and water for 8 weeks. The Results showed that body weight had significantly increased in the OVX group compared to the SH group (p<0.05) and was not different among the OVX-GEN and OVX-ES groups and the OVX group. The liver and uterus weights in the OVX groups were slighter than those in SH group (p<0.05). The kidney weight in the OVX groups was decreased compared to in that in the SH group but was not significantly different among all OVX groups. Femoral length in the OVX groups was longer than in the SH group and was not different. Rats in the OVX groups had higher creatinine levels than those in the SH group and hydroxyproline level did not differ significantly among any of the groups. Serum ALP activity and Ca in bone, feces, urine and serum did not change among the groups. Bone mineral density (BMD) decreased in the OVX groups compared to the SH group and was slightly increased by feeding genistein (p>0.05). Breaking force and stiffness did not change by ovariectomy and feeding genistein. Hence, these results suggested that estrogen may affect bone mineralization in growing rats and that genistein be may involved in the prevention of bone loss. However, more studies are needed to identify the proper mechanism of genistein and bone formation.
