This study determined the effect of ovarian status at the beginning of the modified Double-Ovsynch program on reproductive performance in dairy cows. In the study, 1,302 cows were treated with a modified Double-Ovsynch program at 56 days after calving. This program comprises administering gonadotropin-releasing hormones (GnRH), prostaglandin F2α (PGF2α) 10 days later, GnRH 3 days later, GnRH 7 days later, and GnRH 56 h later, followed by timed artificial insemination (TAI) 16 h later. At the beginning of the program, cows were categorized according to the size of the largest follicle and the presence of a corpus luteum (CL) in the ovaries as follows: 1) small follicle (<5 mm, SF group, n = 100), 2) medium follicle (8-20 mm, MF group, n = 538), and 3) large follicle (≥25 mm, LF group, n = 354) without a CL, or 4) the presence of a CL (CL group, n = 310). The pregnancies per AI after the first TAI were analyzed by logistic regression using the LOGISTIC procedure, and the logistic model included the fixed effects of the herd size, parity, body condition score (BCS) at the first TAI, TAI period, and ovarian status. A larger herd size, higher BCS at the first TAI, and TAI period with no heat stress increased (p < 0.05) the probability of pregnancy per AI after the first TAI. However, ovarian status at the beginning of the program did not affect (p > 0.05) the pregnancies per AI (ranges of 37.9% to 42.9%). These results show that the modified Double-Ovsynch program can be used effectively while maintaining good fertility regardless of the ovarian status in dairy herds.
The relationship between palpable ovarian structure and milk progesterone levels were determined in 144 dairy cows. Depending on the ovarian structure and diseases were divided into two groups, Group I (absence of functional luteal tissue in ovary and <2ng/ml in milk progesterone levels) and Group II(presence of functional luteal tissue in ovary and ${\geq}2ng/ml$ in milk progesterone levels) 1. Among 69 cows of group I, dysfunction of ovary, atropy of ovary, follicle is ovary, follicular cyst and corpus luteum albicans were 17(11.8%), 19(13.2%), 14(9.7%), 3(2.1%) and 16 cows(11.1%), and among 75 cows of group II, corpus luteum A, B and C were 16(11.1%), 17(11.8%) and 42 cows(29.2%), respectively. 2. In Group I, milk progesterone concentrations were <1ng/ml in 55 cows(79.9%). Conversely in Ggroup II, milk progesterone concentrations were ${\geq}4ng/ml$ in 55 cows(73.3%). 3. The mean(${\pm}SE$) concentrations of milk progestsrone in the Group I and II were $1.62{\pm}0.45$ and $7.64{\pm}0.68ng/ml$, respectively, and CR test showed the difference in milk progesterone concentrations between the two groups to be statistically significant(p<0.01). 4. The mean(${\pm}SE$) concentration of milk progesterone in cows with corpus luteum A, B and C were $8.11{\pm}1.83$, $8.48{\pm}1.30$ and $7.12{\pm}0.82ng/ml$, respectively, there was no significant relationship between palpable corpora luteum structure and milk progesterone concentration. 5. The accuracy of ovarian diagnosis was 82.6 and 20.2% in the Group I and II, respectively, and Chi-square test showed the difference in accuracy between the two groups to be statistically significant (p<0.001). 6. The agreement between the rectal palpation and milk progesterone concentrations in ovarian disease was 50%.
