볼락 (Sebastes inermis) 난병 유사구조의 전자현미경적 기재 (Electron Microscopical Description on the Egg Stalk-like Structure of the Rockfish, Sebastes inermis (Teleostei: Scorpaenidae))

  • 볼락의 난소는 낭상형이며, 다수의 난소소엽으로 구성되어 있었다. 난황축적 이전의 난모세포들은 난소소엽의 결체성 조직에 매몰되어 관찰되었다. 난모세포는 성장하면서 난소강으로 돌출되어 난병 유사구조를 가지고 포도송이와 같은 구조를 나타냈다 난소 소엽내에 매몰되어있는 직경 약 20$\mu$m 크기의 초기 난모세포에서 난황과립과 방사대는 관찰되지 않았다 직경 약 80$\mu$m의 초기 난모세포의 난병 유사구조 세포질에서는 전자밀도가 높은 난황과려들이 다수 관찰되었으나, 방사대의 pore canal system에서 음세포작용은 관찰되지 않았다. 이 시기에 난병 유사구조에서는 미세섬유, 발달된 소포체와 미토콘드리아, 전자밀도가 낮은 과립상 물질 그리고 활성화된 multivesicular body들이 관찰되었다. 이러한 결과들은 볼락의 난병 유사구조는 난모세포의 부착과 초기 난모세포의 외재적 난황축적에 관여함을 시사하는 것이라고 할 수 있다.

조기난소부전증 여성에서 난소단백질에 대한 순환항체에 관한 연구 (Circulating Antibodies directed toward Ovarian Proteins in Women with Premature Ovarian Failure)

  • The purpose of this study is to identify the presence of the circulating antibodies directed toward ovarian proteins{antiovarian antibodies, AOA) and the nature of antigenic ovarian structure by comparing the binding activities to 4 types of ovarian proteins, particulated and solubilized forms of pig ovarian and granulosa cell membranes in sera of patients with premature ovarian failure(POF) and to evaluate the usefulness of circulating AOA as a follow up tool after treatment. Measurements of AOA were performed by enzyme linked immunosorbent assay(ELISA) in sera of 58 patients with POF, 51 had normal chromosomes and 7 had X chromosome abnormalities. Sera of 21 natural menopausal women and 17 castrated women were also tested and sera of 32 healthy premenopausal women were served as controls. ELISA reactivities against particulated porcine granulosa cell membrane proteins was the greatest among 4 different ovarian proteins. Fifteen(29%) of 51 POF patients with normal chromosome and 1(14.3%) of 7 POF patients with X chromosome abnormalities had AOA while none of 32 controls and 21 natural menopausal women and 17 castrated women had AOA. One POF patient with 47, XXX was identified AOA positive. The ELISA reactivities were followed up monthly up to 5 months in 4 AOA positive POF patients after estrogen-progestin{E-P) therapy. There was a decreasing tendency of the ELISA reactivities in all these patients after E-P therapy and two of them converted to AOA negative. These data suggest that antigenic structure may be components of granulosa cell membrane and the determination of circulating AOA may be useful in the follow up after treatment in patients with autoimmune POF.

소라, Batillus cornutus의 난소구조 및 난자형성과정 (Ovarian Structure and Oogenesis of the Spiny Top Shell, Batillus cornutus (Lightfoot, 1786) (Gastropoda: Turbinidae))

