A histochemical study of argentaffin endocrine cells in the gastrointestinal tract of ovariectomized rats

  • The regional distributions and frequencies of argentaffin endocrine cells in gastrointestinal (GI) tract of osteoporotic Sprague-Dawley rat induced by ovariectomy were studied by Masson-Hamperl silver stain. The experimental animals were divided into two groups, one is non-ovariectomized group (Sham) and the other is ovariectomized group (OVX). Samples were collected from each part of GI tract (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) at 10th week after ovariectomy or sham operation. Argentaffin cells were detected throughout the entire GI tract with various frequencies regardless of ovariectomy except for the rectum of OVX in which no cells were detected. Most of these argentaffin cells in the mucosa of GI tract were generally spherical or spindle in shape (open type cell) while cells showing round in shape (close type cell) were rarely found in gland regions. Significant decrease of argentaffin cells was detected in OVX compared to that of Sham except for the fundus and jejunum. However, in the fundus and jejunum, argentaffin cells in OVX showed similar frequency compared to that of Sham. In conclusion, the endocrine cells are the anatomical units responsible for the production of gut hormones that regulate gut motility and digestion including absorption, and a change in their density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion. Ovariectomy induced severe quantitative changes of GI argentaffin endocrine cell density, and the abnormality in density of GI endocrine cells may contribute to the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis.

Difference in Spinal Fusion Process in Osteopenic and Nonosteopenic Living Rat Models Using Serial Microcomputed Tomography

  • Objective : To identify and investigate differences in spinal fusion between the normal and osteopenic spine in a rat model. Methods : Female Sprague Dawley rats underwent either an ovariectomy (OVX) or sham operation and were randomized into two groups: non-OVX group and OVX group. Eight weeks after OVX, unilateral lumbar spinal fusion was performed using autologous iliac bone. Bone density (BD) was measured 2 days and 8 weeks after fusion surgery. Microcomputed tomography was used to evaluate the process of bone fusion every two weeks for 8 weeks after fusion surgery. The fusion rate, fusion process, and bone volume parameters of fusion bed were compared between the two groups. Results : BD was significantly higher in the non-OVX group than in the OVX group 2 days and 8 weeks after fusion surgery. The fusion rate in the non-OVX group was higher than that in the OVX group 8 weeks after surgery (p=0.044). The bony connection of bone fragments with transverse processes and bone formation between transverse processes in non-OVX group were significantly superior to those of OVX group from 6 weeks after fusion surgery. The compactness and bone maturation of fusion bed in non-OVX were prominent compared with the non-OVX group. Conclusion : The fusion rate in OVX group was inferior to non-OVX group at late stage after fusion surgery. Bone maturation of fusion bed in the OVX group was inferior compared with the non-OVX group. Fusion enhancement strategies at early stage may be needed to patients with osteoporosis who need spine fusion surgery.

  • Effect of Gamisamul-tang(GS) on body weight, uterine weight, splenial weight, serum and estrogen receptor-α (ER-α) in kidney were investigated in ovariectomized(OVX) rats. Female Sprague -Dawley rats with a body weight of 220 g were undergone ovariectomy or sham operation. The ovariectomy caused significant decrease in serum levels of HDL-cholesterol and in uterine weight compared to those of sham-operated animals. while caused significant increase in serum levels of total cholesterol, LDL-cholesterol, triglyceride, body weight and in splenial weight. In contrast, administration of OVX rats with the GS significantly elevated serum levels of HDL-cholesterol and uterine weight, but significantly depressed serum levels of total cholesterol, LDL-cholesterol, triglyceride, body weight and in splenial weight. ER-α localized in the kidney vessel endothelial cells and its expression was increased by ovariectomy and decreased by administration of OVX rats with the GS. Our results suggest that a possible protective effect of as against hyperlipidemia in postmenopausal cardiovascular disease and as increase the estrogen-estrogen receptor romp/ex in the kidney vessel endothelial cells.

Effects of Aqueous Extracts of Cynanchum wilfordii in Rat Models for Postmenopausal Hot Flush

  • Menopausal hot flushes (HFs), which manifest as a transient increase in skin temperature, occur most frequently in postmenopausal women, and sometimes negatively influence daily life. We investigated the effect of an aqueous extract of Cynanchum wilfordii (CWW) in a rat model of menopausal HFs, where tail skin temperature (TST) is increased after the rapid estrogen decline induced by ovariectomy. Ten-week-old female rats were ovariectomized and treated with CWW for 1 week. We measured TST and rectal temperatures (RT) and investigated serum estradiol. The TST in ovariectomized (OVX) rats was significantly elevated after ovariectomy compared with control rats, whereas the RT in OVX rats was not elevated. Administration of CWW (200 mg/kg/d for 7 days, p.o.) significantly improved the skin temperature increase in OVX rats. The lower level of serum estradiol in OVX rats was significantly increased by supplying E2, but it was not affected by CWW. The present study indicates a need for future research involving treatment with high concentrations of C. wilfordii and measurement over 24 h.