This research investigates the fine structural as well as the morphological changes of the mouse ovarian tissue after irradiation of various dose rates of 6 MeV LINAC radiation. The normal structure of the ovarian tissue is consisted of various stages of follicles including primordial and growing follicles, and ovarian stromal connectives. When we observed the ovarian tissues irradiated with a dose rate of 200 cGy/min using light and electron microscopes, granular cells in growing follicles are in irregular shape unlike normal follicles. Small segments of cells scattered in follicular antrum among granular cells. We could observe neutrophils and macrophages around the segments, which means the cells already got in the process of decease owing to the effects radiation. With coincident to the increase of the dose rate of x-ray irradiation as 400 or 600 cGy/min, the mature follicles appeared as an irregular form and the granular cells surrounding oocyte also deformed comparing to their normal counterparts. The granulosa cells within mature follicle are already occurred necrotic change and apoptosis. The nuclei in some cells got so fragmented that the segments formed the shape of a horseshoe or scattered in small and condensed pieces. All the cells at a granular layer irradiated with a dose rate of 600 cGy/min show typical characteristics of apoptosis. The neutrophils involved in inflammatory reaction appear evidently in follicular antrum of growing follicles, and macrophage scattered with residual and apoptotic bodies.
This study was to examine expression of the recombinant full-length adiponectin (recombinant adiponectin) in insect ovarian cell culture system and to characterize structural properties of the recombinant adiponectin secreted in medium. Gene construct encoding the recombinant adiponectin contained N-terminal collagen-like domain (110 Amino Acids, AAs), C-terminal globular domain (137 AAs) and C-terminal peptides for detection with V5 antibody (26 AAs included adaptor peptide) and purification using the 6xHis tag (6 AAs). The approximate molecular weight of the product (monomer) was 35 kDa. Molecular mass species of the expressed recombinant adiponectin were monomer (~35 kDa), dimer (~70 kDa), trimer (~105 kDa) and hexamer (~210 kDa). The major secreted species were the LMW forms, such as monomer, dimer, and trimer. There was MMW of hexamer as minor form. HMW multimers (~300 kDa) were shown as a tracer or not detected on the SDS-PAGE in several experiments (data not shown). The multimer forms in this study were not compatible to those in animal or human serum and adipose tissue by other researcher's study in which the major multimer forms were HMW. By protein denaturing experiments with reducing reagent (${\beta}$-MeOH), anionic detergent (SDS) and heat ($95^{\circ}C$) on the SDS-PAGE, not all adiponectin multimers seemed to have disulfide bond linked structure to form multimers. The recombinant adiponectin which expressed in insect ovarian cell culture system seemed to have the limitation as full physiological regulator for the application to animal and human study.
Gonad structure, germ cell development and reproductive cycle of the smallmouth scorpionfish, Scorpaena miostoma were investigated based on histological method. Samples were collected monthly in the vicinity of Suyoung Bay, Pusan, Korea from November 1995 to October 1996. The testis is seminiferous tubule type in internal structure. Seminiferous tubule consists of numerous testicular cysts which contain numerous germ cells in same developmental stage. The ovary consists of several ovarian lamellae originated from ovarian outer membrane. Oogonia originated from the inner surface of the ovarian lamella protrude to the ovarian cavity in oocyte stage, and they are suspended by the egg stalk. Biological minimum size of female and male were 12.5cm in total length. Gonadosomatic index (GSI) of female (3.81) and male (0.23) were the highest in October. Reproductive cycle was classified into the following successive stages: in female, growing stage $(May\~August)$, maturation stage $(September\~October)$, ripe and spawning stage $(November\~December)$, recovery and resting stage $(January\~April)$, and in male, growing stage $(June\~August)$, maturation stage $(September\~October)$, ripe and spent stage $(November\~January)$ and recovery and resting stage $(February\~May)$.
Premature ovarian failure (POF) is a long-term adverse effect of chemotherapy treatment. However, current available treatment regimens are not optimal. Emerging evidence suggests that bone marrow-derived mesenchymal stem cells (BMSCs) could restore the structure and function of injured tissues, but the homing and restorative effects of BMSCs on chemotherapy injured ovaries are still not clear. In this study, we found that granulosa cell (GC) apoptosis induced by cisplatin was reduced when BMSCs were migrated to granulosa cells (GCs) in vitro. Chemotherapy-induced POF was induced by intraperitoneal injection of cisplatin in rats. BMSCs labeled with enhanced green fluorescent protein (EGFP) were injected into the rats via the tail vein to investigate the homing and distribution of BMSCs in vivo. The number of BMSCs in the ovarian hilum and medulla was greater than in the cortex, but no BMSCs were found in the follicles and corpus lutea. In addition, the BMSCs treatment group's antral follicle count and estradiol levels increased after 30 days, compared with the POF group. Hence, our study demonstrates that intravenously delivered BMSCs can home to the ovaries, and restore its structure and function in POF model rats.