  • 소라, Batillus cornutus의 난소구조와 난모세포 발달단계를 광학현미경과 투과전자현미경 (TEM) 을 이용하여 관찰하였다. 전라남도 완도군 연안에서 다이버에 의해 채집된 소라는 자웅이체로서 난소는 소화맹낭부에서 꼬리돌기 끝까지 간췌장의 외측을 싸고 발달되어 있었으며, 성숙 시기에 난소는 녹색을 나타내었다. 난소의 내부 구조는 다수의 난소소엽으로 구성되어 있었으며, 난소소엽은 상피세포와 간충결합조직으로 구성되어 있었다. 난원세포는 큰 핵과 전자밀도가 높은 인을 포함하고 있었다. 난황형성 전기의 난모세포는 세포질에 전자밀도가 낮았고, 작은 구형의 난황 과립이 산재해 있었다. 난황형성개시기의 난모세포는 난병에 의해 난소소엽과 연결되어 있었으며, 투과전자현미경 관찰 결과, 세포질에는 잘 발달된 골지체, 조면소포체, 미토콘드리아와 같은 세포소기관과 전자밀도와 크기가 다양한 난황과립이 다수 존재하였다. 이들 난황과립의 전자밀도와 크기는 전단계의 난모세포보다 증가하였다. 후기 난황형성활성기의 난모세포의 난막 두께는 약 4.4 ${\mu}m$였다. 성숙한 난모세포는 세포질에 전자밀도가 높은 단백질성의 난황과립과 전자밀도가 낮은 지질성의 난황과립으로 가득 차 있었으며 두께 약 6.5 ${\mu}m$인 난막에 의해 둘러싸여 있었다.

Comparison of QSAR Methods (CoMFA, CoMSIA, HQSAR) of Anticancer 1-N-Substituted Imidazoquinoline-4,9-dione Derivatives

  • Comparison studies of the Quantitative Structure Activity Relationship (QSAR) methods with new imidazo-quinolinedione derivatives were conducted using Comparative Molecular Field Analysis (CoMFA), Comparative Molecular Similarity Indices Analysis (CoMSIA), and the Hologram Quantitative Structure Activity Relationship (HQSAR). When the CoMFA crossvalidation value, q2, was 0.625, the Pearson correlation coefficient, r2, was 0.973. In CoMSIA, q2 was 0.52 and r2 was 0.979. In the HQSAR, q2 was 0.501 and r2 was 0.924. The best result was obtained using the CoMSIA method according to a comparison of the calculated values with the real in vitro cytotoxic activities against human ovarian cancer cell lines.

Ovarian teratoma removed by laparoscopic ovariectomy in a dog

  • An elective laparoscopic ovariectomy on a healthy dog revealed a cystic structure in the left ovary. The surgical procedure was successful. Histopathological examination showed the presence of a teratoma adjacent to the ovary. To the best of the authors' knowledge, this is the first reported case of an ovarian teratoma removed by laparoscopic ovariectomy in a dog by using a multiport laparoscopic ovariectomy technique.

  • Eleven wrasse species inhabit the coastal waters of Jeju Island, Korea. They are the target of leisure fishing and are considered good eating. We investigated the distribution of standard length (SL) by sex of wrasse in Jeju coastal waters for Halichores poecilopterus, H. tenuispinis, Pseudolabrus japonicus, and Pteragogus flagellifera. A cross-section of the ovary showed the ovarian cavity and ovarian lamellae containing oocytes. A cross-section of the testis showed many lobules containing spermatogonia and spermatocytes. A cross-section of a gonad undergoing sex reversal showed the regression or reduction of oocytes and some spermatocytes located in the ovarian lamellae. A cross-section of a sex-reversed testis showed the primary structure of the ovary, with spermatocytes distributed in the epithelium of the lamellae, and reformed seminiferous ducts in the basement lamellae. Inhibitory effect of ginsenglactone A from Panax ginseng on the tube formation of human umbilical vein endothelial cells and migration of human ovarian cancer cells

  • Background: Here, we aimed to assess the inhibitory effect of a new compound from Panax ginseng on the migration of human ovarian cancer cells and tube formation of human umbilical vein endothelial cells (HUVECs). Methods: A new compound, ginsenglactone A (1), was isolated from ginseng roots, together with seven known compounds (2-8). Spectroscopic data were used to elucidate the chemical structure of 1. The tubular structure formation in HUVECs was assessed by Mayer's hematoxylin staining. The migration of A2780 cells was evaluated using the scratch wound healing assay. Results: HUVECs treated with 1 had the statistically significant decrease in tubular structure formation compared to the HUVECs treated with compounds 2-8. This effect was enhanced by co-treatment with inhibitors for phosphatidylinositol 3-kinase (PI3K) (LY294002) and extracellular signal-regulated kinase (ERK) (U0126). Treatment with 1 decreased the expression of phosphorylation of ERK, PI3K, vascular endothelial growth factor receptor2 (VEGFR2), Akt, and mammalian target of rapamycin (mTOR). In addition, the ability of A2780 cells to cover the scratched area were also decreased. This effect was enhanced by co-treatment with U0126. Lastly, treatment with 1 decreased the phosphorylation of ERK, matrix metalloproteinase-9 (MMP-9), and MMP-2. Conclusion: These results suggest that ginsenglactone A is a potential inhibitor of HUVEC tubular structure formation and A2780 cellular migration, which may be helpful for understanding its anticancer mechanism.