Interleukin-10-Producing B Cells Help Suppress Ovariectomy-Mediated Osteoporosis

  • Osteoporosis is prevalent in elderly women and it may cause dental implant failure. In particular, estrogen deficiency in postmenopausal women leads to higher rates of osteoporosis prevalence. Immune cell-mediated effects involving the development of osteoporosis have been studied previously; however, the role of IL-10-producing regulatory B (B10) cells in osteoporosis is largely unclear. Here, we examined the role of B10 cells in osteoporosis. C57BL/6 mice were subjected to ovariectomy (OVX). Fifteen weeks after OVX surgery, the first molar of the right maxillary was extracted, and twenty-four weeks after OVX surgery, serous progression of osteoporosis was observed in the alveolar bone. Moreover, the proportion of CD19+CD5+CD1dhigh regulatory B cells, B10, and CD4+CD25+FoxP3+ regulatory T cells from the spleen of OVX mice decreased during the progression of osteoporosis, compared to controls. In contrast to regulatory cells, IL-17-producing Th (Th17) cell levels were increased in OVX mice. Adoptive transfer of B10 cells to OVX mice led to a decrease in Th17 cell abundance and inhibited the development of osteoporosis in the alveolar bone from OVX mice. Thus, our results suggest that B10 cells may help suppress osteoporosis development.

Preliminary Study on Effect of Lactiplantibacillus plantarum on Osteoporosis in the Ovariectomized Rat

  • Osteoporosis is a growing global health concern primarily associated with decreased estrogen in postmenopausal women. Recently, some strains of probiotics were examined for potential anti-osteoporotic effects. This study intended to evaluate the impacts of Lactiplantibacillus plantarum MGE 3038 strain (MGE 3038) in ovariectomized rats. For this purpose, twelve weeks old female Wistar rats (n=21; 250-300 g) were divided into 3 groups; ovariectomy (OVX) group, OVX/MGE 3038 group and Sham group (control). In these groups; two went through respective OVX and one had daily MGE 3038 administration through oral gavage. Prior to 16 weeks after OVX, we collected blood samples and extracted the tibiae. We scanned the extracted tibiae by in-vivo micro-computed tomography (micro-CT) and evaluated pathology by hematoxylin and eosin (H&E) and Masson's trichrome staining. The serum levels of C-telopeptide of type I collagen (CTX), osteocalcin (OC), and the receptor activator of nuclear factor-κB ligand (RANKL) were examined. The OVX/MGE 3038 group showed increases in bone mineral density, trabecular bone volume, trabecular number, and trabecular thickness (Tb.Th), and a decrease in trabecular spacing than the OVX group. However, OVX/MGE 3038 group and control group were measurably comparable in Tb.Th. Micro-CT, H&E, and Masson's trichrome findings exhibited increased preservation and maintenance of trabecular bone structure in the OVX/MGE 3038 group in comparison to the OVX group. In serum, the levels of CTX, OC and RANKL were significantly different between the OVX and OVX/MGE 3038 groups. Taken together, L. plantarum MGE 3038 could be helpful for the treatment of osteoporosis.

The Change of Bone Metabolism in Ovariectomized Rats : Analyses of MicroCT Scan and Biochemical Markers of Bone Turnover

  • Objective : The purpose of this study was to verify the appropriateness of ovariectomized rats as the osteoporosis animal model. Methods : Twelve female Sprague-Dawley rats underwent a sham operation (the sham group) or bilateral ovariectomy [the ovariectomy (OVX) group]. Eight weeks after operations, serum biochemical markers of bone turnover were analyzed; osteocalcin and alkaline phosphatase, which are sensitive biochemical markers of bone formation, and C-terminal telopeptide fragment of type I collagen C-terminus (CTX), which is a sensitive biochemical marker of bone resorption. Bone histomorphometric parameters and microarchitectural properties of 4th lumbar vertebrae were determined by micro-computed tomographic (CT) scan. Results : The OVX group showed on average 75.4% higher osteocalcin and 72.5% higher CTX levels than the sham group, indicating increased bone turnover. Micro-CT analysis showed significantly lower bone mineral density (BMD) (p=0.005) and cortical BMD (p=0.021) in the OVX group. Furthermore, the OVX group was found to have a significantly lower trabecular bone volume fraction (p=0.002). Conclusion : Our results showed that bone turnover was significantly increased and bone mass was significantly decreased 8 weeks after ovariectomy in rats. Thus, we propose that the ovariectomized rat model be considered a reproducible and reliable model of osteoporosis.