Inefficiency of in vivo gene transfer using currently available vectors reflects a major hurdle in cancer gene therapy. Both viral and non-viral approaches have been described to improve gene transfer efficiency but suffer from a number of limitations. Here we tested an adenovirus carrying the small peptide ligand derived from heregulin${\beta}$ EGF-like domain onto fiber, the adenoviral capsid protein, to deliver transgene to ovarian cancer cells which overexpress ErbB, the cognate receptors for heregulin. The attachement of 53 amino acids to fiber didn't affect on the fiber's trimer structure that is critical for the viral entry to cells. The fiber-modified adenovirus can mediate entry and expression of a ${\beta}$-galactosidase into cancer cells in an increased efficiency compared the unmodified adenovirus. Particularly, the gene transfer efficiency was improved up to 5 times in OVCAR3 cells, an ovarian cancer cell line. Such transduction systems hold promise for delivering genes to ErbB receptor overexpressing cancer cells, and could be used for future cancer gene therapy.
We screened a small molecular library that was designed and independently synthesized in vitro and found a new drug (MY-03-01) that is active against ovarian cancer. We established that MY-03-01 effectively inhibited SKOV-3 cell survival in a dose-dependent manner, based on cell viability rates, and that it not only induced SKOV-3 apoptosis by itself, but also did so synergistically with paclitaxel. Secondly, when MY-03-01 was applied at $40{\mu}M$, its hemolytic activity was less than 10%, compared with the control, and there was almost no damage to nor mal cells at this concentration. In addition, we used DAPI staining and flow cytometry to show that MY-03-01 could significantly induce apoptosis of SKOV-3 cells. Finally, we found that MY-03-01 likely induced SKOV-3 apoptosis by activating caspase3 and caspase9 through the mitochondrial pathway.
In order to know morphological changes on the female genital organs by Ivermectin(IVM) administration, the histopathological observation was carried out in the organs of rat and mouse treated with the overdose of IVM. In the microscopical findings of the uterus, there were many mitotic figures, epithelial hyperplasia and papillary foldings in the endometrial surface. The increased prevalance of uterine glands, uterine epithelia and glands hyperplasia were markedly presented on diverse patterns adenoma-like structure and single nodular or multiple polyp-like adenoma. In ovary, primary and mature follicles were decreased in number, and hypoplasia of ovarian follicles, atretic follicles, follicular cysts and ovarian atropy were observed. It was considered that IVM administration resulted in follicular hypoplasia and atropy of ovary, and hyperplasia of uterine gland and endometrial surface epithelium might be transformed to neoplasia of glandular structures.
The present study was performed to analyze the expression of LH genes in the rat ovary. Expression of LH subunit genes in the rat ovary was demonstrated by amplification of ovarian RNA by RT-PCR. The ovarian $LH_\beta$ transcripts contained at least two parts of the published cDNA structure, the pituitary exons 1, 2 and 3 and the part of testicular ex on 1 in the major trancripts form in rat testis. Using RIA, significant amount of LH-like molecules were detected in crude ovarian extracts, and the competition curves with increasing amount of tissue extracts were parallel with those of standard peptide, indicating that the ovarian immunoreactive LH-like material is similar to authentic pituitary LH molecule. The administration of PMSG to immature rats resulted in a sharp decrease of the ovarian LH contents after 24 h post-injection. In conclusion, these findings demonstrate that genes for LH subunits are expressed in the rat ovary, and suggest that LH can playa central role in regulation of female reproduction with both endocrine (by pituitary LH) and auto- and/or para-crine (by ovarian LH) manner.