Novel Anti-Angiogenic and Anti-Tumour Activities of the N-Terminal Domain of NOEY2 via Binding to VEGFR-2 in Ovarian Cancer

  • The imprinted tumour suppressor NOEY2 is downregulated in various cancer types, including ovarian cancers. Recent data suggest that NOEY2 plays an essential role in regulating the cell cycle, angiogenesis and autophagy in tumorigenesis. However, its detailed molecular function and mechanisms in ovarian tumours remain unclear. In this report, we initially demonstrated the inhibitory effect of NOEY2 on tumour growth by utilising a xenograft tumour model. NOEY2 attenuated the cell growth approximately fourfold and significantly reduced tumour vascularity. NOEY2 inhibited the phosphorylation of the signalling components downstream of phosphatidylinositol-3'-kinase (PI3K), including phosphoinositide-dependent protein kinase 1 (PDK-1), tuberous sclerosis complex 2 (TSC-2) and p70 ribosomal protein S6 kinase (p70S6K), during ovarian tumour progression via direct binding to vascular endothelial growth factor receptor-2 (VEGFR-2). Particularly, the N-terminal domain of NOEY2 (NOEY2-N) had a potent anti-angiogenic activity and dramatically downregulated VEGF and hypoxia-inducible factor-1α (HIF-1α), key regulators of angiogenesis. Since no X-ray or nuclear magnetic resonance structures is available for NOEY2, we constructed the three-dimensional structure of this protein via molecular modelling methods, such as homology modelling and molecular dynamic simulations. Thereby, Lys15 and Arg16 appeared as key residues in the N-terminal domain. We also found that NOEY2-N acts as a potent inhibitor of tumorigenesis and angiogenesis. These findings provide convincing evidence that NOEY2-N regulates endothelial cell function and angiogenesis by interrupting the VEGFR-2/PDK-1/GSK-3β signal transduction and thus strongly suggest that NOEY2-N might serve as a novel anti-tumour and anti-angiogenic agent against many diseases, including ovarian cancer.

Characterization of a conjugated polysuccinimide-carboplatin compound

  • Carboplatin, an advanced anticancer drug with excellent efficacy against ovarian cancer, was developed to alleviate the side effects that often occur with cisplatin and other platinum-based compounds. Our study reports the in vitro characteristics, viability, and activity of cells expressing the inducible nitric oxide synthase (iNOS) gene after carboplatin was conjugated with polysuccinimide (PSI) and administered in combination with other widely used anticancer drugs. PSI, which has promising properties as a drug delivery material, could provide a platform for prolonging carboplatin release, regulating its dosage, and improving its side effects. The iNOS gene has been shown to play an important role in both cancer cell survival and inhibition. Herein, we synthesized a PSI-carboplatin conjugate to create a modified anticancer agent and confirmed its successful conjugation. To ensure its solubility in water, we further modified the structure of the PSI-carboplatin conjugate with 2-aminoethanol groups. To validate its biological characteristics, the ovarian cancer cell line SKOV-3 and normal ovarian Chinese hamster ovary cells were treated with the PSI-carboplatin conjugate alone and in combination with paclitaxel and topotecan, both of which are used in conventional chemotherapy. Notably, PSI-carboplatin conjugation can be used to predict changes in the genes involved in cancer growth and inhibition. In conclusion, combination treatment with the newly synthesized polymer-carboplatin conjugate and paclitaxel displayed anticancer activity against ovarian cancer cells but was not toxic to normal ovarian cancer cells, resulting in the development of an effective candidate anticancer drug without severe side effects.