Effects of Ovariectomy on Insulin Resistance and β-Cell Function and Mass

  • The prevalence of type-2 diabetes increases remarkably in post-menopausal women, possibly because insulin secretion fails to compensate for the insulin resistance induced in various tissues by estrogen insufficiency. However, this has not been fully defined. Therefore, the present study investigated whether an ovariectomy (OVX) would increase insulin resistance and decrease the $\beta$-cell function and mass in female rats with and without a $90\%$ pancreatectomy (Px). Female rats aged 15 weeks were divided into four groups: 1) OVX + Px, 2) SOVX (sham operation of OVX) + Px, 3) OVX + SPx (sham operation of Px), and 4) SOVX + SPx, and given a $30\%$ fat diet for 8 weeks. At the end of the experimental period, the islet function and insulin resistance were determined using a hyperglycemic clamp and a euglycemic hyperinsulinemic clamp, respectively. The OVX only increased the body weight in the SPx rats, which was partially related to the food intake. Yet, the OVX did increase the peripheral insulin resistance, while the Px increased this resistance further. The OVX and Px both exacerbated the islet function, as measured by the insulin secretion pattern, while delaying and decreasing the first-phase insulin secretion. The OVX only decreased the proliferation of $\beta$-cells in the Px rats, while increasing apoptosis in both the Px and SPx rats. As a result, the OVX decreased the $\beta$-cell mass in the Px rats, but increased the mass in the SPx rats. In conclusion, an OVX was found to accelerate the development and progression of diabetes by increasing the insulin resistance and decreasing the $\beta$-cell mass. Therefore, menopause can be a risk factor for type-2 diabetes, mainly due to a deceased proliferation of $\beta$-cells.

난소절제 쥐에서 비타민 D 첨가식이가 골밀도와 골무기질 함량에 미치는 영향 (The Effects of Vitamin D Supplementation on Bone Mineral Density and Bone Mineral Content in Ovariectomized Rats)

  • The effects of vitamin $D_{3}$ supplementation on bone mineral density and bone mineral content in ovariectmized rats were investigated. Forty female Sprague-Dawley rats (body weight 180 g) were divided into two group, ovariectomy and Sham groups, which were each randomly divided into two subgroups that were fed casein and casein supplemented with vitamin $D_{3}$ diets. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXlmus (GE Lunar Co, Wisconsin, USA) in spine and femur on 6 weeks after feeding. Osteocalcin and urinary DPD crosslinks value were measured as markers of bone formation and resorption. The body weight gain and food efficiency ratio (FER) were higher in OVX groups than in Sham groups regardless of diets. Serum Ca concentration and urinary Ca excretion were higher in vitamin $D_{3}$ supplemented group than in casein group in Sham and OVX. Crosslinks values were higher in OVX groups than in Sham groups. Spine BMD and femur BMD of ovariectomy group were significantly lower than Sham groups, however vitamin $D_{3}$ supplemented groups were significantly higher than control groups in Sham and OVX. In conclusion, dietary vitamin $D_{3}$ supplementation on ovariectimized rats were significantly increased bone mineral density and bone mineral content in spine and femur.

난소 절제 흰쥐에서 카페인 첨가가 혈액과 간 지질 농도에 미치는 영향 (Effects of Caffeine Supplementation on Lipid Concentrations of Serum and Liver in Ovariectomized Rats)

  • The purpose of this study was to examine the impact of caffeine on lipid concentrations in ovariectomized rats. Forty-four rats (75${\pm}$5 g) were randomly divided into ovariectomy(ovx) and sham groups, each of which were randomly divided into two subgroups that were fed the control diet or a caffeine-supplemented diet (0.03%/diet). The experimental groups were classified into 1) sham-control, 2) sham-caffeine, 3) ovx-control and 4) ovx-caffeine groups. All rats had been fed on experimental diet and deionized water ad libitum for 6 weeks. Triglyceride concentration and total cholesterol were measured in serum and liver. Serum HDL-cholesterol was measured. Serum LDL-cholesterol and atherogenic index were calculated. Weight gain and food efficiency ratio were higher in ovx groups than in sham groups (p<0.05). Caffeine intake did not affect weight gain and food efficiency ratio. Mean food intake was significantly increased in ovx-caffeine group than other three groups. There were no significant differences in serum triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol and atherogenic index between the control diet group and caffeine diet group. In addition, hepatic triglyceride and cholesterol level was unaffected by caffeine intake or ovariectomy. Therefore, we suggest that caffeine (0.03%/diet) intake may not have adverse effects on serum lipid level in ovariectomized rats.
